Several pyrazolo[3,4-b]pyridine (3,4) and pyrazolo-[3,4-d]pyrimidine (5-13) derivatives were prepared using 5-amino-l-(5-ethyl-5H-1,2,4-triazino[5,6-b]-indol-3-yl)-lH-pyrazole-4-carbonitrile (2) . The pyridine derivatives 3 and 4 were obtained by reaction of 2 with malononitrile and ethyl cyanoacetate, respectively, while pyrimidine analogs 5-13 were synthesized cither by a one-step or multi-step sequence.  相似文献

The reaction of cuprous acetylides with aryl halides bearing a nucleophilic ortho substituent provides a versatile route to heterocyclic substances. The present work portrays the ease with which polyheterosystems can be constructed with this reaction. The synthesis of 2-substituted 7-iodofuro[3,2-c]pyridines, 2-substituted furo[3,2-b]pyridines, and 3-substituted lH-thieno-[3,4-b]-2-pyran-l-ones (thiaisocoumarins) is described. A series of 3-methyl-1,4,6-triaryl-1H-pyrazolo[3,4-b]pyridines was synthesized via the reaction of 3-methyl-1-phenyl-1H-pyrazol-5-amine and α,β-unsaturated ketones in ionic liquid without any catalyst. This protocol has the advantages of easier work-up, milder reaction conditions, high yields and environmentally benign procedure.  相似文献

A series of 4,7-dihydro-4-oxo-1H-pyrrolo[2,3-b]pyridine-5-carboxylic acids was synthesized from ethyl 5-methyl(or 5H)-2-aminopyrrole-3-carboxylate. The starting pyrroles were obtained by reaction of carbethoxyacetamidine with bromoacetone or chloroacetaldehyde. One compound ( 10 ) showed antibacterial activity in vitro.  相似文献

A series of 2-substituted-7-alkyl-4,7-dihydro-4-oxothiazolo[5,4-b]pyridine-5-carboxylic acids were synthesized. Antibacterial activity was tested in vitro. None of the new compounds prepared showed any antibacterial activity in vitro against the strains tested.  相似文献

Pyrazolo[3,4-b]pyridines are a group of heterocyclic compounds presenting two possible tautomeric forms: the 1H- and 2H-isomers. More than 300,000 1H-pyrazolo[3,4-b]pyridines have been described which are included in more than 5500 references (2400 patents) up to date. This review will cover the analysis of the diversity of the substituents present at positions N1, C3, C4, C5, and C6, the synthetic methods used for their synthesis, starting from both a preformed pyrazole or pyridine, and the biomedical applications of such compounds.  相似文献

The synthesis and characterization of a number of 3-aryl-6H-isoxazolo[3,4-d]pyrazolo[3,4-b]pyridines and 3-aryl-6H-isoxazolo[5,4-d]pyrazolo[3,4–6]pyridines from common precursors, 5-benzoyl-4-chloro-1H-pyrazolo-[3,4-b]pyridines, has been described. The structures were determined by unambiguous chemical synthesis and by isolation and 13C nmr analysis of some key, isolated, intermediates. The ability of these compounds to displace [3H]flunitrazepam from CNS binding sites was also observed.  相似文献

Complementary approaches to the synthesis of the title compounds 1 are described. Metallation of 3,5-di-bromo-2-methoxypyridine ( 5b ) by bromine/lithium exchange gave selectively the 3-lithio intermediate 6 which was trapped with substituted cinnamaldehydes 7 , providing allylic alcohols 8 in good yields. Methyl ether cleavage and concomitant cyclization occurred on exposure to concentrated hydrobromic acid in hot acetic acid. The resulting 2-phenyl-2H-pyrano[2,3-b]pyridines were hydrogenated over Raney nickel to the title compounds which had antiviral activity. Alternatively, 1 were synthesized by Heck reaction of appropriately substituted 3-halo-2-methoxypyridines ( 5 or 24 ) with vinyl carbinol 15 to furnish ketones 16 or 26 which, upon reduction of the carbonyl group, were cyclized directly to 1 .  相似文献
