Background: Khella (Ammi visnaga Lam.) fruits (Apiaceae) are rich in furanochromones, mainly khellin and visnagin, and are thus incorporated in several pharmaceutical products used mainly for treatment of renal stones. Methods: The objective of this study was to compare the yield of khellin and visnagin obtained using different conventional solvents and supercritical fluid extraction (SCFE) with carbon dioxide (containing 5% methanol as co-solvent). Water, acetone and ethanol (30% and 95%) were selected as conventional solvents. Results: Highest extract yield was obtained from 30% ethanol (15.44%), while SCFE gave the lowest yield (4.50%). However, the percentage of furanochromones were highest in SCFE (30.1%), and lowest in boiling water extract (5.95%). HPLC analysis of conventional solvent extracts showed other coumarins that did not appear in supercritical fluid extraction chromatogram due to non-selectivity of solvent extraction. Ammi visnaga extracts as well as standard khellin and visnagin were tested for their cytotoxic activity using sulforhodamine B assay on breast cancer (MCF-7) and hepatocellular carcinoma (Hep G2) cell lines. Results revealed a strong cytotoxic activity (IC50 < 20 µg/mL) for the SCFE and standard compounds (khellin and visnagin) (IC50 ranging between 12.54 ± 0.57 and 17.53 ± 1.03 µg/mL). However, ethanol and acetone extracts had moderate cytotoxic activity (IC50 20–90 µg/mL) and aqueous extract had a weak activity (IC50 > 90 µg/mL). Conclusions: Thus, supercritical fluid extraction is an efficient, relatively safe, and cheap technique that yielded a more selective purified extract with better cytotoxic activity.  相似文献

Tea bags including fruits of Ammi visnaga L. are used in Egypt as remedy for the treatment of kidney stones. Our study focuses on developing simple and rapid method utilising HPLC for quantitative estimation of khellol glucoside (KG), khellin (KH) and visnagin (VS) simultaneously. Their concentrations were determined in A. visnaga L. fruits at different developmental stages and in pharmaceutical formulations together with following up them during shelf life. Separation was accomplished using HPLC. Perfect resolution between KG, KH and VS was possible through using a mobile phase consisting of water:methanol:tetrahydrofuran (50:45:5, v/v/v). Peaks were detected at 245 nm. The suggested method for the determination of KG, KH and VS was successful in determining the analytes of interest without any interference of other compounds and matrix. All validation parameters were satisfactory and the procedure was relatively easy and fast as extracts are evaluated without previous steps of purification.  相似文献

A rapid and sensitive CEC method with methacrylate ester‐based monolithic column has been developed for separation and determination of five coumarins (byakangelicin, oxypeucedanin hydrate, xanthotoxol, 5‐hydroxy‐8‐methoxypsoralen and bergapten) in Angelica dahurica extract. Surfactant sodium desoxycholate (SDC) was introduced into the mobile phase as the pseudostationary to dynamically increase the selectivity of analytes instead of increasing the hydrophobicity of stationary phase. In addition, other factors, pH of phosphate buffer, ACN content and applied voltage, for instance, have also an obvious effect on the resolution but little on the retention time. Satisfactory separation of these five coumarins was achieved within 6 min under a 30:70 v/v ACN–buffer containing 20 mM sodium dihydrogen phosphate (NaH2PO4) and 0.25 mM SDC at pH 2.51. The RSDs of intraday and interday for relative peak areas were less than 3.0% and 4.7%, respectively; and the recoveries were between 87.5% and 95.0%. The LODs were lower than 0.15 μg/mL and the LOQs were lower than 0.30 μg/mL, respectively, while calibration curves showed a good linearity (r2 > 0.9979). Finally, five target coumarins from the crude extracts of A. dahurica were separated, purified, and concentrated by D‐101 macroporous resin, and were successfully separated and quantitatively determined within 6 min.  相似文献

Dimethylacetamide (DMA) is a solvent used in the preparation of intravenous busulfan, an alkylating agent used in blood or marrow transplantation. DMA may contribute to hepatic toxicity, so it is important to monitor its clearance. The aim of this study was to develop an HPLC‐UV assay for measurement of DMA in human plasma. After precipitation of plasma proteins with acetonitrile followed by dilution (1:4) with water, the extract was injected onto the HPLC and detected at 195 nm. Separation was performed using a Cogent‐HPS 5 μm C18 column (250 × 4.6 mm) preceded by a Brownlee 7 μm RP18, pre‐column (1.5 cm × 3.2 mm). The mobile phase was 25 mm sodium phosphate buffer (pH 3), containing 2.5% (v /v) acetonitrile and 0.0005% (v /v) sodium‐octyl‐sulfonate. Using a flow rate of 1 mL/min, the retention times of DMA and the internal standard (IS), 2‐chloroacetamide, were 9.5 and 3.5 min, respectively. Peak area ratio (DMA:IS) was a linear function of concentration from 1 to 1000 μg/mL. There was excellent intraday precision (<5% for 5–700 μg/mL DMA), accuracy (<3% deviation from the true concentration) and recovery (74–98%). The limits of detection and quantification were 1 and 5 μg/mL, respectively. In eight children who received intravenous busulfan, DMA concentrations ranged from 110 to 438 μg/mL.  相似文献

A rapid and effective method was developed for the qualitative and quantitative analysis of the major chemical constituents in Angelicae pubescentis radix by ultra high performance liquid chromatography with photodiode array detection and quadrupole time‐of‐flight tandem mass spectrometry. The chromatographic separation was achieved on an ACQUITY UHPLC BEH C18 column (2.1 × 100 mm, 1.7 μm). Nine phenolic acids, 30 coumarins, bisabolangelone, and adenosine were identified by quadrupole time‐of‐flight tandem mass spectrometry. All calibration curves exhibited good linearity (r > 0.9996) within the linear ranges. The relative standard deviation calculated for intraday and interday precision, stability, and accuracy were <5%. The mean recovery ranged from 95.8 to 106%. The overall limits of detection and quantification were 0.025–0.160 and 0.100–0.560 μg/mL, respectively. Discriminant analysis was investigated as a method for evaluating the quality of the samples with 100% correction in their classification. The results demonstrated that the developed method could successfully be used to differentiate samples from different regions and could be a helpful tool for detection and confirmation of the quality of traditional Chinese medicines.  相似文献

Khellin, 4,9-dimethoxy-7-methyl-5H-furo[3,2-g]chromen-5-one, the most biologically active furochromone present in Ammi visnaga fruits indigenous to Egypt. A procedure for radioiodination of khellin with 125I is carried out via an electrophilic substitution reaction. The reaction parameters studied were khellin amount, pH of the reaction mixture, reaction time, temperature, different oxidizing agents and different organic media to optimize the conditions for the labeling of khellin and to obtain a high radiochemical yield of the 125I-khellin (125I-Khel). Using 3.7 MBq of Na125I, 100 μg (0.96 mM) of khellin as substrate, 100 μg (1 mM) of chloramine-T as oxidizing agent in ethanol at 60 °C for 10 min, a maximum radiochemical yield of 125I-Khel (78 %) was obtained. The specific activity of 125I-Khel was 3 MBq/0.5 mM. The biological distribution in normal mice indicates that radioiodinated khellin is a novel agent for urinary tract infection imaging.  相似文献

A high‐throughput PRiME (process, robustness, improvements, matrix effects, ease of use) sample purification procedure was developed to simplify the multiple steps of traditional SPE in extracting the malachite green and leucomalachite green in Chinese softshell turtle (Pelodiscus sinensis). The sample loading volume, extracting solvent type, and pH value of the employed PRiME hydrophilic‐lipophilic balance cartridge for sample purification were optimized to be 3 mL, acetonitrile, and pH 5, respectively. In comparison with traditional SPE, the PRiME process is cost‐effective, solvent‐saving, and simple to operate, which only consists of a passing through step without traditional sorbent conditioning and impurity washing. Afterward, eluate was analyzed by ultra‐performance liquid chromatography‐tandem mass spectrometry, and the proposed method was validated for linearity (R2 > 0.9992), intraday precision (2.44–3.22%), interday precision (3.28–6.58%), sensitivity (LOD ≤ 0.18 μg/kg and, LOQ ≤ 0.60 μg/kg), and recovery (88.7–94.1%, RSD < 6.79%). The results indicated that the PRiME technique can simplify the sample preparation procedure by avoiding the tedious steps, such as conditioning, washing, etc. It would be of significant interest for environmental and food safety applications in the market of Chinese softshell turtle and related products.  相似文献

A high-performance liquid chromatographic method was developed for separation of the furochromone fraction and for determination of visnagin inAmmi visnagahairy root cultures. Lyophilized samples were extracted with chloroform:methanol (1:1, v/v) and purified on solid-phase extraction cartridges. HPLC analyses were performed on a Eurospher 100-C8Knauer column and the mobile phase was 29:28:526:417 (v/v/v/v) acetonitrile:tetrahydrofuran:30 mM citric acid (pH 3.0):methanol. Quercetin was used as internal standard. Peaks were identified by addition of authentic standards and/or by diode-array detection.  相似文献

A selective and sensitive liquid chromatography tandem mass spectrometry method was developed for the simultaneous determination of salviaflaside and rosmarinic acid in rat plasma. Sample preparation was carried out through liquid–liquid extraction with ethyl acetate using curculigoside as internal standard (IS). The analytes were determined by selected reaction monitoring operated in the positive ESI mode. Chromatographic separation was performed on an Agilent Eclipse Plus C18 column (100 × 4.6 mm, 1.8 μm) with a mobile phase consisting of methanol–water–formic acid (50:50:0.1, v/v/v) at a flow rate of 0.3 mL/min. The run time was 1.9 min per sample and the injection volume was 5 μL. The method had an LLOQ of 1.6 ng/mL for salviaflaside and 0.94 ng/mL for rosmarinic acid in plasma. The linear calibration curves were fitted over the range of 1.6–320 ng/mL for salviaflaside and 0.94–188 ng/mL for rosmarinic acid in plasma with correlation coefficients (r2) >0.99. Intra‐ and inter‐day precisions (relative standard deviation) were < 13.5%, and accuracies (relative error) were between −8.6% and 14.5% for all quality control samples. The method was validated and applied to the pharmacokinetics of salviaflaside and rosmarinic acid in plasma after oral administration of Prunella vulgaris extract to rats.  相似文献

During a search for bioactive compounds from indigenous plants,Pimpinella monoica (Umbelliferae) was found to contain furocoumarin, isopimpenellin (3) and five biogenetically related furocoumarins viz khellin (1), visnagin (2), visamminol (4), ammiol (5) and khellol (6). Labelled (i) and (2) were isolated from [1−14C]- and [2−14C]-acetates. Labelling pattern, determined by degradation of biosynthesised compounds, establishes the polyketide origin of their aromatic and pyrone rings while the furan ring originates via an acetate-mevalonate pathway. The plant also utilises glycine and leucine as substratevia acetate. Biotransformation of [3−3H]-visnagin to (6) but not to (2) was also observed.  相似文献

Time-resolved infrared spectroscopy (TRIR) and density functional theory (DFT) calculations were used to directly observe and assign the vibrational spectra of the triplet states of visnagin and khellin, and to investigate their electron-transfer chemistry. The TRIR spectra of triplet visnagin and triplet khellin, and of their radical cations and anions, were obtained upon 266 nm laser flash photolysis in acetonitrile and in deuterated acetonitrile. The radical cations were observed in the presence of chloranil, and the radical anions were formed in the presence of NaI and KSCN. The TRIR spectra are in good agreement with the calculated vibrational spectra. We did not observe the related neutral radicals by TRIR spectroscopy upon laser flash photolysis (LFP) of khellin in the presence of hydroquinone, but we found evidence for the formation of semiquinone and neutral visnagin radicals upon LFP of visnagin and hydroquinone.  相似文献

For the first time simple, rapid, and systematic methods have been established for preparative isolation and purification of coumarin compounds in an important traditional Chinese Medicine, Radix Angelica dahurica, and for simultaneous determination of several of the compounds in the medicine. Bergapten, imperatorin, and cnidilin, three of the biologically active coumarin compounds, were isolated from the chloroform-soluble fraction of the ethanol extract of Radix Angelica dahurica. After further purification by open column ODS chromatography the purified components were simultaneously determined, with two other coumarins (osthole and isoimperatorin), by reversed phase high-performance liquid chromatography (RP-HPLC) on a C18 column, with methanol–water, 66:34 (v/v), as mobile phase at a flow rate of 0.8 mL min−1. The compounds were detected by UV absorption at 310 nm. Calibration plots for all the coumarins had correlation coefficients close to unity. Limits of detection (S/N = 3) were <92 ng mL−1 and limits of quantification (S/N = 10) were <259 ng mL−1. Mean recovery of the coumarins was in the range 96.7–101.9% and the intra-day and inter-day precision, as relative standard deviation, was <2.3 and <2.9%, respectively. This simple, sensitive, and reproducible method can be used for quality control of Radix Angelica dahurica.  相似文献

A rapid, simple and sensitive ultra-performance liquid chromatography–tandem mass spectrometry (UHPLC–MS/MS) method has been developed to quantify fenoprofen, a nonsteroidal anti-inflammatory drug in human plasma for a pharmacokinetic study in healthy subjects. Owing to high levels of protein binding, protein precipitation followed by solid-phase extraction was employed for the extraction of fenoprofen and fenoprofen-d3 (used as internal standard) from 200 μL human plasma. Separation was performed on a BEH C18 (50 × 2.1 mm, 1.7 μm) column using methanol−0.2% acetic acid in water (75:25, v/v) under isocratic elution. Electrospray ionization was operated in the negative mode for sample ionization. Ion transitions used for quantification in the selected reaction monitoring mode were m/z 241/197 and m/z 244/200 for fenoprofen and fenoprofen-d3, respectively. Under the optimized conditions, fenoprofen showed excellent linearity in the concentration range 0.02–20 μg/mL (r2 ≥ 0.9996), adequate sensitivity, favorable accuracy (96.4–103.7%) and precision (percentage coefficient of variation ≤4.3) with negligible matrix effect. The validated method was successfully applied to a pharmacokinetic study of fenoprofen in healthy subjects. The significant features of the method include higher sensitivity, small plasma volume for processing and a short analysis time.  相似文献

Abstract— Longwave UV irradiation of a frozen aqueous solution of khellin and thymine resulted in the formation of a 2+2 photoadduct between the 2,3 double bond of khellin and the 5', 6' double bond of thymine. This behaviour is analogous to that of the furocoumarins and serves as a mechanism to describe the genotoxicity of the furochromones khellin and visnagin. These two compounds are phototoxic towards bacteria and fungi and inhibit mitosis and cause gross chromosomal changes in Chinese hamster ovary (CHO) cells in near UV light. For comparative purposes, 8-methoxypsoralen was also tested and was found to be more active than visnagin which, in turn, was more active than khellin.  相似文献

A rapid and sensitive assay based on ultra‐high performance liquid chromatography with electrospray ionization tandem mass spectrometry was established and validated for the simultaneous determination of cichoric acid, chlorogenic acid, quinic acid, and caffeic acid in rat plasma after oral administration of Echinacea purpurea extract using butylparaben as the internal standard. Samples were pretreated by liquid–liquid extraction with ethyl acetate. The separations for analytes were performed on an ACQUITY UPLC HSS C18 column (1.8 μm 2.1 × 100 mm) using a gradient elution program with acetonitrile/10 mM ammonium acetate (pH 5.6) at a flow rate of 0.3 mL/min. The analytes were detected in multiple reaction monitoring mode with negative electrospray ionization. The lower limit of quantification of each analyte was not higher than 10.85 ng/mL. The relative standard deviation of the intraday and interday precisions was less than 14.69%. The relative errors of accuracies were in the range of –13.80 to 14.91%. The mean recoveries for extraction recovery and matrix effect were higher than 80.79 and 89.98%, respectively. The method validation results demonstrated that the proposed method was sensitive, specific, and reliable, which was successfully applied to the pharmacokinetic study of four components after oral administration of Echinacea purpurea extract.  相似文献

In this work, we show that gallic acid can significantly inhibit the chemiluminescence (CL) intensity of carbon quantum dots (CQDs)‐enhanced K3Fe(CN)6–luminol system. Under optimum conditions, the decrease in the CL intensity is proportional to the concentration of gallic acid over the range 0.01–1.0 μM, and the detection limit is 1.0 nM. The relative standard deviation of repeated intraday and interday determinations of gallic acid was 1.2–4.2%. This method was successfully applied to the determination of gallic acid in food samples, with recoveries in the range 94.0–103.0%. A possible mechanism of CL is discussed.  相似文献

A method for extracting coenzyme Q10 (CoQ10) from Artemia was developed. 1 g of fresh Artemia was incubated with 75 % acetic acid at (30 ± 2)°C for 24 h, followed by three consecutive extractions with a mixture of 5 mL of hexane and 5 mL of ethanol, then analysis by a validated high-performance liquid chromatography with a diode-array detector. The calibration curve for CoQ10 was linear in a range of 1–50 μg mL?1. The limits of detection and quantification were 0.3 μg mL?1 and 1.1 μg mL?1, respectively. Mean recoveries were 94–100 % with a high precision of below 10 %. The method developed was found to be simple, efficient and the time required for releasing CoQ10 from Artemia was short. The method provides not only low energy consumption but is also practical for industrial applications.  相似文献

A simple, rapid, and efficient ultrasound‐assisted emulsification microextraction method followed by gas chromatography mass spectrometry in selected ion monitoring mode was developed for the determination of organochlorine pesticides in honey samples. The type and volume of organic extraction solvent, pH, effect of added salt content, and centrifuging time and speed were investigated. Under the optimum extraction conditions, 30 μL of 1, 2‐dibromoethane (extraction solvent) was immersed into an ultrasonic bath for 1 min at 40°C. The limits of detection and quantification for all target pesticides were 0.003–0.06 and 0.01–0.2 ng/g, respectively. The extraction recovery was 91–100% and the enrichment factors were 168–192. The relative standard deviation for the method was <6% for intraday (n = 6) and <8% for interday precision (n = 4). The proposed method was successfully applied for the analysis of organochlorine pesticides in honey samples.  相似文献

The naturally occuring furanochromones khellin and visnagin have received considerable attention, largely because of their vasodilatory properties and of their ability (particularly that of khellin) to induce skin pigmentation upon ultraviolet light treatment of patients suffering from vitiligo. There are conflicting statements in the literature on whether or not they are capable of cross-linking DNA photochemically. Supercoiled and linear pBR322 DNA was used to probe this reaction. The results showed that both khellin and, to a greater extent, visnagin photosensitized DNA cross-linking. In addition, both photosensitizers induced extensive DNA cleavage.  相似文献

A high‐throughput and environmentally friendly method based on 96‐well plate thin‐film microextraction was established to determine 14 fungicides in grapes and grape juice using liquid chromatography–tandem mass spectrometry. The thin‐film microextraction optimized method consisted of 60 min of extraction at pH 6.0 with the addition of sodium chloride (2–5%). Acetonitrile/water in the ratio of 8:2 was used for desorption analytes for 60 min. Evaluation of different extractive phases showed that polyacrylonitrile–polystyrene–divinylbenzene was the optimum coating. The linearity of the method was good in the range of 0.01–0.5 μg/mL for 14 fungicides with determination coefficients (R2) from 0.990 to 0.999, which indicated good linearity for both the grape juice and grape matrixes. The limit of detection was in the range of 0.002–0.01 μg/mL. The limit of quantitation was in the range of 0.01 mg/kg according to the minimum fortified level. The average absolute recoveries of the 14 fungicides ranged from 75.0 to 118.3%. The intraday relative standard deviation (n = 4) and interday relative standard deviation (n = 4) were 5.6–13.0% and 1.6–6.4%, respectively. This study showed that this method can be used for analyzing 96 samples in parallel, and the sample preparation time was approximately 2.0 min per sample. In addition, this approach offers a green and low‐cost sample pretreatment technique for future analyses.  相似文献
