The influence of cationic poly(diallyldimethylammonium chloride) on the morphology and phase behavior of anionic phospholipid vesicles was investigated using differential scanning calorimetry, fluorescent microscopy and light scattering technique. A wide range of polymer concentration has been examined for the first time. The polycation can bind electrostatically to the vesicles to compensate, neutralize and reverse the vesicular charge, depending on the molar ratio of cationic to anionic group R. For R<1, charge compensation weakened the electrostatic repulsion between the lipid molecules, leading to formation of polymer-modified vesicles, each with an increased number of bilayers. The bilayer exhibits a rising main phase transition temperature from a gel to liquid crystalline state. This behavior persisted until R≈1 around the neutralization condition, where the complexes became largest and precipitate. With R>1, charge reversal took place, the complex size reduced. Interestingly, the main phase transition temperature was found for the first time to shift back towards the original value in the absence of polymer for large enough R. Although the thermal behavior was nearly independent of the polymer molecular weight, the complex morphology could be different.  相似文献

We report on the gel-to-fluid phase transition behavior of unilamellar vesicles formed with 1,2-dimyristoyl-sn-phosphatidylcholine (14:0 DMPC). We have interrogated the gel-to-fluid transition temperature of these bilayer structures using the chromophore perylene incorporated in their nonpolar region. We observe a discontinuous change in the reorientation time of perylene sequestered within the bilayer at the known melting transition temperature of 14:0 DMPC, 24 degrees C. The perylene reorientation data reveal a local viscosity of 14.5 +/- 2.5 cP in the gel phase, and 8.5 +/- 1.5 cP in the fluid phase. We have also incorporated small amounts of 1,2-dimyristoleoyl-sn-glycero-3-phosphocholine (14:1 DMPC) into these unilamellar vesicles and find that the melting transition temperature for these bilayers varies in a regular manner with the amount of 14:1 DMPC present. These data demonstrate that very little "contaminant" is required to cause a substantial change in the gel-to-fluid transition temperature, even though these contaminants do not alter the viscosity of the bilayer sensed by perylene, either above or below the melting transition.  相似文献

Submonolayer thin films of a three-ring bent-core (or banana-shaped) compound, m-bis(4-n-octyloxystyryl)benzene (m-OSB), were vacuum-deposited on a mica surface, and a spontaneous transition from monolayer films to bilayer crystals was observed at room temperature, which was ascribed to the specific molecular shape and polar layered packing of the bent-core molecules [Tang et al. J. Phys. Chem. B 2004, 108 (34), 12921-12926]. The crystal nucleation and growth from the monolayer films as well as the melting phase transition from the bilayer crystals were investigated using atomic force microscopy (AFM). It was shown that after initial nucleation, the crystal growth was achieved through three pathways: direct absorption of molecules from monolayer films, molecular cluster diffusion, and quasi-Ostwald ripening. When annealing the bilayer crystals at elevated temperatures, morphological change from a bilayer to a monolayer was observed, and some new islands with fingerlike patterns were formed during this process, which resulted from a diffusion-controlled growth of the molten molecules. In general, the high-resolution AFM in combination with the molecularly thin m-OSB films provided us with direct visualization of nucleation, crystal growth, melting, and film morphology evolution on the mesoscopic scale, which are of fundamental interest from the theoretical viewpoint and are of central importance for the control of interfacial properties in practical applications.  相似文献

Cholesterol/phospholipid interactions in hybrid bilayer membranes   总被引:1,自引:0,他引:1  
The interactions between cholesterol and saturated phospholipids in hybrid bilayer membranes (HBMs) were investigated using the interface-sensitive technique of vibrational sum frequency spectroscopy (VSFS). The unique sensitivity of VSFS to order/disorder transitions of the lipid acyl chains was used to determine the main gel to liquid crystal phase transition temperature, Tm, for HBMs of binary cholesterol/phospholipid mixtures on octadecanethiolate self-assembled monolayers. The phase transition temperature and the breadth of the transition were shown to increase with cholesterol content, and the phase boundaries observed in the cholesterol/phospholipid HBMs were comparable to the published phase diagrams of binary cholesterol/phospholipid vesicles. A thermodynamic assessment of the cooperative units of the HBM phase transitions revealed the presence of <10 nm diameter domains that were independent of the cholesterol composition.  相似文献

Occurrence of two-dimensional chain melting phase transition in foam bilayers was established for the first time. Microscopic horizontal foam bilayers [Newton black films (NBF)] were investigated by the microinterferometric method of Scheludko-Exerowa. The foam bilayers were formed from water-ethanol solutions of dimyristoylphosphatidylcholine (DMPC) and dipalmitoylphosphatidylcholine (DPPC) and egg phosphatidylcholine (Egg PC) and samples of amniotic fluid (AF) at different temperatures. The influence of temperature on the foam bilayer thickness h(w) and on the critical concentration Cc for formation of foam bilayer was studied. It was shown that in the range of the main phase transition the temperature dependence of h(w) and C(c) changed specifically in the case of DMPC and DPPC foam bilayers. The thickness of the foam bilayers increased with decreasing temperature in the range of the main phase transition due to the melting of hydrocarbon tails of phospholipid molecules. These changes took place at the temperatures of the bulk chain-melting phase transitions, as determined by differential scanning calorimetry (DSC) for both aqueous, and water/ethanol DMPC, DPPC, and DPPC dispersions. An effect of the 'disperse medium' on h(w) was found for foam bilayers from DPPC. The results that foam bilayers could have different thickness at different temperatures disproved the current concept that NBF acquired constant thickness at concentrations higher than C(el,cr). The data for Cc were analysed on the basis of the hole-nucleation theory of bilayer stability of Kashchiev and Exerowa. This theory considered the amphiphile bilayer as a two-dimensional ordered system with short-range molecular interactions between the first neighbour molecules (as in a crystal). The short-range molecular interactions were presented by the parameter binding energy Q of an amphiphile molecule in the bilayer. The binding energy Q of two neighbouring phospholipids was calculated for the gel (30-60 kT) and liquid crystalline state (16-18 kT) of the bilayers from DMPC, DPPC, Egg PC, AF. Concentration/temperature phase diagram of DPPC foam bilayers that defined regions of gaseous (ruptured), gel and liquid crystalline foam bilayers were drawn. The values of Q obtained for various samples were very close and vary from 5.3 x 10(-20) to 9.4 x 10(-20) (approx. 13-22 kT) which indicated that in all cases the foam bilayers were in liquid-crystalline state. This is an important result since the parameter studied-threshold concentration (threshold dilution) is crucial for a very successful assessment of the risk for respiratory distress syndrome (RDS) in newborns and could be employed in medicine for assessment of other respiratory disturbances. It is to be expected that foam bilayers from phospholipids could be used as a model for investigation of short-range forces in biological structures, of interaction between membranes, etc.  相似文献

MPEG–PCL diblock copolymers consisting of methoxy polyethylene glycol (MPEG, 750 g/mol) and poly(?‐caprolactone) (PCL) were synthesized by ring‐opening polymerization. Aqueous solutions of the synthesized diblock copolymers were prepared by dissolving the MPEG–PCL diblock copolymers at concentrations in the range of 0–20 wt %. When the PCL molecular weight was 3000 or greater, the polymer was only partially soluble in water. As the temperature was increased from room temperature, the diblock copolymer solutions showed two phase transitions: a sol‐to‐gel transition and a gel‐to‐sol transition. The sol‐to‐gel phase transition temperature decreased substantially with increasing PCL length. The sol–gel–sol transition with the increase in temperature was confirmed by monitoring the viscosity as a function of temperature. The temperature ranges of the phase transitions measured by the tilting method were in full agreement with those determined from the viscosity measurements. The maximum viscosity of the copolymer solution increased with increasing hydrophobicity of the diblock copolymer and with increasing copolymer concentration. X‐ray diffraction (XRD) and differential scanning calorimetry (DSC) analyses revealed that the diblock copolymers exhibited crystalline domains that favored the formation of an aggregated gel because of the tight aggregation and strong packing interactions between PCL blocks. Scanning electron micrographs of the diblock copolymer solutions in the sol state showed interconnected polyhedral pore structures, whereas those of the gel state revealed a fibrillar‐like morphology. Atomic force microscope (AFM) studies of the sol and gel surfaces showed that the sol surface was covered with fine globular particles, whereas the gel surface was covered with particles in micron‐scale irregular islets. These findings are consistent with uniform mixing of the diblock copolymer and water in the sol state, and aggregation of PCL blocks in the gel state. In conclusion, we confirm that the MPEG–PCL diblock copolymer solution exhibited a sol–gel–sol transition as a function of temperature. © 2006 Wiley Periodicals, Inc. Aspirin and 1,2-dimyristoyl-sn-glycero-3-phosphoethanolamine (DMPE) are deposited from their alcoholic mixed solution onto highly oriented pyrolytic graphite (HOPG) by spin coating. The film structure and morphology are characterized by atomic force microscopy (AFM). The barely soluble DMPE forms a highly oriented stripe phase as a result of its one-dimensional epitaxy with the HOPG lattice. The bilayer stripe pattern exposes the cross section of the lipid bilayer lamellae and enables the direct visualization of the molecular interactions of drug or biological molecules with either the hydrophobic or the hydrophilic part of the phospholipid bilayer. The bilayer pattern affects the aspirin molecular deposition and aggregation. AFM shows that the aspirin molecules prefer to deposit and aggregate along the aliphatic interior part of the bilayer pattern, giving rise to parallel dimer rods in registry with the underlying pattern. The nonpolar interactions between aspirin and the phospholipid bilayer are consistent with the lipophilic nature of aspirin. The bilayer pattern not only stabilizes the rodlike aggregate structure of aspirin at low aspirin concentration but also inhibits crystallization of aspirin at high aspirin concentration. Molecular models show that the width of the DMPE aliphatic chain interior can accommodate no more than two aspirin dimers. The bilayer confinement may prevent aspirin from reaching its critical nucleus size. This study illustrates a general method to induce a metastable or amorphous form of an active pharmaceutical ingredient (API) by chemical confinement under high undercooling conditions. Metastable and amorphous solids often display better solubility and bioavailability than the stable crystalline form of the API.  相似文献

Thermoresponsive polymer layers offer the possibility of preparing smart surfaces with properties that are switchable through a phase transition, usually close to the lower critical solution temperature of the polymer. In particular, poly( N-isopropylacrylamide) (pNIPAM) has gained a great deal of attention because it has such a phase transition in a physiologically interesting temperature range. We have prepared ultrathin thermoresponsive coatings by grafting pNIPAM on a plasma-CVD-deposited, poly(ethylene oxide)-like polymer substrate that was activated in an Ar plasma discharge to initiate the grafting. The presence and integrity of pNIPAM was verified by XPS and ToF-SIMS, and a dramatic change in the wettability during the phase transition was identified by temperature-dependent contact angle measurements. The transition from the hydrated to the collapsed conformation was analyzed by temperature-dependent QCM measurements and by AFM. An unusual, reversible behavior of the viscoelastic properties was seen directly at the phase transition from the swollen to the collapsed state. The phase transition leads to a switching from protein repulsion to a state that allows the adsorption of proteins.  相似文献

The effects of adsorption of two kinds of proteins on the membrane characteristics of liposomes were examined at pH 7.4 in terms of adsorption amounts of proteins on liposomes, penetrations of proteins into liposomal bilayer membranes, phase transition temperature, microviscosity and permeability of liposomal bilayer membranes, using positively charged lysozyme (LSZ) and negatively charged bovine serum albumin (BSA) as proteins and negatively charged L-alpha-dipalmitoylphosphatidylglycerol (DPPG) liposomes. The saturated adsorption amount of LSZ was 720 g per mol of liposomal DPPG, while that of BSA was 44 g per mol of liposomal DPPG. The penetration of LSZ into DPPG lipid membranes was greater than that of BSA. The microviscosity in the hydrophobic region of liposomal bilayer membranes increased due to adsorption (penetration) of LSZ or BSA, while the permeability of liposomal bilayer membranes increased. The gel-liquid crystalline phase transition temperature of liposomal bilayer membranes was not affected by adsorption of LSZ or BSA, while the DSC peak area (heat of phase transition) decreased with increasing adsorption amount of LSZ or BSA. It is suggested that boundary DPPG makes no contribution to the phase transition and that boundary DPPG and bulk DPPG are in the phase-separated state, thereby increasing the permeability of liposomal bilayer membranes through adsorption of LSZ or BSA. A possible schematic model for the adsorption of LSZ or BSA on DPPG liposomes was proposed.  相似文献

Atomic force microscopy (AFM) has been used to characterize the formation of a phospholipid bilayer composed of 1,2-dimyristyl-sn-glycero-3-phosphocholine (DMPC) at a Au(111) electrode surface. The bilayer was formed by one of two methods: fusion of lamellar vesicles or by the combination of Langmuir-Blodgett (LB) and Langmuir-Schaefer (LS) deposition. Results indicate that phospholipid vesicles rapidly adsorb and fuse to form a film at the electrode surface. The resulting film undergoes a very slow structural transformation until a characteristic corrugated phase is formed. Force-distance curve measurements reveal that the thickness of the corrugated phase is consistent with the thickness of a bilayer lipid membrane. The formation of the corrugated phase may be explained by considering the elastic properties of the film and taking into account spontaneous curvature induced by the asymmetric environment of the bilayer, in which one side faces the gold substrate and the other side faces the solution. The effect of temperature and electrode potential on the stability of the corrugated phase has also been described.  相似文献

The miscibility, spherulite growth kinetics, and morphology of binary blends of poly(β‐hydroxybutyrate) (PHB) and poly(methyl acrylate) (PMA) were studied with differential scanning calorimetry, optical microscopy, and small‐angle X‐ray scattering (SAXS). As the PMA content increases in the blends, the glass‐transition temperature and cold‐crystallization temperature increase, but the melting point decreases. The interaction parameter between PHB and PMA, obtained from an analysis of the equilibrium‐melting‐point depression, is −0.074. The presence of an amorphous PMA component results in a reduction in the rate of spherulite growth of PHB. The radial growth rates of spherulites were analyzed with the Lauritzen–Hoffman model. The spherulites of PHB were volume‐filled, indicating the inclusion of PMA within the spherulites. The long period obtained from SAXS increases with increased PMA content, implying that the amorphous PMA is entrapped in the interlamellar region of PHB during the crystallization process of PHB. Organotin compounds are widely distributed toxicants. They are membrane‐active molecules with broad biological toxicity. We have studied the interaction of tributyltin and triphenyltin with phosphatidylserine model membranes using differential scanning calorimetry, infrared spectroscopy and X‐ray diffraction techniques. Organotin compounds produced a broadening of the gel to the liquid‐crystalline phase transition of the phospholipid and a shifting of the phase transition temperature to lower values. Infrared spectroscopy experiments showed that tributyltin exerted a fluidizing effect on the apolar part of the bilayer, and that both tributyl‐ and triphenyltin interact with the interfacial region of the bilayer, making the carbonyl groups less accessible to water. As seen by X‐ray diffraction experiments, organotin compounds were unable to change the bilayer macroscopic organization of the phospholipid, but they were able to reduce the long‐range order of the multibilayer system and to disorder the packing of the phospholipid molecules. The observed interaction between organotin compounds and phosphatidylserine membranes promotes physical perturbations that could affect membrane function and may mediate some of their toxic effects. We have performed small-angle (SAXS) and wide-angle X-ray scattering (WAXS) measurements on the lamellar phase and on large unilamellar vesicles (LUVs) of DC(13)PC in the temperature range corresponding to the anomalous swelling regime of multibilayer systems, adjacent to the chain melting transition, and across the transition. Our SAXS measurements indicate that on cooling from the L(alpha) phase, a uniform progressive swelling of the lamellar system to anomalous distances, starting approximately 2 degrees C above the main transition, is followed by a region of coexistence, covering the width of the transition ( approximately 0.6 degrees C). Across the transition region, a progressively increasing volume fraction of gel phase with a constant P (beta') interlamellar distance coexists with a decreasing amount of nongel phase that keeps on swelling to longer distances. Along both the swelling and the transition regions, anomalies in the specific heat are observed revealing a two-step process. Simultaneous WAXS experiments show a progressive "density" increase along the swelling region, constituting a direct spectroscopic evidence of an "evolving membrane" approaching the transition in a bulk real system. Calorimetric and densitometric measurements on LUVs are also presented, together with WAXS results, that show the existence of a double step main transition in a single component nanosized closed bilayer.  相似文献

用原子力显微镜、小角X射线散射和31PNMR分别对液晶态磷脂酰乙醇胺脂质体和LB膜结构进行了研究.用原子力显微镜观察到了液晶态脂质体的立方相和双层膜共存的结构图像.研究结果表明,两相共存的状态与双亲性分子的结构、浓度以及介质的组分和pH等因素有关.用小角X射线散射和31PNMR研究发现,在DEPE液晶态中,钠盐诱导形成Q229(Im3m)立方相.DEPE液晶态分别在37.5℃出现Lβ→Lα可逆相变,在63.5℃出现Lα→H可逆相变.  相似文献

Barbaloin is a bioactive glycosilated 1,8-dihydroxyanthraquinone present in several exudates from plants, such as Aloe vera, which are used for cosmetic or food purposes. It has been shown that barbaloin interacts with DMPG (dimyristoylphosphatidylglycerol) model membranes, altering the bilayer structure (Alves, D. S.; Pérez-Fons, L.; Estepa, A.; Micol, V. Biochem. Pharm. 2004, 68, 549). Considering that ESR (electron spin resonance) of spin labels is one of the best techniques to monitor structural properties at the molecular level, the alterations caused by the anthraquinone barbaloin on phospholipid bilayers will be discussed here via the ESR signal of phospholipid spin probes intercalated into the membranes. In DMPG at high ionic strength (10 mM Hepes pH 7.4 + 100 mM NaCl), a system that presents a gel-fluid transition around 23 degrees C, 20 mol % barbaloin turns the gel phase more rigid, does not alter much the fluid phase packing, but makes the lipid thermal transition less sharp. However, in a low-salt DMPG dispersion (10 mM Hepes pH 7.4 + 2 mM NaCl), which presents a rather complex gel-fluid thermal transition (Lamy-Freund, M. T.; Riske, K. A. Chem. Phys. Lipids 2003, 122, 19), barbaloin strongly affects bilayer structural properties, both in the gel and fluid phases, extending the transition region to much higher temperature values. The position of barbaloin in DMPG bilayers will be discussed on the basis of ESR results, in parallel with data from sample viscosity, DSC (differential scanning calorimetry), and SAXS (small-angle X-ray scattering).  相似文献

The role of phospholipid bilayers in controlling and reducing frictional forces between biological surfaces is investigated by three complementary experiments: friction forces are measured using a homemade tribometer, mechanical resistance to indentation is measured by AFM, and lipid bilayer degradation is controlled in situ during friction testing using fluorescence microscopy. DPPC lipid bilayers in the solid phase generate friction coefficients as low as 0.002 (comparable to that found for cartilage) that are stable through time. DOPC bilayers formed by the vesicle fusion method or the adsorption of mixed micelles generate higher friction coefficients. These coefficients increased through time, during which the bilayers degraded. The friction coefficient is correlated with the force needed to penetrate the bilayer with the AFM tip. With only one bilayer in the contact region, the friction increased to a similar value of about 0.08 for the DPPC and DOPC. Our study therefore shows that good mechanical stability of the bilayers is essential and suggests that the low friction coefficient is ensured by the hydration layers between adjacent lipid bilayers.  相似文献

We have used small-angle X-ray scattering and calorimetric methods to investigate the temperature-dependent phase behavior of ternary systems of phospholipid (DMPC), amphiphilic PEO-PPO-PEO block copolymer (Pluronics P85), and water. It is shown that a relatively small amount of block copolymers ( approximately 3 wt %) results in a lamellar-to-cubic phase transition. Still, both the bilayer-characteristic main transition, associated with chain melting, and the pretransition, associated with in-plane modulations, are preserved for copolymer concentrations up to 50-70 wt %, indicating the preservation of a bilayer type of lipid organization also within the cubic phase. The main transition splits up into two transitions upon the addition of copolymers, one resembling the high cooperativity of the main transition and one broad transition which may reflect complex formation with the copolymers. Parallel studies incorporating poly(ethylene glycol) into the DMPC multilamellar vesicles do not give analogous structural changes. It is concluded that the major effect on the molecular scale of adding PEO-PPO-PEO block copolymers is not only due to the hydration of the membrane but also due to the incorporation of the PPO block into the bilayer structure.  相似文献

A simple method for modifying a polymer surface to induce lipid bilayer formation by vesicle fusion is described. A silicate gel was prepared by condensation of tetraethyl orthosilicate (TEOS) in the presence of acid. When applied to a poly(methylmethacrylate) substrate, either a rough or a smooth layer could be produced, depending on the method used for the application. The smooth surface induced formation of a supported lipid bilayer by fusion of lipid vesicles; the rough silicate surface induced adsorption of a vesicle layer. A high-frequency acoustic waveguide device was used to follow the initial adsorption of vesicles, the transition from a vesicle layer to a bilayer, and the formation of a complete bilayer; the time required to form a bilayer was determined as a function of lipid concentration in suspension. The presence of a bilayer on the smooth silicate surface was confirmed by fluorescence recovery after photobleaching. An additional procedure is described to modify a gold surface to induce bilayer formation.  相似文献

We have investigated the physical properties of proton conducting polymer membranes based on a protic ionic liquid (IL). Properties such as ionic conductivity, melting point of the polymer phase, and glass transition temperature of the liquid phase are studied as a function of IL/polymer ratio and temperature. We observe an increased thermomechanical stability of the membrane with increasing polymer content. However, there is a concomitant decrease in the conductivity with increasing polymer content. This decrease is larger than what can be expected from the dilution of the conducting IL by the insulating polymer matrix. The origin of this decrease can be caused both by the morphology of the membrane and by interactions between the polymer matrix and the ionic liquid. We find a change in the glass transition temperature and in the temperature dependence of the conductivity with increasing polymer content. Both effects can be related to the physical confinement of the IL in the polymer membrane.  相似文献

Earlier work showed that heating causes poly(diethylsiloxane) to undergo a first-order transition from a semicrystalline solid to a more mobile viscous—crystalline material. The latter is composed of two phases and analogies between polymer and liquid crystal morphology and behavior have been made. The viscous—crystalline phase in PDES appears to be unique since the literature is devoid of other documented examples. In this study, spin—lattice and spin—spin relaxation times were measured over a wide temperature range. They show a glass transition at 138°K, a crystal—crystal transition at 206°K, and a transition around 250°K which results from translational motion of the polymer chains with respect to each other. This motion is observed in the amorphous phase at a lower temperature than in the crystalline phase. Translational motion in the crystalline phase is observed on melting of the crystallites. The spin—spin data permitted monitoring of the molecular motions in each phase and the data suggest that these phases exert some influence on the molecular motions of each other. The viscous—crystalline phase in PDES may represent a unique model for studying and understanding “precrystalline” behavior and structure in amorphous solids.  相似文献
