本文采用拉曼光谱研究了Ni(Ⅱ)离子在高温和酸性条件下对鸡蛋清溶菌酶淀粉样纤维化动力学的影响.利用蛋白质三级和二级结构的拉曼光谱指针,检测分析了Ni(Ⅱ)离子对蛋白质三级结构展开和二级结构转化影响的浓度效应.结果证实了金属离子在动力学的加速作用.值得注意的是,通过对酰胺Ⅰ谱带的光谱分析并结合ThT荧光分析都表明,Ni(Ⅱ)离子对于形成有组织β-片层结构的淀粉样纤维有抑制作用,而在组装成其他无序结构的聚集体时,则会表现出显著的促进作用.本文为金属介导的蛋白质纤维化过程研究提供了参考信息. 相似文献

探针磨损是影响原子力显微镜图像质量的关键因素之一。为了研究原子力显微镜(AFM)探针针尖磨损问题,选取超平表面(R_q0.5 nm)作为测试样品,采用材料表面的粗糙度(R_q)评估探针针尖磨损效率。在较低的探针悬臂目标振幅比例(10%),较低反馈回路设定值比例(10%),较低扫描速度(1.0 Hz)以及适中I-gain(1.7)的测试条件下,探针的磨损最小,可以有效地延长探针的使用寿命。最后利用扫描电镜对比了不同测试条件下探针针尖的形貌,推测了探针针尖在连续测试中磨损与断裂的机理。 相似文献

β-Amyloid(Aβ) plaques and intracellular neurofibrillary lesions in the brain are markers of Alzheimer's disease(AD). The ability to safely decrease Aβ concentrations is potentially important as a preventive strategy for AD. The interactions between vanillin and Aβ polypeptide were investigated via fluorescence spectroscopy and atomic force microscopy(AFM). The results of fluorescence and synchronous spectroscopies illustrate that the intrinsic fluorescence of tyrosine(Tyr) residues in Aβ1-42 aggregates can be quenched strongly upon the formation of vanillin-Aβ1-42 complex. Thioflavine T(ThT)-induced fluorescence changes indicated that Aβ1-42 aggregates could be disaggregated by vanillin, and the AFM images of Aβ1-42 enunciated the depolymerization of Aβ1-42 aggregates by vanillin in a dose-dependent manner. Vanillin may be a potential pharmacological agent for the treatment of AD. 相似文献

The 8-hydroxyquinoline neodymium(Ndq3) organic thin films deposited on the cleaned indium/tin oxide (ITO) at different deposition rates with the same vacuity (133.3×10-5 Pa) were revealed by atomic force microscopy (AFM). Organic devices with one layer of Ndq3 as the e-type conductive material at different deposition rates sandwiched between ITO and aluminum electrodes have been fabricated. respectively. A novel roundish objects were observed directly by means of atomic force microscopy to be formed on dimyristoylphosphatidylcholoine (DMPC) Langmuir-Blodgeet bilayer films. 用原子力显微镜(AFM)对不同浓度下细胞色素C的分子形态,以及加入蛋白质降聚和变性剂脲后的形态变化进行了考察。实验结果显示,在50μmol/L的低浓度溶液中细胞色素C分子主要以直径为3nm的类球形单体形式存在。浓度增大,细胞色素C分子发生聚集,且随着浓度的进一步增大,细胞色素C分子倾向于形成更大的聚集体。浓度为200μmol/L时,聚集体分子间相互缠绕,形成链状结构。浓度低于0.8mol/L的脲的加入基本不影响细胞色素C分子的形态。加入较高浓度的脲,细胞色素C聚集体的聚集数降低,聚集体分子间没有明显的链状结构。 相似文献

Self-assembled monolayer of two azobenzene-derived alkanethiols with different terminal alkyl chain lengths (shortly as C8C3SH and C1C4SH) on gold were studied using atomic force microscopy. The C8C3SH SAMs showed a very regular packing structure with a lattice constant of a=0.57±0.04 nm, b=0.73±0.06 nm and θ=120±10°, mainly domianted by the terminated alkyl chains. On the other hand, the C1C4SH SAMs exhibited a number of different packing domains on the gold surface. The closest packing lattice was a=0.37±0.04 nm, b=0.47±0.02 nm, and θ=,107±6°, most probably representing the aggregation of azobenzene chromophoren. Another two typical lattices observed were a=0.65±0.03 nm, b=0.72±0.03 nm, θ=120±2°, and a=0.47±0.03 nm.b=0.47±0.03 nm.θ=100±10°, respectively. The packing structure of C1C4SH monolayers on gold is believed to be dominated by the azobenzene chromophore. 相似文献
