采用低压液相色谱法制备西红花有效成分西红花甙。最佳分离、制备条件 :填料为键合正丁烷的苯乙烯 二乙烯苯聚合物 ,其粒度为 57~ 76μm ;制备柱为 50cm× 1 .5cm ;纯化柱为 60cm×0 .80cm ;流动相为甲醇 水溶液 ,梯度洗脱范围是 50 / 50~ 95/ 5(V/V) ;样品量为 2 0mL( 1 0 0g/L) ;工作压力为 2× 1 0 5Pa。在上述条件下 ,制备了 5种西红花甙 ,经过高效液相色谱法测定 ,其纯度均在 97%以上。其中 ,西红花甙 1和西红花甙 5的纯度在 99%以上。此方法具有操作简便 ,制备纯度高且运行成本低等优点 ,在西红花对照品的制备中具有广泛的应用前景  相似文献

以E-1,4-二溴-2-丁烯和亚磷酸三乙酯为起始物, 通过Wittig-Horner反应合成化合物2,7-二甲基-2,4,6-辛三烯-1,8-二醛; 然后以E-2-甲基-2-丁烯酸为起始物经过溴代、酯化合成E-2-甲基-2-丁烯酸甲酯-4-膦酸二乙酯. 两化合物通过Wittig-Horner反应生成西红花酸二甲酯. 西红花酸二甲酯水解得西红花酸.  相似文献

西红花为鸢尾科植物番红花(Crocus sativus L)的干燥柱头.具活血化瘀,凉血解毒,解郁安神等功效.采用原子吸收分光光度法(AAS)测定西红花中部分金属元素的含量,通过对其宏量元素及微量元素含量的测定结果,分析了西红花中金属元素及其生理作用与药用功效之间的关系.随着元素医学的不断发展,金属元素与中药药效关系及对生物体系的作用有待研究.  相似文献

本文系统研究了硅胶和几种国产吸附树脂对西红花提取物溶液的吸附与洗脱性能,包括树脂的筛选,洗脱剂的选择,并研究了pH,温度和西红花提取物的浓度对西红花提取在SZ-1吸附树脂上的吸附影响,结果表明,SZ-1吸附树脂对西红花提取物的吸附能力较强,洗脱能力为最好,其吸附性能和洗脱性能均优于常用色谱固定相硅胶,SZ-1吸附树脂的静态吸附容量为1.04g/g,动态吸附容量为0.5-0.6g/g,洗脱率可达98%以上,且吸附速度快,易于洗脱和再生。  相似文献

建立表面增强拉曼光谱法快速检测西红花中诱惑红、日落黄、苋菜红、胭脂红、酸性橙Ⅱ5种酸性色素的方法,对于阳性样品采用液相色谱-串联质谱法确证。称取西红花样品0.1 g于15 mL离心管中,加入5 mL 50%甲醇水溶液,振荡30 min,以4 000 r/min离心10 min,用一次性吸管取出上清液于15 mL离心管中,于55℃氮吹至1 mL左右,加水至10 mL,涡旋混匀,作为样品溶液。取200μL胶体金表面增强试剂于拉曼样品池中,加入50μL凝聚剂Ⅰ(酸性色素试剂盒中试剂)混匀,再加入50μL样品溶液,混匀,于5 min内放入拉曼光谱仪检测。5种色素在西红花中的检出限:诱惑红、日落黄和苋菜红均为0.05 g/kg、胭脂红为0.5 g/kg、酸性橙Ⅱ为1 g/kg。该方法操作简单、快捷、准确,能够实现西红花中5种酸性色素的快速检测。  相似文献

使用密度泛函理论B3LYP方法,对西红花酸二甲酯的结构、电子吸收光谱、热力学性质进行理论计算研究,并基于Tomasi的极化统一场模型(PCM)讨论溶剂效应.结果显示,溶剂对西红花酸二甲酯的前线分子轨道特征几乎无影响,溶剂作用使该分子的最大吸收波长红移约42 nm,红移程度与溶剂极性无关.西红花酸二甲酯分子的气态热力学性质与温度的关系式分别为:Cm p=189.782+0.925T,S m=457.503+1.366T,H m=1164.899+0.195T+4.671×10-4T2,Gm=1164.899-0.263T-8.989×10-4T2.298 K时,西红花酸二甲酯分子的气态标准摩尔生成焓和标准摩尔生成自由能分别为-1180.67和-733.23 kJ·mol-1.  相似文献

建立高通量的高效液相色谱法同时检测15种西红花中非法添加色素,对于阳性样品采用液相色谱–质谱联用法确证。采用Poroshell 120 EC–C18色谱柱(100 mm×4.6 mm,2.7 μm)分离,以甲醇–0.02 mol/L乙酸铵溶液(用氨水调pH至5.9±0.05)进行梯度洗脱,流量为1.2 mL/min,柱温为35 ℃,以450 nm和500 nm双波长检测。缓冲盐pH值和流动相流量对色谱条件影响大,需严格控制。该方法重现性好,各物质分离度均大于1.5,标准曲线的线性相关系数r为0.998~1.000,样品高、中、低浓度的加标回收率为70.1%~93.1%,测定结果的相对标准偏差为1.2%~6.6%(n=6),检出限为0.01~0.05 mg/kg。该方法精密度良好,分析时间短,分析结果稳定,可用于中药材西红花中15种非法添加色素的快速筛查和含量测定,同时能为相关部门制定监督抽检中药材染色掺伪的补充检验方法。  相似文献

Background: Nanotechnology application has widespread use in many products. Copper nanoparticles (CuNPs) are widely used in industrial applications. The present study was conducted to investigate the effect of the ethanolic saffron extract (ESE) as a natural antioxidant on the hepatotoxicity induced by CuNPs in male mice. Methods: The characterization of CuNPs was determined using ultraviolet–visible absorption spectroscopy, particle size analysis, zeta potential, Fourier-transform infrared spectroscopy, and electron microscope. The effect of saffron on the hepatotoxicity induced by CuNPs in mice was evaluated by evaluating the survival rate of the mice, oxidative stress, antioxidant capacity, DNA evaluation, as well as its effect on the histology and transmission electron microscope of the liver. Results: The results revealed that all parameters were affected in a dose-dependent manner by CuNPs. These effects have been improved when the treatment of CuNPs is combined with ethanolic saffron extract. Conclusions: We can conclude that saffron and its bioactive crocin portion can prevent CuNP-induced oxidative liver damage. This substance should be useful as a new pharmacological tool for oxidative stress prevention.  相似文献

Saffron is a kind of expensive spice and therapeutic agent. In order to establish a set of methods to evaluate the quality of saffron, High Performance Liquid Chromatography/Mass Spectrometry (HPLC/MS) were used to identify the apocarotenoids extracted from saffron stigmas. TOPSIS method was applied to evaluate the qualities of saffron. The conditions of HPLC/MS and chemical fingerprints of saffron had been established. The 14 chromatographic peaks and 13 main apocarotenoids of saffron with high reproducibility, stability and specificity were confirmed. The qualities of 40 kinds of saffron from different countries and regions were ranked.  相似文献

Despite the beneficial health properties shown by Lebanese saffron, its qualitative and quantitative composition has never been investigated before. In the present study, NMR spectroscopy, together with antioxidant activity assays, were applied to evaluate the chemical composition of saffron samples of different geographical origins (Lebanon, Italy, Iran, and India) and to categorize the Lebanese saffron for the first time. The distinction between Lebanese saffron and that produced in other countries was attributed to its higher linolenic and linoleic fatty acids, glucose and picrocrocin contents. Moreover, spices produced in three different regions of the Lebanese territory have been clearly differentiated. Saffron cultivated in the Qaa region displayed a high glucose, fatty acids and polyphenols content, whereas Hermel saffron exhibited the largest rate of picrocrocin and glycosylated carotenoids. Finally, samples from Baalbeck showed lower rates for the majority of metabolites. Moreover, Lebanese saffron showed a high antioxidant activity in ABTS and DPPH assays. A low dose of saffron extract (10 µg/mL) inhibited the growth of human lung adenocarcinoma cells, probably due to the high polyphenolic content. This study highlights the quality and peculiarity of Lebanese saffron cultivated in Northern Beqaa district and allows for a good discrimination between spices produced in relatively close territory.  相似文献

A high-performance liquid chromatographic method previously developed (Lozano et al. in J Chromatogr A 830:477–483, 1999) for simultaneous detection, identification and quantification of the secondary metabolites in commercial saffron was extended for the detection of adulterated saffron prepared by adding styles colored with the natural colorants extracted from saffron petals, safflower, madder and red beet. The chromatograms of the methanol-water (50%, v/v) extracts of pure and adulterated saffron were obtained at the assayed wavelengths, 402 (or 254), 260 and 535 (or 440) nm and then by applying two-way analysis of variance (ANOVA) to the obtained data the presence of the styles colored with the colorant of safflower (>14.3%), styles colored with the colorant of madder (>9.1%) and styles colored with the colorant of red beet (>14.3%) in saffron were significantly detected. But the detection of adulterated saffron prepared with the colorant of saffron petals was not successful.  相似文献

An HPLC method to check components and purity in commercial saffron by photodiode array detection has been developed. The method was suitable for the standard analysis of commercial saffron. Therefore, 10 saffron metabolites responsible for the taste, flavour and colour were identified and quantified with high selectivity, precision and accuracy. Also, some artificial colorants, which can be used as adulterants, were also detected and identified. Three different saffron types were studied and their metabolite concentrations determined at different wavelengths.  相似文献

Background: Colorectal cancer (CRC) is one of the most common cancers worldwide. One of its subtypes is associated with defective mismatch repair (dMMR) genes. Saffron has many potentially protective roles against colon malignancy. However, these roles in the context of dMMR tumors have not been explored. In this study, we aimed to investigate the effects of saffron and its constituents in CRC cell lines with dMMR. Methods: Saffron crude extracts and specific compounds (safranal and crocin) were used in the human colorectal cancer cell lines HCT116, HCT116+3 (inserted MLH1), HCT116+5 (inserted MSH3), and HCT116+3+5 (inserted MLH1 and MSH3). CDC25b, p-H2AX, TPDP1, and GAPDH were analyzed by Western blot. Proliferation and cytotoxicity were analyzed by MTT. The scratch wound assay was also performed. Results: Saffron crude extracts restricted (up to 70%) the proliferation in colon cells with deficient MMR (HCT116) compared to proficient MMR. The wound healing assay indicates that deficient MMR cells are doing better (up to 90%) than proficient MMR cells when treated with saffron. CDC25b and TDP1 downregulated (up to 20-fold) in proficient MMR cells compared to deficient MMR cells, while p.H2AX was significantly upregulated in both cell types, particularly at >10 mg/mL saffron in a concentration-dependent manner. The reduction in cellular proliferation was accompanied with upregulation of caspase 3 and 7. The major active saffron compounds, safranal and crocin reproduced most of the saffron crude extracts’ effects. Conclusions: Saffron’s anti-proliferative effect is significant in cells with deficient MMR. This novel effect may have therapeutic implications and benefits for MSI CRC patients who are generally not recommended for the 5-fluorouracil-based treatment.  相似文献

The difficult cultivation of the saffron plant (Crocus Sativus L.) make the spice of the same name made from its dried stigmas very valuable. It is estimated that some 75,000 blossoms or 225,000 hand-picked stigmas are required to make a single pound of saffron, which explains why it is the world's most expensive spice. The aim of this study was to identify ways of increasing the fertility and production of saffron. For this purpose, the treatment of saffron bulbs with a synthetic growth hormone--a mixture of Polystimulins A6 and K--and two different microorganism based materials--biohumus or vermicompost and Effective Microorganisms (EM)--in four different ways (hormone alone, biohumus alone, EM alone and EM+biohumus) was investigated to determine whether these treatments have any statistically meaningful effects on corms and stigmas. It has been shown that EM + biohumus was the most effective choice for improved saffron cultivation.  相似文献

The metabolite profiling of saffron (Crocus sativus L.) from several countries was measured by using ultra-performance liquid chromatography combined with high resolution mass spectrometry (UPLC-HR MS). Multivariate statistical analysis was employed to distinguish among the several samples of C. sativus L. from Greece, Italy, Morocco, Iran, India, Afghanistan and Kashmir. The results of this study showed that the phytochemical content in the samples of C. sativus L. were obviously diverse in the different countries of origin. The metabolomics approach was deemed to be the most suitable in order to evaluate the enormous array of putative metabolites among the saffron samples studied, and was able to provide a comparative phytochemical screening of these samples. Several markers have been identified that aided the differentiation of a group from its counterparts. This can be important for the selection of the appropriate saffron sample, in view of its health-promoting effect which occurs through the modulation of various biological and physiological processes.  相似文献

The present study evaluated the reliability of the ISO/TS 3632-2 UV-Vis spectrometric method for saffron classification, making experiments on saffron samples to which were added increasing concentrations of common saffron spice adulterants (safflower, marigold and turmeric). The results showed that the ISO/TS 3632-2 method is not able to detect addition of up to 10-20%, w/w, of saffron adulterants. For additions from 20 to 50%, w/w, of the three adulterants, saffron was classified in a wrong category; addition of higher than 50%, w/w, determined variations in the investigated parameters that did not allow identification of the product as "saffron". In all cases, the method did not permit the recognition of the nature of the adulterant. On the contrary, the specificity of the HPLC/PDA/MS technique allowed the unequivocal identification of adulterant characteristic marker molecules that could be recognized by the values of absorbance and mass. The selection of characteristic ions of each marker molecule has revealed concentrations of up to 5%, w/w, for safflower and marigold and up to 2% for turmeric. In addition, the high dyeing power of turmeric allowed the determination of 2%, w/w, addition using exclusively the HPLC/PDA technique.  相似文献
