1-Carbethoxy-2,4,6,8-tetraaryl-3,7-diazabicyclo[3.3.1] nonan-9-ones (1, 2) were synthesized and their 1H and 13C NMR data are reported. Chemical shifts and spectral assignments for 2,4,6,8-tetrakis(4-chlorophenyl)-3,7-diazabicyclo[3.3.1]nonan-9-one (3), 2,4,6,8-tetraphenyl-3-thia-7-azabicyclo[3.3.1]nonan-9-one (4) and 2,4,6,8-tetraaryl-3,7-diazabicyclo[3.3.1]nonanes (5-7) are also included.  相似文献

The reduction of 8-methyl-5-methylsulfanylmethyl-3-thiabicyclo[3.3.1]non-7-en-6-one with aluminum hydride, lithium tetrahydridoaluminate, or lithium tris(tert-butoxy)hydridoaluminate in tetrahydrofuran gave 8-methyl-5-methylsulfanylmethyl-3-thiabicyclo[3.3.1]non-7-en-6-ol. 8-Methyl-5-methylsulfanylmethyl-3-thiabicyclo[3.3.1]nonan-6-ol was obtained by reduction of the title compound with sodium tetrahydridoborate in pyridine or dimethylformamide. The reaction of 8-methyl-5-methylsulfanylmethyl-3-thiabicyclo[3.3.1]non-7-en-6-one with hydroxylamine hydrochloride afforded the corresponding oxime.  相似文献

Using the 1H NMR spectroscopic method it has been shown that 7-alkoxyalkyl-3-oxa-7-azabicyclo[3.3.1]nonan-9-ones and 7-alkoxyalkyl-3-oxa-7-azabicyclo[3.3.1]nonanes exist in deuterochloroform solution in a double chair conformation. 7-(3-Butoxypropyl)-3-oxa-7-azabicyclo[3.3.1]nonan-9-ol is a 1:1 mixture of the two stereoisomeric alcohols. One of them exists in a double chair conformation having an equatorial hydroxyl group with relation to the piperidine ring and the other in a chair-boat conformation having an axial hydroxyl group which involves an intramolecular hydrogen bond with the unshared electron pair of the nitrogen atom.  相似文献

The ring opening in the 1-benzyl-5,7-dimethyl-6-oxo-1-azonia-3-azaadamantane chloride under the treatment with excess aqueous alkali led to a stereoselective formation of anti-1,5-dimethyl-7-benzyl-3-formyl-3,7-diazabicyclo[3.3.1]nonan-9-ol whose structure was established by means of X-ray diffraction analysis and NMR spectroscopy. A reaction mechanism was suggested involving an intramolecular redox hydride transfer.  相似文献

Catalytic hydrogenation of 3-benzyl- and 3-tert-butoxycarbonyl-3-azabicyclo[3.3.1]nonan-9-one oximes over Raney nickel gave the corresponding 3-substituted 3-azabicyclo[3.3.1]nonan-9-amines which were converted into amides via reactions with acetyl and chloroacetyl chlorides and maleic and succinic anhydrides, into Schiff bases by condensation with benzaldehyde and 4-chlorobenzaldehyde, and into isothiocyanates by treatment with thiophosgene in the presence of K2CO3. 3-Benzyl- and 3-tert-butoxycarbonyl-3-azabicyclo-[3.3.1]nonan-9-yl isothiocyanates readily reacted with methanol, aniline, and sodium azide to produce methyl thiocarbamate, thiourea, and dihydrotetrazole-5-thione derivatives having a 3-azabicyclo[3.3.1]nonane fragment.  相似文献

The synthesis of bicyclic phosphorus–nitrogen (PN) compounds containing the rigid bicyclo[3.3.1]nonan-9-one framework was attempted using the Mannich condensation reaction of four different phosphorinanone classes with amines and aldehydes. Three different isomers of 4-tert-butyl-2,6-di(methoxycarbonyl)-3,5-bis(p-dimethylaminophenyl)-4-phosphacyclohexanone were obtained from the Michael addition reaction of tert-butylphosphine with 2,4-di(methoxycarbonyl)-1,5-bis(p-dimethylaminophenyl)penta-1,4-dien-3-one. The reaction of the all-equatorial isomer with methylamine and formaldehyde produced the bicyclic PN compound 7-tert-butyl-1,5-di(methoxycarbonyl)-6,8-bis(p-dimethylaminophenyl)-3-methyl-3-aza-7-phosphabicyclo[3.3.1]nonan-9-one. The identical Mannich reaction of the enol tautomer also yielded the same product, as well as the PN compound 4-tert-butyl-6-methoxycarbonyl-5-(p-dimethylaminophenyl)-2-methyl-2-aza-4-phosphacyclohexanone and the E/Z isomers of 3-(p-dimethylaminophenyl)methyl-2-propenoate. The newly synthesised 3-aza-7-phosphabicyclo[3.3.1]nonan-9-one PN compound adopts a chair–chair conformation both in solution and the solid state.  相似文献

A number of new 1,3-diazaadamantane derivatives containing quinoline fragments on C2 have been synthesized by condensation of 1,5-dialkyl-3,7-diazabicyclo[3.3.1]nonan-9-one, 1,5-dimethyl-3,7-diazabicyclo-[3.3.1]nonan-9-ol, and 1,5-dimethyl-3,7-diazabicyclo[3.3.1]nonane with 2-oxo-1,2-dihydroquinoline-3-carbaldehyde, 2-chloro- and 2-iodoquinoline-3-carbaldehyde, and quinoline-2-carbaldehyde.  相似文献

New 7-alkoxyalkyl-3-thia-7-azabicyclo[3.3.1]nonan-9-ones were synthesized by the double Mannich cyclization of tetrahydrothiopyran-4-one with suitable alkoxyalkylamines and paraformaldehyde in acetous methanol. Wolff-Kishner decarbonylation of these bicyclic ketones gave 7-alkoxyalkyl-3-thia-7-azabicyclo[3.3.1]nonanes. The reduction of 7-alkoxyalkyl-3-thia-7-azabicyclo[3.3.1]nonan-9-ones by alkali metal hydride complexes leads to a mixture of two stereoisomeric secondary alcohols, which are epimers at C(9). Abstract

A series of N-alkyl-3-thia-7-azabicyclo[3.3.1]nonan-9-ones and derivatives have been prepared from the reaction of an appropriate 4-thianone, an aldehyde and an amine in a Mannich type condensation. Reduction of the ketones via Wolff-Kishner conditions produced bicyclic systems with the methylene bridging group. Addition of Grignard reagents to the ketones did yield the expected alcohols. Conformational analysis of the systems was performed via diagnosis of the 1H and 13C NMR spectra. A single crystal X-ray diffraction analysis of 2,2,4,4-tetramethyl-6,8-diphenyl-3-thia-7-azabicyclo[3.3.1]nonan-9-one was accomplished and revealed a boat conformer for the nitrogen-containing ring and a chair conformer for the sulfur-containing ring. The ketone crystallized in the triclinic space group P1 (Z=2) with a=6.216 (3), b=10.507 (5), c=15.335 (6) Å, α=86.64 (4), β=86.66 (4), and γ=98.51 (5)° [Vol=986.55 Å3].  相似文献

Condensation of 3-substituted 3-azabicyclo[3.3.1]nonan-9-ones with hydroxylamine and hydrazine hydrate gave the corresponding oximes, hydrazones, and azines. Reductive amination of the title compounds in the presence of sodium triacetoxyhydridoborate led to the formation of 3-substituted 3-azabicyclo[3.3.1]nonan-9-amines which were converted into the corresponding dihydrochlorides by treatment with dry hydrogen chloride. Treatment of 3-tert-butoxycarbonyl derivatives with HCl under analogous conditions was accompanied by elimination of the tert-butoxycarbonyl group to produce 3-azabicyclo[3.3.1]nonan-9-amine dihydrochlorides.  相似文献

An improved method for the synthesis of 4-azatricyclo [5.2.2.04,8] undecan-11-one (1) is reported. The synthesis involves hydrogenolysis of 8-hydroxyethyl-2-azabicyclo [3.3.1] nonan-7-one 3 followed by intramolecular alkylation of the resulting secondary amine 7. The required azabicyclic alcohol 3 was obtained by oxidative cyclization with mercuric acetate of the α-alkylated methyl 4-piperidineacetoacetate 5.  相似文献

[reaction: see text] The facile stereoselective syntheses of endo-8-hydroxybicyclo[3.3.1]nonan-2-one and endo-7-hydroxybicyclo[3.2.1]octan-2-one, featuring an alpha-amino acid catalyzed intramolecular aldolization of sigma-symmetric substrates, are described. A high enantioselectivity and a high catalytic efficiency have been exhibited by (4R,2S)-tetrabutylammonium 4-TBDPSoxy-prolinate in the aldolization of 3-(4-oxocyclohexyl)propionaldehyde to give highly enantiomerically enriched (1S,5R,8R)-8-hydroxybicyclo[3.3.1]nonan-2-one.  相似文献

Bromination of 3-isopropyl-7-methyl- and 3-isopropyl-7-bromomethyl-3-borabicyclo[3.3.1]nonane leads to corresponding 3-(2-bromo-2-propyl) derivatives, which, on treatment with alcohols or pyridine as well as on heating, undergo the Matteson-Pasto rearrangement to convert into 3-X-4,4,8-trimethyl- and 3-X-4,4-dimethyl-8-bromomethyl-3-borabicyclo[4.3.1]decane (X = Br, OR). Interaction between triethylamine and 3-(2-bromo-2-propyl)-7-methyl-3-borabicyclo[3.3.1]nonane is accompanied by dehydrobromination leading to 3-isopropenyl-7-methyl-3-borabicyclo[3.3.1]nonane. Carbonylation of 3,4,4,8-tetramethyl-3-borabicyclo[4.3.1]decane at 140°C is accompanied by migration of two alkyl groups from the boron to the carbon atom, and subsequent oxidation with H2O2 produces 1-(2-hydroxy-2-methyl-1-propyl)-3-acetonyl-5-methyl-cyclohexane. Under more forcing conditions (180-195°C), the third alkyl group also migrates to give, after oxidation, a mixture of isomeric 3,4,4,8-tetramethylbicyclo[4.3.1]decan-3-ols. 3-n-Butoxy-4,4-dimethyl-8-bromomethyl-3-borabicyclo[4.3.1]decane, on treatment with Lì, undergoes cyclization to afford 4,4-dimethyl-3-borahomoadamantane, carbonylation and subsequent oxidation of which gave 4,4-dimethylhomoadamantan-3-ol.  相似文献

The reaction of bicyclo[3.3.1]nonane-2,6-dione with diazomethane in situ does not lead to the homologous bicyclo[4.3.1]decane-2,7-dione, but mainly to tricyclo[4.4.0.02,9]decan-9-ol-5-one. The structure of the latter was confirmed by the proton NMR spectra measured with an addition of Eu(DPM)3, A mixture of tricyclo[4,4.0.02,9]decan-9-ol-5-one and bicyclo[4.3.1]decane-2,7-dione results when solutions of diazomethane are used. The reaction of bicyclo[3.3.1]nonane-2,6-dione monoethyleneacetal with diazomethane in situ yields predominantly bicyclo[4.3.1]decane-2,7-dione. Under the same conditions bicyclo[3.3.1]nonan-2-one gives with diazomethane in situ only bicyclo[4.3.1]decan-2-one.  相似文献
