Summary 3(Bis(alkylthio)methylene)-3H-isobenzofuran-1-ones2a–e and 2-(bis(alkylthio)methylene)-3(2H)-benzofuranone derivatives4a–c are obtained by reaction of phthalides1a–d or 3(2H)-benzofuranone (coumaranone3), respectively, with carbon disulfide under basic conditions followed by alkylation. The reaction behaviour of the new compounds2 and4 is investigated. 2-((2-Dimethylthio-1-oxo)ethyl)benzoic acid N,N-dimethylamide (7a) and 2-((2-dimethylthio-1-oxo)ethyl)-benzoic acid 2-methylpiperidide (7b) are surprisingly formed instead of the methylthio substitution products by treatment of2a with the corresponding amine in the presence of aluminum chloride.

The geometric parameters, the charge distribution, and the energetics of N-methyl-2-(N-ethylanilino)-3-(indol-1-yl)-and N-methyl-2-(N-ethylanilino)-3-(indol-3-yl)maleimides and their conjugated acids were studied by density functional theory calculations at the B3LYP/6-31G(d) level. The mechanism of the tandem hydride transfer/cyclization sequence, which occurs after protonation of N-methyl-2-(N-ethylanilino)-3-(indol-1-yl)-and N-methyl-2-(N-ethylanilino)-3-(indol-3-yl)maleimides, was analyzed. The investigation of the potential energy surface for the tandem hydride transfer/cyclization of the iminium cation that formed upon protonation revealed that the hydride transfer followed by intramolecular cyclization at position 7 of the indole fragment in N-methyl-2-(N-ethylanilino)-3-(indol-1-yl)maleimide is the preferable process, unlike alternative intramolecular cyclization involving the cationic center at the C(2) atom of the indole fragment and the benzene ring of the N-ethylaniline fragment of the indoleninium cation in N-methyl-2-(N-ethylanilino)-3-(indol-3-yl)maleimide. A study of the key intermediates of the assumed reaction mechanism demonstrated that these intermediates are actually stationary points on the potential energy surface (minima and transition states). The novel intermediate 1-(p-fluorophenyl)-2-(2′-pyridyl)ethanol or 2-[2′-(1-hydroxy-1-(p-fluorophenyl)ethyl]pyridine and the corresponding novel dehydration compound 1-(p-fluorophenyl)-2-(2′-pyridyl)ethene or 2-[p-fluorophenylvinyl]pyridine were obtained from the condensation reaction of p-fluorophenylaldehyde and 2-picoline under catalyst-and solvent-free conditions. The intermediate 1-(p-fluorophenyl)-2-(2′-pyridyl)ethanol was obtained at 42 h reaction time and temperature of 120°C, respectively. 1H-NMR, IR spectroscopic data of the 1-(p-fluorophenyl)-2-(2-pyridyl)ethanol clearly showed the presence of the-CH2-CHOH-group. The compound was obtained as a white powder with m.p. 121–122°C and a yield of 8%. For 1-(p-fluorophenyl)-2-(2-pyridyl)ethene, the reaction conditions were similar, but the reaction temperature was increased to yield the double bond in the 1-(p-fluorophenyl)-2-(2′-pyridyl)ethene. At the reaction temperature of 140°C, the compound was a slightly brown powder with a m.p. of 78°C and yield of 18%. 1H-NMR, IR spectroscopic data for the 1-(p-fluorophenyl)-2-(2′-pyridyl)ethene showed the presence of a double bond in trans configuration (-CH=CH-), characteristic of a styrylpyridine.  相似文献

The reaction of o-iodobenzaldehyde, cyanothioacetamide, and N-acetoacetyl-o-toluidine in the presence of piperidine gave piperidinium 4,5-trans-3-cyano-6-hydroxy-4-(2-iodophenyl)-6-methyl-5-(2-methylphenyl)carbamoyl-1,4,5,6-tetrahydropyridine-2-thiolate used in the synthesis of substituted 4,5-trans-2-alkylthiotetrahydropyridines and 2-(2-thiazolyl)acrylonitriles. The structure of 3-(2-iodophenyl)-2-(4-phenyl-2-thiazolyl)acrylonitrile was studied using X-ray diffraction structural analysis.  相似文献

The synthesis and reactions of methyl 2-[3-(trifluoromethyl)phenyl]-4H-furo[3,2-b]pyrrole-5-carboxylate (1a) are described. Upon reaction with methyl iodide, benzyl chloride, or acetic anhydride, this compound gave N-substituted products 1b-d. By hydrolysis of compounds 1a-c, the corresponding acids 2a-c were formed, or by reaction with hydrazine-hydrate, the corresponding carbohydrazides 3a-c were formed. By heating 2-[3-(trifluoromethyl)phenly]-4H-furo[3,2-b]pyrrole-5-carboxylic acid (2a) in acetic anhydride, 4-acetyl-2-[3-(trifluoromethyl)phenyl]furo[3,2-b]pyrrole (4) was formed. By hydrolysis of 4, 2-[3-(trifluoromethyl)phenyl]-4H-furo[3,2-b]pyrrole (5a) was formed, and reactions with methyl iodide or benzyl chloride gave N-substituted products 5b-c. The reaction of 4 with dimethyl butynedioate gave substituted benzo[b]furan 6. Compound 3a reacted with triethyl orthoesters giving 7a-c, which afforded with phosphorus (V) sulphide the corresponding thiones 8a-c. The thiones 8a-c reacted with hydrazine hydrate to form hydrazine derivatives 9a-c. The reaction of triethyl orthoformiate with compounds 9a-c led to furo[2′,3′: 4,5]pyrrolo[1,2-d][1,2,4]triazolo[3,4-f][1,2,4]triazines 10a-c. Hydrazones 11a-c were formed from 3a-c and 5-[3-(trifluoromethyl)phenyl]furan-2-carboxaldehyde. The effect of microwave irradiation on some condensation reactions was compared with “classical” conditions. The results showed that microwave irradiation shortens the reaction time while affording comparable yields.  相似文献

2-Amino-6-aryl-imidazo(2,1-b)-1,3,4-thiadiazoles (1 a-f) were condensed with 1,3-ketocarboxylates e. g. ethyl-butyroacetate, ethyl-benzoylacetate, 4-nitrobenzoylacetate or ethyl-cyclopentanone(2)-carboxylate and ethyl-cyclohexanone(2)-carboxylate directly to imidazo(2,1-b)-1,3,4-thiadiazolo(3,2-a) pyrimidones-(6) (4 a-f, 5 a-f, 6 d, e, 7 a-f, 8 a, d).

In acetic (aqueous or chloroform) solutionp-benzoquinones yield with primary aliphatic amines mainly 2-(N-alkyl-p-hydroxy-anilino)-1,4-benzoquinones besides the 2-alkylamino- and 2,5-bis(alkylamino)-quinones. The methyl-, ethyl-,n-propyl-,n-butylhomologues ofp-benzoquinone and the isomer methylderivatives of toluquinone are described. Their structure were established by spectroscopic (UV/VIS, IR, NMR) methods and by synthesis of the methylderivative3 a fromp-benzoquinone andp-hydroxy-N-methylaniline. The influence of other acids on the reaction was studied.

The reaction of (1S,2S)-2-amino-1-(4-nitrophenyl)-1,3-propanediol with glutaraldehyde has been studied. It has been established on the basis of AM1 and PM3 calculations and 1H NMR spectra recorded in the presence of the shift reagent Eu(fod)3 that (1S,3S,4S,7R,11R)-3-(4-nitrophenyl)-11-aza-2,6-dioxatricyclo[5,3,1,04,11]undecane is formed as the result of the reaction.  相似文献

Zusammenfassung Es wurden eine Anzahl von neuen Kobalt(II)-Komplexen von 4-Benzylamidothiosemicarbazid (BTSC) und 1-(-)Furyl-4-benzylamidothiosemicarbazon (FBTS) von der allgemeinen Zusammensetzung CoL 2 X 2 (L=BTSC, FBTS;X=Cl, Br, I, NO3 and NCS) synthetisiert. Die Untersuchugen erfolgten mittels Elementaranalyse, magnetischer Messungen, Elektronenanregungsspektroskopie und IR-Spektroskopie (einschließlich des fernen IR); aus diesen Messungen ließ sich eine im wesentlichen tetragonale Symmetrie für alle Komplexe dieser Reihe ableiten.
Ligational behaviour of thiosemicarbazide and thiosemicarbazone-(III): Tetragonal complexes of cobalt(II) derived from 4-benzylamidothiosemicarbazide and 1-(-) furyl-4-benzylamidothiosemicarbazone
A number of new complexes of cobalt(II) have been prepared with 4-benzylamidothiosemicarbazide (BTSC) and 1-(-)furyl-4-benzylamidothiosemicarbazone (FBTS) which conform to the general formula CoL 2 X 2 (whereL=BTSC andFBTS andX=Cl, Br, I, NO3 and NCS). These have been characterized by chemical analyses and physical measurements. The tetragonal symmetry has been proposed on the basis of electronic spectral studies for all these complexes. The explanation for the slightly lower magnetic moments for cobalt(II) complexes has been sought in the possible presence of low symmetry component. The tetragonal radial parametersDq(E),Dq(A),Dt andDs and molecular orbital parameters d and d have been evaluated. The S–N, bidentate nature of the ligands and the presence of the various anions in the coordination sphere have been confirmed on the basis of additional Co–N, Co–S and Co–X frequencies in the far infrared spectra of the complexes. The nitrato and thiocyanato groups act as monodentate and are coordinated through oxygen and nitrogen atoms, respectively.

The reaction of 6-(tert-butyl)-3-hydrazino-1,2,4-triazin-5(2H)-one with acetone and benzaldehyde yields the corresponding hydrazones. In the presence of thionyl chloride the benzaldehyde hydrazone is converted to 3-(tert-butyl)-6-phenyl-1,2,4-triazolo[4,3-b]-1,2,4-triazin-2(1H)-one and this can also be formed via the acylation of the starting hydrazinotriazinone using benzoyl chloride.  相似文献

Staudinger 反应是合成β-内酰胺类化合物最重要的方法之一. 3-(噻吩-2-基)-β-内酰胺是一类重要的β-内酰胺类衍生物. 发展了一种从1-(噻吩-2-基)-4,4,4-三氟-1,3-丁二酮和对甲苯磺酰叠氮方便地制备1-(噻吩-2-基)-2-重氮基乙酮的新方法, 利用1-(噻吩-2-基)-2-重氮基乙酮加热分解生成的噻吩-2-基烯酮参与的Staudinger反应合成了一系列3-(噻 吩-2-基)-β-内酰胺衍生物, 并研究了噻吩-2-基烯酮参与的Staudinger反应的立体选择性. 实验结果表明噻吩-2-基烯酮是比苯基烯酮更富电子的Moore烯酮, 其电子性质介于对甲氧基苯基烯酮和对甲基苯基烯酮之间.  相似文献

以2-氨基苯并噻唑类化合物为原料, 通过碱性条件下水解, 与苯甲酸类化合物在PPA(聚磷酸)条件下脱水环化, 部分化合物通过BBr3去甲基化反应, 合成了一系列潜在的针对β-淀粉样蛋白的正电子放射性探针分子前体或作为参比的6-取代苯并噻唑苯胺类化合物, 并用1H NMR, IR, EI/ESI-MS, HRMS, 元素分析等对其进行了表征. 同时测定比较了6种化合物与早老性痴呆(Alzheimer’s disease, AD)死者脑匀浆的亲和力强弱.  相似文献

Summary 3-Aryl-, 3-benzyl-, and 3H-5-(2-aryl-2-oxoethyl)-2-thioxo-4-oxo-1,3-thiazolidines3a–h react with bromine in acetic acid solution to give mixtures of the respective 5-aroylmethylene (E) and (Z) diastereomeric derivatives5 and6. They contain more than 85% of the (E)-diastereomers along with some pure isomers. The intermediacy of the 5-bromo derivatives4 is proven and a plausible route of the reaction is presented. Structures of compounds3–6 are evidenced by analytical and spectral data.

Zusammenfassung 3-Aryl-, 3-Benzyl- und 3H-5-(2-Aryl-2-oxoethyl)-2-thioxo-4-oxo-1,3-thiazolidine3a–h reagieren mit Brom in essigsaurer Lösung zu Gemischen der entsprechenden diastereomeren 5-Arylmethylen-Derivate ((E) und (Z))5 und6. Sie enthalten mehr als 85% des (E)-Diastereomeren. Die intermediäre Natur der 5-Brom-Derivate4 wird bewiesen; ein Reaktionsweg wird vorgeschlagen. Die Strukturen der Verbindungen3–6 werden durch analytische und spektroskopische Daten abgesichert.

The enantiomers of 1-(α-aminobenzyl)-2-naphthol and 2-(α-aminobenzyl)-1-naphthol analogs were separated isothermally on a 3,5-dimethylphenylcarbamoylated β-cyclodextrin-based chiral stationary phase (Cyclobond DMP), with an n-hexane/alcohol modifier as mobile phase. Optimization of the separation was achieved by variation of combinations of the polar mobile phase additives ethanol and methanol. The nature and position of the α-aminobenzyl substituent of the 1- and 2-naphthol analogs influenced the retention and the selectivity.  相似文献

Three procedures were used for the synthesis of -, , and -pyridyl-substituted (5Z)-3-phenyl-5-(pyridylmethylene)-2-thiohydantoins: the reaction of 2-thiohydantoin with the corresponding aldehyde in AcOH in the presence of AcONa, the two-step one-pot synthesis with the use of the same starting compounds, and three-component condensation of aryl isothiocyanate, glycine, and aldehyde in AcOH. Alkylation of the resulting thiohydantoins with iodomethane in the presence of a base afforded the corresponding S-methylated derivatives, viz., 2-methylthio-3-phenyl-5-(pyridylmethylene)-3,5-dihydro-4H-imidazol-4-ones. The structures of (5Z)-3-phenyl-5-(pyridin-2-ylmethylene)-2-thioxoimidazolin-4-one and (5Z)-2-methylthio-3-phenyl-5-(pyridin-2-ylmethylene)-3,5-dihydro-4H-imidazol-4-one were established by X-ray diffraction analysis.__________Published in Russian in Izvestiya Akademii Nauk. Treatment of 2-(tert-butyl)-1,2,3,4-benzotetrazinium tetrafluoroborates with sodium thiocyanate afforded 2-(tert-butylazo)phenyl isothiocyanates 3, which exist in equilibrium with 2-(tert-butyl)-1,2,4-benzotriazine-3(2H)-thiones 3′. The equilibrium depends on the substituents R in the benzene ring: the percentage of the open isomer 3 is about 20% for R = H or Me; for R = Cl or Br, the equilibrium is completely shifted to cyclic isomer 3′. The equilibrium is slow on the time scale of the 1H and 13C NMR experiments. For compounds 3a/3′a (R = H), the spectra at 24 °C show two sets of signals, while those at 0 °C contain only signals for isomer 3′a. Published in Russian in Izvestiya Akademii Nauk. Substituted 2-amino-4-aryl-3-cyano-5-oxo-5,6-dihydro-4H-pyrano[2,3-d]pyrido[3",2":4,5]thieno[3,2-b]pyridines were synthesized by the reactions of 4-hydroxy-1H-thieno[2,3-b;4,5-b]dipyridin-2-ones with arylidenemalononitriles or by the three-component reactions of hydroxythienodipyridinones with aldehydes and malononitrile in DMF in the presence of triethylamine. Methods for syntheses of substituted 3-alkoxycarbonyl-6-amino-4-aryl-2-(3-cyanopyridin-2-ylthiomethyl)-4H-pyrans were developed on the basis of the reactions of 4-(3-cyanopyridin-2-ylthio)acetoacetates and arylidenemalononitriles or aldehydes and malononitrile. Ethyl 4-(3-cyanopyridin-2-ylthio)acetoacetate and 4-methoxybenzylidenecyanothioacetamide were used for the synthesis of 6-(pyridin-2-ylthiomethyl)-3-cyanopyridine-2(1H)-thione.  相似文献
