Large dendrimers, noted G(n)-3(n+2)cage, containing 3(n+2) o-carborane cluster cages MeC(2)B(10)H(10) at their peripheries (n = number of generation noted G(n)) have been synthesized by Huisgen-type azide alkyne Cu(I)-catalyzed dipolar "click" cycloaddition reactions (CuAAC) between an o-carborane monomeric cluster containing an ethynyl group and arene-centered azido-terminated dendrimers G(n)-3(n+2)N(3) of generations 0, 1, and 2. Attempts to synthesize higher-generation dendrimers of this family yielded insoluble materials. The carborane dendrimers G(0)-9cage, G(1)-27cage, and G(2)-81cage have been characterized by (1)H, (13)C, (11)B NMR, elemental analysis, matrix-assisted laser desorption/ionization time of flight (MALDI-TOF) mass spectroscopy, and size exclusion chromatography (SEC) showing low polydispersities, dynamic light scattering (DLS) showing hydrodynamic diameters of 5.7 nm for the G(1)-27cage and the 12.9 nm for the G(2)-81cage. These dendrimers are extremely robust thermally, with 10% mass loss temperatures of 411 °C for the G(0)-9cage, 371 °C for the G(1)-27cage, and 392 °C for the G(2)-81cage. They all showed a strong absorption in the UV region peaking at 258 nm, whereas emission spectra of low intensities were observed between 280 and 480 nm.  相似文献

An efficient strategy for the solid-phase synthesis of azidomethylene inhibitors targeting cysteine proteases is described. The method is highlighted by its compatibility with readily available building blocks, as well as its ability to accommodate different functional groups. [reaction: see text] Various pseudo-oligosacchardies and amino acid glycoconjugates were synthesized via an intermolecular 1,3-dipolar cycloaddition ("click") reaction using easily accessible carbohydrate and amino acid derived azides and alkynes as building blocks. The rapid rise of drug-resistant bacteria is one of the most serious unmet medical needs facing the world. Despite this increasing problem of antibiotic resistance, the number of different antibiotics available for the treatment of serious infections is dwindling. Therefore, there is an urgent need for new antibacterial drugs, preferably with novel modes of action to potentially avoid cross-resistance with existing antibacterial agents. In recent years, increasing attention has been paid to bacterial protein secretion as a potential antibacterial target. Among the different protein secretion pathways that are present in bacterial pathogens, the general protein secretory (Sec) pathway is widely considered as an attractive target for antibacterial therapy. One of the key components of the Sec pathway is the peripheral membrane ATPase SecA, which provides the energy for the translocation of preproteins across the bacterial cytoplasmic membrane. In this review, we will provide an overview of research efforts on the discovery and development of small-molecule SecA inhibitors. Furthermore, recent advances on the structure and function of SecA and their potential impact on antibacterial drug discovery will be discussed.  相似文献

树枝状、梳形、超枝化、星形、及H形等支链结构和环形、多环形等结构的拓扑高分子具有不同于直链结构高分子的独特性能,是一类在材料改性、纳米科技和生物医药等领域有着重要应用前景的新型功能性聚合物材料,日益受到人们的重视,但其复杂的结构导致它们的合成非常繁琐并因此束缚其实际应用.具有定量、快速、高效和高选择性、反应条件温和、产物单一、易于分离等特点的点击反应(click reaction)的应用极大地推动了拓扑结构高分子合成方法的发展.本文就炔/叠氮1,3-偶极环加成反应(CuAAC)、硫醇-烯/炔、胺-丙烯酸酯、Diels-Alder环加成等点击反应应用于树枝状、超枝化、环形、星型等拓扑结构高分子的合成做一简要介绍,并提出了进一步的研究方向.  相似文献

A click chemistry approach for the synthesis of small molecule inhibitor-peptide conjugates to achieve organelle-specific delivery has been developed. Enantio- and diastereoselective syntheses of a substituted oxazolidinone, isoxazoline and pyrazoline as beta-lactam surrogates are described. The substituted heterocycles were designed to incorporate side chains closely resembling those found in the beta-lactam cholesterol absorption inhibitor ezetimibe (1). The translation of DNA sequences into synthetic products is a key requirement of our approach to evolving synthetic molecules through iterated cycles of translation, selection, and amplification. Here we report general linker and purification strategies for sequence-specific DNA-templated synthesis that collectively enable the product of a DNA-templated reaction to be isolated and to undergo subsequent DNA-templated reactions. Using these strategies, we have achieved the first multistep nucleic acid-templated small-molecule syntheses to generate two different molecules. In addition to representing a method for translating DNA templates sequence-specifically into corresponding multistep synthetic products, our findings also provide experimental support for previously proposed models invoking multistep nucleic acid-templated synthesis as mediating the prebiotic translation of replicable information into the earliest functional molecules.  相似文献

Rapid assembly of starch fragment analogues was achieved using "click chemistry". Two Pd-catalyzed reductive transformations of diynes tethered through a silyl ether linkage were developed, where the reaction outcomes were controlled solely by selection of phosphine ligand. We screened Pd precatalysts, ligands, and additives to optimize conditions selective either for reductive cyclization or hydrogenation of this substrate class. Sixteen silyl ether-tethered diynes were prepared and subjected to the best catalyst/ligand combinations for each pathway. Silacyclic dienes and silyl-tethered enyne products of these reactions were elaborated to densely substituted, stereochemically- and appendage-rich, bicyclic and tricyclic small molecules in 1-3 synthetic steps. These studies illustrate how small modifications to a transition-metal catalyst can be used to access a diverse set of small molecules, in a fashion analogous to biosynthetic pathways such as terpene biosynthesis, where minor changes to enzyme structure direct skeletal differentiation.  相似文献

The de novo molecular design program SPROUT has been used in conjunction with the X-ray crystal structure of RNA polymerase (RNAP) from Thermus aquaticus to produce a novel enzyme inhibitor scaffold. A short and efficient synthesis of molecules corresponding to this scaffold has been developed and in keeping with the design predictions, the resulting inhibitors displayed useful levels of inhibition of Escherichia coli RNAP.  相似文献
