Shapes of the U.V. inactivation curves for single and linked double markers in Haemophilus influenzae transforming DNA 总被引:1,自引:0,他引:1

The known inverse square law S= 1/(1+CD)2, relating survival of transforming activity S to ultraviolet (u.v.) dose D, C being a constant, fails for two linked-double genetic markers in H. influenzae. For these double markers the law is S= 1/(1+CD)4, both for u.v. and nitrous acid inactivation. A theoretical model which predicts the inverse square law for singles also predicts the inverse fourth power law for linked doubles, if the possibility of an even number (>0) of genetic recombinations between the markers is allowed for. However, it can be shown rather generally that other models, not specified in exact detail, can lead to the same inverse square law for single markers, and thus to the inverse fourth power law for sufficiently separated linked-doubles. Ability to predict these laws is not, therefore, strong evidence for the validity of any one model. For all these models, the mean number of damaging lesions per unit length of DNA is directly proportional to dose, making (√(1/S)–1) for single markers and (√(1/S)–1) for loosely linked-doubles a reasonable biological measure of inactivating damage in the DNA. 相似文献

Photochemical damage to the retina occurs for prolonged exposures of intense light. Two action spectra exist for this phenomenon. In rat an action spectrum matching the absorption spectrum of rhodopsin was found. In macaque, the susceptibility for photochemical damage decreased continuously from the UV to long visible wavelengths. Later, such a spectrum was also found in rat. In search for critical parameters that determine the shape of the spectrum we gathered all available data on the damage threshold dose for monochromatic radiation and noted the experimental conditions. The rhodopsin action spectrum was found in two sources; the other 16 sources adhered to the short wavelength spectrum. Comparing the conditions we conclude that the critical parameters for the generation of either action spectrum remain elusive. Experiments are suggested to resolve this issue and fill a few gaps in our knowledge. 相似文献

A general Monte Carlo code for the simulation of X-ray fluorescence spectrometers, described in a previous paper is extended to predict the spectral response of instruments employing polarized exciting radiation. Details of the calculation method specific for the correct simulation of photon-matter scatter interactions in case of polarized X-ray beams are presented. Comparisons are made with experimentally collected spectral data obtained from a monochromatic X-ray fluorescence setup installed at a synchrotron radiation source. The use of the simulation code for quantitative analysis of intermediate and massive samples is also demonstrated. 相似文献

The OGG1 proteins are DNA N-glycosylases-apurinic-apyrimidinic lyases that are responsible for the removal of 8-oxo-7,8-dihydroguanine (8-oxoG) base in DNA. The human enzyme (hOGG1) is a monomer of 345 amino acids containing 10 buried tryptophan (Trp) residues that are very sensitive to UVB irradiation. The photolysis quantum yield of these Trp residues is about 0.3 and 0.1 in argon- and air-saturated solutions, respectively. Matrix-assisted laser desorption-ionization-time-of-flight mass spectrometry shows that several cleavage sites are identical under aerobic and anaerobic photolysis of Trp residues; one of them includes the active site. Western blots and polyacrylamide gel electrophoresis indicate that fragments of high molecular size are also formed. In addition to common photochemical paths with argon-saturated solutions, specific reactions occur in air-saturated solutions of hOGG1. The photolysis rate is inhibited by more than 50% on binding of hOGG1 to a 34mer oligonucleotide containing a single 8-oxoG-C base pair. Binding to the oligonucleotide with 8-oxoG-C induced a 20% quenching of the hOGG1 fluorescence, suggesting interaction of nucleic acid bases with the Trp residue(s) responsible for the photolysis. Using 2,6-diamino-4-hydroxy-5-N-methylformamidopyrimidine (Me-FapyG) and 8-oxoG as substrates, it is shown that protein photolysis induces photoinactivation of the DNA N-glycosylase activities. The excision of 8-oxoG is more affected than that of Me-FapyG at the same dose of UVB irradiation under both air and argon conditions. Besides the role of Trp residues, the possible involvement of Cys 253 in the photoinactivation process of hOGG1 is discussed. 相似文献

A model of UV-induced DNA damage in oceanic bacterioplankton was developed and tested against previously published and novel measurements of cyclobutane pyrimidine dimers (CPD) in surface layers of the ocean. The model describes the effects of solar irradiance, wind-forced mixing of bacterioplankton and optical properties of the water on net DNA damage in the water column. The biological part includes the induction of CPD by UV radiation and repair of this damage through photoreactivation and excision. The modeled damage is compared with measured variability of CPD in the ocean: diel variation in natural bacterioplankton communities at the surface and in vertical profiles under different wind conditions (net damage as influenced by repair and mixing); in situ incubation of natural assemblages of bacterioplankton (damage and repair, no mixing); and in situ incubation of DNA solutions (no repair, no mixing). The model predictions are generally consistent with the measurements, showing similar patterns with depth, time and wind speed. A sensitivity analysis assesses the effect on net DNA damage of varying ozone thickness, colored dissolved organic matter concentration, chlorophyll concentration, wind speed and mixed layer depth. Ozone thickness and mixed layer depth are the most important factors affecting net DNA damage in the mixed layer. From the model, the total amplification factor (TAF; a relative measure of the increase of damage associated with a decrease in ozone thickness) for net DNA damage in the euphotic zone is 1.7, as compared with 2.1-2.2 for irradiance weighted for damage to DNA at the surface. 相似文献

Biological action spectra are commonly used to assess health and ecosystem responses to increases in spectral ultraviolet (UV) irradiances resulting from stratospheric ozone (O3) reductions. For each action spectrum, a normalized sensitivity coefficient (the radiation amplification factor [RAF]) can be calculated as the relative increase in biologically active UV irradiance for a given relative decrease in the atmospheric O3 column amount. We use a detailed radiative transfer model to calculate the dependence of RAF on the O3 column amount and the solar zenith angle (and, therefore, implicitly on latitude and season) for several commonly used action spectra. A simple analytical model is used to interpret the results in terms of the semilogarithmic slope of the action spectra in the UV-B and UV-A wavelength ranges. We also show that RAF may be overestimated substantially if the UV-A portion of an action spectrum is significant but is neglected. This is illustrated using several idealized action spectra as well as published action spectra for plant responses to UV irradiation. Generally, if the portion of an action spectrum measured longward of approximately 300 nm spans less than about two orders in magnitude in its sensitivity, significant errors in the estimated RAF may ensue, and the use of this action spectrum in O3-related studies can be compromised. 相似文献

This paper describes a simple technique for the modification of titania with copper to enhance its photocatalytic performance. In addition to the absorption of UV light resulted in band-to-band excitation of electrons, TiO2 grafted with copper species absorbs radiation in the visible region of spectrum, and it is able to completely oxidize volatile organic pollutants both under UV and visible light. The action spectra of pristine and Cu-grafted TiO2 photocatalysts are measured and discussed to elucidate the reasons for appearance of the activity under visible light. 相似文献

The formation of cyclobutane pyrimidine dimers (CPD) and 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxodGuo) was investigated in Chinese hamster ovary cells upon exposure to either UVC, UVB, UVA or simulated sunlight (SSL). Two cell lines were used, namely AT3-2 and UVL9, the latter being deficient in nucleotide excision repair and consequently UV sensitive. For all types of radiation, including UVA, CPD were found to be the predominant lesions quantitatively. At the biologically relevant doses used, UVC, UVB and SSL irradiation yielded 8-oxodGuo at a rather low level, whereas UVA radiation produced relatively higher amounts. The formation of CPD was 10(2) and 10(5) more effective upon UVC than UVB and UVA exposure. These yields of formation followed DNA absorption, even in the UVA range. The calculated relative spectral effectiveness in the production of the two lesions showed that efficient induction of 8-oxodGuo upon UVA irradiation was shifted toward longer wavelengths, in comparison with those for CPD formation, in agreement with a photosensitization mechanism. In addition, after exposure to SSL, about 19% and 20% of 8-oxodGuo were produced between 290-320 nm and 320-340 nm, respectively, whereas CPD were essentially (90%) induced in the UVB region. However, the ratio of CPD to 8-oxodGuo greatly differed from one source of light to the other: it was over 100 for UVB but only a few units for UVA source. The extent of 8-oxodGuo and CPD was also compared to the lethality for the different types of radiation. The involvement of 8-oxodGuo in cell killing by solar UV radiation was clearly ruled out. In addition, our previously reported mutation spectra demonstrated that the contribution of 8-oxodGuo in the overall solar UV mutagenic process is very minor. 相似文献

We critically investigate the claim put forward by Sagripanti and Lytle ([2007] Photochem. Photobiol. 83, 1278-1282) that inactivation of influenza A virus by solar radiation can explain the seasonality of influenza epidemics. We correct an error in the Sagripanti and Lytle paper and show that changes in relative humidity and temperature affect influenza virus inactivation as strongly as variation in solar radiation. Furthermore, it appears unlikely that transmission in outdoor settings plays an important role during influenza outbreaks, because influenza A virus is sensitive to a wide range of environmental factors. 相似文献

Repair of UV induced DNA damage is of key importance to UV-induced skin carcinogenesis. Specific signal transduction pathways that regulate cell cycling, differentiation and apoptosis are found to be corrupted in skin cancers, e.g., the epidermal growth-stimulating Hedgehog pathway in basal cell carcinomas (BCCs). Mutations in genes coding for proteins in these pathways lead to persistent disturbances that are passed along to daughter cells, e.g., mutations in the gene for the Patched (PTCH) protein in the Hedgehog pathway. Thus far only the point mutations in the P53 gene from squamous cell carcinomas and BCCs, and in PTCH gene from BCC of xeroderma pigmentosum (XP) patients appear to be unambiguously attributable to solar UV radiation. Solar UVB radiation is most effective in causing these point mutations. Other forms of UV-induced genetic changes (e.g., deletions) may, however, contribute to skin carcinogenesis with different wavelength dependencies. 相似文献

We analyzed DNA duplexes modified at central guanine residues by monofunctional Ru(II) arene complexes [(eta(6)-arene)Ru(II)(en)(Cl)](+) (arene = tetrahydroanthracene or p-cymene, Ru-THA or Ru-CYM, respectively). These two complexes were chosen as representatives of two different classes of Ru(II) arene compounds for which initial studies revealed different binding modes: one that may involve DNA intercalation (tricyclic-ring Ru-THA) and the other (mono-ring Ru-CYM) that may not. Ru-THA is approximately 20 times more toxic to cancer cells than Ru-CYM. The adducts of Ru-THA and Ru-CYM have contrasting effects on the conformation, thermodynamic stability, and polymerization of DNA in vitro. In addition, the adducts of Ru-CYM are removed from DNA more efficiently than those of Ru-THA. Interestingly, the mammalian nucleotide excision repair system has low efficiency for excision of ruthenium adducts compared to cisplatin intrastrand crosslinks. 相似文献

Iodinated DNA minor groove binding bibenzimidazoles represent a unique class of UVA photosensitizer and their extreme photopotency has been previously characterized. Earlier studies have included a comparison of three isomers, referred to as ortho-, meta- and para-iodoHoechst, which differ only in the location of the iodine substituent in the phenyl ring of the bibenzimidazole. DNA breakage and clonogenic survival studies in human erythroleukemic K562 cells have highlighted the higher photo-efficiency of the ortho-isomer (subsequently designated UV(A)Sens) compared to the meta- and para-isomers. In this study, the aim was to compare the induction and repair of DNA double-strand breaks induced by the three isomers in K562 cells. Further, we examined the effects of the prototypical broad-spectrum histone deacetylase inhibitor, Trichostatin A, on ortho-iodoHoechst/UVA-induced double-strand breaks in K562 cells. Using γH2AX as a molecular marker of the DNA lesions, our findings indicate a disparity in the induction and particularly, in the repair kinetics of double-strand breaks for the three isomers. The accumulation of γH2AX foci induced by the meta- and para-isomers returned to background levels within 24 and 48 h, respectively; the number of γH2AX foci induced by ortho-iodoHoechst remained elevated even after incubation for 96 h post-irradiation. These findings provide further evidence that the extreme photopotency of ortho-iodoHoechst is due to not only to the high quantum yield of dehalogenation, but also to the severity of the DNA lesions which are not readily repaired. Finally, our findings which indicate that Trichostatin A has a remarkable potentiating effect on ortho-iodoHoechst/UVA-induced DNA lesions are encouraging, particularly in the context of cutaneous T-cell lymphoma, for which a histone deacetylase inhibitor is already approved for therapy. Capsular polysaccharide produced by Haemophilus influenzae b (Hib) is the main virulent agent and used as the antigen in the vaccine formulation. In this study, an improved process of polysaccharide purification was established based on tangential flow ultrafiltration using detergents (cocamidopropyl betaine and sodium deoxycholate), two selective ethanol precipitations steps, and extensive enzymatic hydrolysis as strategy. The relative purity (RP) related to protein and nucleic acids were 122??263 and 294??480, respectively, and compatible with the specifications established by the World Health Organization for Hib vaccine, RP????100. These results make this process simple, cheaper, efficient, environmentally friendly, and prone to be scaled up. 相似文献

A new approach to photoprotection is to repair DNA damage after UV exposure. This can be accomplished by delivery of a DNA repair enzyme with specificity to UV-induced cyclobutane pyrimidine dimers into skin by means of specially engineered liposomes. Treatment of DNA-repair-deficient xeroderma pigmentosum patients or skin cancer patients with T4N5 liposome lotion containing such DNA repair liposomes increases the removal of DNA damage in the first few hours after treatment. In these studies, a DNA repair effect was observed in some patients treated with heat-inactivated enzyme. Unexpectedly, it was discovered that the heat-inactivated T4 endonuclease V enzyme refolds and recovers enzymatic activity. These studies demonstrate that measurements of molecular changes induced by biological drugs are useful adjuvants to clinical studies. 相似文献

The reaction of cis-Pt(NH3)2Cl2(cis-DDP)-closed circular DNA adducts with DNA topoisomerse I(topo I) were studied by electron microscopy. We identified unique topoisomers such as a singly-linked catenane (2(1)2), trefoil (3(1), and dimetric catenane (2(1)2), etc., by analysis with electron micrographs. These unique recombination products resulted from cis-DDP-intra-twisting looped DNA adducts by DNA topo I, and the products could be explained a new mechanism based on an odd-even number rule. Our results suggest a new model on the working mechanisms for DNA topology of cis-DDP which enhances the recombination of DNA. Based on our results, we propose the topological idea that the yields of a mini closed circular DNA and pseudo trefoil DNA, etc., can be expected by reaction of cis-DDP-DNA-histone complexes with DNA topo I in the body. 相似文献
