A new approach to constructing an enzyme-containing film on the surface of a gold electrode for use as a biosensor is described. A basic multilayer film (BMF) of (PDDA/GNPs) n /PDDA was first constructed on the gold electrode by electrostatic layer-by-layer self-assembly of poly(diallyldimethylammonium chloride) (PDDA) and gold nanoparticles (GNPs). Glucose oxidase (GOx) was then sorbed into this BMF by dipping the BMF-modified electrode into a GOx solution. The assembly of the BMF was monitored and tested via UV-vis spectroscopy and cyclic voltammetry (CV). The ferrocenemethanol-mediated cyclic voltammograms obtained from the gold electrode modified with the (PDDA/GNPs) n /PDDA/GOx indicated that the assembled GOx remained electrocatalytically active for the oxidation of glucose. Analysis of the voltammetric signals showed that the surface coverage of active enzyme was a linear function of the number of PDDA/GNPs bilayers. This result confirmed the penetration of GOx into the BMF and suggests that the BMF-based enzyme film forms in a uniform manner. Electrochemical impedance measurements revealed that the biosensor had a lower electron transfer resistance (R et) than that of a sensor prepared by layer-by-layer assembly of PDDA and GOx, due to the presence of gold nanoparticles. The sensitivity of the biosensor for the determination of glucose, which could be controlled by adjusting the number of PDDA/GNPs bilayers, was investigated.  相似文献

We studied the electrochemical properties of gold nanoparticles (GNPs) and their complexes with proteins using square-wave voltammetry. Effect of the nanoparticle size and detection procedure was explored upon the oxidation of GNPs on a glassy carbon electrode (GCE). For pre-characterized GNPs of 13, 35 and 78 nm diameter, the oxidation peak potential was + 0.98, + 1.03 and + 1.06 V vs. Ag/AgCl, respectively. The conjugation of GNPs with four different proteins was verified by UV–Vis spectroscopy and atomic force microscopy indicated the formation of protein shells around GNPs. This process hampered the oxidation of GNPs on bare GCE causing pronounced decrease in the current response by an average factor of 72. GCE modification with carbon nanotubes weakly influenced the sensitivity of GNP detection but resulted in a 14.5-fold signal increase averaged for all GNP–protein complexes. The acidic dissolution and electrodeposition of GNPs or their complexes adsorbed on GCE allowed superior signal amplification directly proportional to nanoparticle size. The results are useful for the optimization of voltammetric analysis of GNP–protein complexes and can be extended to the characterization of other metal nanostructures and their complexes with biological components.  相似文献

The separation of three kinds of aminophenol isomers were achieved within 1 min in polyelectrolytes multilayers modified PDMS microchips by layer-by-layer assembly with electrochemical detection (EC). Two polyelectrolytes, poly(dially dimethyl ammonium chloride) (PDDA) and poly(sodium-4-styrene-sulfonate) (PSS) were used to form polyelectrolyte multilayers (PEMs). The surface characteristic of the modified microchip was studied by XPS. The electroosmotic flow (EOF) on PEMs modified PDMS microchips was more stable than that of the native PDMS microchips and the adsorption of samples was greatly reduced on PEMs modified PDMS microchips during the electrophoretic process. The column efficiencies on PEMs modified microchip were increased by 100 times and the signals enhanced by 2 times compared with those of native microchips. The separation conditions such as running buffer pH, running buffer concentration and separation voltage were also optimized.  相似文献

In this study, we describe the use of Tween 20‐capped gold nanoparticles (AuNPs) as selective probes for the extraction of aminothiols from an aqueous solution. Tween 20 molecules noncovalently attached to the surface of AuNPs to form Tween 20–AuNPs were used for the selective extraction of aminothiols through the formation of Au–S bonds. After extraction and centrifugation, the aminothiols were detached from the surface of the AuNPs by adding DTT in a high concentration. We used this probe in combination with CE and UV absorption detection. On‐line concentration and separation of the released aminothiols were performed by using 1.6% v/v poly(diallyldimethylammonium chloride) as an additive in CE. Under optimal extraction and stacking conditions, the LOD at a S/N of 3 were 28, 554, and 456 nM for glutathione (GSH), cysteine (Cys), and homocysteine (HCys), respectively. In comparison with the normal injection without the extraction procedure, approximately 2280‐, 998‐, and 904‐fold improvements in the sensitivity were observed for GSH, Cys, and HCys, respectively. We have validated the application of our method on the basis of the analysis of GSH and HCys in human urine samples. It is believed that this approach has significant potential to be extended to clinical diagnosis.  相似文献

Uniform and stable polymer protected spherical gold nanoparticles were synthesized using glycerol as reducing agent. Further it was observed that the morphology of the particles varied from spherical to triangular prismatic gold nanoparticles when the reaction was changed from normal mode of reflux condition to microwave mode (MW) of heating. Further a brief mechanism relating the formation of prisms with the orientation of polymer and nucleation period has been discussed. Formation of triangular prismatic and spherical gold nanoparticles were characterised using UV–vis spectroscopy, transmission electron microscopy (TEM) and X-ray diffraction (XRD) analysis.  相似文献

Phosphomolybdate, H3PMo12O40, (PMo12)-doped-poly(3,4-ethylenedioxythiophene) (PEDOT) coated gold nanoparticles have been synthesized in aqueous solution by reduction of AuCl4 using hydroxymethyl EDOT as a reducing agent in the presence of polystyrene sulfonate and PMo12. The resulting PMo12-doped-PEDOT stabilized Au nanoparticles are water soluble and have been characterized by UV–visible spectroscopy, scanning electron microscopy and electrochemistry. Glassy carbon electrodes modified with these Au nanoparticles show excellent stability and catalytic activity towards the reduction of bromate in an aqueous electrolyte solution containing 10 mM H2SO4 and 0.1 M Na2SO4.  相似文献

We are reporting a novel green approach to incorporate silver nanoparticles (NPs) selectively in the polyelectrolyte capsule shell for remote opening of polyelectrolyte capsules. This approach involves in situ reduction of silver nitrate to silver NPs using PEG as a reducing agent (polyol reduction method). These nanostructured capsules were prepared via layer by layer (LbL) assembly of poly(allylamine hydrochloride) (PAH) and dextran sulfate (DS) on silica template followed by the synthesis of silver NPs and subsequently the dissolution of the silica core. The size of silver nanoparticles synthesized was 60±20 nm which increased to 100±20 nm when the concentration of AgNO(3) increased from 25 mM to 50 mM. The incorporated silver NPs induced rupture and deformation of the capsules under laser irradiation. This method has advantages over other conventional methods involving chemical agents that are associated with cytotoxicity in biological applications such as drug delivery and catalysis.  相似文献

2,3,5,6-Tetrakis-(morpholinomethyl)hydroquinone (1) is used for the first time in the preparation of gold nanoparticles by the reduction of HAuCl4 in water–methanol medium without using any capping agent. Compound 1 was prepared by Mannich-type aminomethylation of hydroquinone with morpholine. It is characterized by elemental analysis, FT-IR, UV–Vis and mass spectra and finally by single crystal X-ray diffraction. The ratio of HAuCl4 and compound 1 played a vital role in controlling the shape and size of gold nanoparticles. The samples were characterized by Transmission Electron Microscopy (TEM), XRD, FT-IR, UV–Vis measurements. With the increasing amount of gold(III) solution with respect to compound 1, two different morphologies such as self-assembled and spherical gold nanoparticles have been observed. The results indicate that the morphology of gold nanoparticles with different sizes can be controlled by changing the concentrations of compound 1 and gold(III) solution.  相似文献

An electroosmotic flow (EOF)-switchable poly(dimethylsiloxane) (PDMS) microfluidic channel modified with cysteine has been developed. The native PDMS channel was coated with poly(diallyldimethylammonium chloride) (PDDA), and then gold nanoparticles by layer-by-layer technique was assembled on PDDA to immobilize cysteine. The assembly was followed by infrared spectroscopy/attenuated total reflection method, contact angle, EOF measurements and electrophoretic separation methods. EOF of this channel can be reversibly switched by varying the pH of running buffer. At low pH, the surface of channels is positively charged, EOF is from cathode to anode. At high pH, the surface is negatively charged, EOF is from anode to cathode. At pH 5.0, near the isoelectric point of the chemisorbed cysteine, the surfaces of channels show neutral. When pH is above 6.0 or below 4.0, the magnitude of EOF varies in a narrow range. And the modified channel surface displayed high reproducibility and good stability, a good reversibility of cathodic-anodic EOF transition under the different pH conditions was observed. Separation of dopamine and epinephrine as well as arginine and histidine were performed on the modified chip.  相似文献

Poly(dimethylsiloxane) (PDMS) microfluidic channels modified by citrate-stabilized gold nanoparticles after coating a layer of linear polyethylenimine (LPEI) were successfully used to separate dopamine and epinephrine, which were difficult to be separated from baseline in native and hybrid PDMS microchannels. In-channel amperometric detection with a single carbon fibre cylindrical electrode was employed. Experimental parameters of separation and detection processes were optimized in detail. The analytes were well separated within 100 s in a 3.7 cm long separation channel at a separation voltage of +800 V using a 30 mM phosphate buffer solution (PBS, pH 7.0). Linear responses of them were obtained both from 25 to 600 μM with detection limits of 2 μM for dopamine and 5 μM for epinephrine, respectively. The modified PDMS channels have a long-term stability and an excellent reproducibility within 2 weeks.  相似文献

In the current study, gold nanoparticles (AuC6NPs and AuC8NPs) were prepared through sodium borohydride reduction method by using Calix[6]rene and Calix[8]rene as a stabilizing agents. The synthesized AuNPs were screened for cytotoxic, phytotoxic, antifungal and antibacterial activities. The fabricated AuNPs were characterized by UV–visible spectroscopy, atomic force microscopy (AFM) and FTIR spectroscopy. Antibacterial activities of the AuNPs were tested against E. coli and S. aureus. The AuC6NPs were found to be effective against the growth of gram positive bacteria and inhibited the growth of S. aureus. AuC6NPs interact with bacterial cell and damaged cell membrane. Roughness of the bacterial surface and membrane rupture can be clearly observed by AFM images. The AuNPs possess insignificant antifungal activity against Aspergillus niger and Candida albicans. Moreover, AuC8NPs have significant phytotoxicity and moderate cytotoxicity.  相似文献

A novel film of chitosan-gold nanoparticles is fabricated by a direct and facile electrochemical deposition method and its application in glucose biosensor is investigated. HAuCl(4) solution is mixed with chitosan and electrochemically reduced to gold nanoparticles, which can be stabilized by chitosan and electrodeposited onto glassy carbon electrode surfaces along with the electrodeposition of chitosan. Then a model enzyme, glucose oxidase (GOD) is immobilized onto the resulting film to construct a glucose biosensor through self-assembly. The resulting modified electrode surfaces are characterized with both AFM and cyclic voltammetry. Effects of chitosan and HAuCl(4) concentration in the mixture together with the deposition time and the applied voltage on the amperometric response of the biosensor are also investigated. The linear range of the glucose biosensor is from 5.0 x 10(-5) approximately 1.30 x 10(-3) M with a Michaelis-Menten constant of 3.5 mM and a detection limit of about 13 microM.  相似文献

This work is focused on the synthesis of innovative hybrids made by linking gold nanoparticles to protected organometallic Pd(II) thiolate. The organometallic protected Pd(II) thiolate, i.e. trans-thioacetate-ethynylphenyl-bis(tributylphosphine)palladium(II) has been synthesized, in situ deprotected and linked to Au nanoparticles. In this way new hybrid, with a direct link between Pd(II) and Au nanoparticles through a single S bridge, has been isolated. The combination of the organometallic Pd(II) thiol with gold nanoparticles allows the enhancement and tailoring of electronic and optical properties of the new organic-inorganic nano-compound. Single-crystal gold nanoparticles, uniform in shape and size were obtained by applying a modified two-phase method (improved Brust-Schiffrin reaction). In addition, the chemical environment of the Au nanoparticles was investigated and a covalent bonding between Au nanoparticles and the organometallic thiols was observed.  相似文献

We report on the preparation of a new class of polymer hydrogels obtained through the chemical crosslinking reaction of poly(vinyl alcohol) (PVA) and functionalized gold nanoparticles. Carboxylic group functionalized gold nanoparticles were synthesized, dispersed in a PVA matrix and allow to react with the hydroxyl groups of PVA at high temperature. FT-IR and swelling experiments carried out on the cross-linked samples confirmed that the crosslinking reaction took place. This is the first time, to our knowledge, that functionalized nanoparticles are used as chemical crosslinking agents.  相似文献

Crystallization of high density polyethylene (PE) from the melt on model atomically flat solid surfaces decorated with micro- and nanoparticles of gold or NaCl of different size and densities is investigated. The morphology of the contact layer of PE after its detachment from the support is studied using atomic force microscopy (AFM). It is shown that the nucleating and ordering effect of the solid on PE crystallization depends to a large extend on the nanostructure of its surface, in particular on the size of the atomically flat domains and on the presence of nanoscopic obstacles. The minimum size of the flat domain which can significantly influence the PE crystallization is estimated to be of the order of 150 nm.  相似文献

The interest in applying gold nanoparticles is principally based on their monodispersity. Therefore, the synthesis of monodisperse gold nanocrystals has a key role in designing nanomaterials for their specific application. However, it can represent a cutting edge for many laboratories, mainly because it requires a variety of complex techniques for properly characterizing metal nanoparticles with expensive and rugged equipment. Herein, we propose an approach to synthesize nanoparticles with controlled monodispersity that can be adjusted through the factorial experimental plan 32 using response surface modeling (RSM). The particles were characterized on the basis of visual observation, UV–visible spectroscopy, transmission electron microscopy, scanning electron microscopy coupled with energy dispersive X-ray spectroscopy (SEM/EDS) and statistics. Our findings illustrate how factorial design is important to providing accurate projections of nanoparticle production using sodium citrate and borohydride as reducers. The proposed method leverages facile and inexpensive routes to synthesize monodisperse gold nanocrystals without the assistance of time-consuming image-based characterization tools.  相似文献

Glucose oxidase showed direct electrochemical transfer at glassy carbon electrodes immobilized with carbon nanotube‐gold colloid (CNT‐Au) composites with poly(diallydimethylammonium chloride) (PDDA) coatings. The modified electrode (GC/CNT/Au/PDDA‐GOD) was employed for the amperometric determination of glucose. Under optimal conditions, the biosensor displayed linear response to glucose from 0.5 to 5 mM with a sensitivity of 2.50 mA M?1 at an applied potential of ?0.3 V (vs. Ag|AgCl reference).  相似文献

The objective of this work was to investigate the changes in surface morphology associated with thermal degradation of poly(p-phenylene terephthalamide) (PPTA) into chars. To this end, PPTA samples decomposed at several temperatures up to 800 °C were studied on a local scale using atomic force microscopy (AFM) and scanning tunnelling microscopy (STM). Domains with a diameter of 40-50 nm started appearing among PPTA nanofibrils at about 500 °C. At this temperature and above, a film coating the fibre developed. This layer was much less rigid than PPTA, and remained deposited on the fibres, even at high temperatures. At 800 °C, the STM images showed a surface distribution typical of a carbonaceous material, isotropic although somewhat heterogeneous. When an intermediate isothermal step (500 °C, 200 min) was introduced along with heat treatment of PPTA under a constant rate, the material obtained at the end of this step was conductive enough to be studied by STM. Although the coating over the fibres also remained after the isothermal step, it was less homogeneous than in the absence of isothermal treatment. On further heating, the residue exhibited a surface morphology typical of a carbonaceous material, but much more homogeneous and isotropic than in the absence of the isothermal step.  相似文献

The adsorption-desorption of silica nanoparticles(NPs) on poly(ethylene glycol)(PEG) grafted onto gold(Au) substrate was studied by quartz crystal microbalance with dissipation monitoring(QCM-D) technique. The results of frequency and dissipation show that SiO2 NPs can be adsorbed strongly on PEG-SH brushes at pH of 9.6, and a new dense and rigid construction is formed. Adjusting the pH from 9.6 to 12.3 resulted in the desorption of silica NPs from the PEG brushes because of a significant weakening of the hydrogen bond between the silica NPs and PEG chains. In addition, the viscoelastic properties of the system during the adsorption-desorption process were also analyzed via the relationship between the normalized frequency(Δf/n) and mass. And the corresponding atomic force microscopy(AFM) images also exhibit morphological changes during the above process, consistent with the changes in viscoelasticity.  相似文献
