We compare two methods, solid-phase extraction (SPE) and dialysis, commonly used for extraction and concentration of dissolved organic matter (DOM) prior to molecular characterization by electrospray ionization (ESI) and ultrahigh-resolution Fourier transform ion cyclotron resonance mass spectrometry. Spectra of DOM samples from Minnesota and Sweden peatlands that were extracted with styrene divinyl benzene polymer SPE sorbents included ions with formulas that had higher oxygen to carbon (O/C) ratios than spectra of DOM from the same samples after de-salting by dialysis. The SPE method was not very effective in extracting several major classes of DOM compounds that had high ESI efficiencies, including carboxylic acids and organo-sulfur compounds, and that out-competed other less-functionalized compounds (e.g., carbohydrates) for charge in the ESI source. The large abundance of carboxylic acids in the dialysisextracted DOM, likely the result of in situ microbial production, makes it difficult to see other (mainly hydrophilic) compounds with high O/C ratios. Our results indicate that, while dialysis is generally preferable for the isolation of DOM, for samples with high microbial inputs, the use of both isolation methods is recommended for a more accurate molecular representation.  相似文献

A rapid technique for the screening of trace compounds in water by combining solid-phase extraction (SPE) with activated carbon surface-assisted laser desorption/ionization (SALDI) time-of-flight mass spectrometry is demonstrated. Activated carbon is used both as the sorbent in SPE and as the solid in the SALDI matrix system. This eliminates the need for an SPE elution process. After the analytes have been adsorbed on the surfaces of the activated carbon during SPE extraction, the activated carbon is directly mixed with the SALDI liquid and mass spectrometric analysis is performed. Trace phenolic compounds in water were used to demonstrate the effectiveness of the method. The detection limit for these compounds is in the ppb to ppt range.  相似文献

SPE is a commonly applied technique for preconcentration of pesticides from water samples. Microwave‐assisted extraction (MAE) technique is the extraction applied for preconcentration of different compounds from solid samples. SPE coupled with MAE is capable of preconcentrating these compounds from water samples too. This investigation was aimed at improving the efficiency of atrazine, alachlor, and α‐cypermethrin pesticide extraction from the spiked water samples applying SPE followed by MAE. In this way, MAE served for elution of pesticides from C18‐extraction disks with solvent heated by microwave energy. Various elution conditions were tested for their effects on the extraction efficiency of the SPE–MAE combined technique. Several parameters, such as elution solvent volume (mL), elution temperature (°C), and duration of elution (min), affect the extraction efficiency of the SPE–MAE coupled system and need to be optimized for the selected pesticides. In order to develop a mathematical model, 15 experiments were performed in the central composite design. The equation was then used to predict recoveries of the pesticides under specific experimental conditions. Optimization of microwave extraction was accomplished using the genetic algorithm approach. Best results were achieved using 20 mL of ethanol at 60°C. Optimal hold time was 5 min and 24 s. The SPE–MAE combination was also compared with the conventional SPE extraction technique with elution of a nonpolar or a moderately polar compound with nonpolar solvents.  相似文献

A simple off-line solid-phase extraction (SPE) method for isolation of polychlorinated biphenyls (PCBs) and selected organochlorine pesticides (OCPs) from human serum has been developed. The procedure includes denaturation of serum proteins by a mixture of water-1-propanol, application of the sample by aspiration twice repeatedly through the SPE column and elution with a mixture of n-hexane-dichlormethane. After final clean-up the compounds of interest were analysed by gas chromatography with micro-electron capture detection (GC-microECD). The recoveries achieved for PCB congeners using spiked porcine serum samples were 99-120% and for OCPs 88-115%. Relative standard deviations (RSD) ranged from 3 to 7%. The method was applied to real human serum samples and the recoveries of analytes in the serum were proportionally recalculated considering the recovery of the internal standard PCB-174. PCB-103 served as a syringe standard to correct volume of samples analysed. The aim of this study was to develop an effective off-line SPE procedure by optimization of existing SPE methods to supply laborious, solvent- and time-consuming liquid-liquid extraction (LLE) in routine analytical process.  相似文献

固相萃取(SPE)是目前广泛采用的富集水中痕量多环芳烃(PAHs)的前处理方法,具有回收率高、检测限低、不乳化等优点.在操作过程中,提高回收率和控制背景干扰是SPE能否达到测试分析要求的关键技术.从样品的准备、C18柱的活化、样品的抽滤萃取、C18柱的干燥和洗脱、洗脱液的浓缩和保存等方面讨论了SPE富集水中痕量PAHs的影响因素和提高PAHs回收率的方法,以及如何消除背景干扰,并进行了回收率的验证实验.在14个流沙湾海水样品的测试中,除萘-d8外其它4种替代物的回收率均在60%~120%,相对标准偏差RSD≤10%,达到水中PAHs分析测试的要求.  相似文献

Solid phase extraction (SPE) using Sep-Pak? cartridges is one of the techniques used for fractionation of antioxidant compounds in waste of dabai oil extraction (defatted dabai parts). The aim of this study was to determine the phenolic compounds and antioxidant capacity in crude extracts and several SPE fractions from methanolic extract of defatted dabai pulp and peel. Based on SPE, Sep-Pak? cyanopropyl and C18 cartridges were used to fractionate the antioxidant-rich crude extracts into water and methanolic fractions. Analyzed using LC-MS, flavonoids, anthocyanins, saponin derivatives and other unknown antioxidative compounds were detected in the defatted dabai crude extracts and their SPE fractions. Anthocyanins were the major phenolic compounds identified in the defatted dabai peel and detected in most of the SPE fractions. Methanolic fractions of defatted dabai parts embraced higher total phenolics and antioxidant capacity than water fractions. This finding also revealed the crude extracts of defatted dabai peel have the most significant antioxidant properties compared to the methanolic and water fractions studied. The crude extract of defatted dabai parts remain as the most potent antioxidant as it contains mixture of flavonoids, anthocyanins and other potential antioxidants.  相似文献

Characterization of dissolved organic matter (DOM) at the molecular level will greatly improve our understanding of its bio-geochemical role in controlling the fate of contaminants in the environment, and Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR-MS) is the most powerful analytical technique for this purpose. Before FT-ICR-MS analysis, isolation, desalination and concentration of DOM are necessary, and solid-phase extraction (SPE) is the most widely applied pretreatment procedure. However, some molecular information is lost using conventional SPE methods. Here, we propose a novel strategy of SPE enrichment using stepwise elution (SPE-SE). Compounds in DOM were divided into three fractions by this SPE-SE procedure according to their polarity and ionization efficiency. The diversity of DOM molecules identified by ESI-FT-ICR-MS using SPE-SE exceeded those using conventional SPE methods by more than 50%. This method is feasible and has the potential to be used as a pretreatment strategy for complex DOM matrixes prior to ESI-FT-ICR-MS analysis, especially for those rich in nitrogenous molecules, carbohydrates, lipids and/or aromatic compounds.  相似文献

A comparison of SPE cartridges produced in authors laboratory containing silica modified by addition of three functional moieties with standard C-18 and SDVB cartridges was made in terms of their applicability for the isolation of flavor compounds. Compounds found in wine and grapes were used for model mixture, which was spiked into a grape juice. Functionalized phases for SPE were prepared modifying silica gel with alkoxysilanes with different functional groups: (3-(phenylamino)-propyltrimethoxysilane, octyltriethoxysilane and octadecyl-silane. The functionalization was carried out by the dry method, which resulted 5, 10 and 20 weight parts of initial support. Functionalized phases were characterized using FT-IR, elemental analysis and NMR.Performance for new phases compared to “standard” ones (C-18 and SDVB (styrene–divinylbenzene) varied, depending on the group or type of analyzed compound. They were more efficient in extraction of methyl anthranilate and vanilins. For extraction of terpenes, C-6 alcohols, isoprenoids, benzene derivatives and phenols their efficiency was comparable to that of C-18.Functionalized laboratory-made mixed phases are suitable for extraction of flavor compounds from grape juice. They are suitable for extraction of compounds belonging to different chemical classes with the efficiency comparable to C-18 and SDVB phases. The production of such functionalized phases can be easily performed in the laboratory, at a very low cost, comparing to C-18 or SDVB cartridges. This makes the proposed functionalized phases an interesting alternative, in sample preparation for analysis and particularly in preparative/flash chromatography.  相似文献

This paper highlights recent advances in the solid-phase extraction (SPE) of quaternary ammonium herbicides in water, soil, plant and biological samples. After a brief introduction summarizing the properties of quaternary ammonium herbicides and the difficulties involved in measuring them, attention is paid primarily to solid supports used for isolation and concentration, pre-treatments required for the different matrices, and eluents applied for quantitative desorption of these analytes. The determination techniques used after SPE and applications of the proposed SPE methodology are also briefly discussed.  相似文献

This work describes novel devices for electric field-assisted solid phase extraction (E-SPE) and an application was carried out on the antimicrobial marbofloxacin (MAR). Conventional syringe-type SPE cartridges were easily adapted to receive two electrodes that were inserted and positioned below and above the sorbent. The adapted cartridges were coupled to a flow extraction system, which consisted of an electrophoresis power supply, a peristaltic pump and a homemade SPE manifold. These devices were used to apply electric fields during the extraction of MAR from fortified buffer and milk samples. The recovery of MAR was improved (2.3 times) or reduced (4.2 times) in comparison to conventional SPE when the top electrode was used as cathode (E-SPE(?/+)) or anode (E-SPE(+/?)), respectively. The results demonstrated that usual SPE cartridges can be easily and inexpensively adapted for applying an electric field in SPE. It was also concluded that the electric field applied in a usual SPE cartridge can be employed as a new suitable approach to enhance the extraction efficiency of ionic compounds in a complex sample matrix.  相似文献

Solid-phase extraction (SPE) is an effective tool for the preconcentration of trace elements and their separation from various sample constituents. Octadecyl and other alkyl-bonded silica gels are most widely used for these purposes. The fundamentals of the SPE of inorganic ions are reviewed and compared with those of related techniques (liquid-liquid extraction and reversed-phase liquid chromatography). The extraction of ions in the form of chelate compounds, inorganic salts solvated by neutral reagents, and ion-pair compounds is considered. Numerous applications of SPE to the separation and preconcentration of different elements and their species, including on-line combinations with instrumental determination techniques, are described and tabulated.  相似文献

Using caffeic acid and p-hydroxybenzoic acid as templates, two molecularly imprinted polymers (MIPs) were prepared that were used for isolation of polyphenols from olive mill waste water samples (OMWWs) without previous pre-treatment. For the preparation of the caffeic acid MIPs 4-vinylpyridine, allylurea, allylaniline and methacrylic acid were tested as functional monomers, ethylene glycol dimethylacrylate (EDMA), pentaerythritol trimethylacrylate (PETRA) and divinylbenzene 80 (DVB80) as cross-linkers and tetrahydrofuran as porogen. For p-hydroxybenzoic acid 4-vinylpyridine, allylurea and allylaniline were tested as functional monomers, EDMA and PETRA as cross-linkers and acetonitrile as porogen. The performance of the synthesized polymers was evaluated against seven structurally related compounds by means of polymer-based HPLC. The two polymers that presented the most interesting properties were further evaluated by batch rebinding and from the derived isotherms their capacity and binding strength were determined. Using solid-phase extraction (SPE), their ability to recognize and bind the template molecule from an aqueous solution as well as the pH dependence of the binding strength were explored. After establishing the best SPE protocol, an aqueous model mixture of compounds and a raw OMWWs sample were loaded on the two best polymers. The result of the consecutive use of the two polymers on the same sample was explored. It was concluded that acidic conditions favour the recognition abilities of both polymers and that they can be used for a quick and efficient isolation of the polyphenol fraction directly from raw OMWW.  相似文献

A novel molecularly imprinted polymer (MIP) was synthesized by precipitation polymerization with baicalein (BAI) as the template and used as solid‐phase extraction (SPE) adsorbent, aiming at the affinity isolation and selective knockout of BAI from Scutellaria baicalensis Georgi (SB). We used computational simulation to predict the optimal functional monomer, polymerization solvent and molar ratio of template to functional monomer. Characterization and performance tests revealed that MIP exhibited uniform spherical morphology, rapid binding kinetics, and higher adsorption capacity for BAI compared with nonimprinted polymer (NIP). The application of MIP in SPE coupled with high‐performance liquid chromatography to extract BAI from SB showed excellent recovery (94.3%) and purity (97.0%). Not only the single BAI compound, but also the BAI‐removed SB extract was obtained by one‐step process. An SPE method followed by GC-electron capture detection (ECD) with confirmation by MS for the trace determination of four pesticides considered as endocrine-disrupting compounds (EDCs) in natural waters and sediments has been developed. Target analytes, fenarimol, fenvalerate, pendimethalin, and vinclozolin, belong to different chemical groups and are used mainly in agriculture. In the present study, analysis employs an offline SPE step for the extraction of the target analytes from natural waters. Sonication and subsequent SPE clean-up was used for extraction and purification of the sediment samples which were finally treated with activated copper powder. The type of SPE disk, eluents as well as solution parameters including pH value, and concentrations of salts and humic substances were examined for the efficiency of the method. The recoveries of all pesticides were in relatively high levels, ranging from 75 to 97% for waters and 71 to 84% for sediment samples. Both methods were applied to real water and sediment samples and the presence of the tested compounds was investigated.  相似文献

Solid-phase extraction (SPE) for two groups of polyaromatic compounds--polycyclic aromatic hydrocarbons and naphthalenesulfonates--with completely different problems in the extraction process are reviewed. The sorbents used in each case and the different steps of SPE are studied. Particular problems encountered in the SPE of each group are described. Adsorption problems of PAHs which require an organic solvent or a surfactant to be added to the sample are explained. The need of ion-pair solid-phase extraction for extracting naphthalenesulfonates and the influence of the inorganic species in the extraction are discussed. The essential oil and infusion of Salvia officinalis leaves have been widely applied in traditional medicine since ancient times and nowadays subjected to extensive research of their antibacterial, antiviral and cytotoxic properties. This paper shows chemical composition data of S. officinalis leaves essential oil isolated by steam distillation using a Clevenger-type apparatus. Also, the paper presents the chemical content of volatile and semi-volatile compounds of S. officinalis leaves infusion. The volatile and semi-volatile compounds of S. officinalis leaves infusion were isolated by solid-phase extraction (SPE) and liquid-liquid extraction with hexane and dichloromethane. SPE was carried out on 500 mg octadecylsilane (C18) cartridges and elution with dichloromethane. Liquid-liquid extraction was performed with hexane and dichloromethane. The essential oil in dichloromethane and infusion extracts in hexane and dichloromethane were analyzed by gas chromatography coupled with mass spectrometry. The quantitative results obtained by solid-phase extraction and liquid-liquid extraction showed that SPE on C18 performed the highest recovery of the volatile compounds from infusion sample.  相似文献

A fast, sensitive and selective procedure employing a combination of microwave-assisted extraction (MAE) and solid phase extraction (SPE) was applied prior to liquid chromatographic identification and quantification of phenolic compounds in plant materials. MAE has been tested and optimized for the isolation of phenolic acids (gallic, protocatechuic, p-hydroxybenzoic, chlorogenic, vanilic, caffeic, syringic, p-coumaric, ferulic, sinapic, benzoic, m-coumaric, o-coumaric, rosmarinic, cinnamic acids) and 3,4-dihydroxybenzaldehyde, syringaldehyde, p-hydroxybenzaldehyde, and vanillin in various plants. The effects of experimental conditions on MAE efficiency, such as solvent composition, temperature, extraction time, have been studied. The extraction efficiencies were compared with those obtained by computer-controlled, two-step Soxhlet-like extractions. Plant extracts were purified and phenolic compounds were pre-concentrated using SPE on polymeric RP-105 SPE sorbent prior to HPLC analysis. Chromatographic separation was carried out on a Hypersil BDS C18 column using a mobile phase consisted of 0.3% (v/v) acetic acid in water (solvent A) and methanol (solvent B) at flow rate 0.6 ml min−1 and column temperature 30 °C with gradient elution.  相似文献

Our recent on-line solid-phase extraction (SPE) device for micro-liquid chromatography, known as micro-solid-phase extraction (microSPE), was compared with traditional SPE for the analysis, from aqueous samples, of 4 pesticides belonging to different classes. Two different kinds of adsorbents, C18 and graphitized carbon black, were tested. A 2-stage ion trap mass spectrometer, equipped with homemade microflow electrospray ion (ESI) source, was used. Detection limits with a signal-to-noise ratio of 3:1 for both extraction methods were in the range of 0.1 microg/L for all compounds. However, better recoveries were obtained when microSPE traps were used.  相似文献
