1,2,3-Triazolo[4,5-d]-1,2,4-triazolo[4,3-b]pyridazines

Some new 1,2,3-triazolo[4,5-d]-1,2,4-triazolo[4,3-b]pyridazines were prepared starting from the corresponding 1,2,3-triazolo[4,5-d]pyridazines via the formation of the 1,2,4-triazole ring, by condensation of an appropriate monocarbon fragment with the 4-hydrazino substituent and the nitrogen atom in the 5 position of the heterocycle. Condensation of 4-phenylhydrazino substituted derivatives with formic acid gave zwitterionic compounds.     

Novel first-generation dendrimers on calix[4]resorcinol core equipped with multiple triazole units

Novel first-generation dendrimers on the calix[4]resorcinol core with four branches each containing multiple 1,2,3-triazole units have been synthesized in one-step by acid catalyzed condensation of resorcinols with a new aldehyde dendron, namely, 4-{3,5-bis[(1-benzyl-1H-1,2,3-triazol-4-yl)- methoxy]benzyloxy}benzaldehyde (obtained by alkyne–azide cycloaddition). The reaction proceeds stereoselectively to form rccc-diastereoisomers in high yields.

采用超声辐射法,以2-苯基-1,2,3-三唑-4-甲酰肼为原料,合成了3-(2-苯基-1,2,3-三唑-4-基)-5H-4-氧代噻唑[2,3-c].1,2,4-三唑,再与各种芳香醛进行Knoevenagel缩合反应,合成了一系列噻唑烷酮衍生物.所有目标化合物结构经元素分析,IR,1H NMR确证.     

Synthesis of 1-(4-methylsulfone-phenyl)-5-(4-fluoro-phenyl)-5-[14C]-1,2,3-triazole and 1-(4-sulfonamide-phenyl)-5-(4-fluoro-phenyl)-5-[14C]-1,2,3-triazole as novel carbon-14 anticonvulsant

Summary Two 1,2,3-triazole anticonvulsants, 1-(4-methylsulfone-phenyl)-5-(4-fluoro-phenyl)-5-[¹⁴C]-1,2,3-triazole and 1-(4-sulfonamide-phenyl)-5-(4-fluoro-phenyl)-5-[¹⁴C]-1,2,3-triazole, both labeled with carbon-14 in the 5-position were prepared from para-fluoro-benzonitrile-[cyano-¹⁴C].     

Regioselective synthesis of fluoroalkylated [1,2,3]-triazoles by Huisgen cycloaddition

A series of fluoroalkylated 1,4-disubstituted [1,2,3]-triazoles were synthesized by the 1,3-dipolar cycloaddition of fluoroalkylated azides with terminal alkynes in the presence of Cu(I) salt as catalyst at room temperature. All the reactions were performed in highly regioselective with 1,4-disubstituted, no 1,5-disubstituted product was formed. For aryl or alkyl-alkyne, triethylamine should be used as ligand. But for propiolic ester(amide), triethylamine couldn’t be used, otherwise no products was formed. A mechanism of Cu(I) inserting the internal alkyne was suggested.     

1,2,3-Triazolodiazepines. I. Preparation and benzodiazepine receptor binding of 1-benzyl- and 1-phenethyl-1,2,3-triazolo-[4,5-b][1,4]diazepines

Several new 1,2,3-triazolo[4,5-b][1,4]diazepines were prepared starting from 1-benzyl-1 and 1-phenethyl-4,5-diamino-1,2,3-triazole 2 (Scheme 1), by condensation reactions with β-diketones (Scheme 2), β-ketoesters (Scheme 3), and diethyl malonates (Scheme 4). In the first case we obtained compounds 3 and 4 with basic properties, while the ester function condensations gave cyclic amide derivatives 7, 8, 10, 12 and 13 with acid properties. Some N-methyl derivatives 11, 14 and 15 were obtained from the cyclic amide compounds. Most of compounds were tested for their ability to displace [³H]flunitrazepam from bovine brain membranes but no compound showed benzodiazepine receptor binding affinity.     

Synthesis of 3-(2-Phenylquinolin-4-yl)/3-(l-p-Chlorophenyl-5-methyl-1, 2,3-triazol-4-yl)-s-triazolo[3,4-b]-1, 3,4-thiadiazine Derivatives

s-Triazolo[3, 4-b]-1,3,4-thiadiazine derivatives constitute an important class of organic compounds with diverse biological activities. 2-Phenylquinoline and 1,2, 3-triazole derivatives are very attractive heterocyclic systems due to their wide use in medicine, agriculture and industry^[1]. Incorporation of 2-phenylquinoline and 1,2,3-triazole moiety into the 3-position of s-triazolo[3,4-b]-l, 3,4-thiadiazine ring system may enhance their biological activity. In light of the above findings, we synthesized some new s-triazolo[3, 4-b]-1, 3, 4-thiadiazine derivatives 2a-b~6a-b by the condensation of 4-amino-5-mercapto-3-(2-phenylquinolin-4-yl)/3-(1-p-chlorophenyl-5-methyl-1, 2,3-triazol-4-yl)-1, 2, 4-triazoles 1a-b with chloroacetalde-hyde, ω-bromo-ω-(1H-1, 2, 4-triazol-1-yl)acetophenone, chloranil, 2-bromocyclohexanone, 2, 4'-dibromoacetophenone, respectively.     

Synthesis and spectroscopic characterisation of 4,5-dihydro-[1,2,3]triazolo[5,1-f][1,2,4]triazines. A novel condensed heterocyclic ring system

The reaction of the 5-bromomethyl-1-dibenzoylamino-1,2,3-triazole ( 5 ) with aromatic amines gave the corresponding 5-arylaminomethyl-1-benzoylamino-1,2,3-triazoles 9 , which on treatment with phosphorus pentachloride gave the [1,2,3]triazolo[5,1-f][1,2,4]triazine derivatives 10 . Hydrolysis of the amide bond in 9 gave the 1-aminotriazoles 11 , which with ethyl orthoformate gave the [1,2,3]triazolo[5,1-f][1,2,4]triazines 12 , whereas with phosgene the triazolotriazinones 13 were obtained. The spectroscopic characteristics of all the new compounds are also given.     

Synthesis and thermal properties of novel calix[4]arene derivatives containing 1,2,3-triazole moiety via K2CO3-catalyzed 1,3-dipolar cycloaddition reaction

Two calix[4]arene derivatives containing 1,2,3-triazole moiety were synthesized via K2CO3-catalyzed1,3-dipolar cycloaddition reaction between calix[4]arene-based azide and active methylene compounds in good yields.The structures of the two compounds synthesized herein were fully confirmed by 1HNMR,,（13）C NMR,and MS（ESI）.The thermal analysis showed that the mass losses of the synthesized compounds 4 and 5 containing 1,2,3-triazole groups are similar to each other.     

Binding ability of Zn-tetraarylporphyrins with two,four and eight 4-(4-(3,6-bis(t-butyl)carbazol-9-ylphenyl)-1,2,3-triazole end groups towards N-containing substrates of different nature

The binding ability of Zn-tetraarylporphyrins with two, four and eight 4-carbazolylphenyl-1,2,3-triazole end groups towards 1,4-diazabicyclo[2.2.2]octane, pyridine and 1,2,3-triazole in toluene was studied by spectrophotometric and ¹H NMR titration. It was determined that due to a good geometric match of the ligand size to the size of the intramolecular cavities of the porphyrinic receptor, and by the existence of additional π–π and/or hydrogen bonding interactions between the ligand and the triazole fragments of the porphyrin, the Zn-tetraarylporphyrin with eight 4-carbazolylphenyl-1,2,3-triazole end groups could be used as an effective receptor for 1,2,3-triazole and other small heterocycles such as pyridine. Taking into account the fact that binding is accompanied by a clear and easily identifiable response in the UV–vis spectra of the reaction mixture, this metalloporphyrin could be considered as a molecular optical sensing device for small heterocyclic substrates.     

Synthesis of 6-(1-Aryl-5-methyl-1,2,3-triazol-4-yl)-3-(4-Pyridyl)-s-Triazolo[3,4-b]-l,3,4-Thiadiazoles

A survey of literature¹ revealed that s-triazolo[3,4-b]-1,3,4-thiadiazole, an interesting fused system of s-triazole and 1,3,4-thiadiazole rings, has received much attention during recent years on account of its prominent utilizations as antifungal, antiinflammatory, antiviral, analgesic and anthelmintic agents probably resulting from its planner and compact structure. Our earlier work on the synthesis of this class of heterocycles showed antibacterial, herbicidal and plant growth regulative properties^2-3 for the compounds. 1,2,3-Triazole derivatives have found their wide use in medicine, agriculture and industry^4-5. Incorporation of 1,2,3-triazole moiety into the 6-position of this ring system may lead to achieving compounds of better biological activities. In view of the above findings coupled with scanty reports on these condensed compounds carrying 6-heterocyclic groups, we wish to report here the condensation of 4-amino-5-mercapto-3-(4-pyridyl)-l,2,4-triazole(2) with 1-aryl-4-carboxy-5-methyl-1,2,3-triazoles(1_a-j) as a part of our continuing interest in this area.     

Chemical reactivity of [1,2,3]triazolo[1,5-a]- and [1,5-c]-pyrimidinium salts

The chemical reactivity of the [1,2,3]triazolo[1,5-a]- and [1,5-c]-pyrimidinium salts towards morpholine, water and sodium methoxide have been studied. Among others, new 1-aza and 2-azabutadienes substituted by a [1,2,3]-triazole ring were obtained in the course of the opening of the positively charged pyrimidine ring.

Synthesis and Characterization of Compounds Containing 1,2,3-Triazole via Click Reaction and Ag(Ⅰ) Complex

In this work, 1,4-bis(4-phenyl-1,2,3-triazole)benzene, 1,3-bis(4-phenyl-1,2,3-triazole)propane, bis(1-phenyl-1,2,3-triazole)-methylphenylsilane, and 1-ally-4-phenyl-1,2,3-triazole have been designed and synthesized via Click reaction. Fourier transform infrared spectroscopy(FT-IR) and nuclear magnetic resonance spectroscopy(NMR) were used to confirm the compounds' structures. The effect of silicon atom on the optical properties has also been studied. The UV-vis absorption wavelength of silicon-containing compound is about ca. 10 nm red-shifted when compared with that of other three compounds. The fluorescence emission bands of the compounds in CHCl_3 solutions were observed around ca. 440 nm. And the luminescent coordination compound, namely [AgL1?NO_3?3H_2O]n, based on the ligand 1-allyl-4-phenyl-1,2,3-triazole has been prepared. In addition, this complex exhibits a 1 D chain structure. The crystal structure has been determined by single-crystal X-ray diffraction, and the optical properties have been investigated by fluorescence spectrum. In summary, our work may provide new materials with luminescent property which is potentially useful in material fields.     

The synthesis of [1,2,3]-triazole-based bent core liquid crystals via microwave-mediated ‘Click Reaction’ and their mesomorphic behaviour

A series of [1,2,3]-triazole-based liquid crystal compounds were synthesised via a fast and efficient microwave-mediated ‘Click Reaction’ and their phase behaviour is presented. Most of the 1,4-diaryl-[1,2,3]-triazole compounds exhibited a nematic phase with broad temperature ranges and relatively low transition temperatures compared to previously examined materials. Structural variations, involving regio-isomers and lateral halogenation, gave rise to different phase behaviour of the two families of triazole-based compounds.     

Efficient multicomponent synthesis of highly substituted [1,2,3]triazolo[1,5-a]pyrimidines

Abstract  

An efficient and facile multicomponent procedure for the synthesis of highly substituted [1,2,3]triazolo[1,5-a]pyrimidines by reaction of equimolar amounts of aromatic aldehyde, CH-acid, and 5-amino-1,2,3-triazole in refluxing dimethylformamide is described.     

The synthesis of 1-biphenyl-4-alkyl-[1,2,3]-triazoles and their mesomorphic behaviour

A series of 1-biphenyl-4-alkyl-[1,2,3]-triazole-based liquid crystal compounds were synthesised via a fast and efficient microwave mediated ‘Click Reaction’ and their phase behaviour is presented. Most of the 1-(3-fluorobiphenyl)-4-alkyl-[1,2,3]-triazole compounds exhibited a smectic A phase with relatively low transition temperatures compared to previously examined aryl-triazole-based mesogens. Small angle X-ray scattering measurement indicated the existence of deep smectic phase for some cases. Structural variations gave rise to different phase behaviour in these triazole-based compounds.     

Photoisomerization of 2,3,6,7-tetraphenyl-1,2,3-triazolo[1,2-b]-1,2,3-triazole

Irradiation of the title compound brings about consecutive photoisomerizations via derivatives of 1,2,3-triazolyl-2H-azirine, 1,2,3-triazolo[1,2-a]-1,2,4-triazole and 1,2,4-triazolyl-2H-azirine, leading finally to a dihydrotetraazabenz [e] azulene.     

Trimethylsilyl-directed 1,3-dipolar cycloaddition reactions in the solid-phase synthesis of 1,2,3-triazoles

[reaction: see text] A regioselective method for the preparation of 1,5-trisubstituted 1H-1,2,3-triazoles via a 1,3-dipolar cycloaddition of 1-trimethylsilylacetylenes with organoazides is described. Immobilization of the azide on REM resin and subsequent cycloaddition afforded a 2 x 2 x 4 x 3 membered 1,5-disubstituted 1H-1,2,3-triazole library with an average purified yield of 68%.     

Intramolecular hydrogen bonding-assisted cyclocondensation of α-diazoketones with various amines: a strategy for highly efficient Wolff 1,2,3-triazole synthesis

A catalytic and highly efficient Wolff's cyclocondensation of α-diazoketones with aromatic and aliphatic amines has been realized for the first time by utilizing the strategy of an intramolecular hydrogen bonding-activating carbonyl group. This approach successfully solved the challenging problem of poor condensation efficiency in Wolff 1,2,3-triazole synthesis, and constitutes a powerful method for the synthesis of highly functionalized 1,2,3-triazoles.
