Observations of UV-B radiation in the area of the Antarctic Peninsula are described, with the objective to obtain an evaluation of the UV-B enhancements observed during ozone hole episodes, and compare these with equatorial values. The enhancements observed during Southern Hemisphere spring are described in terms of a specific case of enhancement, at the Antarctic peninsula, which has shown a maximum UV-B index of 8.7, in October 1998. The average enhancement between the autumn-unperturbed and spring-perturbed periods results in an UV-B index of 5.4, but with large fluctuations in which much larger indices are produced. These values are compared to indices normally observed in the equatorial region. For measurements obtained with the same kind of instrument at Natal (5.8°S, 35.2°W), the UV-B index varies between 7 and 14, which means that enhanced UV-B indices in the Antarctic Peninsula may become of the same order of magnitude of the lower limit equatorial values.  相似文献

There are numerous strains of Chlorella with a corresponding variety of metabolic pathways. A strain we previously isolated from wastewater in northern Sweden can grow heterotrophically as well as autotrophically in light and has higher lipid contents under heterotrophic growth conditions. The aims of the present study were to characterize metabolic changes associated with the higher lipid contents in order to enhance our understanding of lipid production in microalgae and potentially identify new compounds with utility in sustainable development. Inter alia, the amino acids glutamine and lysine were 7-fold more abundant under heterotrophic conditions, the key metabolic intermediate alpha-ketoglutarate was more abundant under heterotrophic conditions with glucose, and maltose was more abundant under heterotrophic conditions with glycerol than under autotrophic conditions. The metabolite 3-hydroxy-butyric acid, the direct precursor of the biodegradable plastic PHB (poly-3-hydroxy-butyric acid), was also more abundant under heterotrophic conditions. Our metabolomic analysis has provided new insights into the alga’s lipid production pathways and identified metabolites with potential use in sustainable development, such as the production of renewable, biodegradable plastics, cosmetics, and nutraceuticals, with reduced pollution and improvements in both ecological and human health.  相似文献

The species Pseudogymnoascus is known as a psychrophilic pathogenic fungus which is ubiquitously distributed in Antarctica. While the studies of its secondary metabolites are infrequent. Systematic research of the metabolites of the Antarctic fungus Pseudogymnoascus sp. HSX2#-11 led to the isolation of one new pyridine derivative, 4-(2-methoxycarbonyl-ethyl)-pyridine-2-carboxylic acid methyl ester (1), together with one pyrimidine, thymine (2), and eight diketopiperazines, cyclo-(dehydroAla-l-Val) (3), cyclo-(dehydroAla-l-Ile) (4), cyclo-(dehydroAla-l-Leu) (5), cyclo-(dehydroAla-l-Phe) (6), cyclo-(l-Val-l-Phe) (7), cyclo-(l-Leu-l-Phe) (8), cyclo-(l-Trp-l-Ile) (9) and cyclo-(l-Trp-l-Phe) (10). The structures of these compounds were established by extensive spectroscopic investigation, as well as by detailed comparison with literature data. This is the first report to discover pyridine, pyrimidine and diketopiperazines from the genus of Pseudogymnoascus.  相似文献

The objective of this study was to evaluate the growth of green algae Chlorella sp. on wastewaters sampled from four different points of the treatment process flow of a local municipal wastewater treatment plant (MWTP) and how well the algal growth removed nitrogen, phosphorus, chemical oxygen demand (COD), and metal ions from the wastewaters. The four wastewaters were wastewater before primary settling (#1 wastewater), wastewater after primary settling (#2 wastewater), wastewater after activated sludge tank (#3 wastewater), and centrate (#4 wastewater), which is the wastewater generated in sludge centrifuge. The average specific growth rates in the exponential period were 0.412, 0.429, 0.343, and 0.948 day?1 for wastewaters #1, #2, #3, and #4, respectively. The removal rates of NH4–N were 82.4%, 74.7%, and 78.3% for wastewaters #1, #2, and #4, respectively. For #3 wastewater, 62.5% of NO3–N, the major inorganic nitrogen form, was removed with 6.3-fold of NO2–N generated. From wastewaters #1, #2, and #4, 83.2%, 90.6%, and 85.6% phosphorus and 50.9%, 56.5%, and 83.0% COD were removed, respectively. Only 4.7% was removed in #3 wastewater and the COD in #3 wastewater increased slightly after algal growth, probably due to the excretion of small photosynthetic organic molecules by algae. Metal ions, especially Al, Ca, Fe, Mg, and Mn in centrate, were found to be removed very efficiently. The results of this study suggest that growing algae in nutrient-rich centrate offers a new option of applying algal process in MWTP to manage the nutrient load for the aeration tank to which the centrate is returned, serving the dual roles of nutrient reduction and valuable biofuel feedstock production.  相似文献

利用水热釜研究了低温水热预处理过程中高蛋白小球藻的水热降解特性,考察了125-200℃各相产物产率、元素组成、能量回收率和关键元素C、N的分布规律。结果表明,大量的C、N元素富集在水相中,且提高温度(>175℃)有利于脱氨反应进而提高水相中NH3的相对含量。随着预处理温度升高,N在油相产物中分布逐渐增加且在175℃后快速增加;固相产率及能量回收率逐渐降低,但同时其N/C和O/C也降低,表明水热预处理是一个提质过程。借助FT-IR、XPS、TG-FTIR-MS和Py-GC-MS对比分析了原料和藻渣的官能团结构和热降解特性。结果表明,水热预处理过程改变了C、N不同官能团的相对含量,与原料相比,藻渣中C-C相对含量增加,而C-N、C-O等相对含量降低;除了蛋白质-N和季胺类-N,预处理后藻渣中出现了少量吡啶-N。固体藻渣官能团组分含量和结构变化降低了热失重过程NH3和HCN的释放、及快速热解产物中含N杂环化合物的含量。  相似文献

The goal of this study was to determine the optimal pretreatment process for the extraction of lipids and reducing sugars to facilitate the simultaneous production of biodiesel and bioethanol from the marine microalga Chorella sp. With a single pretreatment process, the optimal ultrasonication pretreatment process was 10 min at 47 KHz, and extraction yields of 6.5 and 7.1 (percentage, w/w) of the lipids and reducing sugars, respectively, were obtained. The optimal microwave pretreatment process was 10 min at 2,450 MHz, and extraction yields of 6.6 and 7.0 (percentage, w/w) of the lipids and reducing sugars, respectively, were obtained. Lastly, the optimal high-pressure homogenization pretreatment process was two cycles at a pressure of 20,000 psi, and extraction yields of 12.5 and 12.8 (percentage, w/w) of the lipids and reducing sugars, respectively, were obtained. However, because the single pretreatment processes did not markedly improve the extraction yields compared to the results of previous studies, a combination of two pretreatment processes was applied. The yields of lipids and reducing sugars from the combined application of the high-pressure homogenization process and the microwave process were 24.4 and 24.9 % (w/w), respectively, which was up to three times greater than the yields obtained using the single pretreatment processes. Furthermore, the oleic acid content, which is a fatty acid suitable for biodiesel production, was 23.39 % of the fatty acids (w/w). The contents of glucose and xylose, which are among the fermentable sugars useful for bioethanol production, were 77.5 and 13.3 % (w/w) of the fermentable sugars, respectively, suggesting the possibility of simultaneously producing biodiesel and bioethanol. Based on the results of this study, the combined application of the high-pressure homogenization and microwave pretreatment processes is the optimal method to increase the extraction yields of lipids and reducing sugars that are essential for the simultaneous production of biodiesel and bioethanol.  相似文献

Chemical investigation of the Antarctic lichen-derived fungal strain Acremonium sp. SF-7394 yielded a new amphilectane-type diterpene, acrepseudoterin (1), and a new acorane-type sesquiterpene glycoside, isocordycepoloside A (2). In addition, three known fungal metabolites, (−)-ternatin (3), [D-Leu]-ternatin (4), and pseurotin A (5), were isolated from the EtOAc extract of the fungal strain. Their structures were mainly elucidated by analyzing their NMR and MS data. The absolute configuration of 1 was proposed by electronic circular dichroism calculations, and the absolute configuration of the sugar unit in 2 was determined by a chemical method. The inhibitory effects of the isolated compounds on protein tyrosine phosphatase 1B (PTP1B) were evaluated by enzymatic assays; results indicated that acrepseudoterin (1) and [D-Leu]-ternatin (4) dose-dependently inhibited the enzyme activity with IC50 values of 22.8 ± 1.1 μM and 14.8 ± 0.3 μM, respectively. Marine diatom, strain JPCC DA0580, and marine green microalga strain NKG400014 were selected as high neutral lipid-producers from marine microalgal culture collection toward biodiesel production. These strains were tentatively identified as Navicula sp. and Chlorella sp., respectively, by 18S rDNA analysis. Growth and lipid accumulation conditions of both strains were analyzed by changing nutrient concentrations in growth media and initial illuminance intensity. The highest productivity of fatty acid methyl ester (FAME) reached to 154 mg/L/week for NKG400014 and 185 mg/L/week for JPCC DA0580. Gas chromatography/mass spectrometry analysis indicates that FAME fraction from NKG400014 mainly contained 9-12-15-octadecatrienoate (C18:3) and that from JPCC DA0580 mainly contained methyl palmitate (C16:0) and methyl palmitoleate (C16:1). Furthermore, calorimetric analysis revealed that the energy content of strain was 4,233?±?55 kcal/kg (i.e., 15.9?±?0.2 MJ/kg) for NKG400014 and 6,423?±?139 kcal/mg (i.e., 26.9?±?0.6 MJ/kg) for JPCC DA0580, respectively. The value from JPCC DA0580 was equivalent to that of coal. The strains NKG400014 and JPCC DA0580 will become a promising resource that can grow as dominant species in the open ocean toward production of both liquid and solid biofuels.  相似文献

In this work, a kinetic expression relating light availability in the culture medium with the rate of microalgal growth is obtained. This expression, which is valid for low illumination conditions, was derived from the reactions that take part in the light‐dependent stage of photosynthesis. The kinetic expression obtained is a function of the biomass concentration in the culture, as well as of the local volumetric rate of absorption of photons, and only includes two adjustable parameters. To determine the value of these parameters and to test the validity of the hypotheses made, autotrophic cultures of the Chlorella sp. strain were carried out in a modified BBM medium at three CO2 concentrations in the gas stream, namely 0.034%, 0.34% and 3.4%. Moreover, the local volumetric rate of photon absorption was predicted based on a physical model of the interaction of the radiant energy with the suspended biomass, together with a Monte Carlo simulation algorithm. The proposed intrinsic expression of the biomass growth rate, together with the Monte Carlo radiation field simulator, are key to scale up photobioreactors when operating under low irradiation conditions, independently of the configuration of the reactor and of its light source.  相似文献

Microalgae is a rich source of polyunsaturated fatty acid. This study was conducted to identify and isolate microalgal strain with the potentials for producing polyunsaturated fatty acids (PUFAs) and determine its cytotoxic effect on some cancer cells. The algal strain (Chlorella sp. S14) was cultivated using modified BG-11 media, and algal biomass obtained was used for fatty acid extraction. Gas chromatographic–mass spectrometry was used to identify and quantify the levels of the fatty acid constituents. The total content of monounsaturated fatty acids (1.12%) was low compared to polyunsaturated fatty acids (PUFAs) (52.87%). Furthermore, n-3 PUFAs accounted for (12.37%) of total PUFAs with the presence of α-linolenic acid (2.16%) and cis-11,14,17-eicosatrienoic acid (2.16%). The PUFA-rich extract did not exhibit a cytotoxic effect on normal cells. Treatment with the PUFA-rich extract (150 µg/mL) significantly reduced cell viability in MCF-7 (31.58%) and A549 (62.56%) cells after the 48 h treatment. Furthermore, treatment of MCF-7 with fatty acid extracts (125 and 150 µg/mL) showed a significant reduction in MDA levels, increase in catalase activities and decrease in GSH level compared to untreated cells. However, a slight decrease in MDA level was observed in A549 cells after the 48 h treatment. There are no significant changes in catalase activities and GSH level in treated A549 cells. However, a slight reduction of NO levels was observed in treated MCF-7 and A549 cells. These results indicate the potentials of PUFA-rich extracts from Chlorella sp. S14 to reduce viability and modulate redox status in A549 and MCF-7 cells.  相似文献

Reactive oxygen species (ROS) are involved the damage of living organisms under environmental stress including UV radiation. Cyanobacteria, photoautotrophic prokaryotic organisms, also suffer from increasing UV-B due to the depletion of the stratospheric ozone layer. The increased UV-B induces the production of ROS in vivo detected by using the ROS-sensitive probe 2',7'-dichlorodihydrofluorescein diacetate (DCFH-DA). Ascorbic acid and N-acetyl-L-cysteine (NAC) scavenged ROS effectively, while alpha-tocopherol acetate or pyrrolidine dithiocarbamate (PDTC) did not. The presence of rose bengal and hypocrellin A increased the ROS level by photodynamic action in the visible light. The presence of the herbicide, 3-(3,4-dichlorophenyl)-1,1-dimethyl urea (DCMU), increased ROS production slightly, and ROS formation was greatly enhanced by the addition of methyl viologen due to the fact that this redox system diverts electrons from PSI to oxygen and thus forms ROS. UV-B induces ROS generation by photodynamic action and inhibition of the electron transport by damaging the electron receptors or enzymes associated with the electron transport chain during photosynthesis.  相似文献

In the present work, we explore in photoanodes of dye-sensitized solar cell (DSSC) the co-sensitization of the red protein phycoerythrin extracted from Antarctic algae with spherical silver nanoparticles between 10 and 200 nm and triangular nanoparticles of ~100 nm. We found that the order of addition of the sensitizers matters. Best results achieved for a sequential approach was phycoerythrin used as a dipping solution at the first step and nanoparticles at the second. We found that cell efficiency depends on the protein concentration, although no main differences detected with the nanoparticle shape or size. Our results show an average increase of 25% in the conversion efficiency values in the presence of nanoparticles.  相似文献

UV-B absorbance and UV-B absorbing compounds (UACs) of the pollen of Vicia faba, Betula pendula, Helleborus foetidus and Pinus sylvestris were studied. Sequential extraction demonstrated considerable UV-B absorbance both in the soluble (acid methanol) and insoluble sporopollenin (acetolysis resistant residue) fractions of UACs, while the wall-bound fraction of UACs was small. The UV-B absorbance of the soluble and sporopollenin fraction of pollen of Vicia faba plants exposed to enhanced UV-B (10 kJ m(-2) day(-1) UV-B(BE)) was higher than that of plants that received 0 kJ m(-2) day(-1) UV-B(BB). Pyrolysis gas chromatography-mass spectrometry (py-GC-MS) analysis of pollen demonstrated that p-coumaric acid and ferulic acid formed part of the sporopollenin fraction of the pollen. The amount of these aromatic monomers in the sporopollenin of Vicia faba appeared to increase in response to enhanced UV-B (10 kJ m(-2) day(-1) UV-B(BE)). The detection limit of pyGC-MS was sufficiently low to quantify these phenolic acids in ten pollen grains of Betula and Pinus. The experimental data presented provide evidence for the possibility that polyphenolic compounds in pollen of plants are indicators of solar UV-B and may be applied as a new proxy for the reconstruction of historic variation in solar UV-B levels.  相似文献

A growth-chamber experiment was conducted to evaluate whether ethylenenediurea (EDU), a chemical shown to be protective against ozone pollution, could ameliorate foliar damage induced by ultraviolet-B (UV-B) radiation exposure in 'Roanoke' soybean (Glycine max L.), a UV-B-sensitive cultivar, and whether these effects could be discriminated using fluorescence (F) observations. The experiment had four treatment groups: control; biologically effective UV-B (18 kJ m(-2) day(-1)); EDU (500 micromol mol(-1)); and both UV-B and EDU (UV/EDU). Measurements included photosynthetic pigments, F image system (FIS) images of adaxial surfaces in four spectral regions (blue, green, red and far-red) and F emission spectra of the pigment extracts produced at two excitation wavelengths, 280 nm (280EX) and 380 nm (380EX). Several F ratios from 280EX, 380EX and the FIS images successfully separated the low UV vs high EDU group responses based on means alone, with intermediate values for controls and the combined UV/EDU groups. A UV-B/blue emission ratio, F315/F420 (280EX), was correlated with chlorophyll content (microg cm(-2))(R = 0.88, P < 0.001), as was a ratio of emissions at two UV-A wavelengths: F330/F385 (280EX) (R = 0.87). These two 280EX ratios were also linearly correlated with emission ratios produced by 380EX, such as the far-red/green ratio, F730/F525 (380EX) (R = 0.92, P < 0.001), and clearly distinguished the UV-B and EDU groups separately, and which bracketed the similar intermediate responses of the UV/EDU and control groups. The FIS images additionally captured the following anatomical spatial patterns across the leaf surfaces: (1) emissions of UV-B-irradiated leaves were more uniform but lower in intensity than those of other groups; and (2) emissions of EDU-treated leaves exhibited the greatest variation in spatial patterns because veins had elevated blue F and leaf edges had enhanced red and far-red F. This experiment supports the hypothesis that EDU substantially ameliorated UV-B damage to foliage, a result that relied on the combined use of FIS images and emission spectra.  相似文献

Tributyl phosphate (TBP), a plasticizer and solvent, is used in nuclear fuel reprocessing, generating TBP wastes laden with residual uranium. ACitrobacter sp. accumulated heavy metals via a phosphohydrolase(s) that precipitated metals with inorganic phosphate liberated from an organic phosphate “donor” molecule (TBP). Mutant analysis suggested that TBP hydrolysis was not attributable to a previously documented acid phosphatase (monoesterase). Purified monoesterase had little activity against phospho di- and triesters, had no requirement for Mg2+ or Mn2+, and was EDTA-resistant. Conversely, TBP cleavage by immobilized cells was enhanced by Mg2+, and ininhibited by Mn2+ and EDTA. A separate phosphotri/diesterase was implicated.  相似文献

Laccase was produced in the supernatant of culture of a local isolate of Agaricus sp. obtained from decaying Ficus religiosa wood. The enzyme was produced at a constitutive level when growing the fungus in a nitrogenlimited medium supplemented with either glycerol, glucose, fructose, mannitol, arabinose, maltose, sacch arose, cellulose, or cellobiose. Atwo-to sixfold increase in enzyme specific activity was observed when growing the strain in the presence of straw, xylan, xylose, lignosulfonate, veratryl alcohol, and ferulic and veratric acid. Experimentsare consistent with the existence of an induction control on laccase and the absence of a form of carbon catabolite repression mediated by noninducing carbon sources. Immobilization of the Agaricus sp. on several supports, including polyurethane foam, textilestrips, and straw, resulted in an increase of enzyme production as compared to cultivation in liquid medium.  相似文献

In order to determine the role of lasers in the stress resistance of broad bean (Vicia faba L.) to ultraviolet-B (UV-B) radiation, the embryos in seeds were exposed to He-Ne laser or CO2 laser radiation. Afterwards they were cultivated in Petri dishes in a constant temperature incubator until the lengths of epicotyls were nearly 3 cm. The epicotyls were then exposed to 1.02, 3.03 or 4.52 kJ m(-2) UV-B radiation, respectively, under 70 micromol m(-2) s(-1) photosynthetically active radiation (PAR) in a growth cabinet. Changes in the concentration of malondialdehyde (MDA), ascorbic acid (AsA) and UV-B absorbing compounds (absorbance at 300 nm) were measured to test the effects of laser pretreatment. The results showed that laser pretreatment of embryos enhanced UV-B stress resistance in the epicotyls of the broad bean by decreasing the MDA concentration and increasing the content of AsA and UV-B absorbing compounds. We suggest that those changes in MDA, AsA and UV-B absorbing compounds were responsible for the increase in stress resistance observed in the broad bean. This is the first investigation reporting the use of laser pretreatment to protect the cells of the broad bean from UV-B-induced damage.  相似文献
