An ultrasensitive and simple method for the determination of melamine was developed based on the fluorescence quenching of thioglycolic acid (TGA) capped CdTe quantum dots (QDs) at pH 11.0. In strong alkaline aqueous solution, the selectivity of the method has been greatly improved due to most heavy metal ions show no interference as they are in the precipitation form or in their anion form. Furthermore, CdTe quantum dots have higher quantum yields at higher pH. The method has a wider concentration range and lower detection limit. The influence factors on the determination of melamine were investigated and the optimum conditions were determined. Under optimum conditions, the fluorescence intensity change of TGA coated CdTe quantum dots was linearly proportional to melamine over a concentration range from 1.0 × 10−11 to 1.0 × 10−5 mol L−1 with a correlation coefficient of 0.9943 and a detection limit of 5 × 10−12 mol L−1. The mechanism of fluorescence quenching of the QDs has been proposed based on the infrared spectroscopy information and electrophoresis experiments in presence of melamine under alkaline condition. The proposed method was employed to detect trace melamine in milk powder and pet feeds with satisfactory results.  相似文献

High-quality colloidal CdTe quantum wires having purposefully controlled diameters in the range 5-11 nm are grown by the solution-liquid-solid (SLS) method, using Bi nanoparticle catalysts, cadmium octadecylphosphonate and trioctylphosphine telluride as precursors, and a TOPO solvent. The wires adopt the wurtzite structure and grow along the [002] direction (parallel to the c axis). The size dependence of the effective band gaps in the wires is determined from the absorption spectra and compared to the experimental results for high-quality CdTe quantum dots. In contrast to the predictions of an effective-mass approximation, particle-in-a-box model, and previous experimental results from CdSe and InP dot-wire comparisons, the effective band gaps of CdTe dots and wires of like diameter are found to be experimentally indistinguishable. The present results are analyzed using density functional theory under the local-density approximation by implementing a charge-patching method. The higher-level theoretical analysis finds the general existence of a threshold diameter, above which dot and wire effective band gaps converge. The origin and magnitude of this threshold diameter are discussed.  相似文献

We have developed a novel method for the determination of iodate based on the carboxymethyl cellulose-capped CdS quantum dots (QDs). Factors affecting the iodate detection were investigated, and the optimum conditions were determined. Under the optimum conditions, the relative fluorescence intensity of CdS quantum dots was linearly proportional to IO3 over a concentration range from 1.0 × 10−8 to 1.0 × 10−5 mol L−1 with a correlation coefficient of 0.9987 and a detection limit of 6.0 nmol L−1. Iodide, being oxidized by bromine to form iodate, was detected indirectly. The method was successfully applied to the determination of iodate and total amount of iodine in table salt samples. Multicolor and water-soluble CdTe quantum dots (QDs) were synthesized with thioglycolic acid (TGA) as stabilizer. These QDs have a good size distribution, display high fluorescence quantum yield, and can be applied to the ultrasensitive detection of Pb(II) ion by virtue of their quenching effect. The size of the QDs exerts a strong effect on sensitivity, and quenching of luminescence is most effective for the smallest particles. The quenching mechanism is discussed. Fairly selective detection was accomplished by utilizing QDs with a diameter of 1.6?nm which resulted in a detection limit of 4.7?nmol?L?1 concentration of Pb(II). The method was successfully applied to the determination of Pb(II) in spinach and citrus leaves, and the results are in good agreement with those obtained with atomic absorption spectrometry.

Five colors water-soluble CdTe QDs are synthesized with thioglycolic acid as a stabilizer. These QDs can be applied to the ultrasensitive detection of Pb2+ by virtue of their quenching effect. The size of the QDs exerts a strong effect on sensitivity, and the quenching of luminescence is most effective when the smallest particles are used. CdS quantum dots (QDs) modified with l-cysteine has been prepared by one step. They are water-soluble and biocompatible. To improve CdS QDs stability and interaction between silver ion and functionalized CdS QDs in aqueous solution, some amounts of fresh l-cysteine were added to functionalized CdS solution. Based on the characteristic fluorescence enhancement of CdS QDs at 545 nm by silver ions in the presence of some amounts of fresh l-cysteine, simultaneously, a gradual red shift of fluorescence emission bands of CdS QDs from 545 to 558 nm was observed. A simple, rapid, sensitive and specific detection method for silver ion was proposed. Under optimum conditions, the fluorescence intensity of CdS QDs is linearly proportional to silver concentration from 2.0 × 10−8 to 1.0 × 10−6 mol/L with a detection limit of 5.0 × 10−9 mol/L. In comparison with single organic fluorophores, functionalized CdS quantum dots are brighter, more stable against photobleaching, and don’t suffer from blinking. Furthermore, owing to the fluorescence enhancement effect of CdS QDs by silver ion, the proposed method showed lower detection blank and higher sensitivity. A versatile nanoprobe was developed for trypsin quantification with fluorescence resonance energy transfer (FRET). Here, fluorescence graphene quantum dot is utilized as a donor while a well-designed coumarin derivative, CMR2, as an acceptor. Moreover, bovine serum albumin (BSA), as a protein model, is not only served as a linker for the FRET pair, but also a fluorescence enhancer of the quantum dots and CMR2. In the presence of trypsin, the FRET system would be destroyed when the BSA is digested by trypsin. Thus, the emission peak of the donor is regenerated and the ratio of emission peak of donor/emission peak of acceptor increased. By the ratiometric measurement of these two emission peaks, trypsin content could be determined. The detection limit of trypsin was found to be 0.7 μg/mL, which is 0.008-fold of the average trypsin level in acute pancreatitis patient's urine suggesting a high potential for fast and low cost clinical screening.  相似文献

This paper reports the construction of a simple CdTe quantum dots (QDs)-based sensor with 1,10-phenanthroline (Phen) as ligand, and the demonstration of a novel ligand displacement-induced fluorescence switch strategy for sensitive and selective detection of Cd2+ in aqueous phase. The complexation of Phen at the surface quenches the green photoluminescence (PL) of QDs dominated by a photoinduced hole transfer (PHT) mechanism. In the presence of Cd2+, the Phen ligands are readily detached from the surface of CdTe QDs, forming [Cd(Phen)2(H2O)2]2+ in solution, and as a consequence the PL of CdTe QDs switches on. The detection limit for Cd2+ is defined as ∼0.01 nM, which is far below the maximum Cd2+ residue limit of drinking water allowed by the U.S. Environmental Protection Agency (EPA). Two consecutive linear ranges allow a wide determination of Cd2+ from 0.02 nM to 0.6 μM. Importantly, this CdTe QDs-based sensor features to distinctly discriminate between Cd2+ and Zn2+, and succeeds in real water samples. This extremely simple strategy reported here represents an attempt for the development of fluorescent sensors for ultrasensitive chemo/biodetection.  相似文献

A sensitive and specific immunoassay method for detecting α-fetoprotein (AFP) based on electrogenerated chemiluminescence (ECL) was described. ECL could perform detection for a series of different concentrations of AFP. CdSe quantum dots (QDs) were used as labels and were linked to AFP antibody (anti-AFP, the secondary antibody, Ab2*). Immunoassay was carried out on a modified electrode using a sandwich assay approach, where anti-AFP (Ab1) was covalently bound to the surface of an Au electrode to be allowed to capture AFP specifically. Afterwards, Ab2* was allowed to bind selectively to the captured AFP. The non-specific adsorption was negligible. In the presence of H(2)O(2), the ECL intensity increased with the increase of AFP, which indicated that an immunosensor for AFP was constructed. The detection of AFP based on measuring the ECL intensity of CdSe without the enzyme and mediator can promote the stability of the immunosensor. The linear range of the AFP assay was from 0.002 to 32 ng mL(-1). Furthermore, the immunosensor showed high sensitivity, good precision, stability, and reproducibility and could be used for the detection of real samples with consistent results in comparison with those obtained by the enzyme-linked immunosorbent assay (ELISA) method. The strategy was successfully demonstrated as a simple, cost-effective, specific, and potential method to detect AFP in practical samples.  相似文献

A new sandwich-type electrochemical immunoassay was developed for the detection of human IgG using doubly-encoded and magnetic redox-active nanoparticles as recognition elements on the surface of a glassy carbon electrode modified with anti-IgG on nanogold particles. The recognition elements were synthesized by coating magnetic Fe3O4 nanoparticles with Prussian blue nanoparticles and then covered with peroxidase-labeled anti-IgG antibodies (POx-anti-IgG) on Prussian blue nanoparticles. The immunoelectrode displays very good electrochemical properties towards detection of IgG via using double-encoded magnetic redox-active nanoparticles as trace and hydrogen peroxide as enzyme substrate. Its limit of detection (10 pmol·L?1) is 10-fold better than that of using plain POx-anti-IgG secondary antibodies. The method was applied to the detection of IgG in serum samples, and an excellent correspondence with the reference values was found.  相似文献

A novel trace tag for chemiluminescent (CL) immunoassay was designed by using DNAzyme to functionalize antibody-labeled Au nanoparticles (AuNPs). The trace tag showed an excellent ability to catalyze the oxidation of luminol by hydrogen peroxide, leading to strong CL emission. By coupling the trace tag with a passive mixing accelerated immunoreaction system, a highly sensitive rapid flow-through CL immunoassay method was proposed. Using carcinoembryonic antigen (CEA) as a model analyte, the capture antibody for CEA was immobilized on paramagnetic microspheres, and DNAzyme-anti-CEA antibody functionalized AuNPs were prepared as trace tag. A three-dimensional helical glass tube kept at 37 °C in a water bath was used for passively mixing immunoreagents in a two-step sandwich immunoassay, with which each immunoreaction step could be finished within 150 s. With the help of a magnet, the immunocomplex could conveniently be separated from reactants. Compared with the horseradish peroxidase-based tag, the newly designed trace tag showed obvious signal amplification due to its strong catalytic ability and high loading ratio of DNAzyme on each AuNP. The proposed method showed a linear calibration range from 0.005 to 0.5 ng mL(-1) for CEA detection with a detection limit of 4.1 pg mL(-1) at a signal-to-noise ratio of 3 and acceptable detection reproducibility. The assay results of clinical serum samples were in acceptable agreement with the reference values. The designed immunoassay system with ultrahigh sensitivity provided a programmable and low-cost approach for high-throughput clinical application.  相似文献

In this work, a micro-plate chemiluminescence enzyme immunoassay by antibody-coated for the determination of aflatoxin B1 (AFB1) in agricultural products has been established. Aflatoxin B1 antibody (AFB1-Ab) was adsorbed physically on polystyrene micro-plate hole as solid phase antibody, which took place immunity-reaction between antigen and antibody with AFB1 standard solution or samples by direct competition. Luminol-hydrogen peroxide chemiluminescence system catalyzed by horseradish peroxidase (HRP) with p-iodophenol enhancement was used as signal detecting system. The effects of several factors, including composition and pH of coating solution, dilution ratio and amount of antibody and enzyme labeled antigen, time of antibody-coating, incubation and chemiluminescence reaction, and other relevant variables upon the immunoasaay were studied and optimized. The linear range of proposed method for AFB1 was 0.05-10.0 ng g−1 with a correlative coefficient of −0.9997. The sensitivity of the proposed method was 0.01 ng g−1. The RSDs of intra- and inter-assay were less than 12.2% and 10.0%, respectively. This method has been successfully applied to the evaluation of AFB1 in agricultural products with recoveries of 79.8%, 101.9% and 115.4% for low, middle and high concentration samples, respectively. It shows a good correlation with the commercial available ELISA kit for AFB1 with correlative coefficient of 0.9098 indicating that the established CLEIA method can be used to determine AFB1 in real samples.  相似文献

A simple procedure was developed for in vitro synthesis and characterization of aflatoxin B1-lysine adduct using aflatoxin B1, N-alpha-acetyl lysine and m-chloroperbenzoic acid (MCPBA). At a molar ratio of 1:16 (aflatoxin B1:N-alpha-cetyl lysine), the recovery of adduct was 62%. Analysis of the adduct by thin-layer chromatography showed a single spot (Rf = 0). Absorption spectra of the adduct showed 2 peaks at 275 and 335 nm. Liquid chromatographic (LC) analysis of the AFB1-lysine adduct showed a relative retention time of 2.1 min. Using the same epoxidation procedure, BSA-AFB1 adduct and ovalbumin-AFB1 adduct were synthesized for production of antibodies and as coating antigen, respectively. Control rat serum, spiked with AFB1-lysine adduct and subjected to LC analysis showed a retention time of 2.1 min, which is similar to that of AFB1-lysine reference standard, synthesized. Further, enzymatically hydrolyzed, control rat serum spiked with BSA-AFB1 adduct showed 2 peaks with retention times of 2.1 and 2.7 min. Based on the LC analysis, recovery of BSA-AFB1 in terms of AFB1-lysine adducts was 67 +/- 5%. The major peak (2.1 min) accounted for 72% of the adduct; the second minor peak (2.7 min) accounted for 28% of the total AFB1-lysine adducts formed. Stability studies on the AFB1-lysine adduct synthesized, indicated that it was stable for 1 month. Antibody capture assay showed an absorbance of 0.9 to 1.0 at a dilution of 1:50,000 when ovalbumin-AFB1 was used as a coating antigen. Indirect competitive ELISA showed 50% displacement (IC50) of the antibodies at a concentration of 13 ng AFB1-lysine, whereas the IC50 for AFB1 was 7 ng. The recovery of AFB1-lysine adduct spiked to control rat serum followed by enzymatic hydrolysis and immunoanalysis (indirect ELISA) was 93 +/- 6%. The enzyme immunoassay was validated by a rodent model, in which the animals were exposed to aflatoxin B1 (20 microg AFB1/kg body mass/day). The level of AFB1-lysine adduct in the rat serum was 27.3 +/- 4.37 microg/mg albumin.  相似文献
