Rheumatoid arthritis (RA) is a multifactorial autoimmune disease whose etiopathogenesis is not well understood. Although soluble (s) forms of 4-1BB (s4-1BB) and 4-1BB legand (s4-1BBL) have been detected in the sera of RA patients, their significance is not known. We compared the serum levels of s4-1BB and s4-1BBL in RA patients with those in systemic lupus erythematosus (SLE) and Behcet's disease (BD) patients. Serum levels of s4-1BB and s4-1BBL were significantly higher in RA patients compared with healthy controls, SLE or BD patients, and the abundance was correlated with disease severity in patients with RA. The serum levels of s4-1BB in RA patients were inversely corroborated with 4-1BB expression levels on activated T lymphocytes. In addition, there was a correlation between serum levels of s4-1BB and s4-1BBL. The augmented secretion of s4-1BB and s4-1BBL levels into the serum may reflect the clinical symptoms of RA and levels of s4-1BB and s4-1BBL in sera at the time of diagnosis may be indicative of the severity and outcome of RA.  相似文献

Our recent studies identified a novel pathway of nicotinamide metabolism that involves 4-pyridone-3-carboxamide-1-β-D-ribonucleoside (4PYR) and demonstrated its endothelial cytotoxic effect. This study tested the effects of 4PYR and its metabolites in experimental models of breast cancer. Mice were divided into groups: 4T1 (injected with mammary 4T1 cancer cells), 4T1 + 4PYR (4PYR-treated 4T1 mice), and control, maintained for 2 or 21 days. Lung metastasis and endothelial function were analyzed together with blood nucleotides (including 4PYR), plasma amino acids, nicotinamide metabolites, and vascular ectoenzymes of nucleotide catabolism. 4PYR metabolism was also evaluated in cultured 4T1, MDA-MB-231, MCF-7, and T47D cells. An increase in blood 4PYR in 4T1 mice was observed at 2 days. 4PYR and its metabolites were noticed after 21 days in 4T1 only. Higher blood 4PYR was linked with more lung metastases in 4T1 + 4PYR vs. 4T1. Decreased L-arginine, higher asymmetric dimethyl-L-arginine, and higher vascular ecto-adenosine deaminase were observed in 4T1 + 4PYR vs. 4T1 and control. Vascular relaxation caused by flow-dependent endothelial activation in 4PYR-treated mice was significantly lower than in control. The permeability of 4PYR-treated endothelial cells was increased. Decreased nicotinamide but enhanced nicotinamide metabolites were noticed in 4T1 vs. control. Reduced N-methylnicotinamide and a further increase in Met2PY were observed in 4T1 + 4PYR vs. 4T1 and control. In cultured breast cancer cells, estrogen and progesterone receptor antagonists inhibited the production of 4PYR metabolites. 4PYR formation is accelerated in cancer and induces metabolic disturbances that may affect cancer progression and, especially, metastasis, probably through impaired endothelial homeostasis. 4PYR may be considered a new oncometabolite.Subject terms: Mechanisms of disease, Pathogenesis, Breast cancer  相似文献

The reductive cleavage of the Si-Si bond in 1,1-bis(1-methyl-2,3,4,5-tetraphenyl-1-silacyclopentadiene) [(C(4)Ph(4)SiMe)(2)] (1) with either Li or Na in THF gives the silole anion [MeSiC(4)Ph(4)]- (2). The head-to-tail dimerization of the silole anion 2 gives crystals of the disilatricyclic diallylic dianion [(C(4)Ph(4)SiMe)(2)]-2 (3). The derivatization of 3 (crystals) with bromoethane (gas) under reduced pressure provides [(MeSiC(4)Ph(4)Et)(2)] (4) quantitatively. The reverse addition of 3 in THF to trimethylsilyl chloride, hydrogen chloride, and bromoethane in THF gives 1-methyl-1-trimethylsilyl-1-silole [Me(3)SiMeSiC(4)Ph(4)] (6), 1-methyl-2,3,4,5-tetraphenyl-1-silacyclo-3-pentenyl-1-methyl-1-silole [C(4)Ph(4)H(2)SiMe-MeSiC(4)Ph(4)] (7), and 1-methyl-2,5-diethyl-2,3,4,5-tetraphenyl-1-silacyclo-3-pentenyl-1-methyl-1-silole [C(4)Ph(4)Et(2)SiMe-MeSiC(4)Ph(4)] (8), respectively. The reaction products unambiguously suggest that the silole anion [MeSiC(4)Ph(4)]- is generated by coordination of the chloride ion at the silicon atom in 3 or by the nucleophilic substitution of either chloride or bromide ion at one of two silicon atoms in 3. The quenching reaction of 3 dissolved in THF with water gives 1,2,3,4-tetraphenyl-2-butene, the disiloxane of 1-methyl-2,3,4,5-tetraphenyl-1-silacyclo-3-pentenyl [O(MeSiC(4)Ph(4))(2)] (10) and methyl silicate.  相似文献

4-1BB, a transmembrane molecule, member of the tumor necrosis factor receptor superfamily, is an important costimulatory molecule in the immune response, plays a key role in the clonal expansion and survival of CD8(+) T cells. In this study, we investigated 4-1BB regulation of CD4(+) T cell responses using 4-1BB transgenic (TG) mice that constitutively expressed 4-1BB on mature T cells. We first showed that CD4(+) T cells of 4-1BB TG mice had more sustained proliferative capacity in response to TCR/4-1BB stimulation in vitro compared to WT mice. Secondly, 4-1BB TG mice exhibited a more elevated contact hypersensitivity (CHS) response mediated by CD4+ Th1 cells due to more vigorous expansion of and apoptotic inhibition of CD4(+) T cells. Finally, CD4(+) T cells of 4-1BB TG mice had a heightened capacity for T cell priming. Overall, our results demonstrate the involvement of 4-1BB in CD4(+) Th1 cell responses by regulating the clonal expansion and survival of CD4(+) T cells as seen in CD8(+) T cells.  相似文献

4-Ethyl-1-hydroxy-3-(4-hydroxyphenyl)-2-oxa-1-boranaphthalene (4) is formed in 78% yield from the reaction of 1-(4-methoxyphenyl)-2-phenylbutan-1-one with an of excess boron tribromide in dichloromethane followed by treatment with water. Reaction of 4 with iodine in aqueous sodium hydroxide gives a second oxaboracycle, 3-ethyl-1-hydroxy-3-(4-hydroxybenzoyl)-2,1-benzoxaborolane (5). The X-ray crystal structure determinations of both boron heterocycles are reported. Other new compounds reported are 1-(4-hydroxyphenyl)-2-(1-hydroxyphenyl)-butan-1-one (6), formed by reaction of 4 with alkaline hydrogen peroxide, and 1-(4-hydroxyphenyl)-2-(2-biphenyl)-butan-1-one (8), formed by coupling of 4 with bromobenzene in the presence of Pd(PPh3) 4.  相似文献

Compelling evidence has indicated the vital role of lysine-specific demethylase 4 A (KDM4A), hypoxia-inducible factor-1α (HIF1α) and the mechanistic target of rapamycin (mTOR) signaling pathway in nasopharyngeal carcinoma (NPC). Therefore, we aimed to investigate whether KDM4A affects NPC progression by regulating the HIF1α/DDIT4/mTOR signaling pathway. First, NPC and adjacent tissue samples were collected, and KDM4A protein expression was examined by immunohistochemistry. Then, the interactions among KDM4A, HIF1α and DDIT4 were assessed. Gain- and loss-of-function approaches were used to alter KDM4A, HIF1α and DDIT4 expression in NPC cells. The mechanism of KDM4A in NPC was evaluated both in vivo and in vitro via RT-qPCR, Western blot analysis, MTT assay, Transwell assay, flow cytometry and tumor formation experiments. KDM4A, HIF1α, and DDIT4 were highly expressed in NPC tissues and cells. Mechanistically, KDM4A inhibited the enrichment of histone H3 lysine 9 trimethylation (H3K9me3) in the HIF1α promoter region and thus inhibited the methylation of HIF1α to promote HIF1α expression, thus upregulating DDIT4 and activating the mTOR signaling pathway. Overexpression of KDM4A, HIF1α, or DDIT4 or activation of the mTOR signaling pathway promoted SUNE1 cell proliferation, migration, and invasion but inhibited apoptosis. KDM4A silencing blocked the mTOR signaling pathway by inhibiting the HIF1α/DDIT4 axis to inhibit the growth of SUNE1 cells in vivo. Collectively, KDM4A silencing could inhibit NPC progression by blocking the activation of the HIF1α/DDIT4/mTOR signaling pathway by increasing H3K9me3, highlighting a promising therapeutic target for NPC.Subject terms: Oncogenes, Cancer  相似文献

New carboxylic acid amides containing an N-methylpiperazine fragment were synthesized by reactions of 1-methylpiperazine or 3- and 4-(4-methylpiperazin-1-ylmethyl)aniline with 4-chlorobenzoyl chloride and of 4-methyl-3-nitroaniline with 4-(4-methylpiperazin-1-ylmethyl)benzoyl chloride or benzotriazol-1-yl 4-(4-methylpiperazin-1-ylmethyl)benzoate. 4-Chloro-N-[4-(4-methylpiperazin-1-ylmethyl)phenyl]benzamide reacted with imidazole, quinolin-5-amine, and 2-methylquinolin-5-amine to give substituted 4-amino-N-[4-(4-methylpiperazin-1-ylmethyl)phenyl]benzamides. 4-Methyl-3-nitrophenyl-4-methylpiperazin-1-yl-substituted benzamides were reduced with hydrazine hydrate over Raney nickel to obtain N-(3-amino-4-methylphenyl)-4-(4-methylpiperazin-1-ylmethyl)benzamide as key intermediate in the synthesis of antileukemic agent imatinib and its isomer with alternative position of the amide group, 4-[(3-amino-4-methylphenylamino)methyl]phenyl-(4-methylpiperazin-1-yl)methanone.  相似文献

The considerable interest in substituted isoquinolin-1-ones related to 5-aminoisoquinolin-1-one (5-AIQ) as drugs points to a need for an efficient and straightforward synthesis of the 4,5-disubstituted bicycles. Bromination of 5-nitroisoquinolin-1-one gave 4-bromo-5-nitroisoquinolin-1-one but neither this nor 5-amino-4-bromoisoquinolin-1-one would participate in Pd-catalysed couplings. Protection of the lactam as 1-methoxy- and 1-benzyloxy-4-bromo-5-nitroisoquinolines, however, permitted Stille, Suzuki and Buchwald-Hartwig couplings to take place in high yields, insensitive to electronic demands and severe steric bulk in the arylboronic acids. Lithiation of 4-bromo-1-methoxy-5-nitroisoquinoline and quench with iodomethane gave 1-methoxy-4-methyl-5-nitroisoquinoline in low yield. Demethylation of the 1-methoxy-4-substituted-5-nitroisoquinolines with hydrogen bromide gave 4-substituted-5-nitroisoquinolin-1-ones, whereas hydrogenolytic debenzylation was achieved with simultaneous reduction of the 5-nitro group. 5-Amino-4-(4-trifluoromethylphenyl)isoquinolin-1-one was identified as a new potent and selective inhibitor of poly(ADP-ribose)polymerase-2 (PARP-2).  相似文献

Lithiation-methylation of 3,3-dimethyl-1λ4,3-thiasilinane 1-oxide and 4,4-dimethyl-1λ4,4-thiasilinane 1-oxide under the action of butyllithium or lithium diisopropylamide and methyl iodide was studied. In both cases, monomethylation proceeds selectively α to the sulfoxide group to form 2,3,3-trimethyl-1λ4,3-thiasilinane 1-oxide and 2,4,4-trimethyl-1λ4,4-thiasilinane 1-oxide, respectively. Subsequently, 2,3,3-trimethyl-1λ4,3-thiasilinane 1-oxide undergoes monomethylation into the same α position to give 2,2,3,3-tetramethyl-1λ4,3-thiasilinane 1-oxide, while 4,4-dimethyl-1λ4,4-thiasilinane 1-oxide is dimethylated into the neighboring α’ position to form two stereoisomers of 2,4,4,6-tetramethyl-1λ4,4-thiasilinane 1-oxide with axial-equatorial or equatorial-equatorial methyl groups in the 2 and 6 positions.  相似文献

4-1BB, a member of the tumor necrosis factor receptor (TNFR) superfamily, is a major costimulatory receptor that is rapidly expressed on the surface of CD4+ and CD8+ T cells after antigen- or mitogen-induced activation. The interaction of 4-1BB with 4-1BBL regulates immunity and promotes the survival and expansion of activated T cells. In this study, the expression of 4-1BB and 4-1BBL was examined during regeneration of the murine thymus following acute cyclophosphamide-induced involution. Four-color flow cytometry showed that 4-1BB and 4-1BBL were present in the normal thymus and were preferentially expressed in the regenerating thymus, mainly in CD4+CD8+ double-positive (DP) thymocytes. Furthermore, the CD4loCD8lo, CD4+CD8lo and CD4loCD8+ thymocyte subsets, representing stages of thymocyte differentiation intermediate between DP and single-positive (SP) thymocytes, also expressed 4-1BB and 4-1BBL during thymus regeneration but to a lesser degree. Interestingly, the 4-1BB and 4-1BBL positive cells among the CD4+CD8+ DP thymocytes present during thymus regeneration were TCRhi and CD69+ unlike the corresponding controls. Moreover, the 4-1BB and 4-1BBL positive cells among the intermediate subsets present during thymus regeneration also exhibited TCRhi/int and CD69+/int phenotypes, indicating that 4-1BB and 4-1BBL are predominantly expressed by the positively selected population of the CD4+CD8+ DP and the intermediate thymocytes during thymus regeneration. RT-PCR and Western blot analyses confirmed the presence and elevated levels of 4-1BB and 4-1BBL mRNA and protein in thymocytes during thymus regeneration. We also found that the interaction of 4-1BB with 4-1BBL promoted thymocyte adhesion to thymic epithelial cells. Our results suggest that 4-1BB and 4-1BBL participate in T lymphopoiesis associated with positive selection during recovery from acute thymic involution.  相似文献

Hydroxylamine reacts with 1-alkyl-1,2-dihydro-3,1-benzothiazine-4thiones ( 1 ), giving 1-alky1-3-hydroxy-2,3-dihydro-1H-quinazoline-4-thiones ( 2 ). The same reagent, in neutral medium, converts 1-aryl-1,2-dihydro-3,1-benzothiazine-4-thiones ( 3 ) into 1-aryl-4-hydroxyimino-1,4-dihydro-2H-3,1-benzothiazines ( 4 ). In acidic medium, the same starting materials lead to 1-aryl-3-hydroxy-2-3-dihydro-1H-quinazoline-4-thiones ( 5 ). genrally with some quantity of the isomer 4 . Thiones 2 and 5 , as well as oximes 4 , heated at 200°, decomopose, yielding, in varying proportions, 1H-quinazoline-4-thiones ( 6 or 7 ), 1H-quinazoline-4-ones ( 9 ) and 2,3-dihydro-1H-quinazoline-4-thiones ( 11 ). Reacting with methyliodide, 1H-quinazoline-4-thiones ( 7 ) give 4-methylthioquinazolin-1-ium iodidies ( 12 ) which can be hydrolysed into 1H-quinazolin-4-ones ( 9 ). The latter are also obtained by reacting benzonitrile N-oxide with the corresponding thiones. 1-Aryl-1 H-quinazoline-4-thiones ( 7 ) react readily with nitrogen nucleophiles XNH2 to give 1-aryl-4-imino-1,4-dihydro-quinazolines diversely substituted on the imino group. While thiones 7 are S- methylated by methyl iodide, the corresponding 1-aryl-1H-quinazolin-4-ones (9), with the same reagent, ungergo a N-methylation, yielding 1-aryl-3-methyl-4-oxo-3,4-dihydroquinazolin-l-ium iodides ( 18 ). Olive-green cobalt(III) complexes having the general formula Co4O4(O2CMe)4L4 (1) where L = py (1a), 4-Mepy (1b), 4-Etpy (1c), and 4-CNpy (1d) have been prepared by the H2O2 oxidation of a mixture of Co2+, MeCO2-, and pyridine or 4-substituted pyridines in a 1:2:1 molar ratio in methanol. Spectroscopic and X-ray crystallographic studies show that these complexes contain a tetrameric cubane-like core [Co4(mu3-O)4]4+ where the four cobalt atoms form an approximate tetrahedron with edge lengths of approximately 2.75 A. Each cobalt in the crystallographically determined structure of Co4(mu3-O)4(mu-O2CMe)4L4 in 1a.NaClO4.3.5H2O and 1b.3H2O is hexacoordinate. Infrared spectra of the complexes show characteristic bands near 700, 635, and 580 cm-1 due to the central cubane-like core. 1H NMR spectra of the complexes show that the dissolved species are essentially diamagnetic and also that the complexes maintain their integrity in solution. UV-vis spectra of the green solutions have been interpreted in terms of ligand-field and charge-transfer bands. The electrochemical behavior of the complexes studied by cyclic and differential pulse voltammetric techniques indicates that the [(CoIII)4(mu3-O)4]4+ core present in the complexes undergoes a reversible one-electron oxidation to the [(CoIII)3CoIV(mu3-O)4]5+ core with an E1/2 value below 1 V. This suggests that these complexes of cobalt may be suitable as catalysts for the oxidation of organic compounds. Preliminary investigations indicate that 1a has a role to play in the cobalt-catalyzed aerobic oxidation of neat ethylbenzene and p-xylene.  相似文献

The action of halogens in an alkaline medium on 1, 2, 4-triazoles has given their N-chloro, N-bromo, and N-iodo derivatives. The formation of C-halogen derivatives of some 1, 2, 4-triazoles from their N-halogen derivatives has been studied, and also the action of halogens on 1, 2, 4-triazoles at elevated temperatures. It has been shown that N-bromo-1, 2, 4-triazoles very readily form 1, 2, 4-triazole and 3, 5-dibromo-1, 2, 4-triazole in approximately equimolar proportions. N-Chloro-1, 2, 4-triazole is more stable to the action of heat and forms 3-chloro-1, 2, 4-triazole with a yield of 40%. It was impossible to convert N-iodo-1, 2, 4-triazole into C-derivatives.  相似文献

Air-oxidation of Cp'(4)Fe(4)(HCCH)(2) (Cp' = Cp (1a), C(5)H(4)Me (1b)) in an NH(4)PF(6)/CH(3)CN solution afforded the one-electron oxidized clusters [Cp'(4)Fe(4)(HCCH)(2)](PF(6)). Oxidation of 1a with excess AgBF(4) in THF afforded [1a](BF(4)), while that of 1b with excess AgBF(4) gave [1b](BF(4))(2). The X-ray crystal structure analysis of [1a](BF(4)) revealed that the monocationic cluster retains the butterfly-type Fe(4)(mu4-eta(2):eta(2):eta(1):eta(1)-HCCH)(2) framework similar to that of the neutral cluster. The average Fe-Fe bond length is shorter by 0.029 A than that in the neutral cluster. Electrochemical oxidation of 1a and 1b in 0.1 M NH(4)PF(6)/CH(3)CN solution at +0.30 and +0.25 V versus Ag/10 mM AgNO(3), respectively, afforded the two-electron oxidized clusters [1a](PF(6))(2) and [1b](PF(6))(2). The X-ray crystal structure analysis for [1b](BF(4))(2) shows that the butterfly-type cluster core is retained but shrinks more of those of neutral and monocationic clusters. The four Fe-Fe bonds in [1b](BF(4))(2) are unequivalent: one Fe-Fe bond (2.397(1) A) is apparently shorter than the others (2.439(2)-2.461(2) A).  相似文献

三种带有不同取代基的重氮萘酮(la~1c)在THF和二氧六环中加热分解给出不同的产物。1-重氮-4-萘酮(1a)的热解产物主要是重氮萘酮热解后产生的烯酮卡宾(2a)与环醚开环后形成的聚合物;3-甲基-1-重氮-4-萘酮(1b)的热解产物比较复杂,除冠醚类产物之外,还有烯酮卡宾对四氢呋喃和二氧六环的C-H键的插入反应产物、螺环化合物、2-甲基萘酚以及难以分离的聚合物;3-硝基-1-重氮-4-萘酮(1c)的热解产物主要是聚合物,此外还有少量C-H键的插入反应产物和2-硝基萘酚。对重氮萘酮热解反应的机理作了讨论。  相似文献

Binding constants for tetraphenylborate and tetraphenylphosphonium ions (Ph4B- and Ph4P+) to cyclodextrins (CDs) to give 1:1 host-guest complexes have been measured using capillary electrophoresis. Mobilities of the ions as a function of gamma-CD concentration give binding constants, K, of 1.08 x 10(5) M-1 for Ph4B- and 0.6 x 10(1) M-1 for Ph4P+. This dramatic difference of four orders of magnitude in binding constants is not seen with beta-CD (K = 7.7 x 10(1) M-1 for Ph4B- and 3.7 x 10(1) M-1 for Ph4P+) or dimethyl (DM)-beta-CD (K = 46 x 10(1) M-1 for Ph4B-1 and 7.7 x 10(1) M-1 for Ph4P+). The crystal and hydrodynamic radii of the ions, the latter calculated from their absolute mobilities, indicate that Ph4B- is smaller than the gamma-CD cavity, whereas Ph4P+ is approximately the gamma-CD cavity size. Results suggest that Ph4B- fits exactly into a gamma-CD cavity, with hydrophobic contacts involving several of the phenyl rings, whereas Ph4P+ is too large to enable these multiple contacts to be made. When only a single phenyl ring can fit into the CD cavity, binding strengths are in the order DM-beta-CD > beta-CD > gamma-CD and Ph4B- > Ph4P+. Measurement of electrophoretic mobilities of the complexes shows that their hydrodynamic radii are in the order gamma-CD < beta-CD approximately DM-beta-CD for Ph4B- and gamma-CD > beta-CD approximately DM-beta-CD for Ph4P+.  相似文献
