An efficient method has been successfully developed for the synthesis of oxygen-bridged pyrimidine tricyclic derivatives from salicylaldehyde, ethyl acetoacete, and urea catalyzed by PdO in very good yield. A series of different oxygen-bridged pyrimidine tricyclic derivatives has been synthesized, in which two compounds’ x-ray crystal structure were obtained.  相似文献

Chlorotrimethylsilane‐promoted Biginelli‐type reaction of benzaldehyde, acetoacetic acid derivatives, and various carboxyl‐containing ureas was explored. It was found that the steric load of the urea substituents influenced strongly the reaction outcome; in particular, the method was efficient only in the case of unbranched mono‐substituted ureas bearing either aliphatic or aromatic groups. The method allows performing a one‐pot, protecting group free synthesis of dihydropyrimidines possessing carboxylic functionality.  相似文献

An efficient synthesis of 3,4-dihydropyrimidin-2(1H)-one derivatives using formic acid as catalyst, from aldehydes,β-ketoester and urea(thiourea) without solvent under the irradiation of microwave is described. Compared with the classical Biginelli reaction, this new method has the advantage of good yields (77-94% for aromatic aldehydes) and short reaction time (3-8 min).  相似文献

A simple, efficient and cost-effective method for the synthesis of octahydroquinazolinone derivatives using dimedone, urea/thiourea and aromatic aldehydes using lanthanum oxide as a catalyst under solvent free condition in microwave irradiation is reported. The present method does not involve any hazardous organic solvents. This catalyst has promising features for the reaction response such as the shortest reaction time, excellent product yields, simple work-up procedure and purification of products by non-chromatographic methods.  相似文献

Copper(I) promoted Huigsen 1,3-dipolar cycloaddition of terminal alkyne with azide is utilized as an efficient protocol for the one-pot five-component synthesis of glycoside annulated dihydropyrimidinone derivatives with 1,2,3-triazol linkage accomplished by tert-butyl β-ketoester, arylaldehyde, urea, propargyl alcohol, and glycosyl azide through the combination of transesterification and Biginelli reaction in aqueous medium. This protocol provides an access to generate scaffolds with molecular diversity from readily available starting materials.  相似文献

An efficient synthesis of 3,4-dihydropyrimidin-2-(1H)-one derivatives by one-pot three-component condensation reaction between an alkyl or aryl aldehyde, a 1,3-dicarbonyl compound, and urea or thiourea in the presence of catalytic amount of bromodimethylsulfonium bromide has been developed. The condensation product was obtained in excellent yields in refluxing ethanol and easily separated from the reaction mixture and purified.  相似文献

A simple and efficient method for the synthesis of 3,4-dihydropyrimidin-2-(1H)one and benzo[4,5]imidazo/thioazo[1,2-a]pyrimidine derivatives has been described using N,N′-dichlorobis(2,4,6-trichlorophenyl)urea (CC-2) as a new reagent. 利用二氯代磷酰基异氰酸酯与4,6-二甲氧基-2-氨基嘧啶的加成反应合成了中间体N-(4,6-二甲氧基嘧啶-2-基)-N'-二氯代磷酰基脲(Ⅰ).Ⅰ与2倍的醇或胺反应得到对称双取代磷酰基脲类化合物Ⅱa_Ⅱi;Ⅰ与1倍的胺反应得到氯代磷酰基脲类化合物Ⅲa_Ⅲe,再与1倍的醇反应则得到不对称双取代磷酰基脲类化合物Ⅳa_Ⅳg.生物活性测定结果表明,化合物Ⅱ、Ⅲ和Ⅳ均显示一定除草活性.  相似文献

Sulfamic acid efficiently catalyzes the three‐component condensation reaction of aldehydes, 1,3‐dicarbonyl compounds, and urea/thiourea under solvent‐free conditions to afford the corresponding dihydropyrimidinones and thio‐derivatives in high yields. Silica sulfuric acid is also found to be an efficient catalyst for the Biginelli reaction under solvent‐free conditions. Compared to the classical Biginelli reaction conditions, this new method consistently has the advantage of giving good yields and requiring short reaction times.  相似文献

以羰基衍生物,取代1,3-二羰基化合物,脲和硫脲为原料,经固定化青霉素酰化酶催化的Biginelli反应合成了22个四氢嘧啶类化合物(1a~1v),其中1s和1v为新化合物,其结构经1H NMR, 13C NHR和IR表征。研究了溶剂、反应温度、反应时间和物料比γ[n(苯甲醛) :n(脲) :n(乙酰乙酸乙酯)]对1a产率的影响。结果表明:在最佳反应条件(固定化青霉素酰化酶为催化剂,乙醇为溶剂,γ=1.0 :1.5:1.0,于50 ℃反应6 h)下,1a产率最高(85%)。  相似文献

Palladium catalyzed cascade coupling of substituted urea derivatives and tert-butyl isocyanide for the efficient synthesis of phenylamino-substituted quinazolinones has been developed in moderate to good yields. This method provides a short and alternative approach for the synthesis of quinazolinones derivatives which are valuable compounds with biological and pharmacological potentials. An efficient and straightforward synthesis of polycyclic 1,4-dihydropyridine derivatives has been developed by reacting heterocyclic ketene aminals (HKAs) with 1,3-cyclohexanedione derivatives and a number of substituted salicylaldehyde, respectively, via three-component cascade reactions in water. This strategy provides an efficient and environmentally friendly approach for easy access to various new fused polycyclic 1,4-dihydropyridine derivatives without the need of organic solvents in moderate to good yields.  相似文献

Oxidation of cyclohexanone to ?-caprolactone with stable industrially manufactured hydrogen peroxide derivatives: adduct with urea (urea hydrogen peroxide), sodium perborate, sodium percarbonate (Persol), magnesium monoperphthalate (Dismozon) was studied. Oxidation with urea hydrogen peroxide is the most efficient in hexafluoroisopropanol in the case of preliminary removal of urea in the form of an oxalate. Oxidation with sodium perborate and percarbonate provides high yields in trifluoroacetic acid. The lowest cost process consists in interaction with sodium monoperphthalate (Persol and phthalic anhydride) in an aqueous medium.  相似文献

An effective route to functionalized hydantoin derivatives is described, involving the reaction of a urea derivative resulting from the addition of a primary amine to an arylsulfonyl isocyanate, and an alkyl propiolate or dialkyl acetylenedicarboxylate in the presence of triphenylphosphine. The reactive 1:1 intermediate obtained from the addition of triphenylphosphine to the alkyl propiolate or dialkyl acetylenedicarboxylate was trapped by NH-acids such as the urea derivative to produce functionalized hydantoin derivatives.  相似文献

An efficient and chemoselective sulfonamide N-alkylation of sulfonyl ureas is described. The sulfonyl urea derivatives, prepared in situ by the addition of an amine to an arylsulfonyl isocyanate, are selectively alkylated in excellent yields under neutral and mild conditions by treatment with trialkylphosphite-dimethyl acetylenedicarboxylate at ambient temperature.  相似文献

A rapid, highly efficient and mild green synthesis of 5,5‐diphenylhydantoin derivatives was achieved from the reaction of symmetrical or unsymmetrical benzil derivatives with urea in the presence of ethanolic KOH under ultrasound irradiation. This simple method affords 5,5‐diphenylhydantoin derivatives at room temperature in short reaction time with high yield and purity. An efficient methodology for the synthesis of 6-styrenylpyrone in a four-step sequence is reported. The reactions were performed under catalyst-free conditions with mild and affordable reagents to produce 5,6-dehydrokawain and fluorinated derivatives in good to excellent overall yields (72–86%) on a multigram scale.
