A simple and sensitive chemiluminescence (CL) procedure is proposed for the determination of cyromazine (CYR) using flow injection technique. CYR has strong enhancing effect on the CL reaction of diperiodatoargentate-(III) complex (DPA) in H2SO4 medium. The CL intensity with solid phase extraction (SPE) technique and with and without using online ion exchange resin column (IERC; OH-form) was proportional to the concentration of CYR over the range 0.1–200, 10–1000 and 2–2500 µg L–1 (R2 = 0.9974, 0.9980 and 0.9990, n = 7 each), respectively. Under the conditions, the limits of detection (S/N = 3) 0.029, 2.5 and 0.5 µg L–1, relative standard deviations (n = 3) 1.9–3.6%, 1.4–2.7% and 1.0–3.0% and sample throughputs were 120, 80 and 120 h–1. The effect of reagents concentration, flow rate, sample loop volume, photomultiplier voltage and IERC length was optimised. The mean results for natural water samples analysed by the proposed method were not significantly different at 95% confidence limit with the previously reported HPLC method. Interference from chloride ions could be eliminated by using SPE procedure or incorporating an in-line IERC. The CL mechanism of DPA–H2SO4–CYR system was also discussed briefly.  相似文献

A rapid and sensitive method for the analysis of three catecholamines by capillary electrophoresis(CE)with directchemiluminescence(CL)detection is described.The detection limits(S/N=3)were 1.3*10-8g/mL for isoprenaline,1.0*10-8g/mL for epinephrine and 2.8*10-8g/mL for dopamine.The proposed method was successfully applied to theanalysis of catecholamines in urine samples of cigarette smokers and nonsmokers.The results showed that there is a close relationbetween the release of dopamine in human body fluids and cigarette smoking/nonsmoking.  相似文献

Concentrations of amino acids in a human plasma pool were determined using four independent quantification methods. Orthogonal separation schemes (LC, GC, or GC×GC) and detection systems (triple quadrupole or time-of-flight mass spectrometry) are shown to demonstrate excellent consistency among platforms for quantifying 18 amino acids in NIST Standard Reference Material (SRM) 1950 Metabolites in Human Plasma using a well-characterized isotope dilution (ID) quantification method. Measured levels were consistent with reference values in plasma from the literature. Individual amino acid concentrations in plasma varied by over an order of magnitude ranging from 1.83 μg/g to 28.0 μg/g (7.78 μmol/L to 321 μmol/L). Average variability (coefficient of variation) between experimental amino acid concentrations (excluding cysteine) among all methods was 6.3%. Certified mass fraction values for amino acids in NIST SRM 1950 will be established from statistically weighted means of all experimental results.  相似文献

In this paper, a sensitive flow injection chemiluminescence system luminol–myoglobin was described for determining femtogram nitrite. Nitrite bound myoglobin producing the ferric heme nitrite complexes, which catalyzed the electron transfer of luminol to myoglobin leading to fast chemiluminescence. The chemiluminescence intensity in the presence of nitrite was remarkably enhanced compared with that in the absence of it. Under the optimum reaction conditions the chemiluminescence increment produced was proportional to the concentration of nitrite in the range of 0.05 pg ml 1–1.0 ng ml 1 (R2 = 0.9991), with a detection limit (3σ) of 20.0 fg ml 1. At the flow rate of 2.0 ml min 1, the whole process including sampling and washing could be completed in 0.5 min offering the sampling efficiency of 120 h 1 accordingly, and the relative standard deviation (RSD) was less than 2.60% (n = 5). It was satisfactory for the application to determine nitrite in human urine samples, and the possible mechanism was proposed.  相似文献

A novel flow injection chemiluminescence (FI-CL) sensor for determination of sulfadiazine (SDZ) using core–shell magnetic molecularly imprinted polymers (MMIPs) as recognition element is developed. Briefly, a hydrophilic MMIPs layer was produced at the surface of Fe3O4@SiO2 magnetic nanoparticles (MNPs) via combination of molecular imprinting and reversible stimuli responsive hydrogel. And it provided the MMIPs with excellent adsorption capacity and rapid adsorption rate due to the imprinted sites mostly situated on the surface of MMIPs. Then the prepared SDZ-MMIPs were packed into flow cell to establish a novel FI-CL sensor. The sensor provided a wide linear range for SDZ of 4.0 × 10−7 to 1.0 × 10−4 mol L−1 with a detection limit of 1.54 × 10−7 mol L−1. And the relative standard deviation (RSD) for the determination of 1.0 × 10−6 mol L−1 SDZ was 2.56% (n = 11). The proposed method was applied to determine SDZ in urine samples and satisfactory results were obtained.  相似文献

Chlorophenoxy acid herbicides are intensively applied to get rid of unwanted plants because of their low cost and selectivity. Due to their toxicity, which depends on their chemical form, the European Community has established legal directives to restrict their use and to control their maximum residue levels in several matrices. Determination of chlorophenoxy acids—2,4-dichlorophenoxyacetic acid (2,4-D), 4-chloro-2-methylphenoxyacetic acid (MCPA), 2-(2,4-dichlorophenoxy)propanoic acid (2,4-DP), 2-(4-chloro-2-methylphenoxy)propanoic acid (MCPP), 4-(4-chloro-2-methylphenoxy)butanoic acid (MCPB) and 2-(2,4,5-trichlorophenoxy)propanoic acid (2,4,5-TP) in spiked human urine samples has been carried out by capillary LC, after solid-phase extraction on a column packed with silica C18 restricted-access material. Chromatographic analysis was performed in gradient-elution mode at 25 °C, with injection of 20 μL low-organic-solvent composition herbicide solutions for focusing purposes on the head of the capillary column, and diode array detection at 232 nm. Urine samples collected during 24 h from healthy and unexposed volunteers were spiked in the concentration range 25–150 μg L−1; recoveries obtained were between 66 and 100% (n = 6 for each spiked level) and RSDs (relative standard deviations) were between 1 and 5%. Detection limits in the urine samples from volunteers were between 3.5 and 6.0 μg L−1. The developed methodology has allowed the clean-up and preconcentration of low volumes of untreated human urine without previous treatment, showing the effectiveness of the employed SPE sorbent for extracting the target analytes and ultimately resulting in the reduction of the sample-preparation time.  相似文献

This work presents a sensitive and rapid analytical method for the determination of oxcarbazepine in human plasma and urine samples. A vortex-assisted switchable hydrophilicity solvent-based liquid phase microextraction (VA–SHS–LPME) was used to preconcentrate oxcarbazepine from the samples before the determination by gas chromatography mass spectrometry. The switchable hydrophilicity solvent was synthesized by protonating N,N-dimethylbenzylamine with carbon dioxide to make it totally miscible with an equivalent volume of water. Parameters of the VA–SHS–LPME method including volume of switchable hydrophilicity solvent, concentration/volume of sodium hydroxide and vortex period were systematically optimized. Under the optimum conditions, good linearity ranging from 27.03 to 353.47 μg/kg was obtained for the analyte. Limit of detection and quantitation values were found to be 6.2 and 21 μg/kg (mass base), respectively. The relative standard deviation was calculated as 6.9% for six replicate measurements of the lowest concentration of the calibration plot. Satisfactory recovery results were calculated in the range of 97–100% for human plasma and urine samples spiked at five different concentrations.  相似文献

A nanomaterial comprising Fe3O4-modified hydroxylated multi-walled carbon nanotubes (Fe3O4–MWCNTs–OH) was prepared by a co-precipitation method. Combined with HPLC-photodiode array detector (DAD), Fe3O4–MWCNTs–OH was used to determine brucine in human urine. Some important parameters that could influence extraction efficiency of brucine were optimized, including the extraction time, amounts of Fe3O4–MWCNTs–OH, pH of sample solutions, desorption solvent and desorption time. Under the optimal conditions, the recoveries of brucine from spiked urine samples were between 93.1 and 104.1%, and the relative standard deviations (RSDs) ranged from 3.1 to 5.7%. The correlation coefficient was 0.9997. The limits of detection and quantification were 6 and 21 ng/mL at a signal-to-noise ratio of 3 and 10, respectively. The results indicated that Fe3O4–MWCNTs–OH combined with HPLC–DAD is a promising solid-phase extraction material for the sample pretreatment in the determination of brucine.  相似文献

A field-amplified sample injection (FASI) technique was elaborated for fast and sensitive determination of selected central nervous system drugs in human urine samples. Factors affecting the sensitivity enhancement, such as background electrolyte (BGE) and the analytical matrix composition were optimized and discussed. Pseudo-isotachophoresis (p-ITP) mechanism contribution in preconcentration mechanism was discussed. All separations were performed in uncoated fused silica capillaries 50 μm × 57 cm at 22 kV. The optimized analytical matrix was composed of 0.25 mM HCOOH in 90% (v/v) methanol, while BGE contained 45 mM TRIS/HCl (pH 2.20). The head-column injection was performed in 0.25 mM HCOOH water solution (3 s, 3.45 kPa). Sample was introduced into the capillary by electrokinetic injection (70 s, 5 kV) followed by short BGE plug (3 s, 3.45 kPa). Seven psychiatric drugs (olanzapine, prochlorperazine dimaleate, trifluoperazine dihydrochloride, perphenazine, promazine hydrochloride, clomipramine hydrochloride, and chlorprothixene hydrochloride) were separated in about 6 min. The elaborated method was additionally supported with dispersive liquid–liquid microextraction (DLLME) technique which in summary with FASI provided about 8000–13,000-fold sensitivity enhancement in comparison to the capillary zone electrophoresis (CZE) method with standard hydrodynamic injection (5 s, 3.45 kPa).  相似文献

In-line solid-phase extraction–capillary electrophoresis coupled with mass spectrometric detection (SPE–CE–MS) has been used for determination of 2-ethylidene-1,5-dimethyl-3,3-diphenylpyrrolidine (EDDP), codeine (COD), hydrocodeine (HCOD), and 6-acetylmorphine (6AM) in urine. The preconcentration system consists of a small capillary filled with Oasis HLB sorbent and inserted into the inlet section of the electrophoresis capillary. The SPE–CE–MS experimental conditions were optimized as follows: the sample (adjusted to pH 6.0) was loaded at 930 mbar for 60 min, elution was performed with methanol at 50 mbar for 35 s, 60 mmol L−1 ammonium acetate at pH 3.8 was used as running buffer, the separation voltage was 30 kV, and the sheath liquid at a flow rate of 5.0 μL min−1 was isopropanol–water 50:50 (v/v) containing 0.5% acetic acid. Analysis of urine samples spiked with the four drugs and diluted 1:1 (v/v) was studied in the linear range 0.08–10 ng mL−1. Detection limits (LODs) (S/N = 3) were between 0.013 and 0.210 ng mL−1. Repeatability (expressed as relative standard deviation) was below 7.2%. The method developed enables simple and effective determination of these drugs of abuse in urine samples at the levels encountered in toxicology and doping.  相似文献

A sensitive and selective liquid chromatographic–tandem mass spectrometric (LC–MS–MS) method was developed to determine pantoprazole sodium (PNT) in human urine. After solid-phase extraction with SPE cartridge, the urine sample was analysed on a C18 column (symmetry 3.5 μm; 75 mm × 4.6 mm i.d) interfaced with a triple quadrupole tandem mass spectrometer. Positive electrospray ionization was employed as the ionization source. The mobile phase consisted of acetonitrile–water (90:10, v/v). The method was linear over a concentration range of 1–100 ng mL?1. The lower limit of quantitation was 1 ng mL?1. The intra-day and inter-day relative standard deviation across three validation runs over the entire concentration range was <10.5%. The accuracy determined at three concentrations (8.0, 50.0 and 85.0 ng mL?1 PNT) was within ±1.25% in terms of relative errors.  相似文献

A simple method using solid-phase extraction (SPE) and ultra high-performance liquid chromatography (UHPLC) for the simultaneous determination of β-blockers, isoflavones, and flavonoids in human urine is developed. A statistical central composite design and response surface analysis is used to optimize the separation of the analytes. These multivariate procedures are efficient in determining the optimal separation condition using resolutions and retention time as responses. A gradient elution using a mobile phase consisting of 0.05% trifluoroacetic acid in water and acetonitrile is applied on a Hypersil GOLD column within a short analysis time of 4.5 min. UV detection was used to monitor the analytes. The suggested method was linear in a concentration range from 0.04-20.00 μg/mL, depending on the compound. The limits of detection ranged from 8.9 to 66.2 ng/mL. The precision was lower than 2.74%, and the accuracy was between 0.01-3.65%. The Oasis HLB column, with the highest recoveries, is selected for the pre-concentration step. This present paper reports, for the first time, a method for the simultaneous determination of β-blockers, isoflavones, and flavonoids in human urine samples. Furthermore, the developed method can also be applied to the routine determination of examined compounds concentrations in human urine.  相似文献

Water-soluble 2-hydroxypropyl-β-cyclodextrin (Hp-β-CyD), a cyclic and nonreducing oligosaccharide was used to enclose a hydrophobic guest molecule 1,1′-dimethylferrocene (DMF) to form a water-soluble yellow complex. At high concentrations (300 mM), Hp-β-CyD enclosed up to 100 mM DMF. The yellow complex was electrochemically oxidized (platinum vs Ag/AgCl poised at +450 mV) to form a blue dye, 1,1′-dimethylferricinium (DMF+). This is a one-electron transfer process and the ferricinium cation formed exhibited an absorption peak at 650 nm. The concentrated DMF+ was stable for at least 4 mo at 4°C and insensitive to a wide pH variation (pH 2–11). Application of the novel DMF+ complex as a colorimetric dye for the determination of uric acid in urine was successfully demonstrated. The reaction between the dye and uric acid is almost instantaneous and decrease in absorbance caused by the reduction of 1,1′-dimethylferricinium to 1,1′-dimethylferrocene can be followed at 650 nm. The results obtained agreed well with those of the reference reversed-phase HPLC method.  相似文献

A new method for the enrichment of Strychnos alkaloids in biological samples via liquid-phase microextraction (LPME) based on porous polypropylene hollow fibers in combination with on-line sweeping in micellar electrokinetic chromatography was developed. The calibration curve was linear over the range of 20-200 ng mL-1 for both strychnine and brucine in human urine sample. The detection limits (S/N=3:1) for strychnine and brucine were 1 ng mL-1 and 2 ng mL-1, respectively. The LPME-sweeping method has been successfully applied to the analysis of strychnine and brucine in real urine samples.  相似文献

Catechins, one of the class of flavonoids, are known as very efficient antioxidants. Here we investigated the kinetics of the reactions of three catechins, namely, catechin, epigallocatechin, and epigallocatechin gallate (EGCG) with some oxidants, which are formed in vivo under oxidative stress, hypochlorite, peroxynitrite, and amino acid peroxyl radicals. Stopped-flow spectrophotometry and pulse radiolysis technique with absorption detection were used to observe the formation of intermediate products of oxidized catechins. We found that catechins react with hypochlorite with the rate constant of the order of 105–106 M−1 s−1 at pH 7.4. Experimental kinetic traces of the reaction of EGCG with valine peroxyl radicals were fitted using chemical simulation, and the rate constant of this reaction was found to be 5 × 105 M−1 s−1. The rate constants of the formation of unstable catechin quinones in the reaction with peroxynitrite were comparable to that of spontaneous peroxynitrite isomerization, which indicates that catechins are oxidized indirectly by peroxynitrite. Biological consequences of these reactions are discussed.  相似文献
