5-Amino-2,3,6,7-tetrahydrothiocines were reacted with arylazides yielding 1-aryl-9a-amino-1,3a,4,5,7,8,9,9a-octahydrothiocino[4,5-d]-1,2,3-triazoles which could be deaminated to 1-aryl-1,4,5,7,8,9-hexahydrothiocino-[4,5-d]-1,2,3-triazoles. Similarly, the above enamines yielded, with nitrile oxides, 3-aryl-9a-amino-3a,4,5,8,9,9a-hexahydrothiocino[4,5-d]isoxazoles which were transformed with acids into 3-aril-4,5,8,9-tetrahydrothiocino-[4,5-d]isoxazoles.  相似文献

Starting from the readily available 4-bromomethy-5-carbethoxy 1,2,3-thiadiazole (V), 5-bromomethy-4-carbethoxy-1,2,3-thiadiazole (IX) and ethyl 2-aryl-5-bromomethyloxazole-4-carboxylate (XIV), 4,10-dihydro-10-oxo[1]benzoxepino[3,4-d][1,2,3]thiadiazole (Ia), 4,10-dihydro-10-oxo[1]benzothiepino[3,4-d][1,2,3]thiadiazole (Ib), 4,10-dihydro-4-oxo[1]benzothiepino[4,3-d] [1,2,3]thiazole (II), 2-aryl-4,10-dihydro-4-oxo[1]benzoxepino[4,3-d]oxazoles (XIXa-XIXc) and 2-aryl-4,10-dihydro-4-oxo[1]benzothiepino[4,3-d]oxazoles (XIXd-XIXf) were prepared.  相似文献

Some new 1,2,3-triazolo[4,5-d]-1,2,4-triazolo[4,3-b]pyridazines were prepared starting from the corresponding 1,2,3-triazolo[4,5-d]pyridazines via the formation of the 1,2,4-triazole ring, by condensation of an appropriate monocarbon fragment with the 4-hydrazino substituent and the nitrogen atom in the 5 position of the heterocycle. Condensation of 4-phenylhydrazino substituted derivatives with formic acid gave zwitterionic compounds.  相似文献

The imidazo[4,5-d]-1,2,3-triazine and pyrazolo[3,4-d]-1,2,3-triazine analogues of the potent anticonvul-sant purine, BW 78U79 (9-(2-fluorobenzyl)-6-methylamino-9H-purine, 1 ), were synthesized and tested for anticonvulsant activity. The imidazo[4,5-d]-1,2,3-triazines 11–13 were prepared in four steps from 5-aminoimidazole-4-carboxamide (2) and the pyrazolo[3,4-d]-1,2,3-triazines 18–21 were synthesized starting with 5-amino-1-(2-fluorobenzyl)pyrazole-4-carbonitrile (14) . The intermediate 1,2,3-triazin-4-ones 6 and 16 were converted to the 4-substituted targets via the 4-(4-dimethylaminopyridinium) salts 10 and 17 . Imidazotriazine 11 had potent anticonvulsant activity against maximal electroshock-induced seizures, but its propensity to cause emesis precluded further development.  相似文献

A method was developed for the synthesis of 5-carbethoxy-4-formyl-1,2,3-thiadiazole (I), its isomer (II); 5-benzoyl-4-formyl-1,2,3-thiadiazole (III), and its isomer (IV). It was demonstrated that although compounds I, III and IV with hydrazine gave 7H-1,2,3-thiadiazolo[4,5-d]-pyridazin-7-one (XXII), 7-phenyl-1,2,3-thiadiazolo[4,5-d]pyridazine (XXIII) and 4-phenyl-1,2,3-thiadiazolo[4,5-d]pyridazine (XXV), respectively; however, compound I gave its corresponding hydrazone (XXIV).  相似文献

The reaction of 6-arylidenehydrazino-1,3-dimethyluracils with thionyl chloride in benzene afforded purine, thiazolo[4,5-d]pyrimidine, pyrimido[4,5-e][1,3,4]thiadiazine, pyrazolo[3,4-d]pyrimidine, and [1,2,3]thiadiazolo[4,5-d]pyrimidine derivatives, while the treatment of 6-(benzylidene-1′-methylhydrazino)-1,3-dimethyluracil with thionyl chloride in benzene gave 4-methylpyrimido[4,5-e][1,3,4]thiadiazine and 1-methylpyrazolo-[3,4-d]pyrimidine derivatives. Plausible mechanisms for the formation of these fused pyrimidines are also described.  相似文献

A series of 4-substituted-5-arylthio-1,2,3-selenadiazoles, 4-substituted-5-arylseleno-1,2,3-selenadiazoles and 4-aryloxymethyl-1,2,3-selenadiazoles were synthesized. Pyrrolysis of these compounds afforded the corresponding acetylenes XI, XIII (X = S, Se) and XII, respectively. Oxidation of 4-substituted-5-arylthio-1,2,3-selenadiazoles (XIV) with m-chloroperbenzoic acid gave 4-substituted-5-arylsulfinyl-1,2,3-selenadiazoles (XV) and 4-substituted-5-arylsulfonyl-1,2,3-selenadiazoles (XVI).  相似文献

The reaction of selenium dioxide with o-hydroxyacetophenone semicarbazones gives 4-(2-hydroxyaryl)-1,2,3-selenadiazoles which undergo ready decomposition by the action of potassium carbonate to form benzofuran-2-selenolates. The latter can be alkylated with methyl iodide and benzyl chloride and arylated with 2,4-dinitrochlorobenzene. Intermediate formation of 2-(o-hydroxyphenyl)ethyneselenolate during decomposition of 1,2,3-selenadiazoles was proved by the isolation of methyl o-methoxyphenylethynyl selenide when the substrate was treated with potassium carbonate in the presence of methyl iodide.  相似文献

The synthesis of 1,2-dihydro[1]benzofuro[2,3-d]pyridazin-1-one and 3,4-dihydro [1]benzofuro[2,3-d]pyridazin-4-one was accomplished by the eyclization of appropriately carbonyl-substituted benzofuran derivatives. Another successful synthetic route was provided using 1,2,3,4-tetrahydro[1]benzofuro[2,3-d]pyridazine-1,4 dione and 1,2,3,4-tetrahydro[1]benzofuro[2,3-d]pyridazin-4-one. The structure of a nitrobenzofuropyridazin-4-one was established using nmr and the nuclear Overhauser effect.  相似文献

A method was developed for the synthesis of 1H-1-arylnaphtho[2,3-d][1,2,3]triazole-4,9-dione 2-oxides based on 2-arylamino-3-chloro-1,4-naphthoquinones. The reaction of 1H-1-arylnaphtho[2,3-d][1,2,3]triazole-4,9-dione 2-oxides with an ethanolic alkali solution affords 2-{[1-(aryl)-2-oxido-1H-1,2,3-triazol-4-yl]carbonyl}benzoic acids.  相似文献

Starting from readily available 2-substituted-4-formylthiazoles and selenazoles, a series of 4-(2-aryl-4-selenazolyl)-1,2,3-selenadiazoles I and 4-(2-substituted-4-thiazolyl)-1,2,3-selenadiazoles II were prepared. Pyrolysis of compound II afforded (2-substituted-4-thiazolyl) acetylenes VII. Addition of potassium hydroxide pellets to an alcoholic solution of II gave 2-substituted-1,4-diselenafulvenes VIII. Decomposition of compound II with base followed by the addition of carbon disulfide gave 5-substituted 2-thioxo-1,3-thiaselenoles XI.  相似文献

Derivatives of two new molecular structures, namely, [1,3]thiazino[3,2-a]purine and [1,2,3]triazolo[4,5-d]-[1,3]thiazino[3,2-a]pyrimidine, were synthesized together with other heterocyclic compounds. Retrosynthetic analysis of their molecular skeletons suggested a simple way of obtaining 3,4-dihydro-7,8-diamino-2H,6H-pyrimido[2,1-b][1,3]thiazin-6-one, which is a useful intermediate for their synthesis. This intermediate and the thiazole homologue were obtained directly by reaction of 5,6-diamino-2,3-dihydro-2-thioxo-4(lH)-pyrimidi-none with 1,3- or 1,2-dibromoalkane, respectively.  相似文献

The synthesis of 1,2-dihydro-l-oxopyrazolo[1,5-d]-1,2,4-triaxine was achieved by rerrangment of 2-(5-pyrazolyl)-1,3,4-oxadiazole under alkaline condition. The cyclization of the N-carbethoxyhydrazone of the pyrazole-5-carboxaldehyde gave the 3,4-dihydro-4-oxopyrazolo[1,5-d]1,2,4-triazine. Electrophilic substitutions of the l-pyrazolotriazinome were made either on the lactam nitrogen with methylsulfate, benzylchloride and monochloracetic axid or on the pyrazole ring with bromine. The synthesis of pyrazolo[1,5-d]-1,2,4-triazine was made from teh l-pyrazolotrizinone via the l-pyrazolotrizinone. The methylation of l-pyrazolotrizinone and 8-bromo-l-pyrazolotrizinone afforded N-and S-methyl derivatives. The sysnthesis of 1,2,3,4-tetrahydropyrazolo[1,5-d]-1,2,4-triazine-1,4-diones was axhived by cyclising N-carbethoxyhydrazides of pyrrole-5-carboxylic acid. The structures of the derivatives were determined by 1H-nmr.  相似文献

The synthetic chemistry of the relatively unknown pyridazino [4,5-d]pyridazine ring system has been extended. 1,4-Diaminopyridazino [4,5-d]pyridazine (VIII) has been prepared by two routes, the most interesting of these being the one-step conversion of 4,5-dicyanopyridazine into VIII with hydrazine. Upon nitration VIII gave only the mononitramine (X). Attempts to prepare 1,4-dichloropyridazino [4,5-d]pyridazine gave only 4-chloro-2H-pyridazino [4,5-d]pyridazin-1-one (XII). Pyrimido [4,5-d]pyridazine-1,3-dione (XIV) was prepared from pyridazine-4,5-dicarboxamide (IV). The hydrolysis of 5,8-dichloropyrazino [2,3-d]pyridazine (XV) gave 5-chloropyrazino [2,3-d]pyridazin-8-one (XVII) and likewise the ammonolysis of XV gave 5-amino-8-chloropyrazino [2,3-d]pyridazine (XX). As expected the hydrolysis of 5,8-dibromo-pyrazino [2,3-d]pyridazine (XXI) gave 5-bromopyrazino [2,3-d]pyridazin-8-one (XXII). Attempted catalytic dechlorination of 5-chloropyrazino [2,3-d]pyridazin-8-one (XVII) gave 1,2,3,4-tetrahydropyrazino [2,3-d]pyridazin-5-one (XIX).  相似文献

Some new 1,2,3-triazolo[4,5-e]-1,2,4-triazolo[3, 4 -c]pyrimidmes were prepared starting from the corresponding 1,2,3-triazolo[4,5-d]pyrimidines via the formation of the 1,2,4-triazole ring. Thus suitable hydrazino derivatives 6 were condensed with triethyl orthoformate, triethyl orthoacetate and triethyl orthobenzoate to give the expected tricyclic derivatives 7 , 8 and 9 . Intramolecular cyclization of the ethoxycarbonylhydrazino derivatives 10 gave the tricyclic compounds 11 bearing an hydroxyl group in the 3 position. The v-triazolo-s-triazolopyrimidine derivatives were tested towards the A1 and A2A adenosine receptors in binding assays, but they did not show any receptor affinity.  相似文献

Also, a series of 5-substituted-4-amino-3-(1,2,3-thiadiazol-5-yl)-s-triazoles and 5-(1,2,3-thiadiazolyl)-s-triazolo[3,4-b]-1,3,4-thiadiazoles were prepared.  相似文献

The hydrazone derivatives of 4-benzoyl-1,2,3-triazole can easily be cyclised by reaction with various organic reagents (ortho esters, aldehyde and ketone compounds, phosgene, etc) which result in the incorporation of the introduced reagent's carbon atom into the new six membered ring. The newly created C-N bond of the resulting [1,2,3]triazolo[1,5-d][1,2,4]triazines displays a particular sensitivity due to the electron attracting effect of the triazole ring. Some mechanistic considerations are proposed to account for the observed results.  相似文献
