Iron-platinum alloy nanoparticles (FePt NPs) are extremely promising candidates for the next generation of contrast agents for magnetic resonance (MR) diagnostic imaging and MR-guided interventions, including hyperthermic ablation of solid cancers. FePt has high Curie temperature, saturation magnetic moment, magneto-crystalline anisotropy, and chemical stability. We describe the synthesis and characterization of a family of biocompatible FePt NPs suitable for biomedical applications, showing and discussing that FePt NPs can exhibit low cytotoxicity. The importance of engineering the interface of strongly magnetic NPs using a coating allowing free aqueous permeation is demonstrated to be an essential parameter in the design of new generations of diagnostic and therapeutic MRI contrast agents. We report effective cell internalization of FePt NPs and demonstrate that they can be used for cellular imaging and in vivo MRI applications. This opens the way for several future applications of FePt NPs, including regenerative medicine and stem cell therapy in addition to enhanced MR diagnostic imaging.  相似文献

Conjugated polyelectrolytes (CPEs) are macromolecules with highly delocalized π‐conjugated backbones and charged side chains, which are unique types of active materials, with wide applications in optoelectronics, sensing, imaging, and therapy. By attaching specific groups (e.g., recognition elements, magnetic resonance (MR) contrast agents, gene carriers, and drugs) to the side chain or backbone of CPEs, functionalized CPEs have been developed and used for specific biological applications. In this account, we summarize the recent progress of functionalized CPEs with respect to their synthesis and biomedical applications. Future perspectives are also discussed at the end.  相似文献

Lanthanide complexes have wide applications in biochemical research and biomedical imaging. We have designed and synthesized a new class of macrocyclic lanthanide chelates, Ln/DTPA-PDA-C(n), for cell labeling and magnetic resonance imaging (MRI) applications. Two lipophilic Gd3+ complexes, Gd/DTPA-PDA-C(n) (n = 10, 12), labeled a number of cultured mammalian cells noninvasively at concentrations as low as a few micromolar. Cells took up these agents rapidly and showed robust intensity increases in T1-weighed MR images. Labeled cells showed normal morphology and doubling time as control cells. In addition to cultured cells, these agents also labeled primary cells in tissues such as dissected pancreatic islets. To study the mechanism of cellular uptake, we applied the technique of diffusion enhanced fluorescence resonance energy transfer (DEFRET) to determine the cellular localization of these lipophilic lanthanide complexes. After loading cells with a luminescent complex, Tb/DTPA-PDA-C10, we observed DEFRET between the Tb3+ complex and extracellular, but not intracellular, calcein. We concluded that these cyclic lanthanide complexes label cells by inserting two hydrophobic alkyl chains into cell membranes with the hydrophilic metal binding site facing the extracellular medium. As the first imaging application of these macrocyclic lanthanide chelates, we labeled insulin secreting beta-cells with Gd/DTPA-PDA-C12. Labeled cells were encapsulated in hollow fibers and were implanted in a nude mouse. MR imaging of implanted beta-cells showed that these cells could be followed in vivo for up to two weeks. The combined advantages of this new class of macrocyclic contrast agents ensure future imaging applications to track cell movement and localization in different biological systems.  相似文献

Polylactic acid (PLA) nanoparticles coated with Gd(III)‐based metallosurfactants (MS) are prepared using a simple and rapid one‐step method, flash nanoprecipitation (FNP), for magnetic resonance imaging (MRI) applications. By co‐assembling the Gd(III)‐based MS and an amphiphilic polymer, methoxy poly(ethylene glycol)‐b‐poly(?‐caprolactone) (mPEG‐b‐PCL), PLA cores were rapidly encapsulated to form biocompatible T1 contrast agents with tunable particle size and narrow size distribution. The hydrophobic property of Gd(III)‐based MS were finely tuned to achieve their high loading efficiency. The size of the nanoparticles was easily controlled by tuning the stream velocity, Reynolds number and the amount of the amphiphilic block copolymer during the FNP process. Under the optimized condition, the relaxivity of the nanoparticles was achieved up to 35.39 mM?1 s?1 (at 1.5 T), which is over 8 times of clinically used MRI contrast agents, demonstrating the potential application for MR imaging.  相似文献

Novel bifunctional fluorescent magneto polymeric nanoprobes (FMPNs) were synthesized to provide simultaneous diagnostic information via magnetic resonance imaging (MRI) and optical imaging. FMPNs consist of ultra-sensitive magnetic nanocrystals that function as MR probes combined with Nile Red, which functions as a fluorescent probe. FMPNs were encapsulated by a nano-emulsion method in polyvinyl alcohol (PVA, 87–89% hydrolyzed) through a matrix of polymethyl methacrylate (PMMA). FMPNs exhibited excellent colloidal stability and monodispersity. The production of MR and optical images demonstrated that FMPNs have potential as dual-mode imaging agents.  相似文献

Recently, multimodal nanoparticles integrating dual- or tri-imaging modalities into a single hybrid nanosystem have attracted plenty of attention in biomedical research. Here, we report the fabrication of two types of multimodal micelle-encapsulated nanoparticles, which were systematically characterized and thoroughly evaluated in terms of their imaging potential and biocompatibility. Optical and magnetic resonance (MR) imaging probes were integrated by conjugating DOTA-gadolinium (Gd) derivative to quantum dot based nanomicelles. Two amphiphilic block copolymer micelles, amine-terminated mPEG-phospholipid and amine-modified Pluronic F127, were chosen as the capping agents because of their excellent biocompatibility and ability to prevent opsonization and prolong circulation time in vivo. Owing to their different hydrophobic-hydrophilic structure, the micellar aggregates exhibited different sizes and protection of core QDs. This work revealed the differences between these nanomicelles in terms of the stability over a wide range of pH, along with their cytotoxicity and the capacity for chelating gadolinium, thus providing a useful guideline for tailor-making multimodal nanoparticles for specific biomedical applications.  相似文献

This work is a part of a series on surface modification of materials made of chitosan. This report focused on grafting monomethoxy ethylene glycol oligomers (mPEG) on the surface of chitosan films. The chemical reactions were performed by immersing the films in organic solvent containing aldehyde derivative of mPEG. The presence of ethylene glycol moieties was determined by attenuated total reflectance-infrared spectroscopy (ATR-IR) and nuclear magnetic resonance (NMR). The hydrophobicity of the modified surface, determined by air-water contact angle, decreased when the ethylene glycol derivatives were grafted on the film. The modified films were also subjected to protein adsorption study in order to assess their uses in biomedical applications. It was found that the presence of ethylene glycol units reduced the adsorption of proteins (albumin and lysozyme) on the films. We therefore have shown that manipulation of the interaction between chitosan and bio-macromolecules is possible by chemically modifying the surface of chitosan.  相似文献

Hybrid nanoparticles are of significant interest primarily because of their innate multifunctional capabilities. These capabilities can be exploited when hybrid nanoparticles are used for applications in the biomedical sciences in particular, where they are utilized as multimodal nanoplatforms for sensing, imaging, and therapy of biological targets. However, the realization of their biomedical applications has been difficult, in part because of a lack of high quality hybrid nanoparticles which possess high aqueous colloidal stability and biocompatibility while retaining their multifunctionalities. Here, we present the development of inorganic heterodimer nanoparticles of FePt-Au with multifunctional capabilities including catalytic growth effects, magnetic resonance (MR) contrast effects, optical signal enhancing properties, and high colloidal stability and biocompatibility. Their multimodal capabilities for biological detection are demonstrated through their utilizations in the patterned biochip based detection of avidin-biotin interaction as well as in molecular MR imaging of neuroblastoma cells.  相似文献

Magnetic Fe(2)O(3) nanorods with low cell toxicity were successfully synthesized via a wet-chemical method. In vivo studies with a rabbit model show that the nanorods exhibit excellent T(2) signal enhancement. This work demonstrates that magnetic nanorods may provide a new type of MR enhancement agent for use in biomedical applications.  相似文献

磁粒子成像是基于功能和断层影像技术检测磁性纳米粒子空间分布的示踪方法, 具有正向的对比信号、 较低的组织背景、 无限的组织穿透深度、 非侵入性成像以及无电离辐射等优点, 是近年来一种很有前途的生物医学成像技术. 磁粒子成像信号是通过在无场点切换磁性纳米粒子的磁自旋矢量来产生的. 磁粒子成像的灵敏度和空间分辨率都高度依赖于作为磁粒子成像示踪剂的磁性纳米粒子本身的磁性能, 因此目前的研究主要集中在磁性纳米粒子的设计和合成上. 本文重点介绍了磁粒子成像示踪剂的最新研究进展, 总结了可作为磁粒子成像示踪剂的磁性纳米粒子的种类、 合成方法、 性能以及生物医学应用, 以期为磁粒子成像的未来研究提供参考.  相似文献

In the present study, quantum dot (QD) capped magnetite nanorings (NRs) with a high luminescence and magnetic vortex core have been successfully developed as a new class of magnetic-fluorescent nanoprobe. Through electrostatic interaction, cationic polyethylenimine (PEI) capped QD have been firmly graft into negatively charged magnetite NRs modified with citric acid on the surface. The obtained biocompatible multicolor QD capped magnetite NRs exhibit a much stronger magnetic resonance (MR) T2* effect where the r2* relaxivity and r2*/r1 ratio are 4 times and 110 times respectively larger than those of a commercial superparamagnetic iron oxide. The multiphoton fluorescence imaging and cell uptake of QD capped magnetite NRs are also demonstrated using MGH bladder cancer cells. In particular, these QD capped magnetite NRs can escape from endosomes and be released into the cytoplasm. The obtained results from these exploratory experiments suggest that the cell-penetrating QD capped magnetite NRs could be an excellent dual-modality nanoprobe for intracellular imaging and therapeutic applications. This work has shown great potential of the magnetic vortex core based multifunctional nanoparticle as a high performance nanoprobe for biomedical applications.  相似文献

Summary: The main objective of this short state-of-the art is to report on the preparation of reactive magnetic latexes for biomedical applications. The main advantage of colloidal magnetic particles is their separation upon applying an external permanent magnetic field. Then magnetic particles bearing reactive groups or specific receptors are used in biomedical diagnosis such as immunoassay, molecular diagnosis for specific capture and detection of nucleic acids, viruses extraction and detection, cell sorting and more recently in numerous biotechnological applications. The preparation of magnetic latexes requires the use of well-defined iron oxide nanoparticles in appropriate medium. In this direction, aqueous ferrofluids were largely used to prepare functional magnetic latexes, whereas few approaches have been devoted to the use of organic ferrofluids.  相似文献

We synthesized three types of magnetic iron oxide nanoparticles (MNPs), which were meso-2,3-dimercaptosuccinic acid (DMSA) coated MNPs (DMSA@MNPs, 17.3 ± 4.8 nm, negative charge), chitosan (CS) coated MNPs (CS@MNPs, 16.5 ± 6.1 nm, positive charge) and magnetic nanoparticles agglomerates, formed by electronic aggregation between DMSA@MNPs and CS (CS-DMSA@MNPs, 85.7 ± 72.9 nm, positive charge) respectively. The interactions of these MNPs with Oral Squamous Carcinoma Cell KB were investigated. The results showed that cellular uptakes of MNPs were on the dependence of incubation time, nanoparticles concentration and nanoparticles properties such as surface charge, size, etc. The cellular uptake was enhanced with the increase of incubation time and nanoparticles concentration. Although all MNPs could enter to cells, we observed apparent differences in the magnitude of nanoparticles uptaken. The cellular uptake of CS-DMSA@MNPs by KB cells was the highest and that of DMSA@MNPs was the lowest among the three types of MNPs. The same conclusions were drawn via the reduction of water proton relaxation times , resulting from the different iron load of labeled cells using a 1.5 T clinical MR imager. The finding of this study will have implications in the chemical design of nanomaterials for biomedical applications.  相似文献

Photo‐responsive block copolymer mPEG‐b‐poly(Tyr)‐g‐NB was prepared by introduction of o‐nitrobenzyl ester group into the side chain of amphiphilic poly(ethylene glycol)‐b‐poly(α‐hydroxy acids) (mPEG‐b‐poly(Tyr)) containing pendent alkynyl group via copper‐catalyzed azide‐alkyne cycloaddition reaction. The amphiphilic mPEG‐b‐poly(Tyr) was synthesized via the ring‐opening polymerization of O‐carboxyanhydrides, with monomethoxy poly(ethylene glycol) (mPEG) as macroinitiator. The molecular structure, self‐assembly, and photo‐controlled release of the obtained mPEG‐b‐poly(Tyr)‐g‐NB were thoroughly investigated. mPEG‐b‐poly(Tyr)‐g‐NB could self‐assemble into spherical micelles in water and showed disassembly under UV light irradiation, which was demonstrated by means of UV‐vis spectroscopy, scan electron microscopes, and dynamic light scattering measurement. Fluorescence emission measurements demonstrated that Nile red, encapsulated by micelles, can be released upon UV irradiation. This study provides a convenient way to construct smart poly(α‐hydroxy acids)‐based nanocarriers for controlled release of hydrophobic drugs. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

 Magnetic fluids are used in many fields of application, such as material separation and biomedicine. Magnetic fluids consist of magnetic nanoparticles, which commonly display a broad distribution of magnetic and nonmagnetic parameters. Therefore, upon application only a small number of particles contribute to the desired magnetic effect. In order to optimize magnetic fluids for applications preference is given to methods that separate magnetic nanoparticles according to their magnetic properties. Hence, a magnetic method was developed for the fractionation of magnetic fluids. Familiar size-exclusion chromatography of two different magnetic fluids was carried out for comparison. The fractions obtained and the original samples were also magnetically characterized by magnetic resonance and magnetorelaxometry, two biomedical applications. The size-exclusion fractions are similar to those of magnetic fractionation, despite the different separation mechanisms. In this respect, magnetic fractionation has several advantages in practical use over size-exclusion chromatography: the magnetic method is faster and has a higher capacity. The fractions obtained by both methods show distinctly different magnetic properties compared to the original samples and are therefore especially suited for applications such as magnetorelaxometry. The unique properties of magnetic nanocrystals provide them with high potential as key probes and vectors in the next generation of biomedical applications. Although superparamagnetic iron oxide nanocrystals have been extensively studied as excellent magnetic resonance imaging (MRI) probes for various cell trafficking, gene expression, and cancer diagnosis, further development of in vivo MRI applications has been very limited. Here, we describe in vivo diagnosis of cancer, utilizing a well-defined magnetic nanocrystal probe system with multiple capabilities, such as small size, strong magnetism, high biocompatibility, and the possession of active functionality for desired receptors. Our magnetic nanocrystals are conjugated to a cancer-targeting antibody, Herceptin, and subsequent utilization of these conjugates as MRI probes has been successfully demonstrated for the monitoring of in vivo selective targeting events of human cancer cells implanted in live mice. Further conjugation of these nanocrystal probes with fluorescent dye-labeled antibodies enables both in vitro and ex vivo optical detection of cancer as well as in vivo MRI, which are potentially applicable for an advanced multimodal detection system. Our study finds that high performance in vivo MR diagnosis of cancer is achievable by utilizing improved and multifunctional material properties of iron oxide nanocrystal probes.  相似文献

Hybrid nanomaterials have received voluminous interest due to the combination of unique properties of organic and inorganic component in one material. In this class, magnetic polymer nanocomposites are of particular interest because of the combination of excellent magnetic properties, stability, and good biocompatibility. Organic–inorganic magnetic nanocomposites can be prepared by in situ, ex situ, microwave reflux, co-precipitation, melt blending, and ceramic–glass processing and plasma polymerization techniques. These nanocomposites have been exploited for in vivo imaging, as superparamagnetic or negative contrast agents, drug carriers, heavy metal adsorbents, and magnetically recoverable photocatalysts for degradation of organic pollutants. This review article is mainly focused on fabrication of magnetic polymer nanocomposites and their applications. Different types of magnetic nanoparticles, methods of their synthesis, properties, and applications have also been reviewed briefly. The review also provides detailed insight into various types of magnetic nanocomposites and their synthesis. Diverse applications of magnetic nanocomposites including environmental and biomedical uses have been discussed.  相似文献

在利用自主研发的专利技术制备球形磁性硅胶微球的基础上,对磁性硅胶微球进行表面改性,使其表面分别键合硅羟基、环氧基、邻二醇基和羧基等官能团,并对表面官能团进行了定量研究。以小牛胸腺基因组脱氧核糖核酸(DNA)为模型化合物,研究了核酸在不同表面官能团的磁性硅胶上的吸附和脱附行为,发现表面具有硅醇基的磁球对DNA的回收率最高。将改性后磁性微球应用于玉米DNA的提取,得到了平均长度大于8kb的高纯度基因组DNA。与传统的有机溶剂抽提法相比,基于磁性微球的核酸固相萃取法具有快速简便、省时省力、易于自动化的特点,适合于大规模植物基因组DNA的样品制备。  相似文献

Magnetic resonance (MR) is one of the most versatile and useful physical effects used for human imaging, chemical analysis, and the elucidation of molecular structures. However, its full potential is rarely used, because only a small fraction of the nuclear spin ensemble is polarized, that is, aligned with the applied static magnetic field. Hyperpolarization methods seek other means to increase the polarization and thus the MR signal. A unique source of pure spin order is the entangled singlet spin state of dihydrogen, parahydrogen (pH2), which is inherently stable and long‐lived. When brought into contact with another molecule, this “spin order on demand” allows the MR signal to be enhanced by several orders of magnitude. Considerable progress has been made in the past decade in the area of pH2‐based hyperpolarization techniques for biomedical applications. It is the goal of this Review to provide a selective overview of these developments, covering the areas of spin physics, catalysis, instrumentation, preparation of the contrast agents, and applications.  相似文献
