The equilibrium constants (K) for the inclusion complex formationof -cyclodextrin (-CD) with Methyl Orange (MO) and substituted azoanilinium chlorides were determinedspectrophotometrically. Based on the results, the substituent effecton the inclusion complexation of -CD with azoanilinium chlorides was discussed indetail. Further, the solvent effects on the inclusion complexation of MO with -CD andheptakis(2,6-di-O-methyl)--cyclodextrin (DM--CD) wereexamined in aqueous organic mixtures with water-miscible organic compounds(dimethylsulfoxide, acetonitrile, N,N-dimethylformamide, and acetone). It was found that the K value for the inclusion complexformation with -CD and DM--CD decreases remarkably with increasing ratioof organic solvents, dependent of the surface tension of solvent mixtures.  相似文献

The possibility of the formation of inclusion complexes based on methyl-4-isobutylphenyl ketone, -cyclodextrin (-CD), and o-hydroxy propylated -cyclodextrin in an aqueous phosphate buffer is demonstrated by methods of NMR spectroscopy and cyclic voltammetry. Preparative electroreduction of acetophenone and methyl-4-isobutylphenyl ketone is performed on a lead electrode in aqueous phosphate and acetate buffers in the absence and presence of -CD and the composition of products of electrochemical reactions is ascertained. It is shown that the products of reduction of ketones are corresponding alcohols and pinacones. The possibility of asymmetric electrochemical hydrogenation of ketones in the presence of -CD, which yields relevant chiral alcohols, is shown.  相似文献

In this work the potential of MIMS (Membrane Introduction Mass Spectrometry) for studying the inclusion of small organic guest molecules into cyclodextrin hosts in aqueous medium was investigated. MIMS profiles showed that the inclusion of benzene in cyclodextrins is favored in the following order: -CD HO-propyl--CD> -CD> -CD with equilibrium constants of K -CD = 404; KHO-propyl-CD = 395, K -CD = 335 and K -CD = 210 M-1 at 25 °C. Kinetic experiments suggested that under the conditions employed the inclusion process has a pseudo first-order dependence on the guest benzene concentration with the following order: -CD > -CD HO-propyl--CD> -CD. MIMS inclusion profiles of chlorobenzene and toluene showed that the presence of substituents in benzene makes the inclusion in -CD more difficult. Experiments with ferrocene--CD have also been carried out, showing that the complex rapidly dissociates in water and the resulting free -CD can complex with benzene present in the solution.  相似文献

The inclusion complex of the anti-inflammatory drug, loxoprofen, with -cyclodextrin-(CD), sulfated -CD, and glycerol ether -CD was studied by UV-VIS absorption and 1H-NMR spectroscopy in solution. The inclusion complex of loxoprofen with -CDs was prepared by freeze-drying, and then characterized in the solid state by thermal analysis, X-ray diffraction, FT-IR and FT-Raman spectroscopy, and scanning electron microscopy (SEM). Furthermore, a physical mixture of loxoprofen/-CD (1/1, mol-%) in the solid state was also characterized. The solubility of the loxoprofen increased on addition of -CDs. The solubility enhancement of the loxoprofen with -CDs is in the following order: glycerol ether -CD > sulfated -CD > -CD.  相似文献

Effects of Cyclodextrins on Deodoration of ``Aging Odor'   总被引:2,自引:0,他引:2  
Hexenal, octenal and nonenal are known causes of unpleasant body odor and are present at markedly increased levels in the middle-aged and elderly. The odor of such unsaturated aldehydes is therefore called ``aging odor'. The present study investigated the effects of cyclodextrins (CDs) on deodoration of crotonaldehyde, pentenal, hexenal, heptenal, octenal, nonenal, decenal, undecenal and dodecenal. -, - and -CD formed inclusion complexes with the majority of unsaturated aldehydes in aqueous solution. The -CD inclusion complex contained the highest amount of guest molecule. One molecule of -CD was estimated to include 1 molecule of short chain aldehyde and 2 molecules of long chain aldehyde. Deodorant testing was conducted by headspace gas analysis using sealed vials. All CDs decreased the concentrations of unsaturated aldehyde. With nonenal, the deodorant power of parent CDs was -CD > -CD > -CD, and that of chemically modified CD was Me-CD > HP-CD > G2-CD > MCT-CD. CDs were demonstrated to reduce ``aging odor'. Me-CD was the most effective type of CD for the deodoration of ``aging odor'.  相似文献

Inclusion compounds of ferrocene(FcH) and its derivatives with cyclodextrins(CDs; -CD, -CD, and -CD) were prepared. CD-ferrocene inclusion compounds were obtained in a crystalline state in high yield. -CD and -CD formed 11 stoichiometric inclusion compounds with ferrocene and its derivatives. -CD formed 21 (CD:guest) complexes with ferrocene and the monosubstituted derivatives, but did not form complexes with 1,1-disubstituted derivatives, -CD-FcH and -CD-FcH complexes are thermally stable and do not liberate ferrocene on heating at 100°C in vacuo. The cyclodextrin inclusion compounds were characterized by1H-NMR, IR, UV, and CD spectra. A large positive induced Cotton effect was observed in the case of -CD-FcH complex, while the -CD-FcH complex showed a negative spectrum. The binding mode is discussed. -Cyclodextrin was found to form inclusion complexes in ethylene glycol, diethylene glycol, triethylene glycol, methyl cellosolve, ethyl cellosolve, methyl alcohol, and glycerine solutions. -CD also formed complexes in ethylene glycol solution. The binding of ferrocene by -CD is stronger in ethylene glycol than in dimethyl sulfoxide and dimethyl formamide.  相似文献

Summary Two -cyclodextrin (-CD)-containing polymers have been prepared either by condensation of -CD molecules with a bifunctional reagent or by grafting a -CD derivative on to a linear polymer (polyvinylimidazole). HPLC stationary phases were obtained by adsorption of the -CD polymers on to silica. The ability of these chromatographic supports to resolve racemic mixtures of organic compounds such as amino acid derivatives, phenylhydantoins, barbiturates, and hydroxycoumarin derivatives has been investigated. In an effort to modify the solubility and dissolution rate of the contraceptive steroid, 19-norprogesterone in order to improve its bioavailability, the cyclodextrin complexation approach was chosen. In solution, the complex formation with -cyclodextrin (-CD), hydroxyethyl -cyclodextrin (HE--CD) and hydroxypropyl -cyclodextrin (HP--CD) was confirmed by using solubility, UV, IR and 1H-NMR spectrophotometric techniques. The phase solubility diagrams were categorized as AL-type. The complexing affinity of the CDs investigated were ranked as follows: -CD > HP--CD > HE--CD. The complexation thermodynamic parameters were obtained from the temperature dependence of the dissociation constants. In the solid state, differential scanning calorimetery (DSC) and optical microscopy methods were utilized to characterize the complexes. Dissolution studies showed that such molecularly encapsulated forms offered a marked improvement in the dissolution rate compared to the parent drug.  相似文献

The interactions of nabumetone (NAB) with -cyclodextrin (-CD) and-cyclodextrin (-CD) were studied in aqueous solution by meansof phase-solubility analysis. Solid dispersions of NAB with -cyclodextrin(-CD), -cyclodextrin (-CD), methyl- (M-CD),hydroxypropyl-cyclodextrin (HP-CD) were prepared by coevaporationand kneading and also by coprecipitation in the case of -CD. X-ray diffractometry, thermal analysis and infrared spectroscopy (FTIR) were used to study the possibility of complexation of the drug with the different cyclodextrins. Solid dispersions of nabumetone with -CD showed a remarkable improvement in the dissolution rate of nabumetone.  相似文献

The solubility of nimodipine was measured in aqueous solutions of the following cyclodextrins: -cyclodextrin (-CD), hydroxypropyl--CD (HP--CD), -cyclodextrin (-CD), random substituted methyl--CD (M--CD), three hydroxypropyl--CDs (HP--CD) with mutually different average degree of substitution, and hydroxypropyl--cyclodextrin (HP--CD). From the determined linear solubility diagrams the values of the binding constant K11 of the inclusion complexes of nimodipine with the respective CDs were evaluated. The -CDs efficiently solubilized sparingly soluble nimodipine, the highest value of K11 was found for M--CD (1680 M-1), followed by -CD (550 M-1) and HP--CDs, where the higher degree of substitution lowered K11. Only slight solubilization of nimodipine was observed in the solutions of the -CDs and HP--CD.  相似文献

The effect of -cyclodextrin (-CD) on aqueous hydrolysis of methyl,p-, andm-nitrophenyl salicylates as well as on the Smiles rearrangement ofp-nitrophenyl salicylate was studied. No effect of -CD on the pH-independent rate constant of aqueous hydrolysis of methyl ester was observed, while -CD accelerated aqueous hydrolysis of nitrophenyl esters byca. 10 times. The inclusion of these esters into the cavity of -CD is accompanied by a change in the mechanism of hydrolysis: free ester in the deprotonated form undergoes hydrolysis through the mechanism of intramolecular general base catalysis, while the ester bound to cyclodextrin is hydrolyzed due to the nucleophilic attack of the deprotonated hydroxyl group of -CD at a neutral substrate molecule. The effects of cyclodextrin on the rate constant of borate-catalyzed hydrolysis were interpreted by assuming that the substrate bound to -CD undergoes borate-assisted attack at the deprotonated cyclodextrin hydroxyl group. Aim of the present work was to investigate the effect of somecyclodextrins (CDs) on the solubility and ocular bioavailability of rufloxacin base (RUF), with theultimate goal of developing an ophthalmic formulation. Phase solubility studies of RUF inpH 7.4 buffer were carried out in the presence of -cyclodextrin (-CD),hydroxypropyl--cyclodextrin (HP--CD) and -cyclodextrin(-CD). The effect of hydroxypropyl methylcellulose (HPMC) on RUF solubility was evaluated after heating the solutionscontaining HP--CD at 120 °C.A significant enhancement of RUF solubility was achieved by associatingthe drug with CDs, particularly HP--CD. This CD formed with RUF a less stablecomplex than that formed by -CD, but did not suffer the solubility limitations ofthe parent CD, and showed a higher solubilizing capacity than -CD. Addition of 0.25%(w/v) HPMC to solutions containing HP--CD increased the solubilizing effect of this CD,thus allowing reduction of the amount necessary for solubilization of 0.3% (w/v) RUF.Preliminary pharmacokinetic data in rabbits indicated that theocular bioavailability of 0.3% (w/v) RUF solubilized by HP--CD was higher when compared witha 0.3% (w/v) RUF suspension used as reference.  相似文献

Inclusion complexation of all-trans-retinol, retinal and retinoic acid with -cyclodextrin (-CD) and heptakis(2,6-O-dimethyl)--cyclodextrin (DM--CD) were investigated by means of UV-vis spectroscopy. The association constants (K a) obtained for vitamin A with DM--CD is greater than with -CD. Studies by electron paramagnetic resonance (EPR), differential scanning calorimetry, thermogravimetric analysis, HPLC and NMR showed that radicals produced by thermolysis and photolysis of benzoyl peroxide,t-butyl peroxide and cumene hydroperoxide included in-cyclodextrin (-CD), undergo significant reaction with the-CD. The formation of-CD radicals was observed by EPR. Products formed by addition of radicals to-CD were also observed. Such host:guest radical reactions explain the reported stabilization of peroxides, found with-CD inclusion, as being primarily due to the interruption of chain reactions by trapping of the chain carriers. A small increase in activation barrier for cleavage of the included peroxide in-CD was also observed.  相似文献

The interactions between cyclodextrins and tetrahydroharmane, 1-methyl-1,2,3,4-tetrahydro-9H-pyrido/3,4-b/indole, have been investigated in pH 10.1 aqueous media. Absorption and fluorescence studies, together with lifetime measurements show that tetrahydroharmane forms ground state inclusion complexes with -CD and -CD but not with -CD. Steady state experiments detect only one kind of tetrahydroharmane--CD complex, whereas time resolved measurements reveal the existence of two different types of 1:1 association complexes. The results in -CD are very similar to those in -CD, but the interactions are much weaker.  相似文献

Summary The twelve structural C2 isomers of naphthalene are resolved on a single capillary column using , , or cyclodextrin as the stationary phase. A change in elution order is seen upon going from to and then cyclodextrin which can be related to the size of the cyclodextrin cavity and the stereochemical fit of the isomer. A crown-ether column and a liquid-crystal column were found to be unable to separate all of the isomers.  相似文献

Nifluminic acid (N) is a frequently used anti-inflammatory drug,but its poor aqueous solubility is a disadvantageous property. Inclusion complexation with cyclodextrinderivatives (CDs) affords a possibility to increase its solubility properties. For thispurpose, different CDs were used, primarily hydroxypropyl--CD (HP--CD), to prepareproducts by powder mixing or kneading in four molecular ratios. The dissolution and in vitromembrane diffusion of the products were investigated. The wetting angles of pure N andHP--CD and of the products, and the n-octanol/water partition coefficients were determined.The interaction, leading to complex formation between the components of the products was examined by thermoanalytical methods.  相似文献

The uptake of -, -, -, hexakis-2,6-di-O-methyl--(DM--), heptakis-2,6-di-O-methyl--(DM--) and heptakis-2,3,6-tri-O-methyl-- (TM--) cyclodextrins (CD's) by Na-, Ca-, and Cu-montmorillonites has been studied at 25°C. Each of -, DM--, DM--, and TM--CD's forms a complex with all of these montmorillonites in which the guest molecules are arranged as a monolayer with their open faces parallel to the silicate sheet of montmorillonite. On the other hand, -CD is intercalated only by the Na form and -CD cannot be taken up by any of the clays used. The interlayer spacing of the -CD complex varies from 18.0 to 21.5 Å with replacement of the interlayer cation, but those of the methylated -and -CD complexes remain unchanged, beingca. 21 and 18 Å, respectively. The different behaviour of the parent and methylated CD's is explained in terms of the structural and physicochemical characteristics of the individual CD's and interlayer cations.  相似文献

The formation of inclusion complexes between 4-amino-1,8-naphthalimides and cyclodextrins (CDs) was investigated. The naphthalimides used in the study were 4-amino-1,8-naphthalimide (I) and4-(2-phosphonoethylamino)-N-(2-phosphonoethyl)-1,8-naphthalimide,tetraethylester (II). The CDs employed were -CD, -CD, -CD, HP--CD, HP--CD andHP--CD (HP = hydroxypropyl). Evidence for complex formation was obtained from the changes in the fluorescence spectra of the dyes in the presence of increasing amounts of the CDs. The most striking changes were observed with HP--CD and HP--CD. Treatment of the data using Benesi–Hildebrand plotswas consistent with a 1:1 inclusion model. The determined stabilityconstants were (Keq, M-1): 106 (I:HP--CD, pH = 2.0), 193 (I:HP--CD, pH = 7.0), 113 (I:HP--CD, pH = 7.0), 155(II:HP--CD, pH = 2.0), 121 (II:HP--CD,pH = 7.0), 301 (II:HP--CD, pH = 7.0). It can beconcluded that compound I forms a more stable complex with HP--CD than with HP--CD. Compound II, on the other hand, forms a more stable complex with HP--CD than with HP--CD.  相似文献
