研究Diaion HP–20大孔吸附树脂分离纯化含羞草种子中黄酮类物质的工艺条件。用海南含羞草种子为原料,以含羞草种子中总黄酮吸附量和解吸率为评价指标,考察上样流量、洗脱液种类、柱径比、洗脱液流量等影响因素。Diaion HP–20大孔吸附树脂分离纯化含羞草种子中黄酮类物质的最佳工艺条件:上样流量3 m L/min,洗脱液为40%乙醇溶液,柱径比为1∶10,洗脱液流量为4 m L/min。该方法简单,可行,能够有效分离纯化含羞草种子中黄酮类物质,且大孔吸附树脂可重生,利用率高。  相似文献

用超声法提取昆仑雪菊中黄酮类化合物,通过大孔树脂对提取液进行纯化,纯化物经聚酰胺层析柱梯度洗脱得到初步分离物,通过颜色反应和紫外光谱扫描进行定性分析。设计正交实验得到超声法提取的最佳工艺条件是65%乙醇、料液比为1∶40、超声时间60 min,此条件下提取3次,总黄酮得率可达132.6 mg/g。通过聚酰胺柱层析法分离出五个组分,初步判断它们属于不同的黄酮类物质。超声法提取总黄酮,提取率较高,聚酰胺树脂对黄酮类化合物的分离效果较好。  相似文献

利用超声波辅助提取柠檬汁中总黄酮,分光光度法测定总黄酮类化合物含量,利用单因素实验,找出了最佳提取工艺条件70%乙醇、料液比1∶1.5、超声温度55℃、超声时间60 min。通过对比实验找出了最佳纯化方法二氯甲烷在1∶1.5比例下萃取、大孔树脂吸附18 h、70%乙醇洗脱,洗脱液除醇后所得的黄酮浓度最高达1.851 mg/m L。  相似文献

比较了D-101、D-160、AB-8、NKA-9和聚酰胺等5种吸附树脂对枇杷花总黄酮的吸附及解吸附性能。在静态吸附和动态吸附实验基础上,筛选出效果较好的AB-8树脂进行动态吸附参数的研究。考察了样品液pH值、样品液浓度、洗脱液浓度、上样速度、洗脱速度等对AB-8树脂吸附和解吸效果的影响,确定了AB-8树脂动态吸附枇杷花总黄酮的最佳条件。获得的最佳纯化条件如下,样品液pH值为5.5,样品液浓度为12mg/mL,洗脱液为30%的乙醇水溶液,上样速度为2BV/h,洗脱速度为1BV/h。纯化后样品总黄酮含量达86.7%,比纯化前总黄酮含量高5~6倍。实验结果表明,AB-8树脂可用于分离纯化枇杷花总黄酮。  相似文献

考察了NKA-9、S-8、XDA-1、AB-8、HPD-100、HPD-600 6种大孔吸附树脂对宁夏枸杞总黄酮的吸附和解吸性能,筛选出XDA-1树脂的效果最佳;考察了pH值、样液浓度、流速等对XDA-1树脂静态吸附效果的影响;并进行了动态吸附试验,确定出XDA-1树脂动态吸附枸杞总黄酮的最佳条件:样品液浓度为0.25mg/mL,控制流速为0.5mL/min,样品液pH 5;最佳洗脱条件:洗脱液为80%的乙醇水溶液,用量为35mL (5倍柱床体积).在此条件下,枸杞总黄酮含量从27.2%提高到79.8%,回收率为87.4%,表明XDA-1树脂可用于宁夏枸杞总黄酮的分离纯化.  相似文献

筛选分离纯化昆仑雪菊总黄酮的大孔吸附树脂并建立纯化工艺条件。以大孔吸附树脂对昆仑雪菊总黄酮的吸附量、吸附率和解吸率等为指标,考察了11种型号的大孔吸附树脂进行分离纯化,并确定了该树脂分离纯化最佳工艺参数。结果显示NKA-9型大孔吸附树脂对昆仑雪菊总黄酮分离纯化效果好,其工艺条件为:以pH值为5.0的原料液上柱吸附,70%浓度的乙醇洗脱、洗脱速度3mL·min-1,在此条件下总黄酮的的静态吸附率为95.74%,解吸率为98.9%。  相似文献

考察了4种大孔吸附树脂对仙人掌总黄酮的吸附分离性能,筛选出效果最佳的树脂为AB-8。以总黄酮的吸附量、总黄酮含量和回收率为考察指标,采用紫外分光光度法测定总黄酮。确定了AB-8树脂吸附分离仙人掌总黄酮的工艺条件:上样浓度为15mg/mL,仙人掌总黄酮最大吸附量为18.6mg/mL,吸附流速为5mL/min,洗脱剂为70%乙醇,洗脱剂用量为6倍柱体积,树脂可重复使用3次。经AB-8树脂分离纯化后,总黄酮含量从29%提高到76%,总黄酮回收率为86%。实验结果表明,AB-8树脂可用于仙人掌总黄酮的分离纯化。  相似文献

筛选适合分离纯化楮果总黄酮的大孔树脂并确定最优工艺条件。以静态吸附率和解吸率为指标对8种大孔树脂进行筛选,确定D101树脂的分离纯化效果最佳。通过动态吸附实验考察上样流速、上样溶液pH值、上样溶液浓度、乙醇浓度、洗脱流速、洗脱剂用量等工艺条件对分离纯化效果的影响,确定最优工艺条件如下:上样流速为2BV/h,pH值为6,上样溶液浓度为0.05mg/mL,80%乙醇作洗脱剂,洗脱流速为5BV/h,洗脱剂用量为7.5BV。采用最优工艺条件,楮果总黄酮含量提高至22.26%,产品精制倍数为4.79,表明D101树脂能有效纯化楮果总黄酮。  相似文献

采用温浸法设计四因素三水平正交试验,对黄芪多糖最佳提取工艺进行了优化,结果表明:四因素对黄芪多糖提取的影响顺序为提取温度>提取次数>料液比>提取时间,提取最佳工艺为:料液比1∶6,提取时间90 min,提取温度100℃时提取3次;采用乙醇沉淀法设计三因素三水平正交实验对其最佳分离工艺进行研究,研究发现:三因素三水平对黄芪多糖分离影响顺序为乙醇浓度>乙醇加入量>沉淀时间,分离的最佳工艺为乙醇浓度为90%,加入量5倍体积,沉淀时间4 h;选用AB-8大孔吸附树脂和聚酰胺为吸附剂,不同浓度乙醇为洗脱剂对黄芪多糖最佳纯化工艺进行了探索,确定了最佳纯化工艺为:AB-8大孔吸附树脂吸附,30%乙醇洗脱.这些条件的确定为黄芪的大规模开发和应用奠定了基础.  相似文献

采用溶剂提取法、索氏提取法、循环超声提取法提取火棘果总黄酮,利用静态吸附方法筛选分离火棘果总黄酮的最适大孔树脂,利用动态吸附方法研究最适大孔树脂纯化火棘果总黄酮的条件,并用紫外分光光度法测定其含量。得出结果为循环超声提取时间短、效率高;D101大孔吸附树脂纯化效果最好,最佳工艺为上样浓度为0.899 8g·L-1,上样液pH为4,上样体积5BV,上样速率2.5mL·min-1,用5BV70%乙醇以2.0 mL·min-1速率洗脱,经树脂纯化后总黄酮的纯度由原来的9.00%提高至28.11%。  相似文献

An integrated procedure was developed to extract and purify total flavonoids from Toona sinensis leaves for the first time, in which pressurized liquid extraction was performed in tandem with HPD100 macroporous resin column. Consequently, the total flavonoids can be extracted using 10% EtOH, and the recovery and purity of total flavonoids was 71.05% and 66.60%. Moreover, products of high quality were obtained in an environmentally friendly process with lower consumption of time and solvent. The results demonstrated that the integrated extraction-adsorption procedure was an efficient process for the preparation of total bioactive flavonoids from Toona sinensis leaves.  相似文献

In recent years, green extraction of bioactive compounds from herbal medicines has generated widespread interest. Deep eutectic solvents (DES) have widely replaced traditional organic solvents in the extraction process. In this study, the efficiencies of eight DESs in extracting flavonoids from Acanthopanax senticosus (AS) were compared. Response surface methodology (RSM) was employed to optimize the independent variable including ultrasonic power, water content, solid-liquid ratio, extraction temperature, and extraction time. DES composed of glycerol and levulinic acid (1:1) was chosen as the most suitable extraction medium. Optimal conditions were ultrasonic power of 500 W, water content of 28%, solid-liquid ratio of 1:18 g·mL−1, extraction temperature of 55 °C, and extraction time of 73 min. The extraction yield of total flavonoids reached 23.928 ± 0.071 mg·g−1, which was 40.7% higher compared with ultrasonic-assisted ethanol extraction. Macroporous resin (D-101, HPD-600, S-8 and AB-8) was used to recover flavonoids from extracts. The AB-8 resin showed higher adsorption/desorption performance, with a recovery rate of total flavonoids of up to 71.56 ± 0.256%. In addition, DES solvent could efficiently be reused twice. In summary, ultrasonic-assisted DES combined with the macroporous resin enrichment method is exceptionally effective in recovering flavonoids from AS, and provides a promising environmentally friendly and recyclable strategy for flavonoid extraction from natural plant sources.  相似文献

采用高效制备液相色谱法从荷叶(Nelumbo nucifera Gaertn)中分离制备荷叶黄酮类化合物。用60%乙醇回流提取荷叶,粗提液浓缩后经D-101柱及聚酰胺柱色谱分离,再在Symmetry PrepTM C18柱上分离,以水-乙腈为流动相进行梯度洗脱(流速为5.0 mL/min),得到了3种黄酮类化合物。经紫外光谱、红外光谱、核磁共振及质谱分析,确定该3种物质分别为金丝桃苷、异槲皮苷和紫云英苷。所制备的3种化合物的纯度都在97%以上,其中紫云英苷为首次从荷叶中分离得到。  相似文献

An efficient ultrasound-assisted extraction technique was employed to extract total flavonoids from Sparganii rhizoma. The optimum extraction conditions for the highest yield of total flavonoids were ethanol concentration 53.62%, ultrasonication time 29.41 min and ultrasound power 300 W, which were determined using response surface methodology. The extraction yields of the optimal ultrasound-assisted extraction were higher than using conventional extraction. The crude extract was then purified on a polyamide resin, whereby the flavonoids content in the purified extract increased to 94.62%. The antioxidant activities of the purified flavonoids including DPPH radical scavenging activity, ABTS+ radical scavenging activity, reducing power, hydroxyl radical scavenging activity and superoxide anion scavenging activity, were evaluated in vitro, which suggested that the flavonoids showed significant antioxidant activities. Rutin, kaempferol and formononetin were identified in the extract by comparing relative retention times and UV-Vis spectra with those of reference standards.  相似文献

比较了NKA-9、D311、S-8、HPD600、NKA-2、A型、D140和聚酰胺等8种树脂对杜仲中降血压活性成分京尼平甙酸的吸附及脱附性能,从中筛选出吸附率及脱附率均较高的A型树脂进行试验.确定了最佳工艺条件:杜仲皮粉末用50%乙醇水溶液提取后,95%乙醇沉淀,上清液回收乙醇后,用蒸馏水稀释,调节pH=6~9后用A型树脂吸附,15%乙醇溶液洗脱,流速为1.5ml/min,洗脱液浓缩后冷冻干燥得产品.  相似文献

In the present paper is reported the metnocl for the isolation and extraction of total flavonoids of Epimedium Koreanum Nateai by means of supercritical fluid extraction(SFE). By examining pressure, temperature, amounts of modifier and extraction time, the optimized condition of SFE is confirmed as 30 MPa and 60℃, with 70% ethanol as the modifier. The samples were statically extracted for 30 min, followed bydynamic extraction for 120 min at a flow rate of 6 mL/min. The quantitative analysis of total flavonoids was performed by UV-Vis spectrophotometry. Compared with the conventional method, the SFE method is more efficient, more rapid and more friendly environmentally.  相似文献

The aim of this study was to investigate the antioxidant and free-radical scavenging activity of extract and fractions from various parts of Elsholtzia ciliata. The inflorescences, leaves, stems and roots of E. ciliata were extracted separately and two phenolic component enrichment methods: ethyl acetate-water liquid-liquid extraction and macroporous resin adsorption-desorption, were adopted in this study. The antioxidant activities of water extracts and fractions of E. ciliata were examined using different assay model systems in vitro. The fraction root E (purified by HPD300 macroporous resin) exhibited the highest total phenolics content (497.2 ± 24.9 mg GAE/g), accompanied with the highest antioxidant activity against various antioxidant systems in vitro compared to other fractions. On the basis of the results obtained, E. ciliata extracts can be used potentially as a ready accessible and valuable bioactive source of natural antioxidants.  相似文献
