Pulps obtained from the ethanol/water cooking of sugarcane bagasse were bleached with the xylanase enzyme obtained from the fungus Thermomyces lanuginosus IOC-4145 and with the commercial enzyme Cartazyme HS from Sandoz. By changing the enzyme dose from 4.3 to 36 IU/g of pulp, kappa number and viscosity were maintained when the xylanase from T. lanuginosus was used. On the other hand, by using Cartazyme HS, kappa number decreased by 17%, reaching 35.5. This pulp was further extracted with NaOH without a decrease in viscosity (10 cP), and pulp with a kappa number of 13 was obtained. Xylanases had no significant effect on the ethanol/water pulps.  相似文献

The possibility of using two by-products of the sugar cane industry, molasses and bagasse steam explosion liquor (SEL), for lignin peroxidase (LiP) production by Phanerochaete chrysosporium was investigated. For comparison, the fungus was initially cultivated in synthetic media containing either glucose, sucrose, xylose, or xylan as sole carbon sources. The effect of veratryl alcohol (VA) was also investigated in relation to the enzyme activity levels. Results showed that sucrose was not metabolized by this fungus, which precluded the use of molasses as a carbon source. Glucose, xylose, and xylan promoted equivalent cell growth. Enzyme levels in the absence of VA were lower than 28 UI/L and in the presence of VA reached 109 IU/L with glucose and 85 IU/L with xylose or xylan. SEL was adequate for P. chrysosporium LiP production as LiP activity reached 90 IU/L. When VA was added to this medium, enzyme concentration increased to 155 IU/L.  相似文献

Three strains of the white-rot fungus Panus tigrinus (FTPT-4741, FTPT-4742, and FTPT-4745) were cultivated on sugarcane bagasse prior to kraft pulping. Pulp yields, kappa number, and viscosity of all pulps were determined and Fourier transform infrared (FTIR) spectra from the samples were recorded. The growth of P. tigrinus strains in plastic bags increased the manganese peroxide and xylanase activities. Lignin peroxidase was not detected in the three systems (shaken and nonshaken flasks and plastic bags). FTIR spectra were reduced to their principal components, and a clear separation between FTPT-4742 and the control was observed. Strain FTPT-4745 decayed lignin more selectively in the three systems utilized. Yields of kraft pulping were low, ranging from 20 to 45% for the plastic bag samples and from 12 to 38% for the flask samples. Kappa numbers were 1–18 and viscosity ranged from 2.3 to 6.8 cP.  相似文献

The formation and decay of lignolytic enzymes, along with the generation of other extracellular metabolites in submerged cultures ofPhanerochaete chrysosporium, were studied under different physiological conditions. Whereas lignin peroxidase (LiP) was detectable only in a narrow range of O2 tension and nitrogen concentration, manganese peroxidase (MnP) reached considerable levels over a broad range. The decay of LiP and MnP activities under lignolytic conditions paralleled that of heme and total proteins. The conditions that decrease or suppress LiP or MnP activities resulted in high levels of extracellular protease activity and/or polysaccharides.  相似文献

Brewer’s spent grain (BSG) was evaluated for bleached pulp production. Two cellulose pulps with different chemical compositions were produced by soda pulping: one from the original raw material and the other from material pretreated by dilute acid. Both of them were bleached by a totally chlorine-free sequence performed in three stages, using 5% hydrogen peroxide in the two initial, and a 0.25 N NaOH solution in the last one. Chemical composition, kappa number, viscosity, brightness and yield of bleached and unbleached pulps were evaluated. The high hemicellulose (28.4% w/w) and extractives (5.8% w/w) contents in original BSG affected the pulping and bleaching processes. However, soda pulping of acid pretreated BSG gave a cellulose-rich pulp (90.4% w/w) with low hemicellulose and extractives contents (7.9% w/w and <3.4% w/w, respectively), which was easily bleached achieving a kappa number of 11.21, viscosity of 3.12 cp, brightness of 71.3%, cellulose content of 95.7% w/w, and residual lignin of 3.4% w/w. Alkaline and oxidative delignification of acid pretreated BSG was found as an attractive approach for producing high-purity, chlorine-free cellulose pulp.  相似文献

Lignocellulolytic enzyme activities of selective fungi Daedalea flavida MTCC 145 (DF-2), Phlebia radiata MTCC 2791 (PR), and non-selective fungus Flavodon flavus MTCC 168 (FF) were studied for pretreatment of cotton stalks. Simultaneous productions of high LiP and laccase activities by DF-2 during early phase of growth were effective for lignin degradation 27.83 ± 1.25 % (w/w of lignin) in 20-day pretreatment. Production of high MnP activity without laccase in the early growth phase of PR was ineffective and delayed lignin degradation 24.93 ± 1.53 % in 25 days due to laccase production at later phase. With no LiP activity, low activities of MnP and laccase by FF yielded poor lignin degradation 15.09 ± 0.6 % in 20 days. Xylanase was predominant cellulolytic enzyme produced by DF-2, resulting hemicellulose as main carbon and energy source with 83 % of cellulose recovery after 40 days of pretreatment. The glucose yield improved more than two fold from 20-day DF-2 pretreated cotton stalks after enzymatic saccharification.  相似文献

Pulps obtained from ethanol/water cooking of sugarcane bagasse were treated at different times using xylanase enzyme obtained from Thermomyces lanuginosus IOC-4145 or commercially (Cartazyme HS, Sandoz Products Ltd.). The enzyme dosage was 18 IU per g of dry pulp and the time varied from 4 h to 12 h. When xylanase from T. lanuginosus was used, the kappa number and viscosity improved independently of the processing time used (4 h, 8 h, and 12 h). After chemical evaluation, the obtained pulps were classified using Fourier Transformed Infra-Red Spectroscopy and Principal Component Analysis. The results showed that the first three principal components explained more than 90 % of the total variance of the pulp spectra.  相似文献

Sugarcane bagasse was subjected to lime (calcium hydroxide) pretreatment and enzymatic hydrolysis for second-generation ethanol production. A central composite factorial design was performed to determine the best combination of pretreatment time, temperature, and lime loading, as well as to evaluate the influence of enzymatic loadings on hydrolysis conversion. The influence of increasing solids loading in the pretreatment and enzymatic hydrolysis stages was also determined. The hydrolysate was fermented using Saccharomyces cerevisiae in batch and continuous mode. In the continuous fermentation, the hydrolysates were concentrated with molasses. Lime pretreatment significantly increased the enzymatic digestibility of sugarcane bagasse without the need for prior particle size reduction. In the optimal pretreatment conditions (90 h, 90 °C, 0.47 g?lime/g bagasse) and industrially realistic conditions of hydrolysis (12.7 FPU/g of cellulase and 7.3 CBU/g of β-glucosidase), 139.6 kg?lignin/ton raw bagasse and 126.0 kg hemicellulose in the pretreatment liquor per ton raw bagasse were obtained. The hydrolysate from lime pretreated sugarcane bagasse presented low amounts of inhibitors, leading to ethanol yield of 164.1 kg?ethanol/ton raw bagasse.  相似文献

Development, characterization, and activity studies of nano-assemblies of lignin peroxidase (LiP), and manganese peroxidase (MnP) from Phanerochaete chrysosporium on flat surfaces as well as colloidal particles have been investigated. These assemblies of LiP and MnP were fabricated with polyelectrolytes-poly(ethylenimine) (PEI), poly(dimethyldiallylammonium chloride) (PDDA), and poly(allylamine) (PAH)-using a layer-by-layer self-assembly technique (LbL). Characterization of these assemblies on flat surfaces was monitored using quartz crystal microbalance (QCM), while assemblies on microparticles such as melamine formaldehyde (MF) were carried out with zeta potential analyzer (ZPA). A unique dynamic adsorption-desorption of the enzyme layers is observed during the assembly. All the nano-assemblies of LiP and MnP can effectively oxidize veratryl alcohol (VA) to its aldehyde for an extended period of time. The effect of different polyions and the number of polyion layers on the activities of LiP and MnP nano-assembly was also examined. It is observed that drying of enzyme layer during the assembly and the use of non-aqueous media, such as acetone can significantly reduce the activity of the enzymes. Enzyme activity reaches a minimum when the concentration of acetone is increased to 30%; however, the activity can be restored to its original value by increasing the concentration of aqueous media. Preliminary studies using assemblies of LiP and MnP on MF microparticles further demonstrate the feasibility of developing potential systems for degradation of environmental pollutants.  相似文献

Biobleaching studies using laccase mediator system (LMS) were carried out, under optimized conditions, on two unbleached Eucalyptus globulus kraft pulps, one produced by conventional way, with kappa number of 16.1, and another with kappa number of 14.5, obtained by modified kraft procedure with a high liquor/wood ratio and with black liquor replacement in the middle of the cooking. The pulp properties before and after LMS and alkaline extraction were evaluated in terms of kappa number, hexeneuronic acid content, viscosity, brightness and acid insoluble lignin content.The original milled wood sample and the kraft pulps were characterized by pyrolysis-gas chromatography/mass spectrometry (Py-GC/MS) and thermogravimetry/mass spectrometry (TG/MS). Eucalypt wood lignin produces guaiacol and syringol derivatives during pyrolysis. These lignin products can be detected with high sensitivity using the selected ion chromatograms even in the bleached pulp of low lignin content (about 0.5%). Py-GC/MS revealed that the lignin moieties were similarly altered during biobleaching as during pulping, which is exemplified by the preferential removal of aldehyde groups from the alkyl side groups. Semi-quantitative analysis of the pyrograms indicates that the lignin content of the biobleached pulps is reduced by about half in comparison with the unbleached pulps. The TG/MS results show that the hemicellulose content of wood was strongly modified during pulping resulting in higher thermal stability.  相似文献

Agricultural residues as sugarcane straw and bagasse are burned in boilers for generation of energy in sugar and alcohol industries. However, excess of those by-products could be used to obtain products with higher value. Pulping process generates cellulosic pulps and lignin. The lignin could be oxidized and applied in effluent treatments for heavy metal removal. Oxidized lignin presents very strong chelating properties. Lignins from sugarcane straw and bagasse were obtained by ethanol–water pulping. Oxidation of lignins was carried out using acetic acid and Co/Mn/Br catalytical system at 50, 80, and 115 °C for 5 h. Kinetics of the reaction was accomplished by measuring the UV-visible region. Activation energy was calculated for lignins from sugarcane straw and bagasse (34.2 and 23.4 kJ mol−1, respectively). The first value indicates higher cross-linked formation. Fourier-transformed infrared spectroscopy data of samples collected during oxidation are very similar. Principal component analysis applied to spectra shows only slight structure modifications in lignins after oxidation reaction.  相似文献

The white rot fungus Phanerochaete chrysosporium has been identified to be an environmentally useful microorganism for the degradation of various hazardous pollutants, mainly because of its ligninolytic enzyme system, particularly the lignin peroxidase (LiP) secreted by the fungus. In the present work, the behavior of the fungus in liquid medium due to variation in physico-chemical parameters, i.e., glucose concentration, nitrogen concentration, agitation, etc., was studied. Increment of the initial concentration of glucose in the medium increases the biomass growth and LiP activity, when cultured under controlled conditions. The biomass growth and LiP activity by the fungus was modeled following stochastic approach. The behavior of growth and enzyme activity of the fungus observed from the model were found to be in agreement with the experiments qualitatively.  相似文献

The thermal stabilities of three annual plants (reed, kenaf and Glycyrrihiza glablra L.) are studied. Various types of pulps were prepared from reed stalks by employing the soda, kraft and soda-anthraquinone processes. Their thermal stabilities are reported.It is shown that the thermal stability measured thermogravimetrically depends, to some extent, on the amount of lignin remaining in the raw materials, i.e. the kappa number.The activation energy of the decomposition process and the decomposition temperatures of the pulps are also reported.  相似文献

The pretreatment of sugarcane bagasse with lime (calcium hydroxide) is evaluated. The effect of lime pretreatment on digestibility was studied through analyses using central composite design (response surface), considering pretreatment time, temperature, and lime loading as factors. The responses evaluated were the yield of glucose from pretreated bagasse after enzymatic hydrolysis. Experiments were performed using the bagasse as it comes from an alcohol/sugar factory (non-screened bagasse) and bagasse in the size range from 0.248 to 1.397 mm (screened bagasse) (12-60 mesh). It was observed that the particle size presented influence in the release of fermentable sugars after enzymatic hydrolysis using low loading of cellulase and β-glucosidase (3.5 FPU/g dry pretreated biomass and 1.0 IU/g dry pretreated biomass, respectively).  相似文献

Sugarcane bagasse and straw can be converted into pulps, oils, controlled-release formulations, chelating agents, and composites. This article reviews bagasse and straw conversion efforts in Brazil. Laboratory-scale processes were developed aiming at the integral use of these biomass byproducts. Organosolv pulping and oxidation of lignin are the most promising processes for the rational use of sugarcane residues. Fungal pretreatment and spectroscopic characterization are also discussed.  相似文献

Organosolv (ethanol/water and acetosolv) pulps were treated with Bacillus pumilus xylanase for 4, 8, and 12 h and compared with commercial Cartazyme HS xylanase-treated pulps. Treatment of ethanol/water pulps with B. pumilus xylanase increased viscosity by 40% in 8 h of treatment compared with pulps treated without enzyme. However, acetosolv pulps treated with B. pumilus xylanase lost viscosity. Ethanol/water pulps treated with Cartazyme had a viscosity of 18.5 cP in 4 h of treatment. In the acetosolv pulps treated with commercial enzyme, the loss of viscosity was 20% compared with pulps treated without enzyme. Ethanol/water pulps treated with B. pumilus and Cartazyme had similar effects: a 44% reduction in kappa number for pulps treated with enzyme followed by alkaline extraction compared with pulps treated with alkaline extraction. In acetosolv pulps treated with B. pumilus, the kappa number was from 12 to 18, compared with pulps treated without enzyme, which had a 40% reduction in 4 and 12 h and a 60% reduction in 8 h. Cartazyme-treated acetosolv pulps had a kappa number of 14 in 4 and 8 h of treatment. For 12 h of treatment, the kappa number was 8. Fourier transform infrared spectra of the pulps showed that enzyme-treated pulps had changes in the 1000 cm?1 absorption owing to a C-O bond present in esters. Using principal component analysis, it is possible to differentiate the unbleached pulps and enzyme-treated pulps.  相似文献

Ionic liquid 1-ethyl-3-methylimidazolium acetate ([EMIM]oAc) was employed for the pretreatment of sugarcane bagasse (SCB) and extraction of lignin, a potentially valuable by-product of the biofuel industry. Response surface methodology based on central composite design was exploited and thereby an empirical model, exhibiting a coefficient of determination, R2, of 0.9890, was established to optimize lignin recovery. In particular, a maximum lignin yield, equal to 90.1%, was calculated at the optimal pretreatment conditions, namely time: 120 min, temperature: 140 °C, and ionic liquid to bagasse ratio equal to 20:1 (wt/wt). The presence of guaiacyl and syringyl rings in lignin was confirmed by Fourier transform infrared spectroscopy (FTIR); whereas UV–Vis spectrophotometry showed that both p-coumaric acid and ferulic acid were contained in the lignin. Thermal analysis indicated a maximum decomposition rate of 2%/°C at 265 °C while Gel permeation chromatography analysis revealed that the molecular weight (Mw) of recovered lignin was equal to 1769 g/mol. Comparison of FTIR spectra of pretreated and untreated bagasse showed a negligible presence of lignin in the pretreated samples. Maximum delignification of bagasse after pretreatment was thus ensured. Thermal stability of the ionic liquid towards recyclability was proven by thermogravimetric analysis. The present study established adequate performance of neat and recycled ([EMIM]oAc) with regard to lignin recovery from SCB.  相似文献

This study centred on the analysis of lignin in situ of cloned eucalypt and pine kraft pulps. Trametes versicolor laccase-violuric acid system (LMS) delignifications were performed on a softwood (Pinus pinaster) and a hardwood (Eucalyptus globulus) conventional kraft pulp with an initial kappa number of 34.5 and 15.5, respectively. The LMS treated pulps were then subjected to alkaline extraction stages (E). The kappa number data show that LMS is effective at biodelignifying both softwood and hardwood kraft pulps. However, under the conditions employed in this study, a greater level of biodelignification was obtained with LMS E. globulus (hardwood) than with LMS P. pinaster (softwood), but the amount of lignin removed was higher for the softwood pulp. The original milled wood samples, kraft pulps, biodelignified kraft pulps, and isolated residual lignin and milled wood lignins from the two wood samples have been characterized by pyrolysis-gas chromatography/mass spectrometry. The analysis of the pyrograms indicates that the lignin compositions of the two wood species and corresponding pulps are very different, as expected; however, the knowledge of the chemical mechanisms of delignification is very limited and requires additional work. Analytical pyrolysis is one the few degradative methods for the analysis of biopolymers that has shown a sufficient degree of success.  相似文献

Marine-derived fungi are prone to produce structurally unique secondary metabolites, a considerable number of which display the promising biological properties and/or industrial applications. Among those, ligninolytic enzymes have attracted great interest in recent years. In this work, about 20 strains were isolated from sea mud samples collected in the East China Sea and then screened for their capacity to produce lignin-degrading enzymes. The results showed that a strain, named J63, had a great potential to secrete a considerable amount of laccase. Using molecular method, it was identified as an endophytic fungus, Pestalotiopsis sp. which was rarely reported as ligninolytic enzyme producer in the literature. The production of laccase by Pestalotiopsis sp. J63 was investigated under submerged fermentation (SF) and solid state fermentation (SSF) with various lignocellulosic by-products as substrates. The SSF of rice straw powder accumulated the highest level of laccase activity (10,700 IU/g substrate), whereas the SF of untreated sugarcane bagasse provided the maximum amount of laccase activity (2,000 IU/ml). The value was far higher than those reported by other reports. In addition, it produced 0.11 U/ml cellulase when alkaline-pretreated sugarcane bagasse was used as growth substrate under SF. Meanwhile, the growth of fungi and laccase production under different salinity conditions were also studied. It appeared to be a moderately halo-tolerant organism.  相似文献
