用荧光光谱法研究了甲苯咪唑与牛血清白蛋白结合反应的光谱特征,证实由于非辐射能量转换机制,甲苯咪唑对牛血清白蛋白有较强的荧光猝灭能力,按照Stem—Volmer方程和双倒数方程分析处理实验数据,得到反应的结合常量和热力学参数等。  相似文献

The interactions of two organoplatinum complexes, [Pt(C^N)Cl(dppa)], 1, and [Pt(C^N)Cl(dppm)], 2 (C^N = N(1), C(2')-chelated, deprotonated 2-phenylpyridine, dppa = bis(diphenylphosphino)amine, dppm = bis(diphenylphosphino)methane), as antitumor agents, with bovine serum albumin (BSA) and human serum albumin (HSA) have been studied by fluorescence and UV-vis absorption spectroscopic techniques at pH 7.40. The quenching constants and binding parameters (binding constants and number of binding sites) were determined by fluorescence quenching method. The obtained results revealed that there is a strong binding interaction between the ligands and proteins. The calculated thermodynamic parameters (ΔG, ΔH, and ΔS) confirmed that the binding reaction is mainly entropy-driven, and hydrophobic forces played a major role in the reaction. The displacement experiment shows that these Pt complexes can bind to the subdomain IIA (site I) of albumin. Moreover, synchronous fluorescence spectroscopy studies revealed some changes in the local polarity around the tryptophan residues. Finally, the distance, r, between donor (serum albumin) and acceptor (Pt complexes) was obtained according to F?rster theory of nonradiation energy transfer.  相似文献

The interaction between N‐(4‐ethoxyphenyl)‐N′‐(4‐antipyrinyl)thiourea (EPAT) and bovine serum albumin (BSA) was studied by fluorescence spectroscopy in combination with UV absorption spectroscopy. The intrinsic fluorescence of bovine serum albumin was quenched by EPAT through a static quenching procedure. The binding constants of EPAT with BSA were estimated according to the fluorescence quenching results at different temperatures. The thermodynamic parameters: enthalpy change (ΔH) and entropy change (ΔS) were calculated to be ?10.69 kJ/mol and 42.64 J·mol?1·K?1 according to thermodynamic equations, respectively, and indicating that the binding force was suggested to be mainly a hydrophobic force. The effect of common ions on the binding constant was also investigated. A new fluorescence spectroscopy assay of the proteins was presented in this paper. The determination results of the proteins in bovine serum by means of this method were very close to those obtained using Coomassie Brilliant Blue G‐250 colorimetry.  相似文献

The interactions between potassium perfluorooctanesulfonate (PFOS) and bovine serum albumin (BSA) were studied by fluorescence spectroscopy. The association constants between PFOS and BSA were obtained by fluorescence enhancing and fluorescence quenching respectively. Furthermore, fluorescence quenching was studied at different temperatures, and the binding constant was also determined by the method of fluorescence quenching. According to the thermodynamic parameters, the main binding force could be judged. The experimental results revealed that BSA and PFOS had strong interactions. The mechanism of quenching belonged to dynamic quenching and the main sort of binding force was hydrophobic force. IR-spectra proved the interaction changed the conformation of BSA.  相似文献

The interaction of nicotinamide (NA) and bovine serum albumin (BSA) was studied by fluorescence and absorption spectroscopy at different temperatures. The results revealed that NA caused the fluorescence quenching of BSA through a static quenching procedure. The binding constants K(A), and the number of binding sites n, corresponding thermodynamic parameters DeltaG, DeltaH, DeltaS between NA and BSA at different temperatures were calculated. The primary binding pattern between NA and BSA was interpreted as hydrophobic interaction. In addition, the effect of NA on the conformation of BSA was analyzed using synchronous fluorescence spectroscopy. The binding average distance, r between the donor (BSA) and acceptor (NA) was determined based on the F?rster's theory and it was found to be 3.1 nm.  相似文献

采用荧光猝灭光谱、同步荧光光谱研究了核黄素与牛血清白蛋白(BSA)相互作用的光谱行为。结果发现,在温度为293 K和310 K时核黄素与BSA的结合常数(Kb)分别为4.879×105L.mol-1和1.880×105L.mol-1,结合热力学方程计算得到了对应温度下的热力学参数。结果表明核黄素对BSA有较强的荧光猝灭作用,其荧光猝灭过程属于动态猝灭机制,二者主要靠疏水作用力结合。采用同步荧光光谱探讨了核黄素对BSA构象的影响。  相似文献

本文应用荧光光谱法研究了可可碱(TB)与牛血清白蛋白(BSA)相互作用的光谱特性。测定了18℃、30℃、40℃温度下的结合常数KA分别为1.68×10^4、1.58×10^4、1.45×10^4L/mol,结合位点数咒分别为1.04、1.03、1.03。实验结果表明:TB对BSA内源荧光的猝灭机理主要为静态猝灭;热力学参数探讨其相互作用机理,TB主要以静电力与BSA相互作用;研究了TB对BSA构象的影响,BSA的荧光主要源于色氨酸残基。同时研究了Cu^2+存在下TB与BSA的相互作用。  相似文献

用伊文思蓝(Evans blue, EB)作荧光探针研究了氨苄青霉素(Ampicillin, A)对牛血清白蛋白(Bovine serum albumin, BSA)的竞争反应. 伊文思蓝与牛血清白蛋白作用, 使牛血清白蛋白荧光发生猝灭, 根据Stern-Volmer方程及荧光寿命研究了荧光猝灭的类型及机理. 结果表明, 猝灭类型为静态猝灭, 即伊文思蓝和牛血清白蛋白形成了一种稳定的复合物. 伊文思蓝与牛血清白蛋白的结合常数KBSA-EB=1.122×106 L/mol, 结合点数n=0.9935, 并确定了EB和BSA之间的热力学常数及作用力类型. 当加入氨苄青霉素后, 牛血清白蛋白的相对荧光强度恢复. 这表明氨苄青霉素与伊文思蓝对牛血清白蛋白发生了竞争反应. 探讨了该竞争反应的相关机理, 求出了伊文思蓝与氨苄青霉素的结合常数为KEB-A=7.131×105 L/mol.  相似文献

用荧光光谱法研究了具有抑制人肿瘤细胞活性的含喹唑啉酮的4-(4-氟苯基)哌嗪二硫代甲酸酯与牛血清白蛋白的相互作用。结果表明:在生理条件下,它对牛血清白蛋白的荧光有较强的猝灭作用。根据猝灭结果,求得了不同温度下反应的结合位点数、结合常数及反应热力学参数,并据此确定了它们相互作用的主要形式。  相似文献

The interactions of uracil (U) with bovine serum albumin (BSA) and its complex with bilirubin (BR·BSA) in phosphate buffer at pH 7.4 were studied by fluorescence and electronic spectroscopy. The parameters of the resulting intermolecular complexes (binding constants, quenching rate constants, the radius of the quenching sphere-of-action, etc.) were determined. The interaction of BSA with U occurs through a static quenching of protein fluorescence and has a predominantly hydrophobic character. The effect of U on the conformational changes of the protein molecule was analyzed by synchronous fluorescence spectroscopy. Uracil binds to BR·BSA more efficiently than to the free protein due to the interaction of U with the tetrapyrrole pigment incorporated in the macromolecular complex.  相似文献

通过荧光和紫外光谱法研究了2,4-二硝基苯胺同牛血清白蛋白(BSA)的相互作用. 2,4-二硝基苯胺对牛血清白蛋白的内源荧光具有强烈的猝灭作用. 二者之间形成不发荧光的复合物是导致荧光猝灭的主要原因. 计算了其结合常数和结合位点数. 紫外光谱法进一步证明了其猝灭机理为静态猝灭. 根据能量转移理论计算了作用距离(3.13 nm). 同步荧光的结果表明2,4-二硝基苯胺的存在改变了牛血清白蛋白的分子构象.  相似文献

The interaction between 3,4,5-trimethoxybenzoic acid (TMBA) and bovine serum albumin (BSA) was studied by fluorescence and UV-vis absorption spectroscopy. In the mechanism discussion, it was proved that the fluorescence quenching of BSA by TMBA is a result of the formation of TMBA-BSA complex. Quenching constants were determined using the Stern-Volmer equation to provide a measure of the binding affinity between TMBA and BSA. The thermodynamic parameters DeltaH, DeltaG, DeltaS at different temperatures were calculated, and electrostatic interactions play an important role to stabilize the complex. The distance r between donor (BSA) and acceptor (TMBA) was obtained according to fluorescence resonance energy transfer (FRET).  相似文献

In this article the interaction between methyl violet (MV) and bovine serum albumin (BSA) was studied with spectroscopy. The results indicated that the fluorescence intensity of BSA was quenched strongly by MV through a static quenching procedure. The association constants, the number of binding sites and basic thermodynamic parameters were obtained based on fluorescence quenching data. 运用荧光光谱(FS)、紫外光谱(UV)和超滤(UF)法, 研究了阿魏酸、毛蕊异黄酮及芒柄花素三种抗氧化小分子物质与牛血清白蛋白(BSA)的结合反应. 以Lineweaver-Burk双倒数方程和能量传递原理分别计算了三者与BSA反应的结合常数(KLB)和结合距离(r)以及由热力学参数的计算判断了三种小分子物质与BSA结合的作用力类型; 以Scatchard方程计算了三种小分子与BSA的结合常数(Ka)和结合位点数(n); 并对阿魏酸与毛蕊异黄酮及芒柄花素的竞争结合反应进行了研究. 实验结果表明: 这三种物质能与BSA结合形成非共价复合物, 导致BSA内源性荧光的静态猝灭; 它们与BSA的结合能力随着极性基团的增多、分子体积的增大而减弱; 静电作用和疏水作用可能是它们与BSA结合的主要作用力; 阿魏酸与芒柄花素或毛蕊异黄酮会产生与BSA的竞争结合效应.  相似文献

The interaction between ranitidine hydrochloride (RAN) and bovine serum albumin (BSA) in aqueous solution was investigated by means of fluorescence, synchronous fluorescence, and UV-Vis spectroscopy. The fluorescence of BSA was quenched remarkably by RAN and the quenching mechanism was concluded to be static quenching. The binding constants K and the number of binding sites n were calculated at three different temperatures. The RAN–BSA binding distance was determined to be less than 8 nm, suggesting that energy transfer may occur from BSA to RAN. The interaction process is spontaneous. Based on the obtained thermodynamic parameters, electrostatic forces may play a major role in this process. In addition, the effect of RAN on the conformation of BSA was analyzed using synchronous fluorescence spectra.  相似文献
