A new device combining microdialysis with electrochemical microsensor was developed. It can be applied to monitor the biomolecules in the brain for biological and pharmaceutical research. In this paper, the device was applied to simultaneously determine ascorbic acid (AA) and 5-hydroxyindole-3-acetic acid (5-HIAA) in rat striatum. The microsensor used for the device was poly (sulphosalicylic acid) microsensor, which exhibited a good electrocatalytic effect on oxidization of AA and 5-HIAA. The oxidation currents measured by differential pulse voltammetry (DPV) were linear for AA in the range of 0.02-1.0 mmol l−1, and for 5-HIAA from 0.5 to 10.0 μmol l−1 (r=0.9998 and 0.9991, respectively). The detection limits were calculated to be 0.01 mmol l−1 for AA and 0.25 μmol l−1for 5-HIAA (S/N=3). Studies also showed that co-existing substances in biological fluids did not interfere with AA and 5-HIAA determination when using this microsensor. Since, the substances in the microdialysate are easily oxidized by air, the microdialysate in this device was under the protection of N2. It was found that the concentrations of AA and 5-HIAA in rat striatum were 215±5 and 6.21±0.61 μmol l−1 (mean±S.E.M., n=7), respectively with this device under the protection of N2. In addition, the effect of the nitric oxide donor, sodium nitroprusside (SNP), on 5-HIAA in the rat striatum was investigated. It was found that a high concentration of SNP (1.0 mmol l−1) resulted in a 34% increase in 5-HIAA level.  相似文献

A novel biosensor for determination of d-amino acids (DAAs) in biological samples by using an electrode based on immobilization of a thermostable d-Proline dehydrogenase (d-Pro DH) within an agar gel membrane was developed. The electrode was simply prepared by spin-coating the agar solution with the d-Pro DH on a glassy carbon (GC) electrode.An electrocatalytic oxidation current of 2,6-dichloroindophenol (DCIP) was observed at −100 mV vs. Ag/AgCl with the addition of 5 and 20 mmol L−1d-proline. The current response and its relative standard deviation were 0.15 μA and 7.6% (n = 3), respectively, when it was measured in a pH 8.0 phosphate buffer solution containing 10 mmol L−1d-proline and 0.5 mmol L−1 DCIP at 50 °C. The current response of d-proline increased with increase of the temperature of the sample solution up to 70 °C. The electrocatalytic response at the d-Pro DH/agar immobilized electrode subsequently maintained for 80 days. Finally, the d-Pro DH/agar immobilized electrode was applied to determination of DAAs in a human urine sample. The determined value of DAAs in the human urine and its R.S.D. were 1.39 ± 0.12 mmol L−1 and 8.9% (n = 3), respectively.  相似文献

This work reports a hybrid flow-injection analysis (FIA)/sequential-injection analysis (SIA) method for the rapid enzymatic assay of glucose with soluble glucose oxidase (GOD). The method relies on the sequential injection of segments of the sample and of a solution of enzyme by means of a multi-port selection valve in a flowing water stream. As the two zones are swept downstream, they overlap and merge so that the glucose in the sample is enzymatically oxidised. The generated hydrogen peroxide is merged with an alkaline luminol solution and the chemiluminescence (CL) intensity is monitored and related to the glucose concentration in the sample. The linear range of the method for glucose determination is 0.01-1 mmol L−1, the relative standard deviation is 3.9% at the 0.08 mmol L−1 level (n = 8), the limit of detection at the 2σ level is 4 μmol L−1 glucose and the injection rate is 80 h−1. The method was applied to the analysis of energy drinks and honey with relative errors in glucose determination in the range 100 ± 4.3%. The advantages of the proposed method are the wide linear range, the simple instrumentation used, the low consumption of sample and reagents, the elimination of catalysts and immobilised enzymes and the high sample throughput.  相似文献

High performance capillary electrophoresis (HPCE) was developed for quantitative determination of 18 phenylthiohydantoin (PTH)-amino acids. The influence of electrolyte concentration, pH, organic modifier and applied voltage on HPCE performance was investigated. The HPCE separation of a PTH-amino acids mixture was much improved by adding organic modifier and Tris-boric acid buffer to the run buffer. After optimization of the method, 17 PTH-amino acids in a solution containing 18 PTH-amino acids could be separated using 400 mmol l−1 Tris-boric acid, 1.0 mmol l−1 diethylamine at pH 9.5 adjusted with 0.1 mol l−1 NaOH as a run buffer, voltage of 25 kV was applied, temperature was maintained at 25 °C, detection wavelength was 254 nm. The precision (n = 7) of this method is less than 3.2% (peak area) and 1.1% (migration time) of relative standard deviation (R.S.D.). Linearity was established over the concentration range 50-1000 μM of each derivative, with correlation coefficients (r) ranging between 0.9904 and 0.9993. The detection limits (S/N = 3) range from 2 to 48 μmol l−1. The method was applied to determine amino acids in Sargassum fusiforme, a marine algae collected from Tongtou County of Zhejiang Province in China with satisfactory results.  相似文献

A new sequential injection (SI) system with spectrophotometric detections has been developed for successive determination of protein and glucose. The protein assay is based on ion-association of protein with tetrabromophenolphthalein ethyl ester (TBPE) in the presence of Triton X-100 at pH 3.2. The blue product is monitored for absorbance at 607 nm. For glucose, hydrogen peroxide, generated by the oxidation of glucose in the presence of glucose oxidase immobilized on glass beads packed in a minicolumn, is monitored using iron-catalyzed oxidation reaction of p-anisidine to form a red colored product (520 nm). The SI procedure takes advantage in performing the protein assay during the incubation period for glucose oxidation. Linear ranges were up to 10 mg dL−1 human serum albumin (HSA) with a limit of detection (LOD) (3σ) of 0.3 mg dL−1, and up to 12.5 mg dL−1 glucose with LOD of 0.08 mg dL−1. R.S.D.s (n = 11) were 2.7% and 2.5% (for 1 mg dL−1 and 5 mg dL−1 HSA) and 1.4% (9 mg dL−1 glucose). Sample throughput for the whole assay of both protein and glucose is 6 h−1. The automated system has been demonstrated for the successive assay of protein and glucose in urine samples taken from diabetic disease patients, with good agreement with the other methods. This developed SI system is an alternative automation for screening for diabetic diagnosis.  相似文献

A new, simple and sensitive spectrofluorimetric method for the determination of salicylic acid (λex = 315 nm, λem = 408 nm) using As(III) as a sensitizing reagent has been investigated by measuring the increase of fluorescence intensity of salicylic acid due to the complexation of As(III)-salicylic acid in presence of sodium dodecyl sulfate (SDS) 10−3 M. Under optimum conditions, a significant relationship was obtained between the fluorescence intensity and salicylic acid concentration. A linear calibration curve was obtained in the range 13.8-13812 μg l−1 with product-moment correlation coefficient (R) 0.99985 and detection limit 4.2 μg l−1. The R.S.D. is 2.35% (n = 5).The method was applied successfully to the determination of salicylic acid in human serum.  相似文献

A novel, highly sensitive, selective, and simple kinetic method was developed for the determination of Mo(VI) based on its catalytic effect on the oxidation of 1-amino-2-naphthol-4-sulfonic acid (ANSA) with H2O2. The reaction was followed spectrophotometrically by tracing the oxidized product at 465 nm after 30 min of mixing the reagents. The optimum reaction conditions were: 10 mmol l−1 ANSA, 50 mmol l−1 H2O2, 100 mmol l−1 acetate buffer of pH 5.0 ± 0.05 and at 40 °C. Addition of 200 μg ml−1 diethylenetriaminepentaacetic acid (DTPA) conferred high selectivity for the proposed method. Following the recommended procedure, Mo(VI) could be determined with a linear calibration graph up to 2.5 ng ml−1 and a detection limit, based on the 3Sb-criterion, of 0.027 ng ml−1. The unique sensitivity and selectivity of the implemented method allowed its direct application to the determination of Mo(VI) in natural and industrial waste water. The method was validated by comparison with the standard ETAAS method. Moreover, published catalytic-spectrophotometric methods for the determination of molybdenum were reviewed.  相似文献

Terbium sensitized fluorescence was used as a post-column detection system to develop a simple, sensitive and rapid high-performance liquid chromatographic method for the simultaneous determination of catecholamines norepinephrine (NE), epinephrine (E) and dopamine (DA).Catecholamines were separated by an ion-pair reversed-phase chromatography on a BDS-Hypersil analytical column with a mobile phase of methanol and 50 mmol l−1 acetate buffer (pH 4.7) containing 1.1 mmol l−1 SOS and 0.11 mmol l−1 EDTA (15+85 v/v).Catecholamines and the internal standard (3,4-dihydroxybenzylamine, DHBA) were post-column derivatized by the addition to the eluent of an alkaline solution containing a stoichiometric mixture of terbium(III) chloride and EDTA. Fluorescence detection (λex=300 nm, λem=545 nm) is based on the sensitization of terbium ion fluorescence after complexation with catecholamines.The chemical compatibility between the eluent and the post-column reagent was studied and the analytical characteristics of the method were established. Detection limits found were 1.0×10−8, 4.0×10−8 and 7.0×10−8 mol l−1 for NE, E and DA, respectively. The method has been successfully applied to the determination of catecholamines in urine samples after solid-phase extraction (SPE) pre-treatment. Recoveries from urine spiked with NE (4.0×10−7, 2.0×10−6 and 4.0×10−6 mol l−1), E (8.2×10−8, 4.1×10−7 and 8.2×10−7 mol l−1) and DA (1.0×10−6, 5.0×10−6 and 1.0×10−5 mol l−1) varied from 98 to 100% (mean=99.3%), from 106 to 107% (mean=106.3%) and from 98 to 101% (mean=99.3%), respectively. The between-run precision (relative standard deviation, R.S.D.) for the method for three urine samples at different concentration levels of each catecholamine varied from 3.6 to 7.0%.  相似文献

The aim of present work was to optimize the experimental parameters in single drop microextraction under solution immersion (SDME) and headspace (HS-SDME) extraction modes for the determination of periodate using guaifenesine [3-(2′-methoxyphenoxy)-1,2-propane diol] and norephedrine (phenylpropanolamine) as new and alternative reagents for the Malaprade reaction. The reactions were complete within 5 min resulting in the formation of 2-(2′-methoxyphenoxy)-acetaldehyde and benzaldehyde, respectively. SDME/HS-SDME of oxidation products with 2 μl of anisole or 1 μl of toluene, respectively, has permitted the determination of periodate at μg l−1 concentration levels. The results indicated that HS-SDME (range 0.01-10 mg l−1, r2 = 0.9990; limit of detection 1.55 μg l−1) was more sensitive than SDME (range 0.05-50 mg l−1, r2 = 0.9984; limit of detection 3.42 μg l−1), and was inexpensive, rapid and convenient. Tolerance of excess of iodate has permitted the application of this method in the determination of ethylene glycol in motor oil; the average recovery on spiked sample was 98.6% with R.S.D. of 4.2%.  相似文献

The electrochemical behavior of maltol on a glassy carbon (GC) electrode was investigated. The results were applied to differential pulse voltammetric determination of maltol in beverages pretreated by ultrafiltration. Under the optimum experimental conditions, the linear range is 1×10−5 to 6×10−4 mol l−1 maltol and the relative standard deviation for 0.4 mmol l−1 maltol is 0.6% (n=9). The detection limit was 5 μmol l−1. Furthermore, silica sol-gel film on GC electrode could be used as suitable selective membrane, which integrated selective membrane on the electrode and substituted for the pretreatment of ultrafiltration. Under the above conditions, maltol was determined by semi-differential linear sweep voltammetry at a silica sol-gel modified GC electrode in the concentration range of 5×10−6 to 5×10−4 mol l−1. The detection limit was 2 μmol l−1 and the relative standard deviation for 0.1 mmol l−1 maltol was 0.7% (n=7). The proposed method is of sensitivity, simplicity, rapidness and no contamination. It had been applied to the direct determination of maltol in beverages such as grape wines, drinks and beers without any pretreatment. The results obtained with the present method were satisfactory with those obtained by spectrophotometry. It could be used as a simple and practical method for the determination of the flavor enhancer maltol in beverages.  相似文献

A new method is proposed for the chemiluminescent determination of the pesticide 3-indolyl acetic acid by means of an flow injection analysis system. The chemiluminescence emission is obtained by oxidation of the analyte with Ce (IV) in nitric acid and presence of β-cyclodextrine.The continuous-flow method allows the determination of 159 samples h−1 of 3-indolyl acetic acid in an interval of concentrations over the range 0.5-15.0 mg l−1. The limit of detection was 0.1 μg l−1 and the R.S.D. (n, 17) at 2.0 mg l−1 of the pesticide level was 2.7%. The method was applied to water samples.  相似文献

A flow amperometric enzymatic biosensor for the determination of glucose was constructed. The biosensor consists of a flow reactor based on porous silver solid amalgam (AgSA) and a flow tubular detector based on compact AgSA. The preparation of the sensor and the determination of glucose occurred in three steps. First, a self-assembled monolayer of 11-mercaptoundecanoic acid (MUA) was formed at the porous surface of the reactor. Second, enzyme glucose oxidase (GOx) was covalently immobilized at MUA-layer using N-ethyl-N′-(3-dimethylaminopropyl) carboimide and N-hydroxysuccinimide chemistry. Finally, a decrease of oxygen concentration (directly proportional to the concentration of glucose) during enzymatic reaction was amperometrically measured on the tubular detector under flow injection conditions. The following parameters of glucose determination were optimized with respect to amperometric response: composition of the mobile phase, its concentration, the potential of detection and the flow rate. The calibration curve of glucose was linear in the concentration range of 0.02–0.80 mmol L−1 with detection limit of 0.01 mmol L−1. The content of glucose in the sample of honey was determined as 35.5 ± 1.0 mass % (number of the repeated measurements n = 7; standard deviation SD = 1.2%; relative standard deviation RSD = 3.2%) which corresponds well with the declared values. The tested biosensor proved good long-term stability (77% of the current response of glucose was retained after 35 days).  相似文献

Electrodeposition of Pt-Pb nanoparticles (PtPbNPs) to multi-walled carbon nanotubes (MWCNTs) resulted in a stable PtPbNP/MWCNT nanocomposite with high electrocatalytic activity to glucose oxidation in either neutral or alkaline medium. More importantly, the nanocomposite electrode with a slight modification exhibited high sensitivity, high selectivity, and low detection limit in amperometric glucose sensing at physiological neutral pH (poised at a negative potential). At +0.30 V in neutral solution, the nanocomposite electrode exhibited linearity up to 11 mM of glucose with a sensitivity of 17.8 μA cm−2 mM−1 and a detection limit of 1.8 μM (S/N = 3). Electroactive ascorbic acid (0.1 mM), uric acid (0.1 mM) and fructose (0.3 mM) invoked only 23%, 14% and 9%, respectively, of the current response obtained for 3 mM glucose. At −0.15 V in neutral solution, the electrode responded linearly to glucose up to 5 mM with a detection limit of 0.16 mM (S/N = 3) and detection sensitivity of ∼18 μA cm−2 mM−1. At this negative potential, ascorbic acid, uric acid, and fructose were not electroactive, therefore, not interfering with glucose sensing. Modification of the nanocomposite electrode with Nafion coating followed by electrodeposition of a second layer of PtPbNPs on the Nafion coated PtPbNP/MWCNT nanocomposite produced a glucose sensor (poised at −0.15 V) with a lower detection limit (7.0 μM at S/N = 3) and comparable sensitivity, selectivity and linearity compared to the PtPbNP/MWCNT nanocomposite. The Nafion coating lowered the detection limit by reducing the background noise, while the second layer of PtPbNPs restored the sensitivity to the level before Nafion coating.  相似文献

A method for determination of metabisulfite and hydrosulfite in poultice and decolorant by isotachophoresis was developed. Metabisulfite and hydrosulfite are ionizable oxoanions of sulfur of similar character that can easily be oxidized to sulfates. To protect the analytes from oxidation the solid samples were dissolved in a 1% (w/v) solution of formaldehyde. Hydrosulfite and metabisulfite present in the samples were transformed by the reaction with formaldehyde to stable compounds, hydroxymethanesulfinate and hydroxymethanesulfonate that were determined isotachophoretically without any pretreatment except for sample filtering and degassing. A capillary of 0.4 mm i.d. and 100 mm effective length made of fluorinated ethylene-propylene copolymer was filled with an electrolyte system consisting of 10 mmol L−1 HCl + 11 mmol L−1 imidazole, 0.15% (w/v) hydroxyethylcellulose, pH 6.0 (leading electrolyte) and 5 mmol L−1 benzoic acid + 6 mmol L−1 imidazole, pH 6.5 (terminating electrolyte). Separation was performed at a driving current of 80 μA and for detection current was decreased to 30 μA. Using contactless conductivity detection, the calibration curves in the tested concentration range up to 2.5 mmol L−1 were linear for both metabisulfite and hydrosulfite complexes. The concentration detection limits for metabisulfite and hydrosulfite were 2.9 and 3.4 μmol L−1, respectively. For 1 mmol L−1 concentration, values of R.S.D. (n = 6) were 2.6% for hydrosulfite and 0.8% for metabisulfite. Isotachophoretic determination took about 20 min. The elaborated isotachophoretic procedure is simple to perform, sufficiently sensitive and accurate. In addition to this, low cost of analyses makes the method an alternative procedure to methods used so far for the determination of oxoanions of sulfur.  相似文献

In this paper, the formation of gold nanoparticles (Au NPs) as a result of the thermo-active redox reaction of chlorauric acid (HAuCl4) and glucose in alkaline medium was identified by measuring the plasmon resonance absorption, localized surface plasmon resonance (LSPR), and transmission electron microscopy (TEM) images, for the formation of Au NPs displays characteristic plasmon resonance absorption bands and corresponding LSPR signals. It was found that the resulted LSPR signals could be easily detected with a common spectrofluorometer. With increasing glucose concentration, the LSPR intensity displays linear response with the glucose content over the range from 2.0 to 250.0 μmol l−1. Thus, a novel assay of glucose was established with the limits of determination (3σ) being 0.21 μmol l−1, and the detection of glucose could be made easily in the serum samples of diabetes sufferers. Mechanism investigations showed that the activation energy and molar ratio of the reaction were 34.8 kJ mol−1 and 3:2, respectively.  相似文献

A new rapid and sensitive FI assay is reported for the simultaneous direct spectrophotometric determination of trace Cr(VI) and Cr(III) in real samples. The method is based upon the reaction of Cr(VI) with chromotropic acid (CA) in highly acidic medium to form a water-soluble complex (λmax = 370 nm). Cr(III) reacts with CA only after its on-line oxidation to Cr(VI) by alkaline KIO4. The determination of each chromium species in the sample was achieved by absorbance differences. The calibration curves were linear over the range 3-4000 μg l−1 and 30-1200 μg l−1 for Cr(VI) and Cr(III), respectively, while the precision close to the quantitation limit was satisfactory in both cases (sr = 3.0% for Cr(VI) and 4.0% for Cr(III) (n = 10) at 10 and 50 μg l−1 level, respectively). The method developed proved to be adequately selective and sensitive (cL = 1 and 10 μg l−1 for Cr(VI) and Cr(III), respectively). The application of the method to the analysis of water samples (tap and mineral water) gave accurate results based on recovery studies (93-106%). Analytical results of real sample analysis were in good agreement with certified values.  相似文献

A highly sensitive flow-injection (FI) method with chemiluminescence (CL) detection is used for the determination of l-ascorbic acid. The method is based on the CL reaction of Rhodamine B with cerium(IV) in sulfuric acid media. l-Ascorbic acid is suggested to be a catalyst utilized in the energy-transferred excitation process. The proposed procedure allows quantitation of l-ascorbic acid in the range 3.8×10−13 to 1.0×10−10 mol l−1 with a correlation coefficient of 0.9998 (n=5) and relative standard deviation (R.S.D.) of 0.92% (n=11) at 1.0×10−11 mol l−1. The detection limit (3×blank) was 1.0×10−13 mol l−1. The method is successfully used to determine l-ascorbic acid in fresh vegetables. The possible mechanism of the chemiluminescence in the system is discussed.  相似文献

Nickel hydroxide nanoparticles were successfully electrodeposited on graphite electrode (Gr/NiONP) and employed as a robust non-enzymatic glucose sensor. The results of cyclic voltammetry (CV) and chronoamperometry demonstrated that the Gr/NiONP electrode displayed high electrocatalytic activity toward glucose. The oxidation current is directly related to the glucose concentration from 1 μM to 15 mM. Besides, the glucose sensor displayed high sensitivity (2400 μA mM−1 cm−2) with a detection limit of 0.53 μM (S/N = 3) in basic solution. Moreover, the sensor showed excellent selectivity, reproducibility and stability properties. The relative standard deviation is 1.2% for 10 successive measurements in 16 μM glucose. Interestingly, the signal for glucose was maintained at 95% of its initial value even after 6 months of storage under ambient conditions. Gr/NiONP electrode has also been tested to detect glucose in human serum with satisfactory results.  相似文献

This work reports a sequential-injection analysis (SIA) method for the enzymatic assay of glucose with soluble glucose oxidase (GOD) and on-line sample dilution with chemiluminescence (CL) detection. A zone of sample was aspirated in the holding coil of the SIA manifold and, if necessary, was diluted on-line by means of an auxiliary dilution conduit. Then, a zone of GOD was aspirated adjacent to the sample zone and a stopped-flow period was applied to allow the enzymatic reaction to proceed with production of hydrogen peroxide. Then, zones of a catalyst (Co(II) solution) and alkaline luminol were aspirated into the holding coil. Finally, the flow was reversed and the stacked zones were sent to a flow-cell located in front of a photomultiplier tube (PMT) that monitored the CL intensity. The linear dynamic range was 1 × 10−5-1 × 10−3 mol L−1 glucose, the coefficient of variation at 8 × 10−5 mol L−1 of glucose was sr = 3.1% (n = 8), the limit of detection at the 3σ level was cL = 1 × 10−6 mol L−1 and the sampling frequency was 28 h−1. With on-line dilution by a factor of 1/200, the linear range could be extended up to 0.2 mol L−1 glucose. The advantages of the proposed method are the simple manifold and instrumentation used, the scope for automated on-line dilution, the low consumption of sample and reagents and the elimination of enzyme immobilisation procedures. The method was applied to the analysis of commercial drinks and honey with percent relative errors in glucose determination in the range 100 ± 6.1%.  相似文献

The determination of glucose in beverages is demonstrated using newly developed fluoride selective optical sensing polymeric film that contains aluminum (III) octaethylporphyrin (Al[OEP]) ionophore and the chromoionophore ETH7075 cast at the bottom of wells of a 96-well polypropylene microtiter plate. The method uses a dual enzymatic reaction involving glucose oxidase enzyme (GOD) and horseradish peroxidase (HRP), along with an organofluoro-substrate (4-fluorophenol) as the source of fluoride ions. The concentration of fluoride ions after enzymatic reaction is directly proportional to the glucose level in the sample. The method has a detection limit of 0.8 mmol L−1, a linear range of 0.9-40 mmol L−1 and a sensitivity of 0.125 absorbance/decade of glucose concentration. Glucose levels in several beverage samples determined using the proposed method correlate well with a reference spectrophotometric enzyme method based on detection of hydrogen peroxide using bromopyrogallol red dye (BPR). The new method can also be used to determine H2O2 concentrations in the 0.1-50 mmol L−1 range using a single enzymatic reaction involving H2O2 oxidation of 4-fluorophenol catalyzed by HRP. The methodology could potentially be used to detect a wide range of substrates for which selective oxidase enzymes exist (to generate H2O2), with the high throughput of simple microtiter plate detection scheme.  相似文献
