Ten types of new P-chiral phosphorodiamidite ligands having pyrrolo[1,2-c][1,3,2]diazaphosphol-1-one backbone were designed and prepared. They were preliminarily utilized for iridium-catalyzed asymmetric allylic etherification of cinnamyl carbonate with phenol, where both R- and S-products were obtained with good enantioselectivity (up to 74% ee) by changing the N- and P-substituents of the ligands. The opposite enantioselectivity in iridium-catalyzed allylic substitution was explained by DFT calculations of the energy difference of the pi-allyliridium-phosphorodiamidite intermediates.  相似文献

Cycloaddition reaction between toluamides and benzonitriles was applied to prepare the 3-arylisoquinolines, and their chemical transformation to the dienes 4 was performed. The ring-closing metathesis (RCM) reaction afforded the desired heterocyclic compounds, benzo[3,4]azepino[1,2-b]isoquinolinones 5 in good yield.  相似文献

Bromination of naphtho[1,2-c][1,2,5]thiadiazole ( I ) gives either an addition product, 4,5-dibromo-4,5-dihydronaphtho[1,2-c][1,2,5]thiadiazole ( II ), or a substitution product, 5,6-dibromonaphtho[1,2-c][1,2,5]thiadiazole ( III ), depending on the reaction conditions. Dehydrobromination of II gives 5-bromonaphtho[12-c][1,2,5]thiadiazole ( IV ). Chlorination of I gives the corresponding addition product V or 5-chloronaphtho[1,2-c][1,2,5]thiadiazole ( VI ). Compound VI can also be obtained by dehydrochlorination of V. The nitration of I produces a mixture of isomeric substitution products, 9-nitro VIII and 6-nitronaphtho[1,2-c][1,2,5]thiadiazoles ( VII ).  相似文献

[reaction: see text] Cyclic mixed acetals with pendant diazoketone side chains undergo rearrangement to ether-bridged cycloheptane ring systems on treatment with Cu(hfacac)(2). Stevens [1,2]-shift of an oxonium ylide furnishes the major product (7), in some cases accompanied by minor amounts of a product (8) resulting from [1,2]-shift of a sulfonium ylide. In the subsequent sulfur-triggered cleavage of the bridging ether, the desired bicyclo[5.3.0]heptene was obtained, along with the product of novel S(N)2' attack on the resulting allylic ketal.  相似文献

3-Substituted imidazo[1,2-a]pyridines, imidazo[1,2-a]pyrimidines, and imidazo[1,2-c]pyrimidine were obtained regiospecifically in yields of 35-92% in one pot by reaction of 2-aminopyridines or 2-(or 4-)aminopyrimidines, respectively, with 1,2-bis(benzotriazolyl)-1,2-(dialkylamino)ethanes.  相似文献

Hydrazonoyl bromides 1a-c react with 5-amino-3-phenyl-1H-pyrazole, 5-amino-1H-1,2,4-triazole, 2-aminopyridine, and 2-aminobenzimidazole to afford the corresponding imidazol[1,2-b]pyrazoles 10, imidazo[1,2-b]-1,2,4-triazoles 11, imidazo[1,2-a]pyridines 16, imidazo[1,2-a]pyrimidines 17, and imidazo[1,2-a]benzimidazoles 20, respectively. A series of 1,2-dihydro-5-imidazo[1,2-a]pyridinyl-2(1H)-pyridonones was synthesized and evaluated for positive inotropic activity, 1,2-Dihydro-5-imidazo[1,2-a]pyridin-6-yl-6-methyl-2- oxo-3-pyridinecarbonitrile (11a) hydrochloride monohydrate (E-1020) was found to be a potent and selective inhibitor of phosphodiesterase III and a long-acting, potent, orally active positive inotropic agent. Additional imidazo[1,2-a]pyridin-2-yl (3a), -3-yl (16), -7-yl (20) and -8-yl (24a) compounds were also prepared. Altering the pyridine substitution from the 2-position to the 6-position produced a 2-fold increase in the i.v. cardiotonic potency (ED50) from 52 to 23 micrograms/kg, while substitution at the 3-, 7- or 8-position reduced potency. In the 2-positional isomers, introduction of halogen groups enhanced the activity and 3-chloro-1,2-dihydro-5-(6-fluoroimidazo[1,2-a] pyridin-2-yl)-6-methyl-2(1H)-pyridinone (3u) was the most potent (i.v. ED50 11 micrograms/kg) in this series. E-1020 is presently under development for the treatment of congestive heart failure.  相似文献

The synthesis of 1,2-dihydro-1-oxopyrrolo[1,2-d]-1,2,4-triazines was achieved by rearrangement of 2-pyrrolyloxadiazoles under alkaline conditions or by cyclisation of pyrrole N-ethoxymethylidene hydrazides. The cyclisation of the N-carbethoxy hydrazone of the pyrrole-2-carboxaldehyde gave the 3,4-dihydro-4-oxopyrrolo[1,2-d]-1,2,4-triazine. Electrophilic substitution reactions of the 1- and 4-pyrrolotriazinones were made either on the lactam nitrogen with methyl sulphate, benzyl chloride and monochloroacetic acid or on the pyrrole ring with bromine and nitric acid. Stable 3-acetyl derivatives of imidazo[1,2-a]benzimidazole were synthesized by the action of acetic anhydride on 2,9-disubstituted imidazo[1,2-a]benzimidazole. Methyl- and phenyl-substituted N-(ethoxycarbonyl)-2-azabicyclo[2.2.0]hex-5-enes 6 have been prepared by photoirradiation of appropriately substituted 1,2-dihydropyridines. Torquoselectivity is observed in the synthesis of the 3-endo-methyl- and 3-endo-phenyl-2-azabicyclo[2.2.0]hexenes 6c-e from 2-methyl- and 2-phenyl-1,2-dihydropyridines 5c-e. Products formed upon addition of bromine to 3-endo-, 4-, and 5-methyl- and 3-endo-phenyl-substituted N-(ethoxycarbonyl)-2-azabicyclo[2.2.0]hex-5-enes 6a-f were substituent dependent. For 6a,b, which lack substituents at C(3) or C(5), mixtures of unrearranged dibromides 8a,b and rearranged dibromides 9a,b were obtained. With the 3-endo-substituents in 6c-e, only rearranged dibromides 9c-e were formed; 5-methyl substitution afforded mainly unrearranged dibromide 8f and some allylic bromide 10. Both unrearranged 5-endo,6-exo-dibromo-2-azabicyclo[2.2.0]hexanes 8 and rearranged 5-anti-6-anti-dibromo-2-azabicyclo[2.1.1]hexanes 9 are formed stereoselectively. The dibromoazabicyclo[2.1.1]hexanes 9 have been reductively debrominated to afford the first reported 2-azabicyclo[2.1.1]hexanes 11 with alkyl or aryl substituents at C-3.  相似文献

The synthesis of the title compounds was carried out by cyclization via isocyanate of (E)-4,5-dihydro-10H-benzo[5,6]cyclohepta[1,2-b]-thiophene-10-ylideneacetic acid and 4,5-dihydro-10H-benzo[5,6]cyclohepta[1,2-b]-thiophene-10-ylacetic acid respectively, which were obtained by the Wadsworth-Emmons modification of the Wittig reaction of 4,5-dihydro-10H-10-oxobenzo[5,6]cyclohepta[1,2-b]thiophene and triethyl phosphonacetate. [reaction: see text] The viability of hydroxylamines as nucleophiles in transition-metal-catalyzed allylic substitutions was examined. We have found that the oxygen atom of hydroxylamines having an N-electron-withdrawing substituent (also known as hydroxamic acids) acts as a reactive nucleophile. The palladium-catalyzed O-allylic substitution of hydroxylamines with allylic carbonate afforded the linear hydroxylamines. The selective formation of the branched hydroxylamines was observed in iridium-catalyzed reaction. Regio- and enantioselective allylic substitution of the unsymmetrical phosphates with hydroxylamines was studied by using the iridium complex of chiral pybox ligand. The aqueous-medium reaction with hydroxylamines proceeded smoothly in the presence of Ba(OH)(2).H(2)O to give the branched products with good enantioselectivities.  相似文献

1,2-Diphenylbenzo[b]cyclopenta[e]pyran was pyrylated with 2,6-diphenylpyrylium perchlorate. The structures of the singly and doubly charged 2,6-diphenyl-4-(1,2-di-phenylbenzo[b]cyclopenta[e]-3-pyrania)pyrylium cations are confirmed by data from the electronic absorption spectra.
