Gold(I) N‐heterocyclic carbene (AuI‐NHC) complexes have emerged as potential anticancer agents owing to their high cytotoxicity and stability. Integration of their above unique functions with customized aggregation‐induced emission (AIE) luminogens to achieve specific bioimaging and efficient theranostics to cancer is highly desirable but is rarely studied. Now, a series of novel AuI‐NHC compounds were developed with AIE characteristics. A complex with a PPh3 ligand was selected out as it could achieve both prominent specific imaging of various cancer cells and efficient inhibition of their growth with negligible toxic effects on normal cells due to the targeting binding and strong inhibition towards thioredoxin reductase. This complex could also act as a powerful radiosensitizer to boost the anticancer efficacy with performance superior to that of popularly used auranofin. It holds great potential as a specific and effective theranostic drug in cancer diagnosis and precise therapy.  相似文献

This communication reveals the impact of surface AuI‐thiolate motifs on the AIE properties of Au NCs, by employing a series of water‐soluble glutathione (GSH)‐coordinated Au complexes and NCs as a model ([Au10SR10], [Au15SR13], [Au18SR14], and [Au25SR18]?, SR=thiolate ligand). Spectroscopic investigations show that the emission wavelength of Au NCs is adjustable from visible to the near‐infrared II (NIR‐II) region by controlling the length of the AuI‐SR motifs on the NC surface. Decreasing the length of AuI‐SR motifs also changes the origin of cluster luminescence from AIE‐type phosphorescence to Au0‐core‐dictated fluorescence. A deep understanding on the luminescence property of aggregation‐induced emission (AIE) featured metal nanoclusters (NCs) is highly desired. This paper reports a systematic study on enhancing the luminescence of AIE‐featured Au NCs, which is achieved by Ag doping to engineer the size/structure and aggregation states of the AuI‐thiolate motifs in the NC shell. Moreover, by prolonging the reaction time, the luminescence of the as‐synthesized AuAg NCs could be further tailored from orange to red, which is also due to the variation of the AuI‐thiolate motifs of NCs. This study can facilitate a better understanding of this AIE‐featured luminescent probe and the design of other synthetic routes for this rising family of functional materials.  相似文献

The concept of aggregation‐induced emission (AIE) has been exploited to render non‐luminescent CuISR complexes strongly luminescent. The CuISR complexes underwent controlled aggregation with Au0. Unlike previous AIE methods, our strategy does not require insoluble solutions or cations. X‐ray crystallography validated the structure of this highly fluorescent nanocluster: Six thiolated Cu atoms are aggregated by two Au atoms (Au2Cu6 nanoclusters). The quantum yield of this nanocluster is 11.7 %. DFT calculations imply that the fluorescence originates from ligand (aryl groups on the phosphine) to metal (CuI) charge transfer (LMCT). Furthermore, the aggregation is affected by the restriction of intramolecular rotation (RIR), and the high rigidity of the outer ligands enhances the fluorescence of the Au2Cu6 nanoclusters. This study thus presents a novel strategy for enhancing the luminescence of metal nanoclusters (by the aggregation of active metal complexes with inert metal atoms), and also provides fundamental insights into the controllable synthesis of highly luminescent metal nanoclusters.  相似文献

The incorporation of an N-heterocyclic carbene (NHC) moiety into a self-assembled MII4L6 cage framework required the NHC first to be metallated with gold(i). Bimetallic cages could then be constructed using zinc(ii) and cadmium(ii) templates, showing weak luminescence. The cages were destroyed by the addition of further gold(i) in the form of AuI(2,4,6-trimethoxybenzonitrile)2SbF6, which caused the reversibly-formed cages to disassemble and controllably release the AuI-NHC subcomponent into solution. This release in turn induced the growth of gold nanoparticles. The rate of dianiline release could be tuned by capsule design or through the addition of chemical stimuli, with different release profiles giving rise to different nanoparticle morphologies.  相似文献

The synthesis of tetranuclear gold complexes, a structurally unprecedented octanuclear complex with a planar [AuI8] core, and pentanuclear [AuI4MI] (M=Cu, Ag) complexes is presented. The linear [AuI4] complex undergoes C?H functionalization of carbonyl compounds under mild reaction conditions. In addition, [AuI4AgI] catalyzes the carbonylation of primary amines to form ureas under homogeneous conditions with efficiencies higher than those achieved by gold nanoparticles.  相似文献

The synthesis, reactivity, and potential of well‐defined dinuclear gold complexes as precursors for dual gold catalysis are explored. Using the preorganizing abilities of the ditopic PNHPiPr ( LH ) ligand, dinuclear AuI–AuI complex 1 and mixed‐valent AuI–AuIII complex 2 provide access to structurally characterized chlorido‐bridged cationic species 3 and 4 upon halide abstraction. For 2 , this transformation involves unprecedented two‐electron oxidation of the redox‐active ligand, generating a highly rigidified environment for the Au2 core. Facile reaction with phenylacetylene affords the σ,π‐activated phenylacetylide complex 5 . When applied in the dual gold heterocycloaddition of a urea‐functionalized alkyne, well‐defined precatalyst 3 provides high regioselectivities for the anti‐Markovnikov product, even at low catalyst loadings, and outperforms common mononuclear AuI systems. This proof‐of‐concept demonstrates the benefit of preorganization of two gold centers to enforce selective non‐classical σ,π‐activation with bifunctional substrates.  相似文献

A new class of emissive cyclometallated IrIII−AuI complexes with a bis(diphenylphosphino) methanide bridging ligand was successfully synthesised from the diphosphino complex [Ir(N^C)2(dppm)]+ ( 1 ). The different gold ancillary ligand, a triphenylphosphine ( 2 ), a chloride ( 3 ) or a thiocytosine ( 4 ) did not reveal any significant effect on the photophysical properties, which are mainly due to metal-to-ligand charge-transfer (3MLCT) transitions based on IrIII. However, the AuI fragment, along with the ancillary ligand, seemed crucial for the bioactivity in A549 lung carcinoma cells versus endothelial cells. Both cell types display variable sensitivities to the complexes (IC50=0.6–3.5 μM). The apoptotic pathway is activated in all cases, and paraptotic cell death seems to take place at initial stages in A549 cells. Species 2 – 4 showed at least dual lysosomal and mitochondrial biodistribution in A549 cells, with an initial lysosomal localisation and a possible trafficking process between both organelles with time. The bimetallic IrIII−AuI complexes disrupted the mitochondrial transmembrane potential in A549 cells and increased reactive oxygen species (ROS) generation and thioredoxin reductase (TrxR) inhibition in comparison with that displayed by the monometallic complex 1 . Angiogenic activity assays performed in endothelial cells revealed the promising antimetastatic potential of 1 , 2 and 4 .  相似文献

Metal–metal bonding interactions have been employed as an efficient strategy to generate a number of unique gold(I) metallo‐macrocycles with fascinating functions. The self‐assembly, crystal structure and emission property of novel nest‐like tetramer 14 , namely, {[Au4(μ‐dppm)2(μ‐dctp2?)](BF4)2}4 ? (CH3CN)2 (dppm=bis(diphenylphosphino)methane, dctp2?=N,N′‐bis(dicarbodithioate)‐2,11‐diaza[3.3]paracyclophane) is reported. The complex has been characterized by single‐crystal X‐ray diffraction analysis, 1H NMR spectroscopy, 13C NMR spectroscopy, and CSI‐MS spectrometry. The aggregate demonstrates the sixteen gold(I) atoms are arranged in a ring with a circumference of 50.011(68) Å generated by AuI???AuI attractions. UV/visible and luminescence spectroscopy revealed that this AuI???AuI bonded metallo‐macrocycle exhibited yellow phosphorescence.  相似文献

The zinc finger protein tristetraprolin (TTP) regulates inflammation by downregulating cytokine mRNAs. Misregulation results in arthritis, sepsis and cancer, and there is an interest in modulating TTP activity with exogenous agents. Gold has anti-inflammatory properties and has recently been shown to modulate the signaling pathway that produces TTP, suggesting that TTP may be a target of gold. The reactivity of [AuIII(terpy)Cl]Cl2 with TTP was investigated by UV/Vis spectroscopy, spin-filter inductively coupled plasma mass spectrometry, X-ray absorption spectroscopy and native electrospray ionization mass spectrometry. AuIII was found to replace zinc in the protein active site in the reduced AuI form, with the AuI ion coordinated to two cysteine residues in a linear geometry. The replacement of ZnII with AuI results in loss of both secondary structure and RNA binding function. In contrast, when ZnIITTP is bound to its RNA target, no replacement of ZnII with AuI is observed, even in the presence of excess AuIIIterpy. This discovery of differential reactivity of gold with TTP versus TTP/RNA offers a potential strategy for selective targeting with gold complexes to control inflammation.  相似文献

Materials exhibiting excitation wavelength-dependent photoluminescence (Ex-De PL) in the visible region have potential applications in bioimaging, optoelectronics and anti-counterfeiting. Two multifunctional, chiral [Au(NHC)2][Au(CN)2] (NHC=(4R,5R)/(4S,5S)-1,3-dimethyl-4,5-diphenyl-4,5-dihydro-imidazolin-2-ylidene) complex double salts display Ex-De circularly polarized luminescence (CPL) in doped polymer films and in ground powder. Emission maxima can be dynamically tuned from 440 to 530 nm by changing the excitation wavelength. The continuously tunable photoluminescence is proposed to originate from multiple emissive excited states as a result of the existence of varied AuI⋅⋅⋅AuI distances in ground state. The steric properties of the NHC ligand are crucial to the tuning of AuI⋅⋅⋅AuI distances. An anti-counterfeiting application using these two salts is demonstrated.  相似文献

The use of weak bases and mild conditions is currently the most sustainable and attractive synthetic approach for the preparation of late-transition metal complexes, some of which are widely used in catalysis, medicinal chemistry and materials science. Herein, the use of cuprate, aurate or palladate species for a continuous flow preparation of CuI, AuI and PdII-NHC complexes is reported. All reactions examined proceed under extremely mild conditions and make use of technical grade acetone as solvent. The scalability of the process was exemplified in a multigram-scale synthesis of [Cu(IPr)Cl].  相似文献

Supramolecular ensembles adopting ring‐in‐ring structures are less developed compared with catenanes featuring interlocked rings. While catenanes with inter‐ring closed‐shell metallophilic interactions, such as d10–d10 AuI–AuI interactions, have been well‐documented, the ring‐in‐ring complexes featuring such metallophilic interactions remain underdeveloped. Herein is described an unprecedented ring‐in‐ring structure of a AuI‐thiolate Au12 cluster formed by recrystallization of a AuI‐thiolate Au10 [2]catenane from alkane solvents such as hexane, with use of a bulky dibutylfluorene‐2‐thiolate ligand. The ring‐in‐ring AuI‐thiolate Au12 cluster features inter‐ring AuI–AuI interactions and underwent cluster core change to form the thermodynamically more stable Au10 [2]catenane structure upon dissolving in, or recrystallization from, other solvents such as CH2Cl2, CHCl3, and CH2Cl2/MeCN. The cluster‐to‐cluster transformation process was monitored by 1H NMR and ESI‐MS measurements. Density functional theory (DFT) calculations were performed to provide insight into the mechanism of the “ring‐in‐ring? [2]catenane” interconversions.  相似文献

The compound [(μ‐dppp)(AuCl)2], previously reported to associate intermolecularly in a chain (catena) structure through AuI–AuI interactions (3.316Å), was obtained from gold(III) precursors in a cyclo form with shortened intramolecular AuI—AuI contacts at 3.237Å and a puckered AuPCCCPAu seven‐membered ring. DFT calculations using a large relativistic basis to account for the d10–d10 interaction reproduce the observed molecular structure in the crystal of this “linkage isomer”, including the conspicuous distortion at one of the gold atoms. The chelate complex [(dppp)PtCl2] was crystallized and structurally characterized as the dichloromethane solvate.  相似文献
