A simple, inexpensive and reagent-less colorimetric micro flow analysis (μFA) system was implemented in a polymethyl methacrylate (PMMA) micro fluidic manifold. A T-shaped micro channel on a PMMA chip was fabricated by laser ablation and topped with molded polydimethylsiloxane (PDMS). The fabricated μFA system was integrated with the optical components as detector and applied to the determination of iron in water samples. It is based on the measurement of Fe(III)-nitroso-R salt complex at 720 nm formed by the reaction between Fe(III) and nitroso-R salt in an acetate buffer solution pH 5. The proposed μFA consumed very small amount of reagent and sample, it released waste of less than 2.0 mL h−1. The relative standard deviation (R.S.D.) was less than 2% (n = 11) with the recovery of 98.7 ± 0.12 (n = 5). The linear range for the determination of iron in water samples was over the range of 0.05-4.0 μg mL−1 with a correlation coefficient (r2) of 0.9994. The limit of detection (3σ) and limit of quantitation (10σ) were 0.021 μg mL−1 and 0.081 μg mL−1, respectively with a sample throughput of 40 h−1.  相似文献

A cost-effective sequential injection system incorporating with an in-line UV digestion for breakdown of organic matter prior to voltammetric determination of Zn(II), Cd(II), Pb(II) and Cu(II) by anodic stripping voltammetry (ASV) on a hanging mercury drop electrode (HMDE) of a small scale voltammetric cell was developed. A low-cost small scale voltammetric cell was fabricated from disposable pipet tip and microcentrifuge tube with volume of about 3 mL for conveniently incorporated with the SI system. A home-made UV digestion unit was fabricated employing a small size and low wattage UV lamps and flow reactor made from PTFE tubing coiled around the UV lamp. An in-line single standard calibration or a standard addition procedure was developed employing a monosegmented flow technique. Performance of the proposed system was tested for in-line digestion of model water samples containing metal ions and some organic ligands such as strong organic ligand (EDTA) or intermediate organic ligand (humic acid). The wet acid digestion method (USEPA 3010a) was used as a standard digestion method for comparison. Under the optimum conditions, with deposition time of 180 s, linear calibration graphs in range of 10-300 μg L−1 Zn(II), 5-200 μg L−1 Cd(II), 10-200 μg L−1 Pb(II), 20-400 μg L−1 Cu(II) were obtained with detection limit of 3.6, 0.1, 0.7 and 4.3 μg L−1, respectively. Relative standard deviation were 4.2, 2.6, 3.1 and 4.7% for seven replicate analyses of 27 μg L−1 Zn(II), 13 μg L−1 Cd(II), 13 μg L−1 Pb(II) and 27 μg L−1 Cu(II), respectively. The system was validated by certified reference material of trace metals in natural water (SRM 1640 NIST). The developed system was successfully applied for speciation of Cd(II) Pb(II) and Cu(II) in ground water samples collected from nearby zinc mining area.  相似文献

An analytical procedure with improved sensitivity was developed for cyanide determination in natural waters, exploiting the reaction with the complex of Cu(I) with 2,2′-biquinoline 4,4′-dicarboxylic acid (BCA). The flow system was based on the multi-pumping approach and long pathlength spectrophotometry with a flow cell based on a Teflon AF 2400® liquid core waveguide was exploited to increase sensitivity. A linear response was achieved from 5 to 200 μg L−1, with coefficient of variation of 1.5% (n = 10). The detection limit and the sampling rate were 2 μg L−1 (99.7% confidence level), and 22 h−1, respectively. Per determination, 48 ng of Cu(II), 5 μg of ascorbic acid and 0.9 μg of BCA were consumed. As high as 100 mg L−1 thiocyanate, nitrite or sulfite did not affect cyanide determination. Sulfide did not interfere at concentrations lower than 40 and 200 μg L−1 before or after sample pretreatment with hydrogen peroxide. The results for natural waters samples agreed with those obtained by a fluorimetric flow-based procedure at the 95% confidence level. The proposed procedure is then a reliable, fast and environmentally friendly alternative for cyanide determination in natural waters.  相似文献

A simple, precise, accurate and validated, acetonitrile-free, reverse phase high performance liquid chromatography (HPLC) method is developed for the determination of melamine in dry and liquid infant formula. The separation is performed on a Kromasil C18 column (150 mm × 3.2 mm I.D., 5 μm particle size) at room temperature. The mobile phase (0.1% TFA/methanol 90:10) is pumped at a flow rate of 0.3 mL min−1 with detection at 240 nm. Melamine elutes at 3.7 min. A linear response (r > 0.999) is observed for samples ranging from 1.0 to 80 μg mL−1. The method provides recoveries of 97.2-101.2% in the concentration range of 5-40 μg mL−1, intra- and inter-day variation in <1.0% R.S.D. The limit of detection (LOD) and limit of quantification (LOQ) values are 0.1 μg mL−1 and 0.2 μg mL−1, respectively.  相似文献

A new method based on microcolumn packed with ionic liquid-modified silica combined with flame atomic absorption spectrometry has been developed for the determination of lead in environmental samples. Several factors influencing the preconcentration efficiency of lead and its subsequent determination, such as pH of the sample, flow rate, mass of ionic liquid, and interfering effect, have been investigated. Lead could be quantitatively retained by ionic liquid-modified silica in the pH range of 5-7, and then eluted completely with 3.0 mL 1.0 mol L−1 HCl. The detection limit of this method for lead was 0.7 μg L−1 with preconcentration factor of 185, and the relative standard deviation (RSD) was 4.2% at 0.1 μg mL−1 Pb(II). This method has been applied for the determination of trace amount of lead in NIST standard reference material 2709 (San Joaquin Soil) and river water samples with satisfactory results.  相似文献

A highly sensitive determination of mercury in the presence of Cu(II) using a boron-doped diamond (BDD) thin film electrode coupled with sequential injection–anodic stripping voltammetry (SI–ASV) was proposed. The Cu(II) was simultaneously deposited with Hg(II) in a 0.5 M HCl supporting electrolyte by electrodeposition. In presence of an excess of Cu(II), the sensitivity for the determination of Hg(II) was remarkably enhanced. Cu(II) and Hg(II) were on-line deposited onto the BDD electrode surface at −1.0 V (vs. Ag/AgCl, 3 M KCl) for 150 s with a flow rate of 14 μL s−1. An anodic stripping voltammogram was recorded from −0.4 V to 0.25 V using a frequency of 60 Hz, an amplitude of 50 mV, and a step potential of 10 mV at a stopped flow. Under the optimal conditions, well-defined peaks of Cu(II) and Hg(II) were found at −0.25 V and +0.05 V (vs. Ag/AgCl, 3 M KCl), respectively. The detection of Hg(II) showed two linear dynamic ranges (0.1–30.0 ng mL−1 and 5.0–60.0 ng mL−1). The limit of detection (S/N = 3) obtained from the experiment was found to be 0.04 ng mL−1. The precision values for 10 replicate determinations were 1.1, 2.1 and 2.9% RSD for 0.5, 10 and 20 ng mL−1, respectively. The proposed method has been successfully applied for the determination of Hg(II) in seawater, salmon, squid, cockle and seaweed samples. A comparison between the proposed method and an inductively coupled plasma optical emission spectrometry (ICP-OES) standard method was performed on the samples, and the concentrations obtained via both methods were in agreement with the certified values of Hg(II), according to the paired t-test at a 95% confidence level.  相似文献

A novel method was developed for the determination of amiodarone and desethylamiodarone by high-performance liquid chromatography (HPLC) coupled with chemiluminescent (CL) detection. The procedure is based on the post-column photolysis of the analytes into photoproducts which are active in the tris(2,2′-bipyridyl)ruthenium(III) [Ru(bpy)33+] CL system. Ru(bpy)33+ was on-line generated by photo-oxidation of the Ru(II) complex in the presence of peroxydisulfate. The separation was carried out on a Mediterranea C18 column with isocratic elution using a mixture of methanol and 0.017 mol L−1 ammonium sulfate buffer of pH 6.8. Under the optimum conditions, analytical curves, based on standard solutions, were linear over the range 0.1-50 μg mL−1 for amiodarone and 0.5-25 μg mL−1 for desethylamiodarone. The detection limits of amiodarone and desethylamiodarone were 0.02 and 0.11 μg mL−1, respectively. Intra- and inter-day precision values of 0.9% relative standard deviation (R.S.D.) (n = 10) and 1.6% R.S.D. (n = 15), respectively, were obtained. The method was applied successfully to the determination of these compounds in serum and pharmaceutical formulations.  相似文献

In this paper, the conversion of azoxystrobin in a strongly fluorescent degradation product by UV irradiation with quantitative purposes and its fluorimetric determination are reported for the first time. A multicommuted flow injection-solid phase spectroscopy (FI-SPS) system combined with photochemically-induced fluorescence (PIF) is developed for the determination of azoxystrobin in grapes, must and wine. Grape samples were homogenized and extracted with methanol and further cleaned-up by solid-phase extraction on C18 silica gel. Wine samples were solid-phase extracted on C18 sorbent using dichloromethane as eluent. Recoveries of azoxystrobin from spiked grapes (0.5-2.0 mg Kg−1), must (0.5-2.0 μg mL−1) and wine (0.5-2.0 μg mL−1) were 84.0-87.6%, 95.5-105.9% and 88.5-111.2%, respectively. The quantification limit for grapes was 0.021 mg Kg−1, being within European Union regulations, and 18 μg L−1 and 8 μg L−1 for must and wine, respectively.  相似文献

Hollow fibre liquid-phase microextraction with in situ derivatization using dansyl chloride has been successfully developed for the high-performance liquid chromatography-ultraviolet (HPLC-UV) determination of the biogenic amines (tryptamine, putrescine, cadaverine, histamine, tyramine, spermidine) in food samples. Parameters affecting the performance of the in situ derivatization process such as type of extraction solvent, temperature, extraction time, stirring speed and salt addition were studied and optimized. Under the optimized conditions (extraction solvent, dihexyl ether; acceptor phase, 0.1 M HCl; extraction time, 30 min; extraction temperature, 26 °C; without addition of salt), enrichment factors varying from 47 to 456 were achieved. Good linearity of the analytes was obtained over a concentration range of 0.1–5 μg mL−1 (with correlation coefficients of 0.9901–0.9974). The limits of detection and quantification based on a signal-to-noise ratio of 3–10, ranged from 0.0075 to 0.030 μg mL−1 and 0.03 to 0.10 μg mL−1, respectively. The relative standard deviations based on the peak areas for six replicate analysis of water spiked with 0.5 μg mL−1 of each biogenic amine were lower than 7.5%. The method was successfully applied to shrimp sauce and tomato ketchup samples, offering an interesting alternative to liquid–liquid extraction and solid phase extraction for the analysis of biogenic amines in food samples.  相似文献

Four simple, rapid, accurate, precise, reliable and economical spectrophotometric methods have been proposed for simultaneous determination of salbutamol sulphate (SS), bromhexine hydrochloride (BH) and etofylline (ET) in pure and commercial formulations without any prior separation or purification. They were first derivative zero crossing spectrophotometry (method 1), simultaneous equation method (method 2), derivative ratio spectra zero crossing method (method 3) and double divisor ratio spectra derivative method (method 4). The ranges for SS, BH and ET were found to be 1-35 μg mL−1, 4-40 μg mL−1 and 5-80 μg mL−1. For methods 1 and 2, the values of limit of detection (LOD) were 0.2314 μg mL−1, 0.4865 μg mL−1 and 0.2766 μg mL−1 and the values of limit of quantitation (LOQ) were 0.7712 μg mL−1, 1.6217 μg mL−1 and 0.9221 μg mL−1 for SS, BH and ET, respectively. For method 3, LOD values were 0.3297 μg mL−1, 0.2784 μg mL−1 and 0.7906 μg mL−1 and LOQ values were 0.9325 μg mL−1, 0.9282 μg mL−1 and 2.6352 μg mL−1 for SS, BH and ET, respectively. For method 4, LOD values were 0.3161 μg mL−1, 0.2495 μg mL−1 and 0.2064 μg mL−1 and LOQ values were 0.9869 μg mL−1, 0.8317 μg mL−1 and 0.6879 μg mL−1 for SS, BH and ET. The precision values were less then 2% R.S.D. for all four methods. The common excipients and additives did not interfere in their determinations. The results obtained by the proposed methods have been statistically compared by means of Student t-test and by the variance ratio F-test.  相似文献

A simple and precise square-wave adsorptive cathodic stripping voltammetry (SW-AdCSV) method has been described for simultaneous determination of Mn(II), Cu(II) and Fe(III) in water samples using a carbon paste electrode. In 0.1 mol L−1 acetate buffer (pH 5) containing 50 μmol L−1 of 2-(5′-bromo-2′-pyridylazo)-5-diethylaminophenol (5-Br-PADAP), Mn(II), Cu(II) and Fe(III) were simultaneously determined as metal-complexes with 5-Br-PADAP following preconcentration onto the carbon paste electrode by adsorptive accumulation at +1.0 V (vs. Ag/AgCl/3 M KCl). Insignificant interference from various cations (K+, Na+, Mg2+, Ca2+, Al3+, Bi3+, Sb3+, Se4+, Zn2+, Ni2+, Co2+, Cd2+, Pb2+, V5+, Ti4+ and NH4+), anions (HCO3, Cl, NO3−, SO42− and PO43−) and ascorbic acid was noticed. Limits of detection of 0.066, 0.108 and 0.093 μg L−1 and limits of quantitation of 0.22, 0.36 and 0.31 μg L−1 Mn(II), Cu(II) and Fe(III), respectively, were achieved by the described method. The described stripping voltammetry method was successfully applied for simultaneous determination of Mn(II), Cu(II) and Fe(III) in ground, tap and bottled natural water samples.  相似文献

In pH 2.8-3.8 BR buffer medium, the third generation cephalosporin antibiotics (TGCs) such as ceftazidime (CZD), ceftriaxone (CTRX), cefoperazone (CPZ), and cefotaxime (CFTM) react with palladium(II) (Pd(II)) to form 1:2 yellowish-brown cationic chelates, which further react with 4, 5-dibromofluorescein (DBF) to form 1:3 brown ion-association complexes. As a result, not only the spectra of absorption and fluorescence are changed, but also the resonance Rayleigh scattering (RRS) is enhanced greatly and the new RRS spectra are observed. The four TGCs products have similar spectral characteristics and their maximum RRS wavelengths are all located at 291 nm. The quantitative determination ranges and the detection limits of the four TGCs are 0.0065-1.0 μg mL−1 and 2.0 ng mL−1 for CZD, 0.0070-1.1 μg mL−1 and 2.2 ng mL−1 for CTRX, 0.0090-1.6 μg mL−1 and 2.7 ng mL−1 for CPZ, and 0.014-2.2 μg mL−1 and 4.2 ng mL−1 for CFTM, respectively. The optimum conditions of the reactions and the effects of foreign substances are investigated, and the composition of ion-association complexes is discussed also. Based on the ion-association reaction, a highly sensitive, simple and rapid method has been proposed to the determination of TGCs.  相似文献

A simple chemiluminometric method using flow injection has been developed for the determination of paracetamol (acetaminophen), based on the chemiluminescence produced by the reduction of tris(2,2′-bipyridyl)ruthenium(III). The latter is obtained by oxidation of tris(2,2′-bipyridyl)ruthenium(II) by potassium permanganate in dilute sulphuric acid in the presence of paracetamol. A standard or sample solution was injected into the ruthenium(II) stream (flow rate 1.5 ml min−1) which was then merged with potassium permanganate in dilute sulphuric acid stream (flow rate 0.5 ml min−1). The chemiluminescence intensity is enhanced by the presence of manganese(II) ions. Under the optimum conditions, a linear calibration graph was obtained over the range of 0.3-50.0 μg ml−1 and the detection limit was 0.2 μg ml−1 (s/n = 3). The relative standard deviation of the proposed method calculated from 20 replicate injections of 5.0 μg ml−1 paracetamol was 1.1%. The sample throughput was 90 h−1. The method was successfully applied to the determination of paracetamol in commercial pharmaceutical formulations.  相似文献

We developed a flow injection (FI) method for the determination of thiomersal (sodium ethylmercurithiosalicylate, C9H9HgNaO2S) based on the UV/microwave (MW) photochemical, online oxidation of organic mercury, followed by cold vapor generation atomic fluorescence spectrometry (CVG-AFS) detection. Thiomersal was quantitatively converted in the MW/UV process to Hg(II), with a yield of 97 ± 3%. This reaction was followed by the reduction of Hg(II) to Hg(0) performed in a knotted reaction coil with NaBH4 solution, and AFS detection in an Ar/H2 miniaturized flame. The method was linear in the 0.01–2 μg mL−1 range, with a LOD of 0.003 μg mL−1. This method has been applied to the determination of thiomersal in ophthalmic solutions, with recoveries ranging between 97% and 101%. We found a mercury concentration in commercial ophthalmic solutions ranging between 7.5 and 59.0 μg mL−1.  相似文献

In this paper, an automatic method for the screening of water samples containing Cu(II) was proposed, based on peryoxalate chemiluminescence reaction using coproporphyrin I as fluorophor compound to provide selectivity and a simple flow injection (FI) chemiluminescence detector (CLD). FI system conditions were chosen in order to distinguish samples over or under legislation limit established (50 μg l−1) with high reliability. The detection limit found was 9 μg l−1 and the linear dynamic range was 15-125 μg l−1 of Cu(II). Repeatibility and reproducibility studies gave good precision and accuracy with recovery near 100%. Under these conditions, the method resulted selective and only Fe(II), Fe(III) and Pb(II) could interfere, but at a concentration level higher than their normal concentration in waters. The proposed method was found to be highly reliable for screening purposes and it was successfully applied to the screening of a variety of real water samples.  相似文献

An optical chemical sensor based on immobilization of 2-(5-bromo-2-pyridylazo)-5-(diethylamino)phenol (Br-PADAP) in Nafion membrane is described. The membranes were cast onto glass substrates and were used for the determination of nickel in aqueous solutions by spectrophotometry. The sensor system is highly transparent, mechanically stable and showed no evidence of reagent leaching. The influence of several parameters such as pH, ligand concentration, and type and concentration of regenerating solution were optimized. The sensor system showed good sensitivity in the range 0.5-20 μg ml−1 with a detection limit of 0.3 μg ml−1 Ni(II). The sensor has been incorporated into a home-made flow-through cell for determination of nickel in flowing streams with improved sensitivity, precision and detection limit. The calibration curve in the flow system was linear in the range 0.1-16 μg ml−1 with a detection limit of 0.07 μg ml−1. The sensor is easily regenerated by dilute nitric acid solution. The proposed method was successfully applied to the determination of nickel content in vegetable oil and chocolate samples and the results were compared with those obtained using atomic absorption spectrometry.  相似文献

The utility of a dynamic hollow-fibre liquid phase microextraction method (optimized using a four-variable experimental design and response surface modelling) for extracting dinitrophenolic compounds from human plasma samples was evaluated. The investigated variables were donor phase salt concentration (10-400 mM), donor phase pH (2-6), acceptor phase pH (7-12), and donor/acceptor phase flow rates (30/7.5 to 70/17.5 μL min−1). Four dinitrophenol pesticides were used as model substances at concentrations of 0.1 μg mL−1 in spiked human plasma samples. Extraction efficiencies ranging from 42 to 77% with RSDs below 9 were achieved with the optimized method. The flow rate and acceptor pH were shown to strongly affect the extraction efficiency for all compounds, while the donor phase pH and salt concentration had minor effects. With a well-defined acceptor phase pH and flow rate the system exhibited high robustness. The limits of quantification for the investigated compounds, using the presented extraction method followed by liquid chromatography/electrospray ionization mass spectrometry in selected ion monitoring mode, ranged from 0.05 to 0.1 μg mL−1 plasma.  相似文献

Based on CdTe/CdS quantum dots (CdTe/CdS QDs) fluorescence (FL) reversible control, a new and sensitive FL sensor for determination of anthraquinone (AQ) anticancer drugs (adriamycin and daunorubicin) and herring sperm DNA (hsDNA) was developed. Under the experimental conditions, FL of CdTe/CdS QDs can be effectively quenched by AQ anticancer drugs due to the binding of AQ anticancer drugs on the surface of CdTe/CdS QDs and photoinduced electron transfer (PET) process from CdTe/CdS QDs to AQ anticancer drugs. Addition of hsDNA afterwards brought the restoration of CdTe/CdS QDs FL intensity, as AQ anticancer drugs peeled off from the surface of CdTe/CdS QDs and embedded into hsDNA double helix structure. The liner ranges and the detection limits of FL quenching methods for two AQ anticancer drugs were 0.33-9 μg mL−1 and 0.09 μg mL−1 for ADM and 0.15-9 μg mL−1 and 0.04 μg mL−1 for DNR, respectively. The restored FL intensity was proportional to concentration of hsDNA in the range of 1.38-28 μg mL−1and the detection limit for hsDNA was 0.41 μg mL−1. It was applied to the determination of AQ anticancer drugs in human serum and urine samples with satisfactory results. The reaction mechanism of CdTe/CdS QDs FL reversible control was studied.  相似文献

A flow injection analysis (FIA) method coupled to resonance Rayleigh scattering (RRS) detection for the determination of verapamil hydrochloride (VP) was proposed. In pH 1.0 acidic medium, 12-tungstophosphoric acid (TP) reacted with VP to form an ion-association complex, which resulted in a significant enhancement of RRS intensity. The maximum scattering peak was located at 293 nm. RRS intensity was proportional to the concentration of VP in the range of 0.017-13.0 μg mL−1, and the detection limit (3σ) was 5.1 ng mL−1. The proposed method exhibited satisfactory reproducibility with a relative standard derivation (R.S.D.) of 2.1% for 11 successive determinations of 5.0 μg mL−1 VP. Therefore, a novel method for the determination of VP by FIA-RRS was developed. The optimum reaction conditions and the parameters of the FIA operation such as flow rate, injection volume, reactor length, and so on had been optimized in this paper. The present method had been applied to the determination of VP in serum samples and pharmaceuticals with satisfactory results. The maximal sample throughput in the optimized system was 80 h−1.  相似文献

In this paper, a chemiluminescence (CL) micro-flow system combined with on-line solid phase extraction (SPE) is presented for determination of β-lactam antibiotics (penicillin, cefradine, cefadroxil, cefalexin) in milk. It is based on the enhancement effect of β-lactam antibiotics on the luminol-K3Fe(CN)6 CL system. The micro-flow system was fabricated from two polymethyl methacrylate (PMMA) plates (50 mm × 40 mm × 5 mm) with the microchannels of 200 μm wide and 150 μm deep. C18-modified silica gel was packed into the microchannel (length: 10 mm; width: 1 mm; depth: 500 μm) to serve as SPE device. Extraction and preconcentration of the analytes were carried out using on-line SPE micro-flow system and the selectivity of CL detection was improved. The detection limits were 0.5 μg mL−1 of penicillin, 0.04 μg mL−1 of cefradine, 0.08 μg mL−1 of cefadroxil and 0.1 μg mL−1 of cefalexin. The proposed method was also applied to analyze the β-lactam antibiotics in milk. Experimental results were in good agreement with those obtained by high performance liquid chromatography (HPLC) method with UV detection.  相似文献
