A new methyl viologen-mediated amperometric enzyme electrode sensitive to glucose has been developed using carbon film electrode substrates. Carbon film electrodes from resistors fabricated by pyrolytic deposition of carbon were modified by immobilization of glucose oxidase through cross-linking with glutaraldehyde in the presence of bovine serum albumin. The mediator, methyl viologen, was directly immobilised with the enzyme together with Nafion cation-exchange polymer. The electrochemistry of the glucose oxidase/methyl viologen modified electrode was investigated by cyclic voltammetry and by electrochemical impedance spectroscopy. The biosensor response to glucose was evaluated amperometrically; the detection limit was 20 μM, the linear range extended to 1.2 mM and the reproducibility of around 3%. When stored in phosphate buffer at 4 °C and used every day, the sensor showed good stability over more several weeks.  相似文献

Square wave voltammograms of methyl viologen (MV2+) were recorded at a renewable Nafion-loaded carbon paste electrode immersed in 20 mL of phosphate buffer solution (pH 7.4) contained in a beaker-type electrochemical cell (Pyrex glass; contact area = 33 cm2). From the calibration plot obtained in the concentration range 0.01–2.4 μM, it could be concluded that the maximum surface excess of MV2+ adsorbed on Pyrex glass (0.24 nmol cm−2) was reached at concentrations above 1.4 μM. The adsorption isotherm presented a sigmoidal shape characteristic of attractive interaction. From this plot, it was evaluated that the maximum depletion of MV2+ in solution (28%) did not occur at the lowest concentrations, but in nearly micromolar solutions. This result emphasizes the unavoidable and random errors which are involved when MV2+ solutions are purposely diluted in Pyrex glassware. The MV2+ adsorption proceeds to a less extent on Duran borosilicate glass and/or in acid or alkaline medium.  相似文献

A simple and sensitive flow injection fluorometric method for the determination of ascorbic acid is described. Perylenebisimide-linked nitroxide (PBILN) is used as a fluorescent reagent, which permits the selective determination of ascorbic acid. The fluorescence of the perylenebisimide moiety in PBILN is quenched by the nitroxide moiety, which is linked to the perylenebisimide. When a stream of a solution of ascorbic acid is merged with a stream of PBILN, the ascorbic acid reacts with the nitroxide moiety of PBILN to form hydroxylamine, and the fluorescence properties of the perylenebisimide moiety are recovered. As a result, a peak-shaped fluorescence signal is produced, which can be observed by a fluorescence detector located downstream. Under optimized conditions, a good linear relationship between the concentration of ascorbic acid and peak height in the concentration range from 0.5 to 10 μmol L−1 was found and the detection limit (S/N = 3) was 0.28 μmol L−1. The relative standard deviation for the determination of 4.0 μmol L−1 ascorbic acid samples was 1.0% (n = 5). The proposed method was applied to the determination of ascorbic acid in several soft drink beverages and the analytical results were in good agreement with those obtained using a conventional method.  相似文献

Ascorbic acid may be determined spectrophotometrically at 360 nm based on reduction of vanadotungstophosphoric acid using flow-injection analysis. The carrier stream was distilled water and the reagent streams were buffer solution (pH 3.0), 1.735 × 10?3 M dodecatungstophosphoric acid and 1.735 × 10?3 M sodium vanadate. The injection rate was 80 h?1. The calibration graph was linear up to 80 μg ml?1 ascorbic acid and the relative standard deviation for the determination of 20 μg ml?1 ascorbic acid was 1.5% (n=10). The detection limit was 1.0 μg ml?1 ascorbic acid, based on an injection volume of 250 μl. The system was applied to the determination of ascorbic acid in vitamin C tablets.  相似文献

将在线恒电流电解产生ClO-与Luminol构成了较强的化学发光体系,基于维生素C对该化学发光体系有强抑制作用,结合流动注射技术,建立了测定维生素C的流动注射抑制电化学发光新方法.该方法在维生素C的浓度为2.0×10-10~1.5×10-8 mmol/mL之间分段回归,呈良好的线性,检出限达到5.0×10-11 mmol/mL.  相似文献

Simple, strip-type sensors based on 7,7,8,8-tetracyanoquinodimethane-modified graphite were prepared using screen printing techniques. The electrochemical strips operated at low potentials [50 mV at pH 7.0 or 100 mV at pH 4.8 vs. Ag/AgCl (printed)] and had a sensitivity of 3.5–7.1 μA 1 mmol?1L-ascorbic acid. Determination of ascorbic acid concentration was achieved in 30 s and required samples of ca. 30 μl. The current output of the electrodes was found to be relatively insensitive to variations in pH over the range 5.0–8.5. Between 15 and 35 °C, the temperature coefficient was 2.7% °C?1. The printed electrodes were suitable for single determinations but demonstrated adequate stability for periodic re-use. The ascorbic acid concentration in the juice of fresh fruit was determined using the electrochemical printed electrodes and a commercially available enzymatic test kit. Close agreement was observed between the two methods [r=0.9997 (n=12),slope=0.9798]. The limit of detection using the printed sensor for real samples was calculated as 4mg l?1(22 μM).  相似文献

Chiral CE was successfully applied to the separation and quantification of catechin, epicatechin and ascorbic acid in some commercial drinks and human urine. Analysis involved the separation of analytes in less than 5.0 min at 240 nm with an untreated fused-silica capillary under hydrodynamic injection mode. The running buffer consisted of 50 mM borate buffer with 3 mM beta-CD at pH 8.35. Detection limits for catechin, epicatechin and ascorbic acid were 0.028, 0.011 and 0.004 microg/mL, respectively. Linearity was investigated by selecting the ranges of calibration according to the amount of analytes in urine giving correlation coefficient percent (% r(2)) ranging between 99.4 and 99.6 at 99% confidence level. The maximum urinary excretion of catechin and epicatechin were noted at 2.0 and 4.0 h of the administrated dose. Unchanged catechin, epicatechin and ascorbic acid amounted to about 1.500, 8.696 and 0.003% of the administered dose in the 48.0 h urine collection. The proposed method achieved 99.2% completeness (n = 20) in urine media.  相似文献

A highly sensitive, fast and stable conductometric enzyme biosensor for determination of nitrate in water is reported for the first time. The biosensor electrodes were modified by methyl viologen mediator mixed with nitrate reductase (NR) from Aspergillus niger by cross-linking with glutaraldehyde in the presence of bovine serum albumin and Nafion® cation-exchange polymer. The process parameters for the fabrication of the enzyme electrode and various experimental variables such as pH, the enzyme loading and time of immobilization in glutaralaldehyde vapor were investigated with regard to their influence on sensitivity, limit of detection, dynamic range and operational and storage stability. The biosensor can reach 95% of steady-state conductance value in about 15 s. Linear calibration in the range of 0.02 and 0.25 mM with detection limits of 0.005 mM nitrate was obtained with a signal-to-noise ratio of 3. When stored in 5 mM phosphate buffer (pH 7.5) at 4 °C, the sensor showed good stability over 2 weeks.  相似文献

A sol-gel carbon composite electrode (CCE) has been prepared by mixing a sol-gel precursor (e.g. methyltrimethoxysilane) and carbon powder without adding any electron transfer mediator or specific reagents. It was demonstrated that this sensor can be used for simultaneous determination ascorbic acid, neurotransmitters (dopamine and adrenaline) and uric acid. Direct electrochemical oxidation of ascorbic acid, uric acid and catecholamines at a carbon composite electrode was investigated. The experimental results were compared with other common carbon based electrodes, specifically, boron doped diamond, glassy carbon, graphite and carbon paste electrodes. It was found that the CCE shows a significantly higher of reversibility for dopamine. In addition, in comparison to the other electrodes used, for CCE the oxidation peaks of uric acid, ascorbic acid and catecholamines in cyclic and square wave voltammetry were well resolved at the low positive potential with good sensitivity. The advantages of this sensor were high sensitivity, inherent stability and simplicity and ability for simultaneous determination of uric acid, catecholamines and ascorbic acid without using any chromatography or separation systems. The analytical performance of this sensor has been evaluated for detection of biological molecules in urine and serum as real samples.  相似文献

The possibility of use of phosphoric acid (0.2% v/v, pH 2.1) in the mobile phase and co-existing compounds present in foods as the dissolving agent for the pre-analysis sample stabilization were examined for the routine determination of ascorbic acid (AA) in foods by high-performance liquid chromatography (HPLC) with electrochemical detection (ED). The applied potential was set at 400 mV versus an Ag/AgCl reference electrode. It was demonstrated that 0.2% v/v phosphoric acid was the useful mobile phase and l-methionine was the most effective dissolving agent for the pre-run sample stabilization of AA in foods after comparison with other amino acids and water-soluble vitamins. The proposed method was simple, rapid (retention time @ ca. 4 min), sensitive (detection limit: ca. 0.1 ng per injection (5 μl) at a signal-to-noise ratio of 3), highly selective and reproducible (relative standard deviation (R.S.D.); 2.5% (n=7), between-day R.S.D.: 3.7% (5 days)). The calibration graph of AA was linear in the range of 0.1-12.5 ng per injection (5 μl). Recovery of AA was over 90% by the standard addition method. Relationship between structure of compounds and the stability of AA was also examined.  相似文献

Multilayer thin films composed of poly(allylamine hydrochloride) (PAH) and carboxymethyl cellulose (CMC) have been prepared on the surface of a gold (Au) disk electrode by a layer-by-layer deposition of PAH and CMC and ferricyanide ions ([Fe(CN)6]3−) were confined in the film. [Fe(CN)6]3− ions can be successfully confined in the films from weakly acidic or neutral [Fe(CN)6]3− solutions, while, in basic solution, [Fe(CN)6]3− ion was not confined. The [Fe(CN)6]3− ion-confined Au electrode showed clear redox peaks in the cyclic voltammogram around 0.35 V versus Ag/AgCl. The amounts of [Fe(CN)6]3− ions confined in the films depended on the thickness of the films or the number of layers in the LbL films. The [Fe(CN)6]3− ion-confined Au electrode was used for electrocatalytic determination of ascorbic acid in the concentration range of 1-50 mM.  相似文献

Two sequential injection titration systems with spectrophotometric detection have been developed. The first system for determination of ascorbic acid was based on redox reaction between ascorbic acid and permanganate in an acidic medium and lead to a decrease in color intensity of permanganate, monitored at 525 nm. A linear dependence of peak area obtained with ascorbic acid concentration up to 1200 mg l−1 was achieved. The relative standard deviation for 11 replicate determinations of 400 mg l−1 ascorbic acid was 2.9%. The second system, for acetic acid determination, was based on acid–base titration of acetic acid with sodium hydroxide using phenolphthalein as an indicator. The decrease in color intensity of the indicator was proportional to the acid content. A linear calibration graph in the range of 2–8% w v−1 of acetic acid with a relative standard deviation of 4.8% (5.0% w v−1 acetic acid, n=11) was obtained. Sample throughputs of 60 h−1 were achieved for both systems. The systems were successfully applied for the assays of ascorbic acid in vitamin C tablets and acetic acid content in vinegars, respectively.  相似文献

The present work reports a quercetin-modified wax-impregnated graphite electrode (Qu/WGE) prepared through an electrochemical oxidation procedure in quercetin-containing phosphate buffer solution (PBS), for the purpose of detecting uric acid (UA) in the presence of ascorbic acid (AA). During modification quercetin was oxidized to the corresponding quinonic structure, and in the blank buffer solution the electrodeposited film exhibits a voltammetric response anticipated for the surface-immobilized quercetin. Retarding effect of the film towards the reaction of anionic species was found; therefore the pH of sample solutions was selected to ensure the analyte in molecular form. At suitable pHs the Qu/WGE shows excellent electrocatalytic effect towards the oxidation of both AA and UA, and separates the voltammetric signal of UA from AA by about 280 mV, allowing simultaneous detection of these two species. A linear relation between the peak current and concentration was obtained for UA in the range of 1-50 μM in the presence of 0.5 mM AA, with a detection limit 1.0 μM (S/N = 3). This sensor was stable, reproducible and outstanding for long-term use.  相似文献

This paper describes the simultaneous determination of ascorbic acid (AA), dopamine (DA), uric acid (UA) and xanthine (XN) using an ultrathin electropolymerized film of 2-amino-1,3,4-thiadiazole (p-ATD) modified glassy carbon (GC) electrode in 0.20 M phosphate buffer solution (pH 5.0). Bare GC electrode failed to resolve the voltammetric signals of AA, DA, UA and XN in a mixture. On the other hand, the p-ATD modified electrode separated the voltammetric signals of AA, DA, UA and XN with potential differences of 110, 152 and 392 mV between AA-DA, DA-UA and UA-XN, respectively and also enhanced their oxidation peak currents. The modified electrode could sense 5 μM DA and 10 μM each UA and XN even in the presence of 200 μM AA. The oxidation currents were increased from 30 to 300 μM for AA, 5 to 50 μM for DA and 10 to 100 μM for each UA and XN, and the lowest detection limit was found to be 2.01, 0.33, 0.19 and 0.59 μM for AA, DA, UA and XN, respectively (S/N = 3). The practical application of the present modified electrode was demonstrated by the determination of AA, UA and XN in human urine samples.  相似文献

制备了多壁碳纳米管修饰碳黑微电极,研究了多巴胺(DA)和抗坏血酸(AA)在该修饰电极上的电化学行为.实验表明,在pH 7.0的PBS缓冲溶液中,该修饰电极对DA和从均具有显著的催化氧化作用,AA与DA的氧化电位分别为30 mV和280 mV(vs.SCE).利用二次导数线性扫描伏安法测定,DA与AA的线性范围分别为6.0×10-9~2.0×10-4 mol/L和2.0×10-7~1.0×10-3mol/L,检出限为2.0×10-9mol/L 和1.0×10-7mol/L.方法已用于人工合成样品的分析.  相似文献

A simple and sensitive fluorimetric method for the determination of ascorbic acid (AA) is described. The method is based on the condensation reaction between AA and o-phenylenediamine (OPDA) in the absence of the oxidant. The fluorescence intensity is measured at excitation and emission wavelengths of 360 and 430 nm, respectively. Under optimum condition, a linear relationship is obtained between the fluorescence intensity and the concentration of AA in the range of 0.05-40 μg ml−1. The detection limit is 0.006 μg ml−1, which is obviously lower than that of other fluorimetric methods reported.  相似文献

建立了浮选CuSCN间接测定抗坏血酸(Vc)的方法。研究表明,在NaNO3存在下,Cu(Ⅱ)被抗坏血酸还原生成的Cu(Ⅰ)与SCN-形成CuSCN沉淀而被浮选。结果表明,Vc量在2.64~15.8 mg/L范围内与CuSCN的浮选率呈良好的线性关系,回归方程为:E/%=6.008ρ-3.673,r=0.9962。此方法可用于水果和药片中Vc量测定。  相似文献
