Synthesis and theoretical study of molecularly imprinted monoliths for HPLC   总被引:1,自引:0,他引:1  
Molecularly imprinted monoliths integrate the high permeability of monolithic materials and the high selectivity and affinity of molecularly imprinted polymers (MIP). Thus, in recent years, development of this novel MIP format in HPLC has expanded quickly, particularly use of organic materials. This review focuses on the principal aspects of good practice in polymerization, theoretical studies, and recent developments in molecularly imprinted monoliths. Some thoughts on perspectives of MIP monoliths are also expressed.  相似文献

In this paper, we describe how to prepare a highly selective imprinted polymer by a bulk polymerization technique. We used tramadol as the template, (MAA) as functional monomers, and (EGDMA) as the cross-linker in chloroform as solvent. Results from Fourier Transform Infrared Spectroscopy (FTIR), Thermogravimetric Analysis (TGA), Scanning Electron microscopy (SEM) show that this imprinted sorbent exhibits good recognition and high affinity for tramadol. Selectivity of molecularly imprinted polympers (MIP) was evaluated by comparing several substances with similar molecular structures to that of tramadol. Controlled release of tramadol from MIPs was investigated through in vitro dissolution tests and by measuring the absorbance at λ max of 272 nm by (HPLC-UV). The dissolution media employed were hydrochloric acid pH 3.0 and phosphate buffers, pH 5.0 and 7.4, maintained at 37 and 25 ± 0.5°C. The results show the ability of MIP polymers to control tramadol release. In all cases, the release of MIPs was deferred for a longer time as compared to NMIP. At a pH of 7.4 and 25°C slower release of tramadol imprinted polymer occurred.   相似文献

A voltammetric sensor for albuterol was investigated where we combined the techniques of microfabrication and molecular imprinting to construct on-chip devices using photoirradiation of cross-linkable polymers. We are presenting an electrochemical sensor for the simultaneous determination of dopamine (DA) and uric acid (UA) in the presence of even high concentrations of ascorbic acid (AA). It based on a glassy carbon electrode modified with an electroactive film of polymerized dibromofluorescein. The electrochemical behaviors of DA and UA were studied by cyclic voltammetry using the modified electrode. It exhibits excellent electrocatalytic activity towards the oxidation of the two analytes. Most notably, the oxidation potentials differ by 180 and 200?mV between AA-DA and DA-UA, respectively. Thus, excellent selectivity towards the oxidation of DA and UA in the presence of even high concentrations of AA is accomplished. Under the optimum conditions, the anodic peak currents are linearly related to the concentrations of DA and UA in the range from 0.2 to 200?μmol?L-1 and from 1.0 to 250?μmol?L-1, respectively. The detection limits for DA and UA are 0.03?μmol?L-1 and 0.2?μmol?L-1, respectively (at an S/N of 3). The method has good selectivity and sensitivity and was successfully applied to the simultaneous determination of DA and UA in spiked human serum.

Solid-phase extraction (SPE) with a molecularly imprinted polymer (MIP) as sorbent has been investigated for the clean-up of the broad-spectrum bacteriostatic antibiotic chloramphenicol (CAP) in honey samples. The MIP was prepared by using methacrylic acid (MAA) as functional monomer, ethylene glycol dimethacrylate (EDMA) as cross-linker, chloroform as porogen and CAP as template molecule. The binding behaviour of the template CAP on the MIP was evaluated by high-performance liquid chromatography, and then the MIP was applied as a sorbent in SPE to selectively extract CAP from honey. It was shown that recoveries of nearly 100% of a CAP standard solution and up to 94% from spiked honey samples could be obtained after SPE.  相似文献

Molecular imprinted solid-phase extraction (MISPE) is a well known technique for the selective extraction and pre-concentration of analytes, are present at low levels in chemically complex materials. Herein, water-soluble, molecularly imprinted polymers (MIP) were prepared for solid-phase extraction of pseudoephedrine hydrochloride (PSE), which was monitored at 256 nm by the UV spectroscopy. MISPE conditions were optimized to allow the selective and determination of PSE in aqueous samples and composite materials, such as biological fluids and human urine. MIP was prepared by precipitation polymerization method, using methacrylic acid as a functional monomer and ethylene glycol dimethacrylate as a cross-linking agent in either acetonitrile or chloroform. The results suggest that the obtained MISPE exhibits high affinity for PSE, and the imprinted polymer demonstrates much higher efficiency than a non-imprinted polymer (NIP). The imprinting-induced extraction was confirmed by the determination of recovery values for NIP (4%) and MIP (80%) polymers, respectively. The binding capacity of the MIP for PSE was found of 47.6 mg g−1.  相似文献

A photoresponsive molecularly imprinted polymer was prepared from a di(ureidoethylenemethacrylate)azobenzene monomer, using a methotrexate analogue as template. Photoisomerization of the 3D crosslinked polymer matrix allowed switching the substrate affinity by altering the geometry and spatial arrangement of the receptor binding sites. As a result, controlled release and uptake of the template (or analogous ligands) were obtained.  相似文献

Molecularly imprinted polymers (MIPs) are currently used to provide selectivity in chemical sensors. In this context, a non-covalent bisphenol-A (BPA)-imprinted polymer using 4-vinylpyridine (4-Vpy) as the functional monomer, ethylene glycol dimethacrylate (EGDMA) as crosslinker and a low volatile solvent, triethylene glycol dimethyl ether (TRIGLYME), in combination with a non-reactive linear polymer, poly (vinyl acetate) (PVAc), as porogen, was synthesized with a simple polymerization procedure. Batch rebinding experiments were carried out to evaluate the binding and selectivity properties of the BPA-MIP. The experimental adsorption isotherms were fitted and a heterogeneous distribution of the binding sites was found. The selectivity of MIP demonstrated higher affinity for target BPA and BPA-analogues over other common water pollutants. The adsorption kinetics followed the pseudo-second-order kinetic model so that the specific adsorption in the imprinted cavities by two strong hydrogen bonds could be described as a chemisorption process. The diffusion mechanism was determined by the intra-particle diffusion and Boyd models, both of them revealing that the adsorption was mainly governed by intra-particle diffusion. MIP was shown to be promising for regeneration without significant loss in adsorption capacity.  相似文献

A novel amine-imide type conducting polymer, denoted as poly(PD-BCD), was molecularly imprinted on an indium-tin oxide (ITO) glass, with uric acid (UA) as the template and without any functional monomer. Intending to improve the imprinting efficiency, the polymer content was varied from 0.3 to 0.9 wt% during the preparation of the molecularly imprinted polymer (MIP), thereby varying the thickness of the polymer film; the content of UA as the template was maintained to be the same for all the films. The sensitivities of the thus prepared MIP electrodes were calculated to be more than 3-fold, compared to those of the corresponding non-MIP (NMIP) electrodes, which were obtained through the same method, however, without adding UA during their preparation. A polymer content of 0.6 wt% rendered the best performing MIP electrode, as judged by the imprinting efficiency and sensitivity of the electrode for UA. A linear relationship between steady-state currents and UA concentrations from 0 to 1.125 mM was obtained for both types of the sensors. The sensitivities of the MIP and the NMIP electrodes made with 0.6 wt% of polymer were calculated to be 24.72 and 6.63 μA mM−1 cm−2, respectively. The limit of detection (LOD) for this MIP was found to be 0.3 μM at a signal to noise ratio (S/N) of 3. This MIP electrode was used as a biosensor for the detection of UA in the presence of ascorbic acid (AA) in a sample containing these species in the same concentrations as those in a human serum. The selectivity of MIP electrode is higher than that of NMIP electrode, and the values are 28.76 and 8.85, respectively. The results are substantiated by using cyclic voltammetry (CV), linear sweep voltammetry, amperometry, and scanning electron microscopy.  相似文献

Novel molecularly imprinted polymer nanoparticles were synthesized by precipitation polymerization with sunset yellow as the template and [2‐(methacryloyloxy)ethyl] trimethylammonium chloride as the functional monomer. The molecularly imprinted polymer nanoparticles were characterized by Fourier transform infrared spectroscopy, scanning electron microscopy, transmission electron microscopy, and their specific surface area and thermal stability were measured. The molecularly imprinted polymer nanoparticles had a high adsorption capacity in wide pH range (pH 1–8) for sunset yellow. The adsorption equilibrium only needed 5 min, and the quantitative desorption was very fast (1 min) by using 10.0 mol/L HCl as the eluant. The maximum adsorption capacity of the molecularly imprinted polymer nanoparticles for sunset yellow was 144.6 mg/g. The adsorption isotherm and kinetic were well consistent with Langmuir adsorption model and pseudo‐second‐order kinetic model, respectively. The relative selectivity coefficients of the molecularly imprinted polymer nanoparticles for tartrazine and carmine were 9.766 and 12.64, respectively. The prepared molecularly imprinted polymer nanoparticles were repeatedly used and regenerated ten times without significant absorption capacity decrease.  相似文献

In principle, molecularly imprinted polymer science and technology provides a means for ready access to nano-structured polymeric materials of predetermined selectivity. The versatility of the technique has brought it to the attention of many working with the development of nanomaterials with biological or biomimetic properties for use as therapeutics or in medical devices. Nonetheless, the further evolution of the field necessitates the development of robust predictive tools capable of handling the complexity of molecular imprinting systems. The rapid growth in computer power and software over the past decade has opened new possibilities for simulating aspects of the complex molecular imprinting process. We present here a survey of the current status of the use of in silico-based approaches to aspects of molecular imprinting. Finally, we highlight areas where ongoing and future efforts should yield information critical to our understanding of the underlying mechanisms sufficient to permit the rational design of molecularly imprinted polymers.  相似文献

In this study, molecular imprinting was used to develop a method based on noncovalent interactions for synthesis of a testosterone-specific polymer. The effect of the different template–monomer ratios, the particle sizes of polymers, and chromatographic mobile phases on steroid–polymer interactions are discussed. The polymer obtained was found to interact specifically with testosterone, while other steroids under study were eluted close to the void volume in the HPLC experiments. Batch rebinding studies in acetonitrile were undertaken to quantitatively evaluate the affinity of the polymer for testosterone. During this experiment, the testosterone concentration was measured in two ways: spectrophotometrically and by HPLC on a column with testosterone-specific imprinted polymer synthesized by us. Both methods resulted in similar values of association constants and the number of binding sites. However, the second method has obvious advantages when the analyzed solution contains a mixture of optically dense compounds. The results obtained focus on the two-point binding nature of the imprinted polymer–testosterone interaction and the significant role of hydrogen bonds between the OH group of testosterone and carboxy group of methacrylic acid residues inside specific recognition sites of the imprinted polymer. © 1998 John Wiley & Sons, Inc. Fluorescent conjugated polymers are an attractive basis for the design of low detection limit sensing devices owing to their intrinsic signal amplification capability. A simple and universal method to rationally control or fine-tune the chemodetection selectivity of conjugated polymer materials toward a desired analytical target would further benefit their applications. In a quest of such a method we investigated a general approach to cross-linked molecularly imprinted fluorescent conjugated polymer (MICP) materials that possess an intrinsic capability for signal transduction and have potential to enhance selectivity and sensitivity of sensor devices based on conjugated polymers. To study these capabilities, we prepared an MICP material for the detection of 2,4,6-trinitrotoluene and related nitroaromatic compounds. We found the imprinting effect in this material to be based on analyte shape/size recognition being substantial and generally overcoming other competing thermodynamically determined trends. The described molecularly imprinted fluorescent conjugated polymers show remarkable air stability and photostability, high fluorescence quantum yield, and reversible analyte binding and therefore are advantageous for sensing applications due to the ability to "preprogram" their detection selectivity through a choice of an imprinted template.  相似文献

A new molecularly imprinted polymer (MIP) has been prepared on silica beads using the radical “grafting from” polymerization method for selective extraction of minor contaminant mycotoxin of patulin (PTL). After the introduction of amino groups onto the silica surface with 3-aminopropyltriethoxysilane, azo initiator onto the silica surface was achieved by the reaction of surface amino groups with 4,4′-azobis(4-cyanopentanoic acid). The scale-up synthesis of MIP was then carried out in the presence of 6-hydroxynicotinic acid as template substitute, functional, and cross-linking monomers. The prepared sorbent was characterized using FT-IR spectroscopy, scanning electron microscopy, elemental analysis, and the adsorption–desorption selectivity, and the capacity characteristic of the polymer was investigated by a conventional batch adsorption test and Scatchard plot analysis. The results indicated that coated polymers had specific adsorption to PTL as compared with its co-occurring 5-hydroxymethyl-2-furaldehyde (hydroxymethylfurfural (HMF)), at the same bulk concentration for solution of PTL and HMF, the maximum absorbance in the solid-phase extraction (SPE) method to PTL were 93.97% or 0.654 μg/mg while to HMF they were 76.89% or 0.496 μg/mg. Scatchard analysis revealed that two classes of binding sites were formed in PTL-MIP with dissociation constants of 3.2 × 10−2 and 5.0 × 10−3 mg/mL and the affinity binding sites of 8.029 and 1.364 mg/g, respectively. The recoveries of PTL were more than 90% for the developed MISPE and around 75% for the traditional liquid–liquid extraction in spiked apple juice samples. It was concluded that the method is suitable for the scale-up synthesis of PTL-MIP grafted on silica, and the polymer can be effectively applied as SPE coupled with high-performance liquid chromatography (HPLC) for the determination of PTL in apple juice or other related products.  相似文献

A library of molecularly imprinted polymers (MIPs) was synthesized by radical bulk polymerization using the beta-lactam antibiotic penicillin G as the template. Diversity of the library was obtained by combining various functionalized monomers and cross-linkers and by varying the stoichiometry and the concentration of the components in the prepolymerization mixtures. The library was screened for selectivity to penicillin G by a radioligand binding assay and was compared to a corresponding control library. The best MIP candidate, showing the highest selectivity for penicillin G, was prepared from methacrylic acid and trimethylolpropane trimethacrylate as the functionalized monomer and cross-linker, respectively. Cross-reactivity studies with other beta-lactam antibiotics showed a low cross-reactivity of penicillin V (15%), ampicillin (16%), and amoxicillin (19%). Nafcillin and oxacillin showed less cross-reactivity (<1%). Cross-reaction with a cephalosporin antibiotic (cephapirin) and structurally nonrelated antibiotics (chloramphenicol, tetracycline, dapsone, and erythromycin) was less than 0.01%.  相似文献

The selective extraction of baicalin is important to its quality control especially when the matrices are complicated. In this work, a novel molecularly imprinted polymer was prepared for the selective extraction of baicalin in herbs. The molecularly imprinted polymer was synthesized by the copolymerization of 4‐vinyl pyridine and ethylene glycol dimethacrylate in the presence of baicalin by a precipitation polymerization method. After the optimization of parameters for molecularly imprinted polymer preparation, including the functional monomer, porogen, sampling solvent, and washing solvent, good selectivity was obtained, with an imprinting factor of about 4, which is much better than that achieved by the bulk‐polymerization method. The performances of the prepared molecularly imprinted polymers were systematically investigated, including adsorption kinetics, isotherm experiment, and Scatchard analysis. On the basis of the good adsorptive capability of the prepared molecularly imprinted polymer, it was also applied for the pretreatment of baicalin in Scutellaria baicalensis Georgi. The result showed that most of the matrices were removed and baicalin was selectively enriched.  相似文献

Atrazine contamination of water is of considerable concern because of the potential hazard to human health. In this study, a magnetic molecularly imprinted polymer for atrazine was prepared by the surface‐imprinting technique using Fe3O4 as the core, mesoporous silica as the carrier, atrazine as the template, and itaconic acid as the functional monomer. The magnetic molecularly imprinted polymer was characterized by Fourier‐transform infrared spectroscopy, scanning electron microscopy, X‐ray diffraction, and vibration‐sample magnetometry. The binding properties of the magnetic molecularly imprinted polymer toward atrazine were investigated by adsorption isotherms, kinetics, and competitive adsorption. It was found that the adsorption equilibrium was achieved within 2 h, the maximum adsorption capacity of atrazine was 8.8 μmol/g, and the adsorption process could be well described by the Langmuir isotherm model and pseudo‐second‐order kinetic model. The magnetic molecularly imprinted polymer exhibited good adsorption selectivity for atrazine with respect to structural analogues, such as cyanazine, simetryne, and prometryn. The reusability of the magnetic molecularly imprinted polymer was demonstrated for at least five repeated cycles without a significant decrease in adsorption capacity. These results suggested that the magnetic molecularly imprinted polymer could be used as an efficient material for the selective adsorption and removal of atrazine from water samples.  相似文献

This work reports the preparation of a molecularly imprinted polymer (MIP) for selective catalytic detection of serotonin (5-hydroxytryptamine, 5-HT). The process is based on the synthesis of polymers with hemin introduced as the catalytic center to mimic the active site of peroxidase. The copolymer MIP, containing artificial recognition sites for 5-HT, has been prepared by bulk polymerization using methacrylic acid (MAA) and hemin as the functional monomers, and ethylene glycol dimethacrylate (EGDMA) as the cross-linker. For the determination of 5-HT, a flow injection analysis system coupled to an amperometric detector was optimized using multivariate analysis. The effects of different parameters, such as pH, buffer flow rate, buffer nature, peroxide concentration and sample volume were evaluated. After optimizing the experimental conditions, a linear response range from 1.0 up to 1000.0 μmol L−1 was obtained with a sensitivity of 0.4 nA/μmol L−1. The detection limit was found to be 0.30 μmol L−1, while the precision values (n = 6) evaluated by relative standard deviation (R.S.D.) were, respectively, 1.3 and 1.7% for solutions of 50 and 750 μmol L−1 of 5-HT. No interference was observed by structurally similar compounds (including epinephrine, dopamine and norepinephrine), thus validating the good performance of the imprinted polymer. The method was applied for the determination of 5-HT in spiked blood serum samples.  相似文献
