This study was undertaken to explore gel permeation chromatography (GPC) for estimating molecular weights of proanthocyanidin fractions isolated from sainfoin (Onobrychis viciifolia). The results were compared with data obtained by thiolytic degradation of the same fractions. Polystyrene, polyethylene glycol and polymethyl methacrylate standards were not suitable for estimating the molecular weights of underivatized proanthocyanidins. Therefore, a novel HPLC-GPC method was developed based on two serially connected PolarGel-L columns using DMF that contained 5% water, 1% acetic acid and 0.15 M LiBr at 0.7 ml/min and 50 °C. This yielded a single calibration curve for galloyl glucoses (trigalloyl glucose, pentagalloyl glucose), ellagitannins (pedunculagin, vescalagin, punicalagin, oenothein B, gemin A), proanthocyanidins (procyanidin B2, cinnamtannin B1), and several other polyphenols (catechin, epicatechin gallate, epicallocatechin gallate, amentoflavone). These GPC predicted molecular weights represented a considerable advance over previously reported HPLC-GPC methods for underivatized proanthocyanidins.  相似文献

通过偶氮反应将氨基苯硼酸共价连接到聚苯乙烯载体上,制备了硼酸基聚苯乙烯载体.研究了实验条件对载体吸附果糖能力及载体分离柱分离效果的影响,确定了最佳洗脱条件.以长径比为175:1的分离柱,于室温下用pH6.1的二次水以0.16mL/min的流速进行洗脱,成功地将42%的果葡糖分离,果糖纯度达到99%.  相似文献

Analysis of glucose and other carbohydrates are often performed by use of normal phase HPLC methods with acetonitrile as major eluent coupled with evaporative light-scattering detector (ELSD) or by use of anion-exchange ion chromatography (IC) methods with NaOH as eluent coupled with pulsed amperimetric electrochemical detector. In this work, a novel method for the determination of carbohydrates by IC in conjunction with a self-regenerating suppressor and an ELSD detector was investigated. Three carbohydrates (glucose, fructose, and sucrose) were separated using a KOH eluent generator to avoid the effect of carbon dioxide absorption in the alkaline eluent. Due to the use of the suppressor, non-volatile components were removed and a low salt background (K+ approximately 0.070 microg/mL) can be obtained so the suppressed eluent could directly go into an ELSD detector without obvious interference of inorganic salts. After examining the changes in retention and resolution, an optimized method was established (for IC: using 32 mM KOH as the eluent at a flow rate of 1 mL/min; for ELSD: operated at 95 degrees C, 4.0 bar nitrogen with a gas flow rate of 2.0 L/min) and the linearity, reproducibility, and the limit of detection (LOD) for the three carbohydrates were further evaluated. Regression equations revealed acceptable linearity (correlation coefficients=0.994-0.998) across the working-standard range (100-1000 microg/mL for glucose and sucrose, 150-1000 microg/mL for fructose) and LODs of glucose, fructose, and sucrose were 93, 126, and 90 microg/mL, respectively. This method has successfully been applied to the determination of the three carbohydrates in carbonated cola drinks and fruit juices. The recoveries were between 95 and 113% (n=3) for different carbohydrates.  相似文献

A molybdate-reducing bacterium has been locally isolated. The bacterium reduces molybdate or Mo6+ to molybdenum blue (molybdate oxidation states of between 5+ and 6+). Different carbon sources such as acetate, formate, glycerol, citric acid, lactose, fructose, glucose, mannitol, tartarate, maltose, sucrose, and starch were used at an initial concentration of 0.2% (w/v) in low phosphate media to study their effect on the molybdate reduction efficiency of bacterium. All of the carbon sources supported cellular growth, but only sucrose, maltose, glucose, and glycerol (in decreasing order) supported molybdate reduction after 24 h of incubation. Optimum concentration of sucrose for molybdate reduction is 1.0% (w/v) after 24 h of static incubation. Ammonium sulfate, ammonium chloride, valine, OH-proline, glutamic acid, and alanine (in the order of decreasing efficiency) supported molybdate reduction with ammonium sulfate giving the highest amount of molybdenum blue after 24 h of incubation at 0.3% (w/v). The optimum molybdate concentration that supports molybdate reduction is between 15 and 25 mM. Molybdate reduction is optimum at 35 °C. Phosphate at concentrations higher than 5 mM strongly inhibits molybdate reduction. The molybdenum blue produced from cellular reduction exhibits a unique absorption spectrum with a maximum peak at 865 nm and a shoulder at 700 nm. The isolate was tentatively identified as Serratia marcescens Strain Dr.Y6 based on carbon utilization profiles using Biolog GN plates and partial 16s rDNA molecular phylogeny.  相似文献

A new bioprocess for production of sorbitol and gluconic acid from two low-cost feedstocks, inulin and cassava starch, using a commercially available enzyme was proposed in this study. The commercial glucoamylase GA-L NEW from Genencor was found to demonstrate a high inulinase activity for hydrolysis of inulin into fructose and glucose. The glucoamylase was used to replace the expensive and not commercially available inulinase enzyme for simultaneous saccharification of inulin and starch into high titer glucose and fructose hydrolysate. The glucose and fructose in the hydrolysate were converted into sorbitol and gluconic acid using immobilized whole cells of the recombinant Zymomonas mobilis strain. The high gluconic acid concentration of 193 g/L and sorbitol concentration of 180 g/L with the overall yield of 97.3 % were obtained in the batch operations. The present study provided a practical production method of sorbitol and gluconic acid from low cost feedstocks and enzymes.  相似文献

建立了高效液相色谱法同时分析菊芋发酵液中的乙醇和有机酸的方法.采用HPLC有机酸分析柱, 流动相为0.01 mol/L H2SO4,流速为0.5 mL/min, 以紫外和示差折光检测器作为双通道检测手段,同时对克鲁维酵母菊芋发酵液中的柠檬酸、α-酮戊二酸、葡萄糖、丙酮酸、果糖、琥珀酸、乳酸、甘油、乙酸、乙醇进行了定量分析,本方法的回收率为95.8%~109.6%;RSD为0.33%~4.0%,结果表明,本方法简单、快速、准确,适用于监控克鲁维酵母发酵产物并指导整个发酵过程条件的优化.  相似文献

In combination with non-corrosive and low-toxic boric acid, AlCl3 · 6H2O was found to be effective for the synthesis of 5-hydroxymethylfurfural (5-HMF) from glucose. In this work, a 5-HMF yield of ≈ 60 % was obtained at 170°C for 40 min in a H2O/THF biphasic solvent mixture. An addition of NaCl not only improved the partition coefficients but also inhibited by-product formation. THF was identified as an ideal extraction solvent in biphasic systems containing C4 solvents. However, low concentration of ZnCl2, CoCl2 · 6H2O, MnCl2 · 4H2O, NiCl2 · 6H2O, FeCl3 · 6H2O were not suitable for the catalyst system, while ZrOCl2 · 8H2O, InCl3 · 4H2O showed high activity for the reaction. Boric acid increased the amount of Lewis acid sites in the reactive phase and enhanced the isomerization of glucose to fructose. A mechanism of the AlCl3 · 6H2O and boric acid catalyzed glucose dehydration reaction was proposed to proceed through the isomerization of glucose to fructose followed by the transformation of fructose to 5-HMF.  相似文献

建立了同时分析鲜红枣中果糖、葡萄糖和蔗糖的高效液相色谱方法.利用鲜红枣样品进行添加回收实验,获得方法对果糖、葡萄糖和蔗糖测定的平均回收率分别为93.5%、91.8%和89.8%,变异系数3.27%、2.34%、5.86%.结果表明:采用高效液相色谱示差分析方法定量检测鲜红枣中果糖、葡萄糖和蔗糖快速、简便.同时得出方法的检出限为:果糖5.73×10-3mg/mL,葡萄糖5.22×10-3mg/mL,蔗糖3.59×10-3mg/mL,测定结果符合食品检测要求.  相似文献

For optimum fermentation, fermenting xylose into acetic acid by Clostridium thermoaceticum (ATCC 49707) requires adaptation of the strain to xylose medium. Exposed to a mixture of glucose and xylose, it preferentially consumesxylose over glucose. The initial concentration of xylose in the medium affects the final concentration and the yield of acetic acid. Batch fermentation of 20 g/L of xylose with 5g/L of yeast extract as the nitrogen source results in a maximum acetate concentration of 15.2 g/L and yield of 0.76 g of acid/g of xylose. Corn steep liquor (CLS) is a good substitute for yeast extract and results in similar fermentation profiles. The organism consumes fructose, xylose, and glucose from a mixture of sugars in batch fermentation. Arabinose, mannose, and galactose are consumed only slightly. This organism loses viability on fed-batch operation, even with supplementation of all the required nutrients. In fed-batch fermentation with CSL supplementation, d-xylulose (an intermediate in the xylose metabolic pathway) accumulates in large quantities.  相似文献

A low, but significant, fraction of the carbohydrate portion of herbaceous biomass may be composed of fructose/fructosyl-containing components (“fructose equivalents”); such carbohydrates include sucrose, fructooligosaccharides, and fructans. Standard methods used for the quantification of structural-carbohydrate-derived neutral monosaccharide equivalents in biomass are not particularly well suited for the quantification of fructose equivalents due to the inherent instability of fructose in conditions commonly used for hemicellulose/cellulose hydrolysis (>80% degradation of fructose standards treated at 4% sulfuric acid, 121°C, 1 h). Alternative time, temperature, and acid concentration combinations for fructan hydrolysis were considered using model fructans (inulin, β-2,1, and levan, β-2,6) and a grass seed straw (tall fescue, Festuca arundinacea) as representative feedstocks. The instability of fructose, relative to glucose and xylose, at higher acid/temperature combinations is demonstrated, all rates of fructose degradation being acid and temperature dependent. Fructans are shown to be completely hydrolyzed at acid concentrations well below that used for the structural carbohydrates, as low as 0.2%, at 121°C for 1 h. Lower temperatures are also shown to be effective, with corresponding adjustments in acid concentration and time. Thus, fructans can be effectively hydrolyzed under conditions where fructose degradation is maintained below 10%. Hydrolysis of the β-2,1 fructans at temperatures ≥50°C, at all conditions consistent with complete hydrolysis, appears to generate difructose dianhydrides. These same compounds were not detected upon hydrolysis of levan, sucrose, or straw components. It is suggested that fructan hydrolysis conditions be chosen such that hydrolysis goes to completion; fructose degradation is minimized, and difructose dianhydride production is accounted for.  相似文献

We examined the deboronation reaction of 4‐mercaptophenylboronic acid (4MPBA) via fructose and glucose on silver surfaces by means of surface‐enhanced Raman scattering (SERS) at the excitation wavelengths of 488, 514, and 633 nm. The SERS spectra on silver nanoparticles clearly exhibited specific spectral signatures of thiophenol (TP) peaks, indicating a deboronation reaction of 4MPBA on the surfaces, whereas no strong TP peaks were observed on gold nanoparticles. The vibrational bands at 417, 999, 1021, and 1574 cm?1 in the Ag SERS spectra could correspond to the in‐plane aromatic ring modes in TP. X‐ray photoelectron spectroscopy also supported the surface reaction on Ag by referring the B1s peaks at ~193 eV. The ratiometric Raman measurements of the band at 1574 cm?1, with respect to that at 1587 cm?1, revealed fructose and glucose quantification in the concentration range of 1–10 mm . We did not identify such changes for mannose, sucrose, and sialic acid. The SERS peaks of 4MPBA on roughened Ag plates also exhibited TP bands to show the time‐dependent spectral change. Our findings indicate that the deboronation of 4MPBA and conjugation with fructose and glucose may be facilitated efficiently on silver surfaces for their quantification. A novel boronic acid and carboxyl-modified glucose molecularly imprinted polymer were prepared through suspension polymerization, which is based on 1.0 mmol glucose as a template, 1.2 mmol methacrylamidophenylboronic acid, and 6.8 mmol methacrylic acids as monomers, 19 mmol ethyleneglycol dimethacrylate, and 1 mmol methylene-bis-acrylamide as crosslinkers. The prepared glucose-molecularly imprinted polymer had a particle size of 25–70 μm, and was thermally stable below 215°C, with a specific surface area of 174.82 m2/g and average pore size of 9.48 nm. The best selectivity between glucose and fructose was 2.71 and the maximum adsorption capacity of glucose- molecularly imprinted polymer was up to 236.32 mg/g which was consistent with the Langmuir adsorption model. The similar adsorption abilities in six successive runs and the good desorption rate (99.4%) verified glucose-molecularly imprinted polymer could be reused. It was successfully used for extracting glucose from cellulose hydrolysis. The adsorption amount of glucose was 2.61 mg/mL and selectivity between glucose and xylose reached 4.12. A newly established chromatography (glucose-molecularly imprinted polymer) mediated hollow fiber membrane method in time separated pure glucose from cellulose hydrolysates on a large scale, and purified glucose solution with a concentration of 3.84 mg/mL was obtained, which offered a feasible way for the industrial production of glucose from cellulose hydrolysates.  相似文献

A side-by-side diffusion cell was used to study the permeability of glucose through a temperature- and pH-sensitive poly(N-isopropylacrylamide-co-methacrylic acid) hydrogel membrane. At fixed temperature (37°C), lowering pH in one side of the cell induced hydrogel volume collapse and strongly attenuated glucose permeation across the membrane. Hysteresis was observed in the glucose permeability versus pH characteristic.  相似文献

Zirconyl chloride upon hydrolysis in water to form Zr(OH)+ has been found to react to form a fluorescent derivative with not only a ketose such as fructose but also a hexose such as glucose and the disaccharide sucrose. When reaction conditions such as a temperature of 99°C and a time of 60 min are used, detection limits below 1 μg/mL are possible. All three zirconyl–sugar derivatives show very similar absorbance and fluorescence spectra, indicating a common mechanism involving formation of an enediol which can be complexed with ZrOH+ is likely. Because the reactivity order is glucose < sucrose < fructose, the reaction can be made selective for fructose at a lower reaction temperature and time such as 60°C at 5 min. Because interference from ascorbic acid and caffeine is also avoided, the fluorescent determination of fructose in soft drink samples after simply a dilution step is possible. We have also employed this reaction for flow injection analysis (FIA) using a polystyrene–divinylbenzene-packed HPLC column as a mixing device. Using a 0.01 M HClO4 with 1% zirconyl chloride carrier, we obtained a linear calibration curve from 2 to 30 μg/mL with a correlation coefficient of 0.994. A detection limit less than 2 μg/mL was possible. A comparison of results for the FIA of soft drinks with the enzymatic method involving fructose-5-dehydrogenase confirmed the FIA method was quite specific for fructose.  相似文献

利用乙醇、果糖和葡萄糖在波数为14000-7500cm^-1范围内的吸收值,在近红外谱区用偏最小二乘法(PLS)对体系进行建模,并通过内部和外部样品校验确认最佳数据预处理方法。结果表明,乙醇在0.05-0.25L/L,果糖在0.01-0.05g/mL及葡萄糖在0.005-0.009g/mL的范围内外部校验模型较好的分别是在PLS中所用的一阶偏导(平滑点数是5)、二阶偏导(平滑点数是5)和直线消除法的数据预处理方法所建立的模型,其外部校验预测结果的相对误差在2%左右。此方法具有可同时测定不同样品,简便快速及成本低等优点。  相似文献

A green analytical method was developed for the analysis of sugar-based depilatories. Three independent partial least squares (PLS) regression models were built for the direct determination of glucose, fructose and maltose without any sample pretreatment based on their attenuated total reflectance - Fourier transform infrared (ATR-FTIR) spectra. The models showed adequate prediction capabilities with root-mean-square-errors of prediction ranging from 7.04 to 12.55 mg sugar g−1 sample. As a reference procedure, gradient liquid chromatography with on-line infrared detection, employing background correction based on cubic smoothing splines, was used. The analysis revealed changes in the sugar concentration due to the formulation process as compared to information on the ingredients provided by the manufacturers. Although fructose, glucose and sucrose were declared to be used for the production of depilatories, in the final products only fructose, glucose and maltose were determined. This fact was attributed to pH and temperature conditions employed during the production process as well as to the use of glucose syrup instead of crystalline glucose. The present ATR-FTIR-PLS method enables an accurate, cheap and fast determination without solvent consumption or toxic waste generation and offers therefore a green screening alternative to methods employing chromatographic techniques.  相似文献

The effect of amaranth seeds on the lipid profile, glucose level, protein metabolism and selected trace element (Na, K, Ca and Mg) levels were determined in high-fructose fed Wistar rats. Fructose addition to rat fodder caused changes mainly in the blood lipid profile, particularly manifesting in an increased triglyceride level within all subsequent pairs of rat groups, and ranged between 85% and 112%. Administration of amaranth seeds to rats did not inhibit the increase of triglyceride induced by fructose. There was an increase in glucose concentration of between 3% and 14%. Uric acid concentrations also increased in all groups (30-37%), while changes in creatinine levels were varied. Fructose addition to fodder also brought about a significant decrease in alkaline phosphatase activity (9-20%).  相似文献

Sulfonic acid-functionalized heterogeneous catalysts have been evaluated in the catalytic dehydration of C(,monosaccharides into 5-hydroxymethylfurfural(HMF) using dimethyl sulfoxide(DMSO)as solvent.Sulfonic commercial resin Amberlyst-70 was the most active catalyst,which was ascribed to its higher concentration of sulfonic acid sites as compared with the other catalysts,and it gave 93 mol%yield of HMF from fructose in 1 h.With glucose as the starting material,which is a much more difficult reaction,the reaction conditions(time,temperature,and catalyst loading) were optimized for Amberlyst-70 by a response surface methodology,which gave a maximum HMF yield of 33 mol%at 147 °C with 23 wt%catalyst loading based on glucose and 24 h reaction time.DMSO promotes the dehydration of glucose into anhydroglucose,which acts as a reservoir of the substrate to facilitate the production of HMF by reducing side reactions.Catalyst reuse without a regeneration treatment showed a gradual but not very significant decay in catalytic activity.  相似文献

Cholesterol-lowering activity of the major polyphenols in grape seed   总被引:1,自引:0,他引:1  
The major polyphenols in grape seed have been shown to have beneficial health effects in the prevention of dyslipidemia and cardiovascular diseases. In this present study, we investigated the cholesterol-lowering activity of three major polyphenolic compounds found in grape seed. The results showed that gallic acid, catechin, and epicatechin significantly inhibited pancreatic cholesterol esterase in a concentration-dependent manner. Moreover, they bound to taurocholic acid, taurodeoxycholic acid, and glycodeoxycholic acid at levels ranging from 38.6% to 28.2%. At the concentration of 0.2 mg/mL, gallic acid, catechin, and epicatechin reduced the formation of cholesterol micelles 27.26 ± 2.17%, 11.88 ± 0.75%, and 19.49 ± 3.71%, respectively. These findings clearly demonstrate that three major polyphenolic compounds present in a particular grape seed have cholesterol-lowering activity by inhibiting pancreatic cholesterol esterase, binding of bile acids, and reducing solubility of cholesterol in micelles which may result in delayed cholesterol absorption.  相似文献

A spectrophotometric method for determining sucrose is proposed. Sucrose is hydrolyzed by invertase into glucose and fructose. Then, glucose is oxidized in presence of glucose oxidase and the produced hydrogen peroxide reacts with phenol-4-sulfonic acid sodium salt and 4-aminoantipyrine in presence of peroxidase, yielding a pink dye with an absorption maximum at 505 nm. This method was validated following the EURACHEM and VAM project guidelines for method validation. Trueness, precision, robustness, sensitivity and linearity were considered. The method was applied to the determination of sucrose in green and roasted coffee beans. A comparison with the HPLC method with pulsed amperometric detection was carried out.  相似文献
