Phytochemistry of genus Gentiana. XXII. Identification of new O-glucosides of mangiferin in Gentiana asclepiadea L. Two new O-glucosides of mangiferin (mangiferin-7-O-β-D -glucoside ( 1 ) and mangiferin-6-O-β-D -glucoside ( 2 )) have been isolated from the leaves of Gentiana asclepiadea L . This is the first full structure elucidation of naturally occurring O-glucosides of C-glucosylxanthones. The known C-glucosylflavone, saponarin ( 5 ) was also identified.  相似文献

Phytochemistry of genus Gentiana XXI: The cinnamoyl-C-glucosylflavones and their O-glucosides in Gentiana punctata L . Three new ( 1 – 3 ) and three previously identified ( 4 – 6 ) cinnamoyl-C-glucosyl-flavones have been isolated from the leaves of Gentiana punctata L . The structures of the new compounds were established as: trans-cafeoyl-2″-iso-orientin-4′-O-β-D -glucoside ( 1 ), trans-feruloyl-2″-isovitexin-4′-O-β-D -glucoside ( 2 ) and trans-feruloyl-2″-isovitexin ( 3 ). Isoscoparine ( 8 ) and O-β-D -glucosyl-2″-iso-orientin ( 7 ) were also isolated and identified.  相似文献

New cinnamoyl-C-glucosyl-flavones Nine flavonic compounds: isoorientin ( 1 ), isovitexin ( 2 ), isoorientin-4′-O-β-D -glucoside ( 3 ), isovitexin-4′-O-β-D -glucoside ( 4 ), luteolin-7-O-β-D -glucoside ( 5 ), trans-cafeoyl-2′′-isoorientin ( 6 ), trans-feruloyl-2′′-isoorientin ( 7 ), trans-p-coumaroyl-2′′-isoorientin ( 8 ), p-O-β-D -glucosyl-trans-cafeoyl-2′′-isoorientin ( 9 ) and three xanthones: gentioside ( 10 ), isogentisine ( 11 ), mangiferin ( 12 ), have been identified from leaves of Gentiana X marcailhouana RY . Phytochemistry of genus Gentiana, XXIV. New C-glycosylflavones from the leaves of Gentiana asclepiadea L. p-Hydroxybenzoyl-2″-isoorientin-4′-O-β-D-glucoside ( 1 ) was isolated from the leaves of Gentiana asclepiadea L. by means of column chromatography. This is the first occurrence in nature of a p-hydroxybenzoate of isoorientin. p-Hydroxybenzoyl-2″-isoorientin ( 2 ) was also shown to be present in the same plant.  相似文献

Neomangiferin, a natural xanthone derivative bearing both O- and C-glucosides, was isolated from the leaves of Gentiana asclepiadea L. and has shown potential anti-diabetic activity. We describe herein the first semi-synthesis of neomangiferin from the natural C-glucoside mangiferin and glucose. The developed synthesis presents a facile protection strategy using Jurd’s method to distinguish the different phenolic hydroxyl groups. Following this strategy, the regioselective protection of 1,3,6-hydroxyl groups was accomplished and neomangiferin was prepared by glycosylation under the phase-transfer catalysis conditions.  相似文献

Five flavone C-glucosides [6-C-β-D -glucopyranosyl-tricine ( 1 ) or isopyrenine; 7-O-β-D -glucopyranosyl-6-C-β-D -glucopyranosyltricine ( 2 ) or isopyrenine-7-O-glucoside; isoscoparine-7-O-glucoside ( 3 ); 7-O-β-D -glucopyranosyl-6-C-β-D -glucopyranosylacacetine ( 4 ) and isovitexine-7-O-glucoside ( 5 ) or saponarine] as well as the flavonol glucoside isoquercitrine ( 9 ) have been isolated chromatographically, using polyamide and cellulose columns, from the aerial parts of Gentiana pyrenaica L . 1 and 2 are new compounds and the first glycoflavones trisubstituted on cycle B. One new iridoid glucoside, 4″‐Oβ‐D ‐glucosyl‐6′‐O‐(4‐Oβ‐D ‐glucosylcaffeoyl)linearoside ( 1 ), and two new secoiridoid glucosides, 6′‐O‐acetylsweroside ( 2 ) and 6′‐O‐acetyl‐3′‐O‐[3‐(β‐D ‐glucopyranosyloxy)‐2‐hydroxybenzoyl]sweroside ( 3 ), were isolated from the dried roots of Gentiana manshurica (Gentianaceae), together with 11 known ones, including one iridoid glucoside, five secoiridoid glucosides, and five triterpenes. The structures of the new compounds were determined on the basis of detailed spectroscopic analyses and acidic hydrolysis.  相似文献

Several naturally occurring xanthone-1-O-glycosides have been synthesized in order to study monoamine oxidase (MAO) inhibition structure-activity relationships. The syntheses also confirmed the structures as 1-β-D -glucosyloxy-3-hydroxy-5-methoxyxanthone (Canscora decussata SCHULT .), 1-O-β-primeverosyl-3, 7, 8-trimethoxyxanthone (decussatin-1-O-primeveroside, Gentiana verna L .) and 1-O-β-primeverosyl-3, 8-dimethoxy-7-hydroxyxanthone (gentiacaulein-1-O-β-primeveroside, Gentiana verna L .).  相似文献

To identify the structures of flavonoid glycosides in bee pollen collected from rapeseed plants (Brassica napus L.), we utilised an approach that combined liquid chromatography–diode array detector–electrospray ionization–mass spectrometry (LC–DAD–ESI–MS) and nuclear magnetic resonance (NMR) technology with a step-wise separation strategy. We identified four constituents of high purity in rape bee pollen samples: (1) quercetin-3-O-β-D-glucosyl-(2→l)-β-glucoside, (2) kaempferol-3, 4′-di-O-β-D-glucoside, (3) 5, 7, 4′-trihydroxy-3′-methoxyflavone-3-O-β-D-sophoroside and (4) kaempferol-3-O-β-D-glucosyl-(2→l)-β-D-glucoside. This study will also provide useful reference standards for qualification and quantification of four flavonoid glycosides in natural products.  相似文献

Two new xanthone-O-glycoside, the 1,3,5-trihydroxy-xanthone-8-O-β-D -glucopyranoside ( 3 ), the 1,5-dihydroxy-3-methoxyxanthone-8-O-β-D -glucopyranoside ( 4 ), have been isolated from the leaves of Gentiana campestris L . by means of column chromatography on polyamid. One new and six known pentacyclic triterpenoids, 1 and 4 – 9 , resp., along with two new and three known iridoid glycosides, 2 and 3 , and 10 – 12 , resp., were isolated from the roots of Gentiana dahurica (‘Qin‐Jiao’). The new structures were elucidated by means of spectroscopic and chemical methods as 1β,2α,3α,24‐tetrahydroxyursa‐12,20(30)‐dien‐28‐oic acid ( 1 ), 6′‐O‐acetylgentiopicroside ( 2 ), and 3′‐acetylgentiopicroside ( 3 ). Isolated compounds were tested for their cytotoxicity against the MCF‐7 human breast cancer cell line using the MTT assay. Among them, triterpenoids 2α,3β,24‐trihydroxyurs‐12‐en‐28‐oic acid ( 6 ) and 3β,24‐dihydroxyurs‐12‐en‐28‐oic acid ( 9 ) were found to have moderate cytotoxic effects with IC50 values of 20.9 and 21.7 μg/ml, respectively. Additionally, the chemotaxonomic significance of the identified secondary metabolites is briefly discussed.  相似文献

A new ellagic acid derivative, 3,3′-dimethylellagic acid-4′-O-(6″-galloyl)-β-d-glucoside, named runcinatside (5), together with four known compounds 3,3′-dimethylellagic acid (1), 3,3′,4′-trimethylellagic acid (2), 3,3′-dimethylellagic acid-4′-O-β-d-glucoside (3) and 3-methylellagic acid-4′-O-α-l-rhamno-pyranoside (4), was isolated from the roots of Polygonum runcinatum Buch.-Ham. ex D.Don Var. sinense Hemsl and the structures of these compounds were established by spectroscopic methods and comparison with previously reported data. All compounds showed antioxidant activities in vitro and compound 5 possessed the highest activity.  相似文献

Summary The aim of this work was to compare the chemical composition of the underground parts (roots and rhizomes) ofGentiana cruciata L.,Gentiana pneumonanthe L., andGentiana asclepiadea L.— the three gentians native to Hungary—with that of the widely used stomachicGentiana lutea L., to determine which of the three Hungarian species could be used as a substitute forGentiana lutea in pharmaceutical preparations. The four gentians were compared by means of RPHPLC with diode-array detection (DAD) and electrospray ionization-mass spectrometric detection (ESI-MSD). The quantities of the lead compounds, the secoiridoid-glycosides, in 220 samples of the underground parts of gentians originating from several locations in Hungary, were determined by a more economical RPHPLC-DAD method. The occurrence of the characteristic compounds investigated—bitter principles and xanthones—in the underground parts of the speciesGentiana asclepiadea L. suggest it might be a potential replacement forGentiana lutea L. in pharmaceutical products. High-speed counter-current chromatography (HSCCC) was applied to the preparative isolation and purification of peonidin 3-O-(6-O-(E)-caffeoyl-2-O-β-D -glucopyranosyl-β-D -glucopyranoside)-5-O-β-D -glucoside ( 1 ), cyanidin 3-O-(6-O-p-coumaroyl)-β-D -glucopyranoside ( 2 ), peonidin 3-O-(2-O-(6-O-(E)-caffeoyl-β-D -glucopyranosyl)-6-O-(E)-caffeoyl-β-D -glucopyranoside)-5-O-β-D -glucopyranoside ( 3 ), peonidin 3-O-(2-O-(6-O-(E)-feruloyl-β-D -glucopyranosyl)-6-O-(E)-caffeoyl-β-D -glucopyranoside)-5-O-β-D -glucopyranoside ( 4 ) from purple sweet potato. Separation of crude extracts (200 mg) from the roots of purple sweet potato using methyl tert-butyl ether/n-butanol/acetonitrile/water/trifluoroacetic acid (1:4:1:5:0.01, v/v) as the two-phase solvent system yielded 1 (15 mg), 2 (7 mg), 3 (10 mg), and 4 (12 mg). The purities of 1 – 4 were 95.5%, 95.0%, 97.8%, and 96.3%, respectively, as determined by HPLC. Compound 2 was isolated from purple sweet potato for the first time. The chemical structures of these components were identified by 1H NMR, 13C NMR and ESI-MSn.  相似文献

The stereospecific cis-hydroxylation of 1-(2,3-dideoxy-β-D -glyceropent-2-enofuranosyl)thymine (1) into 1-β-D -ribofuranosylthymine (2) by osmium tetroxide is described. Treatment of 2′,3′-O, O-isopropylidene-5-methyl-2,5′-anhydrouridine (8) with hydrogen sulfide or methanolic ammonia afforded 5′-deoxy-2′,3′-O, O-isopropylidene-5′-mercapto-5-methyluridine (9) and 2′,3′-O, O-isopropylidene-5-methyl-isocytidine (10) , respectively. The action of ethanolic potassium hydroxide on 5′-deoxy-5′-iodo-2′,3′-O, O-isopropylidene-5-methyluridine (7) gave rise to the corresponding 1-(5-deoxy-β-D -erythropent-4-enofuranosyl)5-methyluracil (13) and 2-O-ethyl-5-methyluridine (14) . The hydrogenation of 2 and its 2′,3′-O, O-isopropylidene derivative 4 over 5% Rh/Al2O3 as catalyst generated diastereoisomers of the corresponding 5-methyl-5,6-dihydrouridine ( 17 and 18 ).  相似文献

By means of column chromatography on polyamide, we have isolated from the leaves of Gentiana campestris L. a new xanthone-O-glucoside, the 3,4-dimethoxy-5, 8-dihydroxy-xanthone-1-O-β-D-glucopyranoside ( 2 ) and its aglucone, the 3,4-dimethoxy-1,5,8-trihydroxy-xanthone ( 1 ).
