离子液体中脂肪酶催化拆分外消旋烯丙酮醇反应 总被引:2,自引:0,他引:2
以离子液体为溶剂,考察了溶剂类型、水活度、温度、 pH值和共溶剂等因素对脂肪酶催化拆分外消旋烯丙酮醇(R,S)-4-羟基-3-甲基-2-(2-丙烯基)-2-环戊烯-1-酮反应的影响,并与常用于外消旋烯丙酮醇拆分的有机溶剂乙酸乙烯酯进行了比较. 结果表明,在离子液体[bmim]PF6中脂肪酶的催化性能较好,酶初始反应速率为18.48 μmol/(g·min), 半衰期为74.53 h, 高于在乙酸乙烯酯中的相应值(9.18 μmol/(g·min)和64.29 h). 但离子液体中拆分反应的转化率低于在乙酸乙烯酯中的转化率,可以通过向离子液体中补加酰基供体来提高外消旋烯丙酮醇的转化率. 两种反应介质中最佳酶反应条件均为水活度0.17, 温度40 ℃和pH=7, 但加入共溶剂后,离子液体中脂肪酶催化拆分外消旋烯丙酮醇的效率降低,而在乙酸乙烯酯中则有所提高. 相似文献

用聚乙烯醇包埋、硫酸钠固定法对根霉( Rhizopussp.Bc0-09)菌体细胞进行了固定化,所得凝胶具有良好的机械性和稳定性. 对固定化细胞催化( R,S)-环氧丙醇丁酸酯水解反应的性能进行了研究. 对pH值的影响考察结果表明, 30 ℃下, 反应初速度在pH 值为5.5 时最大; 酶的立体选择性在pH值为5.5~6.0 时最高, 对映体比率( E值)为57. 对温度的影响考察结果表明,pH 值为5.5, 反应初速度在55 ℃时最大; 酶的立体选择性随温度升高而下降. 固定化细胞具有较好的操作稳定性. 相似文献

Bacillus sp. DL-2 was isolated from the deep sea of the Western Pacific and further utilized as novel biocatalysts to efficiently asymmetrically hydrolyze (±)-1-phenylethyl acetate. After the optimization of hydrolytic reactions, chiral chemicals (R)1-phenylethanol and (S)-l-phenylethyl acetate were obtained with high optical purities (96% and 99.8%, respectively). Our research is about the asymmeric hydrolysis of (±)-1-phenylethyl acetate using whole-cell biocatalysts. In addition, the optical purity of (S)-l-phenylethyl acetate generated through the kinetic resolution of (±)-1-phenylethyl acetate using the whole-cells of Bacillus sp. DL-2 was the highest report so far. Using the whole cells of deep sea bacterium Bacillus sp. DL-2 as the biocatalysts is an enviromnentally friendly method and will play critical roles in industrial asymmetric synthesis. 相似文献

By using acetonitrile as the sole nitrogen source, a microbial strain with high nitrilase activity, named as Alcaligenes sp. ECU0401, was newly isolated from soil, which could enantioselectively transform racemic mandelonitrile into (R)-(-)-mandelic acid, with an enantiomeric excess of >99.9%. 相似文献
