The two-dimensional Quantitative Structure-Activity Relationship (2D-QSAR) models have been developed to estimate and predict the inhibitory activities of a series of HEPT analogues against HIV-1 by using quantum chemical parameters and physicochemical parameters. The best model of three parameters yields r = 0.908, r^2A = 0.800 and s = 0.467 based on stepwise multiple regression (SMR) method. The stability of the model has been verified by t-test, and the results show that the model has perfect robustness. The predictive power of QSAR models has been tested by Leave-One-Out (LOO) and Leave-Group(regularly random set)-Out(LGO) procedure Cross-Validation methodology. The r^2cv of 0.755 and r^2pred of 0.759 were obtained, respectively.  相似文献

Summary.  A series of 6-naphthylmethyl substituted HEPT analogues bearing a β-carbonyl and a terminal phenyl ring or ester groups on the N-1 side chain of uracil were synthesized, and the in vitro anti-HIV activity was evaluated. Most of these HEPTs were considerably less potent and selective or inactive, only a few compounds showed moderate or high activity against HIV-1. The results demonstrated that the anti-HIV-1 activity of 6-naphthylmethyl substituted HEPT analogues was diminished or eliminated when the β-oxygen of N-1 side chain was replaced by a carbonyl group.  相似文献

Analogues of MKC-442 capable of undergoing Michael addition reactions were synthesised in order to investigate the activity against the HIV-1 mutant (Y181C). An improved activity was postulated on the basis of a possible covalent binding to the mercapto group of Cys181. Lithiation of the C-6 position of 1-ethoxymethyl-5-ethyl-1H-pyrimidine-2,4-dione (5) was followed by reaction with alpha,beta-unsaturated aldehydes and oxidation of the alcohols formed to give the alkenoyl analogues 1a-3a. Analogues 1b-3b containing an allyloxymethyl group in the N-1 position instead of the ethoxymethyl group could not be synthesised due to isomerisation of the allylic group during the metallation reaction. The NMR data for compounds 1a-3a showed a hindered rotation, which was more pronounced for the 6-cyclohexenylcarbonyl derivative 3a than for the propenyl derivatives 1a and 2a. Moderate activity against wild type HIV-1 was observed for the alcohol 8 and the ketones 2a-3a. However, no activity was observed against the Y181C mutant.  相似文献

A Three-Dimensional Quantitative Structure-activity Relationship (3D-QSAR) model that correlates the biological activities with the chemical structures of a series of Glucose-6-phosphatase inhibitors, exemplified by the 4,5,6,7-tetrahydrothienopyridines derivatives, was established by means of comparative molecular field analysis (CoMFA). The resulting leave-one-out cross-validated value (q2=0.600) and non-cross-validated value (r2=0.956) indicate that the obtained pharmacophore model indeed mimics the steric and electrostatic environment, where inhibitors bind to the enzyme. Furthermore, the developed model also possesses promising predictive ability as discerned by the testing on the external test set. The analysis of the CoMFA contour map, which reveal how steric and electrostatic interactions contribute to inhibitors' bioactivities, provide us with the important information to understand the molecular nature of inhibitor-enzyme interactions and to aid in the design of more potent Glucose-6-phosphatase inhibitors.  相似文献

6-chloro-5-ethyl-, n-propyl- and isopropyluracils 5a-c were efficiently prepared from the corresponding 5-alkybarbituric acids 3a-c via treatment with phosphorus oxychloride and N,N-dimethylaniline to yield the corresponding 5-alkyl-2,4,6-trichloro-pyrimidines 4a-c, which were selectively hydrolyzed by heating in 10% aqueous sodium hydroxide for 30 minutes. The reaction of compounds 5a-c with 1-substituted piperazines yielded the corresponding 5-alkyl-6-(4-substituted-1-piperazinyl)uracils 6a-j. The target 8-alkyltetrazolo[1,5-f]pyrimidine-5,7(3H,6H)-diones 7a-c were prepared via the reaction of 5a-c with sodium azide. Compounds 6a-j and 7a-c were tested for in vitro activities against a panel of Gram-positive and Gram-negative bacteria and the yeast-like pathogenic fungus Candida albicans. Compound 6h displayed potent broad-spectrum antibacterial activity, while compound 6b showed moderate activity against the Gram-positive bacteria. All the tested compounds were practically inactive against Candida albicans.  相似文献

Two methods for the preparation of dialkylsubstituted dipyridazino[4,3‐b:3′,4′‐e][1,4]diazepinones and dipyridazino[3,4‐b:3′,4′‐e][1,4]diazepinones are reported. The new compounds were evaluated for their inhibitory effects against the replication of human immunodeficiency virus [HIV‐1(IIIB) and HIV‐2(ROD)] in MT‐4 cells.  相似文献
