Solid-state dye-doped polymers are attractive alternatives to the conventional liquid dye systems. In this paper, the third order nonlinear optical property of Sudan III, an azo dye in solvents and dye doped polymer films measured using the Z-scan technique is reported. Multiple diffraction rings were observed when the dye doped in both the liquid and solid media was exposed to a diode-pumped Nd:YAG laser at 532nm.  相似文献

The creation of complex three-dimensional (3D) microfluidic systems has attracted significant attention from both scientific and applied research communities. However, it is still a formidable challenge to build 3D microfluidic structures with arbitrary configurations using conventional planar lithographic fabrication methods. Here, we demonstrate rapid fabrication of high-aspect-ratio microfluidic channels with various 3D configurations in glass substrates by femtosecond laser direct writing. Based on this approach, we demonstrate a 3D passive microfluidic mixer and characterize its functionalities. This technology will enable rapid construction of complex 3D microfluidic devices for a wide array of lab-on-a-chip applications.  相似文献

Formaldehyde (CH2O) is an important intermediate species in combustion processes and it can through laser-induced fluorescence measurements be used for instantaneous flame front detection. The present study has focussed on the use of the third harmonic of a Nd:YAG laser at 355 nm as excitation wavelength for formaldehyde, and different dimethyl ether (C2H6O) flames were used as sources of formaldehyde in the experiments. The investigations included studies of the overlap between the laser profile and the absorption lines of formaldehyde, saturation effects and the potential occurrence of laser-induced photochemistry. The technique was applied for detection of formaldehyde in an internal combustion engine operated both as a spark ignition engine and as a homogenous charge compression ignition engine.  相似文献

Laser nitridation of a pure iron (Fe) surface was conducted using a focused pulsed Nd:YAG laser under a nitrogen atmosphere, and the effects of nitrogen gas pressure, laser power, and repetition number of laser shots on the surface characteristics were analyzed using XPS. The laser-irradiated surface consisted of the topmost surface layer of Fe oxynitride (FeOxNy) and the underlayer beneath, which mainly comprised Fe nitride (Fe4N). The topmost surface layer is a post-formed layer due to the oxidation of the nitride layer. The thickness of the underlayer corresponding to the original nitride layer drastically increased under nitrogen gas at atmospheric pressure. Increasing the repetition number of laser shots enhanced layer thickness up to 5 shots, after which no change was observed. Moreover, the layer thickness increased monotonically with increasing laser power. Nitridation through pulsed laser irradiation was likely predominated by the melting and resolidification of a specific surface area, as well as the convection of nitrogen therein. Thickness variation under various conditions can be explained appropriately using this assumption.  相似文献

We have studied the combination of fourth-harmonic (266 nm) and fundamental (1064 nm) Nd:YAG laser pulses of the same irradiance. On a metallic target (Al), a sequence of ultraviolet (UV) and near-infrared (NIR) pulses produces deeper craters and can lead under certain conditions to analyte signal enhancements larger than those obtained with a NIR–NIR sequence. Compared to a single NIR pulse, signal enhancements by factors of approximately 30 for the Si I 288.16-nm line and 100 for the Al II 281.62-nm line were observed with double pulses of the same total energy. This effect correlates with a substantial increase in plasma temperature, with ionic lines and lines having a higher excitation energy experiencing a larger enhancement. Moreover, the optimal pulse separation is found to be larger for ionic than for neutral lines (∼3 compared to ∼0.1 μs). Another finding of this study concerns the combination of two different wavelengths (266 and 1064 nm) in a single ‘mixed-wavelength’ pulse, a scheme that also leads to an enhanced laser-induced breakdown spectroscopy (LIBS) sensitivity. It is proposed that the double-pulse and mixed-wavelength approaches are both capable of temperature and signal enhancement for the same reason: a larger portion of laser energy is absorbed in the plasma region containing the analyte atoms, instead of being absorbed at the sample surface or in the atmosphere.  相似文献

A precise understanding of individual cellular processes is essential to meet the expectations of most advanced cell biology. Therefore single-cell analysis is considered to be one of possible approach to overcome any misleading of cell characteristics by averaging large groups of cells in bulk conditions. In the present work, we modified a newly designed microchip for single-cell analysis and regulated the cell-adhesive area inside a cell-chamber of the microfluidic system. By using surface-modification techniques involving a silanization compound, a photo-labile linker and the 2-methacryloyloxyethyl phosphorylcholine (MPC) polymer were covalently bonded on the surface of a microchannel. The MPC polymer was utilized as a non-biofouling compound for inhibiting non-specific binding of the biological samples inside the microchannel, and was selectively removed by a photochemical reaction that controlled the cell attachment. To achieve the desired single-macrophage patterning and culture in the cell-chamber of the microchannel, the cell density and flow rate of the culture medium were optimized. We found that a cell density of 2.0 × 10(6) cells/ml was the appropriate condition to introduce a single cell in each cell chamber. Furthermore, the macrophage was cultured in a small size of the cell chamber in a safe way for 5 h at a flow rate of 0.2 μl/min under the medium condition. This strategy can be a powerful tool for broadening new possibilities in studies of individual cellular processes in a dynamic microfluidic device.  相似文献

The capability of LA-ICP-MS for determination of trace impurities in transparent quartz glasses was investigated. Due to low or completely lacking absorption of laser radiation, laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) proves difficult on transparent solids, and in particular the quantification of measurement results is problematic in these circumstances. Quartz glass reference materials of various compositions were studied by using a Nd:YAG laser system with focused laser radiation of wavelengths of 1064 nm, 532 nm and 266 nm, and an ICP-QMS (Elan 6000, Perkin Elmer). The influence of ICP and laser ablation conditions in the analysis of quartz glasses of different compositions was investigated, with the laser power density in the region of interaction between laser radiation and solid surface determining the ablation process. The trace element concentration was determined via calibration curves recorded with the aid of quartz glass reference materials. Under optimized measuring conditions the correlation coefficients of the calibration curves are in the range of 0.9–1. The relative sensitivity factors of the trace elements determined in the quartz glass matrix are 0.1–10 for most of the trace elements studied by LA-ICP-MS. The detection limits of the trace elements in quartz glass are in the low ng/g to pg/g range.  相似文献

Porous monolithic polymers have been prepared by photoinitiated polymerization of mixtures consisting of 2-hydroxyethyl methacrylate, ethylene dimethacrylate, UV-sensitive free radical initiator and porogenic solvent within channels of specifically designed microfluidic chips and used as micromixers. Substituting azobisisobutyronitrile with 2,2-dimethoxy-2-phenylacetophenone considerably accelerated the kinetics of the polymerization. Mixtures of cyclohexanol and 1 -dodecanol and of hexane and methanol were used, respectively, to control the porous properties and therefore the mixing efficiency of the device. The performance of the monolithic mixers has been tested by pumping aqueous solutions of two fluorescent dyes at various flow rates and monitoring the point at which the boundary of both streams completely disappears. Best results were achieved with a monolithic mixer containing very large irregular pores.  相似文献

Laser irradiation on hard tissue has produced a resistant surface that is likely to prevent caries. In this study, human enamel and dentine were exposed to nanosecond pulsed Nd:YAG laser with energy densities of 20-40 J/cm(2) and pulse width of 6 ns inducing chemical changes in these tissues. Infrared analysis of human dental enamel and dentine was performed using the KBr method (2mg sample/300 mg KBr). A correlation between non-lased and lased spectra was performed that gives an indication of the changes in organic and inorganic compounds after laser-tissue interaction. Spectra of teeth simultaneously show the inorganic and organic parts of the tissue. The principal bands: amide bands A, I, II, and III from the collagen-matrix, phosphate from the mineral content, and carbonate bands were identified. The normalized area of peak versus peak position was determined. Changes of the bands attributed to the collagen matrix were verified after Nd:YAG irradiation. The present results suggest a chemical modification of organic and mineral compounds by laser. The spectral results indicated an alteration in the absorption bands relative to, essentially, organic compounds.  相似文献

This paper describes a general procedure for the formation of hydrogels that contain microfluidic networks. In this procedure, micromolded meshes of gelatin served as sacrificial materials. Encapsulation of gelatin meshes in a hydrogel and subsequent melting and flushing of the gelatin left behind interconnected channels in the hydrogel. The channels were as narrow as approximately 6 microm, and faithfully replicated the features in the original gelatin mesh. Fifty micrometre wide microfluidic networks in collagen and fibrin readily enabled delivery of macromolecules and particles into the channels and transport of macromolecules from channels into the bulk of the gels. Microfluidic gels were also suitable as scaffolds for cell culture, and could be seeded by human microvascular endothelial cells to form rudimentary endothelial networks for potential use in tissue engineering.  相似文献

Nanopowder of yttrium aluminum garnet (YAG, Y3Al5O12) doped with neodymium ions (Nd:YAG) was prepared in the water/cetyltrimethylammonium bromide/1-butanol/n-heptane system. Aluminum, yttrium, and neodymium nitrates were used as starting materials, and ammonia was used as a precipitating agent. Coprecipitate hydroxide precursors where thermally treated at 900 degrees C to achieve the garnet phase. The starting system with and without reactants was characterized by means of the small-angle neutron scattering technique. The system, without reactants, is constituted by a bicontinuous structure laying near the borderline with the lamellar phase region. The introduction of nitrates stabilizes the bicontinuous structure, while the presence of ammonia induces a transformation from the bicontinuous phase to a lamellar phase. Nd:YAG nanopowder was characterized by wide-angle X-ray scattering, transmission electron microscopy, gas adsorption, and photoluminescence spectroscopy. By comparison with a sample prepared by the conventional coprecipitation method, the obtained Nd:YAG nanopowder is constituted by smaller crystalline nanoparticles showing a lower tendency to agglomerate. In addition, the nanoparticles present a well-defined spherical shape. Photoluminescence spectroscopy confirms that the doping Nd3+ ions substitute Y3+ ions in the YAG crystalline lattice. The Nd3+ lifetime value, obtained from the luminescence decay curves, was 286 +/- 10 micros, higher than the single-crystal value (255 micros) and much higher than the nanopowder value obtained by the conventional coprecipitation method (75 micros).  相似文献

Recently excavated roman amphorae were submitted to instrumental neutron activation analysis for the determination of Ca, Sc, Cr, Fe, Co, Sb, Ba, La, Ce, Eu, Yb, Lu, Hf and Th. Data reduction was carried out by identifying first the elements useful for discrimination purposes and then by submitting the data to cluster analysis. A microfluidic traveling-wave electrophoresis (TWE) system is reported that uses a locally defined traveling electric field wave within a microfluidic channel to achieve band transport and separation. Low voltages, over a range of -0.5 to +0.5 V, are used to avoid electrolysis and other detrimental redox reactions while the short distance between electrodes, ~25 μm, provides high electric fields of ~200 V cm(-1). It is expected that the low voltage requirements will simplify the future development of smaller portable devices. The TWE device uses four interdigitated electrode arrays: one interdigitated electrode array pair is on the top of the microchannel and the other interdigitated electrode array pair is on the microchannel bottom. The top and bottom substrates are joined by a PDMS spacer that has a nominal height of 15 μm. A pinched injection scheme is used to define a narrow sample band within an injection cross either electrokinetically or hydrodynamically. Separation of two dyes, fluorescein and FLCA, with baseline resolution is achieved in less than 3 min and separation of two proteins, insulin and casein is demonstrated. Investigation of band broadening with fluorescein reveals that sample band widths equivalent to the diffusion limit can be achieved within the microfluidic channel, yielding highly efficient separations. This low level of band broadening can be achieved with capillary electrophoresis, but is not routinely observed in microchannel electrophoresis. Sample enrichment can be achieved very easily with TWE using a device with converging electric field waves controlled by two sets of independently controlled interdigitated electrodes arrays positioned serially along the microchannel. Sample enrichment of 40-fold is achieved without heterogeneous buffer/solvent systems, sorptive, or permselective materials. While there is much room for improvement in device fabrication, and many capabilities are yet to be demonstrated, it is anticipated that the capabilities and performance demonstrated herein will enable new lab-on-a-chip processes and systems.  相似文献

The application of medical lasers in treating pigmented lesions has rapidly developed over the past decade. In both clinical and cosmetic application, melanin is targeted in pigmented areas and destroyed by the mechanism of selective photothermolysis. When laser radiation passes through superficial pigmented tissue, energy will be further reduced by dermal collagen scattering and absorption. Non-pigmented dermal fibroblasts will be exposed to co-incidental laser irradiation at lower energy levels. Biostimulation of dermal fibroblasts by low energy laser is reported in this paper. The Q-switched frequency doubled Nd:YAG 532nm laser used in clinical laser therapy was used in this study. Sublethal laser fluence was determined at 0.8J/cm(2) and used to stimulate normal human fibroblasts in monolayer culture. The results showed that there was no significant difference in collagen synthesis between the stimulated fibroblasts and controls. However, significant delay in collagen remodeling activity was demonstrated in the irradiated group by measuring fibroblast populated collagen lattice (FPCL) contraction. The stimulation of SCF, HGF and b-FGF gene expression was determined by RT-PCR analysis and demonstrated to vary between cases. Two out of six cell lineages that showed stronger responses to laser stimulation on SCF, HGF and b-FGF gene expressions were used to prepare conditioned media. The conditioned media from irradiated groups showed significant increase in SCF and b-FGF content and stimulated SK-mel-3 melanoma cells to synthesize more melanin in vitro. These results suggest that sublethal laser stimulation of fibroblasts may cause post-laser hyperpigmentation through production of melanogenic stimulatory cytokines. The degree of stimulation of SCF, HGF and b-FGF production varied between individual cell lineages, which may reflect the true variation of post-laser hyperpigmentation in clinical practice.  相似文献

采用共沉淀方法,以碳酸氢铵(NthHC03)为沉淀剂制备得到Nd:YAG前驱物,将其在不同的温度下煅烧并采用红外光谱(FT-IR)、X射线衍射(XRD)、透射电镜(TEM)、荧光光谱分析等技术对样品进行了表征。在900~1000℃下煅烧前驱物,出现YAP和YAM中间相并对共沉淀法中间相出现的原因进行了分析;获得纯YAG晶相的煅烧温度为1100℃,晶粒粒度分布约为40~80nm,具有良好的分散性。光谱研究表明Nd:YAG纳米粉体的发光性能良好,其红外光谱及荧光光谱与单晶无明显的差异。  相似文献

The aim of the present study was to form a nitride layer on a titanium (Ti) substrate through a compact laser-nitriding system comprising a focused pulsed Nd:YAG laser and nitrogen gas blow. To obtain a high-quality layer, the effects of pulse frequency and gas flow rate on the surface characteristics were investigated by using plasma emission analysis as well as X-ray analyses. Optical emission spectra from the laser-induced plasma mainly consisted of ionic Ti lines, and their intensities when the pulse frequency was 15 Hz were much higher than those for 8 Hz. Similarly, the reflections from the δ-TiN phase in the X-ray diffraction (XRD) pattern were enhanced when using 15 Hz. On the other hand, the flow rate of nitrogen gas blow had a significant effect on the thickness of the thin oxide layer that formed above the nitride layer. Using a lower flow rate resulted in the formation of a thicker oxide layer. The higher pulse frequency and the faster flow rate were beneficial for obtaining a higher-quality layer because of the enhancement of nitridation and the suppression of oxidation, respectively.  相似文献

On-site detection methods for DNA have been demanded in the pathophysiology field. Such analysis requires a simple and accurate method, rather than high-throughput. This report describes a novel microfluidic analysis method and its application for simple sequence-selective DNA detection. The method uses a microchannel device with a serpentine structure. Sequence-specific binding of probe DNA can be detected at one side of the microchannel. This method is capable of sequence-specific detection of DNA with high accuracy. Single base mutations can also be analyzed. Combination of laminar stream and laminar secondary flow in the microchannel enable specific detection of probe-bound DNA.  相似文献

Plastics are increasingly being used for the fabrication of Lab-on-a-Chip devices due to the variety of beneficial material properties, affordable cost, and straightforward fabrication methods available from a range of different types of plastics. Rapid prototyping of polydimethylsiloxane (PDMS) devices has become a well-known process for the quick and easy fabrication of microfluidic devices in the research laboratory; however, PDMS is not always an appropriate material for every application. This paper describes the fabrication of thermoset polyester microfluidic devices and masters for hot embossing using replica molding techniques. Rapid prototyped PDMS molds are convienently used for the production of non-PDMS polymeric devices. The recessed features in the cast polyester can be bonded to a second polyester piece to form an enclosed microchannel. Thermoset polyester can withstand moderate amounts of pressure and elevated temperature; therefore, the cast polyester piece also can be used as a master for embossing polymethylmethacrylate (PMMA) microfluidic systems. Examples of enclosed polyester and PMMA microchannels are presented, and we discuss the electroosmotic properties of both types of channels, which are important for analytical applications such as capillary electrophoresis.  相似文献
