Series of novel furan/thiophene and piperazine‐containing 1,2,4‐triazole Mannich bases and bis(1,2,4‐triazole) Mannich bases have been conveniently synthesized via Mannich reaction with triazole Schiff bases, various piperazine derivatives, and formaldehyde as intermediates in good yields. Their structures were characterized by melting points, 1H NMR, 13C NMR, IR and elemental analysis. The preliminary bioassay showed that most compounds exhibited significant in vitro and in vivo fungicidal activity against several test plant fungi. Among 32 new compounds, the trifluoromethyl‐containing compounds showed superior activity than the methyl‐containing ones. Several compounds, such as F8 , F9 , F10 , G5 , H7 , H8 , I3 and I4 , were comparable with some commercial fungicides against different fungi during the present study and could be further structurally optimized. Meanwhile, several compounds showed good herbicidal activity against Brassica campestris at 100 µg/mL and KARI inhibitory activity at 200 µg/mL. However, compounds exhibited poor insecticidal activity against oriental armyworm at 200 µg/mL in the preliminary studies. The research results will provide useful information for the design and discovery of new agrochemicals with novel heterocyclic structures.  相似文献

42 novel diacylhydrazine derivatives containing furan were synthesized by the reaction of 5‐substituted phenyl‐2‐furoyl chloride with substituted benzohydrazide in anhydrous dichloromethane under reflux. Their structures were confirmed by IR, 1H NMR, MS and elemental analysis. Insecticidal and antitumor activity of these new compounds was evaluated. The preliminary bioassays showed that the target compounds exhibited larvicidal activity to the Mythimna separate, some of them exhibited good or moderate larvicidal activity. At the concentration of 0.1%, compounds I2 , I4 , I5 , and III1 showed 95.0%, 90.0%, 95.0% and 95.0% larvicidal activity to Mythimna separate respectively. Some compounds such as I2 , I4 , I5 and III1 showed the typical IGRs' activity, which could induce the larvae premature, abnormal, and lethal larval molt. Results of anticancer activity indicated that some compounds exhibited potential activity against some human cancer cell lines, for example, I1 and IV showed good activity to the BGC‐823 and the inhibitory rates were 60.86% and 61.94% respectively at 10 µmol/L.  相似文献

In order to find novel bleaching herbicide lead compounds, a series of novel 3‐aryl‐4‐substituted‐5‐[3‐(trifluoromethyl)phenoxy]‐1,2,4‐triazoles were designed and synthesized by the multi‐step reactions. N‐(Arylformamido)phenylthioureas undergo ring closure in the presence of sodium hydroxide to generate 3‐aryl‐4‐substituted‐4H‐[1,2,4]triazol‐5‐thiols 1 , which reacted with methyl sulfate in the presence of K2CO3 to give 3‐aryl‐5‐methylsulfanyl‐4‐substituted‐4H‐[1,2,4]triazoles 2 . The target compounds 4 were synthesized by the oxidation of 2 in the presence of H2O2 and Na2WO4, followed by the substitution with 3‐(trifluoromethyl)phenol in moderate to good yields. Their structures were confirmed by IR, 1H NMR, EI–MS, and elemental analyses. The preliminary bioassay indicated that some of them displayed moderate to good selective herbicidal activity against Brassica campestris L at the concentration of 100 µg/mL. Compounds 4c and 4i possessed 75.0% and 82.6% inhibition against Brassica campestris L at the concentration of 100 µg/mL. However, the target compounds 4 showed weak herbicidal activity against Echinochloa crus‐galli at the concentration of 100 and 10 µg/mL.  相似文献

根据官能团的组合,设计合成了硫(醇)代N-对甲苯基/苯基磺酰基氨基羧酸酯类化合物,这些化合物的结构1H NMR, 13C NMR, MS 和 HRMS证实,初步的实验结果表明:在10μg/mL浓度下,化合物4a, 4b, 4d, 5c, 5d, 5g, 6b和 6d显示对PC12细胞缺氧损伤具有显著的保护作用,化合物4c, 5b和 6c显示对PC12细胞缺氧损伤具有一定的保护作用;在5μg/mL浓度下,4d和 6d显示对PC12细胞缺氧损伤具有一定的保护作用;初步的实验也表明:在10μg/mL浓度下,4c, 5a, 5c, 5d, 5e和 6b显示对PC12细胞具有一定的促分化作用。  相似文献

A series of novel 2‐(4‐(4‐chlorophenyl)‐1H‐pyrazol‐3‐yl)‐5‐(Aryl)‐1,3,4‐oxadiazoles were synthesized by unexpected aromatization during oxidative cyclization of 4‐(4‐chlorophenyl)‐4,5‐dihydro‐1H‐pyrazole‐3‐carbohydrazones using chloramine‐T as an oxidant. The hydrazones were derived from 4‐(4‐chlorophenyl)‐4,5‐dihydro‐1H‐pyrazole‐3‐carbohydrazide and various substituted aldehydes. The structure of the synthesized compounds was confirmed by FTIR, 1H NMR, 13C NMR, and mass spectral data. The synthesized compounds were evaluated for their antitubercular and antioxidant activities. All the compounds 4a , 4b , 4c , 4d , 4e , 4f , 4g , 4h and 5a , 5b , 5c , 5d , 5e , 5f , 5g , 5h showed good antitubercular activity against Mycobacterium tuberculosis (minimum inhibitory concentration = 25 µg/mL for 4f and 4g , 50–100 µg/mL for the rest). However, all the compounds exhibited poor antioxidant activity against 1,1‐diphenyl‐2‐picryl‐hydrazil free radical.  相似文献

A series of novel N‐(4‐(2‐aryloxythiazol‐5‐yl)but‐3‐yn‐2‐yl)benzamide derivatives were designed and synthesized. Their structures were identified by 1H NMR and elemental analyses. Preliminary bioassays indicated that some title compounds provided >80% control of Sclerotinia sclerotiorum at 50 µg/mL and >70% herbicidal activities against B. campestris at 100 µg/mL. A new series of tetrahydropyrido[4,3‐d]dihydropyrimidine‐2‐thiones ( 3a–3x ) were designed and synthesized. Their structures were confirmed by 1H NMR, IR, MS and elemental analysis, and the conformation of compound 3j was confirmed by X‐ray diffraction. Preliminary bioassays indicated that most of the target compounds presented good antiproliferative activities against leukemic K562 cells, ovarian cancer HO‐8910 cells and liver cancer SMMC‐7721 cells in vitro. Among them the compounds 3i and 3m afford the best activity, the IC50 of them were 3.22 and 3.65 µg/mL against leukemic K562 cells, respectively, which were lower than the anticancer drug of clinical practice 5‐FU (IC50=8.56 µg/mL). Preliminary mechanism of action studies revealed that compound 3i caused DNA fragmentation and activated caspase‐3/7 in leukemic K562 cells.  相似文献

A series of novel 1‐(3‐chloropyridin‐2‐yl)‐N‐substituted‐5‐(trifluoromethyl)‐pyrazole carboxamide derivatives TC1 , TC2 , TC3 , TC4 , TC5 , TC6 , TC7 , TC8 , TC9 , TC10 , TC11 were synthesized and characterized by IR, 1H NMR, 13C NMR, MS, and elemental analysis. All the target compounds were tested in vitro for their antibacterial activities and antifungal activities. The preliminary bioassays indicated that compound TC6 exhibited excellent activity against Xanthomonas oryzae (94.9% and 84.9%) at different concentrations (200 µg/mL and 100 µg/mL), which was higher than that of Bismerthiazol (94.6% and 64.0%), respectively. A series of novel ethyl 4‐(methyl or trifluoromethyl)‐2‐(2‐(substituted phenoxy)acetamido)thiazole‐5‐carboxylates 7a , 7b , 7c , 7d , 7e and 8f , 8g , 8h , 8i , 8j , 8k , 8l , 8m , 8n , 8o , 8p , 8q , 8r were synthesized, and their structures were confirmed by IR, 1H‐NMR, MS spectra and elemental analysis. The results of preliminary bioassays show that some of the title compounds exhibit moderate to good herbicidal activities. Compared with the fluorine free compounds 7a , 7b , and 7e , the compounds bearing fluorine 8g , 8j , and 8q showed higher herbicidal activities with 70–100% inhibition against Capsella bursa‐pastoris, Amaranthus restroflexus, and Eclipta prostrata at the dosage of 150 g/ha, which indicated that the trifluoromethyl on the thiazole ring was beneficial for the herbicidal activity. Furthermore, compounds 8f , 8g , 8h , 8i , 8j , 8k , 8l , 8m , 8n , 8o , 8p , 8q , 8r were tested for fungicidal activity against Pseudoperonospora cubensis at 500 µg/mL. Compounds 8f and 8q showed the best fungicidal activity with more than 80% inhibition.  相似文献

In order to study the potential bioactivities of 4‐amino tetramic acid derivatives, the 4‐amination products of pyrrolidine‐2,4‐diones ( 5 ) and 4‐hydroxy‐2‐oxo‐2,5‐dihydro‐1H‐pyrrole‐3‐carboxylates ( 4 ) were prepared. The 4‐amination of 5 took place in high yield when catalyzed by acetic acid, whereas the 4‐amination of 4 was achieved through a 4‐ethoxy intermediate, which was prepared by acidic etherification. Their herbicidal, fungicidal, insecticidal, and antitumor activities were tested. The bioassays showed that two of the compounds exhibited good herbicidal activity against dicot Arabidopsis thaliana at a concentration of 10 µg/mL, and one compound gave instinct fungicidal activity against Pythium sp. at a concentration of 2 µg/mL.  相似文献

合成了10个未见文献报道的1-(5-(2-氯苯基)-3-(2,4-二氯苯基)-4,5-二氢-N-吡唑肟酯类衍生物,并经过元素分析、HRMS、核磁共振氢谱对其结构进行了表征。对新合成的化合物进行了初步抗Bacillus subtilis, Staphylococcus aureus, Escherichia coli 和 Pseudomonas aeruginosa生物活性测试,结果表明:化合物7c 和7f对供试病菌具有较好的体外杀灭活性,其MIC值达到1.562 μg/mL;化合物7c ,7d和7f 具有中等的抑制DNA回旋酶活性(IC50 = 1.6~2.5 µg/mL)。在生物活性结果的基础上对系列化合物的构效关系进行了初步的探讨。  相似文献

An environmentally benign method has been developed for the synthesis of novel chiral thiourea derivatives in high yields in ionic liquid [Bmim]PF6. The ionic solvent can be recovered and reused without any loss of its activity. The target compounds were characterized by elemental analysis, IR, 1H NMR and 13C NMR spectral data. According to the preliminary bioassay, some of the chiral thiourea analogues exhibited moderate in vivo antiviral activities against TMV at a concentration of 500 mg/L. Title chiral compound 3i was found to possess good in vivo protection, inactivation and curative activities of 57.0%, 96.4% and 55.0%, respectively against TMV with an inhibitory concentration at 500 mg/L. The title chiral compound 3i revealed better inactivation effect on TMV (EC50=50.8 µg/mL) than Ningnanmycin (EC50=60.2 µg/mL).  相似文献

Some new quinolone condensed s‐triazine derivatives endowed with different heterocycles and 4‐aminobenzonitrile moiety has been synthesized and examined for their bioactivities against eight bacteria (Staphylococcus aureus, Bacillus cereus, Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae, Salmonella typhi, Proteus vulgaris, and Shigella flexneri), two fungi (Aspergillus niger, Candida albicans) by using agar streak dilution method, and Mycobacterium tuberculosis H37Rv by using Lowenstein and Jensen MIC method. Upon preliminary biological screening, it was observed that the majority of the compounds were found to possess a significant broad spectrum antimicrobial (MICs: 6.25–25 µg/mL) and antitubercular (MIC: 12.5 µg/mL) potential. Hence, anti‐HIV activity against two types of HIV viral strains [HIV‐1 (IIIB) and HIV‐2 (ROD)] has been carried out using the MTT assay. From this bioassay, we have identified some potent inhibitors acting as anti‐HIV‐1 agents (IC50: 4.45 µg/mL) with promising therapeutic index of 16 for analogue 7 h. The structural assignments of the new products were carried out on the basis of IR, 1H NMR, 13C NMR spectroscopy, and elemental analysis.  相似文献

通过类Mannich 反应“一锅法”合成了10个未见文献报道的含吡啶基的环状α-氨基膦酸酯, 其结构经1H NMR, 31P NMR, IR, MS和元素分析确证. 测定了该系列化合物的杀虫、杀菌和除草活性, 结果表明, 所测化合物的杀虫活性不太好, 但所有化合物在药液浓度为1.0×10-4 g/mL 时都具有较好的除草活性, 部分化合物在5.0×10-5 g/mL时显示出良好的杀菌活性.  相似文献

A synthesis, characterization, and antimicrobial study of a novel benzodipyran analog of chloramphenicol was carried out. Structure–antimicrobial activity relationship study indicates that benzodipyran analog of chloramphenicol was most active against Gram‐positive bacteria, Staphylococcus aureus, and the fungal strains Rhizoctonia bataticola and Penicillium even at minimum inhibitory concentration 10 µg/mL and showed moderate activity in other bacterial and fungal organisms. All the compounds synthesized during the present investigation were characterized by IR, 1H NMR, 13C NMR, ESI‐MS, and elemental analysis.  相似文献

In the present work, we synthesized a series of [1,2,4]triazolo[3,4‐b][1,3,4]thiadiazole derivatives ( 6a , 6b , 6c , 6d , 6e , 6f and 7a , 7b , 7c , 7d , 7e , 7f ) by using simple starting materials, namely, β‐amino acids and different aromatic acid hydrazides. The newly synthesized compounds were characterized by mass, IR, 1H, and13C‐NMR spectral data analysis. The newly synthesized compounds were tested for their antimicrobial activities and antioxidant properties. Compound 6c was a potent microbial agent particularly against Staphylococcus aureus (MIC 3.12 µg/mL) and Candida albicans (MIC 6.25 µg/mL) when compared with the reference drugs ciprofloxacin and fluconazole, respectively. The antioxidant activity of the synthesized compounds was also evaluated by 1,1‐diphenyl‐2‐picryl hydrazyl, nitric oxide, and hydrogen peroxide radical scavenging methods. Compounds 6c , 6f , 7c , and 7f showed good radical scavenging activity due to the presence of electron‐donating group on phenyl ring.  相似文献

Abstract

A novel series pyrrolo[3,2-b]pyridine-3-carboxamide linked 2-methoxypyridine derivatives have been designed, synthesized and confirmed by FT-IR, 1H NMR, 13C NMR, 19F NMR, MS, and elemental analysis. The synthesized compounds were screened for their antitubercular activity using microplate alamar blue assay method and antibacterial activity. Among the tested compounds, 4- fluorophenyl (8m), 4- chlorophenyl (8n) and 4-methoxyphenyl (8i) showed potent anti-TB activity (3.12?µg/mL) in comparison with reference drug, Pyrazinamide ((3.12?µg/mL). In addition, all compounds were docked into DprE1 (PDB code: 4KW5) to explore their binding interactions at the active site. The compounds exhibited essential key interactions as that of reported DprE1 inhibitors and hence, the synthesized compounds may be considered as molecular scaffolds for antitubercular activity. Compounds, 4-chlorophenyl (8n) and 4-flurophenyl (8m) showed significant antibacterial activity against Escherichia coli and Staphylococcus aureus strains. In silico prediction of toxicities, druglikeness and drug score profiles of the tested compounds are promising.  相似文献

A series of novel 2‐substituted methlthio‐5‐(4‐amino‐2‐methylpyrimidin‐5‐yl‐)‐1,3,4‐thiadiazole derivatives were synthesized, characterized and evaluated for antiviral activities against tobacco mosaic virus (TMV). The preliminary biological results indicated that most compounds exhibit excellent antiviral activity against TMV in vivo. Among these compounds, compounds 9c , 9i , and 9p displayed the similar curative effect against TMV (EC50 = 287.05–322.47 µg/mL) to that of the commercial agent Ningnanmycin (EC50 = 301.83 µg/mL). In particular, compound 9d demonstrated the best curative effect against TMV (EC50 = 266.21 µg/mL), which was better than that of commercial Ningnanmycin.  相似文献

A series of compounds 5‐amino‐2‐ethylmercapto‐4‐phenyl‐6‐subistitutedthieno[2,3‐d]pyrimidines ( 8a–d ), 4‐chloro‐7‐ethylmercapto‐9‐phenylpyrimido[5′,4′:4,5]thieno[3,2‐d]triazine ( 9 ), and 2‐ethylmercapto‐8‐oxo‐4‐phenyl‐7‐(4‐chlorophenyl)pyrimido [4′,5′:4,5]thieno[2,3‐d]pyrimidine ( 10 ) were synthesized and their structures were confirmed by 1H NMR , 13C NMR, and MS . All compounds were evaluated for their IC50 values against three cancer cell lines (MCF ‐7, HUH ‐7 and BHK ) and WISH cells. The IC50 of compound ( 8d ) was calculated for each cell line. Interestingly, the IC50 for the normal human amnion WISH cell line was much higher (723 µg/mL) than those found for the tumor cell lines BHK (17 µg/mL), HUH ‐7 (5.8 µg/mL), and MCF ‐7 (8.3 µg/mL). The proliferation inhibition of normal (WISH ) and tumor (BHK , HUH ‐7, and MCF ‐7) cells by compound ( 8d ) was investigated using MTT assay, and the IC50 was calculated after 48 h of treatment for each cell line.  相似文献

The novel fungicidal agents, (E )‐5‐[1‐(2‐oxo‐1‐oxaspiro[4,5]dec/non‐3‐en‐3‐yl)ethylidene]‐2‐aminoimidazolin‐ 4‐one derivatives, were designed and synthesized in moderate to excellent yields in four steps using α ‐hydroxyketone and diketene as raw materials and characterized by HR‐ESI‐MS , 1H NMR and X‐ray diffraction. The preliminary bioassay showed that some of these compounds, such as 5e , 6a , 6e , and 7 h exhibit 87.8%, 91.3%, 89.9% and 87.8% inhibition rates against Sclerotinia scleotiorum , 3b , 3c , 4c and 7 h exhibit 96.4%, 92.5%, 90.3% and 76.9% inhibition rates against Phytophthora capsici at the concentration of 50 µg/mL , respectively. These compounds exhibited significant fungicidal activities against S. scleotiorum and P. capsici with EC50 values of 2.56–11.60 µg/mL , and compounds 6e and 7 h exhibited weak inhibition against the spore germination of S. scleotiorum , while the spore germination of P. capsici was strongly inhibited by compound 7 h solution. Scanning electron microscopy (SEM ) and transmission electron microscopy (TEM ) observation indicated that compound 7 h had a significant impact on the structure and function of the hyphal cell wall of P. capsici mycelium.  相似文献
