We describe an easy and inexpensive way to provide a highly defined Gaussian shaped laser spot on target of 5 μm diameter for imaging mass spectrometry using a commercial MALDI TOF instrument that is designed to produce a 20 μm diameter laser beam on target at its lowest setting. A 25 μm pinhole filter on a swivel arm was installed in the laser beam optics outside the vacuum ion source chamber so it is easily flipped into or out of the beam as desired by the operator. The resulting ion images at 5 μm spatial resolution are sharp since the satellite secondary laser beam maxima have been removed by the filter. Ion images are shown to demonstrate the performance and are compared with the method of oversampling to achieve higher spatial resolution when only a larger laser beam spot on target is available.

Matrix assisted laser desorption ionization imaging mass spectrometry (MALDI IMS) has the ability to provide an enormous amount of information on the abundances and spatial distributions of molecules within biological tissues. The rapid progress in the development of this technology significantly improves our ability to analyze smaller and smaller areas and features within tissues. The mammalian eye has evolved over millions of years to become an essential asset for survival, providing important sensory input of an organism’s surroundings. The highly complex sensory retina of the eye is comprised of numerous cell types organized into specific layers with varying dimensions, the thinnest of which is the 10 μm retinal pigment epithelium (RPE). This single cell layer and the photoreceptor layer contain the complex biochemical machinery required to convert photons of light into electrical signals that are transported to the brain by axons of retinal ganglion cells. Diseases of the retina, including age-related macular degeneration (AMD), retinitis pigmentosa, and diabetic retinopathy, occur when the functions of these cells are interrupted by molecular processes that are not fully understood. In this report, we demonstrate the use of high spatial resolution MALDI IMS and FT-ICR tandem mass spectrometry in the Abca4 –/– knockout mouse model of Stargardt disease, a juvenile onset form of macular degeneration. The spatial distributions and identity of lipid and retinoid metabolites are shown to be unique to specific retinal cell layers.

Mass spectrometry imaging (MSI) provides the ability to detect and identify a broad range of analytes and their spatial distributions from a variety of sample types, including tissue sections. Here we describe an approach for probing neuropeptides from sparse cell cultures using matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) MSI—at single cell spatial resolution—in both MS and tandem MS modes. Cultures of Aplysia californica neurons are grown on an array of glass beads embedded in a stretchable layer of Parafilm M. As the membrane is stretched, the beads/neurons are separated physically and the separated beads/neurons analyzed via MALDI TOF MS. Compared with direct MS imaging of samples, the stretching procedure enhances analyte extraction and incorporation into the MALDI matrix, with negligible analyte spread between separated beads. MALDI tandem MSI using the stretched imaging approach yields localization maps of both parent and fragment ions from Aplysia pedal peptide, thereby confirming peptide identification. This methodology represents a flexible platform for MSI investigation of a variety of cell cultures, including functioning neuronal networks.  相似文献

MALDI mass spectrometry imaging (MSI) enables analysis of peptides along with histology. However, there are several critical steps in MALDI MSI of peptides, 1 of which is spectral quality. Suppression of MALDI matrix clusters by the aid of ammonium salts in MALDI experiments is well known. It is asserted that addition of ammonium salts dissociates potential matrix adducts and thereafter decreases matrix cluster formation. Consequently, MALDI MS sensitivity and mass accuracy increase. Up to our knowledge, a limited number of MALDI MSI studies used ammonium salts as matrix additives to suppress matrix clusters and enhance peptide signals. In this work, we investigated the effect of ammonium phosphate monobasic (AmP) as alpha‐cyano‐4‐hydroxycinnamic acid (α‐CHCA) matrix additive in MALDI MSI of peptides. Prior to MALDI MSI, the effect of varying concentrations of AmP in α‐CHCA was assessed in bovine serum albumin tryptic digests and compared with the control (α‐CHCA without AmP). Based on our data, the addition of AmP as matrix additive decreased matrix cluster formation regardless of its concentration, and specifically, 8 mM AmP and 10 mM AmP increased bovine serum albumin peptide signal intensities. In MALDI MSI of peptides, both 8 and 10 mM AmP in α‐CHCA improved peptide signals especially in the mass range of m/z 2000 to 3000. In particular, 9 peptide signals were found to have differential intensities within the tissues deposited with AmP in α‐CHCA (AUC > 0.60). To the best of our knowledge, this is the first MALDI MSI of peptides work investigating different concentrations of AmP as α‐CHCA matrix additive to enhance peptide signals in formalin‐fixed paraffin‐embedded (FFPE) tissues. Further, AmP as part of α‐CHCA matrix could enhance protein identifications and support MALDI MSI‐based proteomic approaches.  相似文献

We have investigated the use of a Gaussian beam laser for MALDI Imaging Mass Spectrometry to provide a precisely defined laser spot of 5 μm diameter on target using a commercial MALDI TOF instrument originally designed to produce a 20 μm diameter laser beam spot at its smallest setting. A Gaussian beam laser was installed in the instrument in combination with an aspheric focusing lens. This ion source produced sharp ion images at 5 μm spatial resolution with signals of high intensity as shown for images from thin tissue sections of mouse brain.

Mass spectrometry imaging by Fourier transform ion cyclotron resonance (FT-ICR) yields hundreds of unique peaks, many of which cannot be resolved by lower performance mass spectrometers. The high mass accuracy and high mass resolving power allow confident identification of small molecules and lipids directly from biological tissue sections. Here, calibration strategies for FT-ICR MS imaging were investigated. Sub-parts-per-million mass accuracy is demonstrated over an entire tissue section. Ion abundance fluctuations are corrected by addition of total and relative ion abundances for a root-mean-square error of 0.158?ppm on 16,764 peaks. A new approach for visualization of FT-ICR MS imaging data at high resolution is presented. The ??Mosaic Datacube?? provides a flexible means to visualize the entire mass range at a mass spectral bin width of 0.001?Da. The high resolution Mosaic Datacube resolves spectral features not visible at lower bin widths, while retaining the high mass accuracy from the calibration methods discussed.  相似文献

采用沉淀法和色谱分离法将渤海某油田稠油分离成沥青质、胶质、剩余分3个组分。采用负离子电喷雾技术(ESI)结合高分辨傅立叶变换离子回旋共振质谱仪(FT-ICR MS)研究了该稠油及各组分的分子组成。结果表明,剩余分中有较少的极性杂原子化合物能被负离子ESI电离,如N_1,N_1O_1,O_1和O_2类,其等效双键数(DBE)较小。胶质和原油中极性化合物有相同的杂原子类型,包括N_1,N_1O_1,N_1O_2,N_2,N_2O_2S_1,O_1,O_2,其中胶质和原油中N_1,O_1,O_2类化合物的DBE-碳数分布图相似。沥青质中富集高缩合度且多杂原子的酸性化合物,如含杂原子N_,S的氧化程度高的化合物(N_2O_1,N_1O_3,S_1O_3)及O_3类化合物,这些物质具有较高的界面活性,易吸附在界面上促进界面张力降低和增强界面膜强度,从而有利于乳状液稳定存在。N_1O_1,N_1O_2,N_1O_3类化合物,N_2O_1类化合物可能分别是N_1化合物,N_2化合物的氧化降解产物;随着氧化降解程度增加,降解产物的极性明显增强。  相似文献

A possibility of using tryptamine as a reactive matrix for the analysis of non-polar carbonyl compounds by matrix-assisted laser desorption/ionization (MALDI) mass spectrometry has been shown. Presence of a terminal primary amine group in the tryptamine molecule predetermines the formation of Schiff bases from aliphatic and alicyclic carbonyl compounds. No additional matrix compounds are necessary to register MALDI mass spectra, because the excess of the derivatization agent plays the role of a matrix. MALDI mass spectra demonstrate high efficiency of desorption/ionization of the derivatives. To discover reactive matrices, a set of aromatic primary amines (mainly substituted anilines) has been tested, but they have not demonstrated matrix properties.  相似文献

High resolution mass spectrometry (HRMS) was successfully applied to elucidate the structure of a polyfluorinated polyether (PFPE)-based formulation. The mass spectrum generated from direct injection into the MS was examined by identifying the different repeating units manually and with the aid of an instrument data processor. Highly accurate mass spectral data enabled the calculation of higher-order mass defects. The different plots of MW and the nth-order mass defects (up to n = 3) could aid in assessing the structure of the different repeating units and estimating their absolute and relative number per molecule. The three major repeating units were -C2H4O-, -C2F4O-, and -CF2O-. Tandem MS was used to identify the end groups that appeared to be phosphates, as well as the possible distribution of the repeating units. Reversed-phase HPLC separated of the polymer molecules on the basis of number of nonpolar repeating units. The elucidated structure resembles the structure in the published manufacturer technical data. This analytical approach to the characterization of a PFPE-based formulation can serve as a guide in analyzing not just other PFPE-based formulations but also other fluorinated and non-fluorinated polymers. The information from MS is essential in studying the physico-chemical properties of PFPEs and can help in assessing the risks they pose to the environment and to human health.

样品采取索氏抽提,抽提液依次经多段混合硅胶柱、氧化铝柱和硅胶柱净化后,采用同位素稀释法和高分辨气相色谱/高分辨质谱联用仪(HRGC/HRMS)对其中的17个2,3,7,8-氯取代二噁英(PCDD/Fs)同系物进行了测定。研究结果表明,用本法4次分析二噁英标准溶液,其结果的RSD<7.1%;回收率可达62%~103%;标准参考样品的分析结果与标准值基本吻合,3次实验结果的RSD≤15%;仪器检出限为2,3,7,8-TCDF 0.1 pg,2,3,7,8-TCDD 0.2 pg,OCDD 0.8 pg;测定某降尘样品中二噁英,计算得降尘通量为14.02 pgTEQ m-2day-1。  相似文献

Resolution improvements in time-of-flight instrumentation and the emergence of the Orbitrap mass spectrometer have researchers using high resolution mass spectrometry to determine elemental compositions and performing screening methods based on the full-scan data from these instruments. This work is focused on examining instrument performance of both a QTOF and a bench-top Orbitrap. In this study, the impact of chromatographic resolution on mass measurement accuracy, mass measurement precision, and ion suppression is examined at a fundamental level. This work was extended to a mixture of over 200 pesticides to determine how well two different software algorithms componentized and correctly identified these compounds under different sets of chromatographic conditions, where co-elution was expected to vary markedly.  相似文献

该文基于心磷脂标准品的甲基化反应,建立了高灵敏度高通量分析甲基化心磷脂的液相色谱 - 高分辨质谱方法。由于甲基化后的心磷脂具有特征性的碎片,因此建立了包含780个分子特征片段的模拟数据库用于脂质鉴定。衍生化后,CL(14∶0/14∶0/14∶0/14∶0)信号提高了10倍,定量下限(LOQ)达到10 ng/mL。将该方法应用于小鼠衰老过程中心磷脂的调控研究,通过Tracefinder本地二级数据库共鉴定了43个心磷脂分子,其中21个心磷脂在衰老的大脑中有差异变化。结果发现:含有FA(20∶4)、FA(22∶6)长链多不饱和脂肪酸的心磷脂在衰老过程中呈下降趋势。高分辨质谱平台为甲基化的心磷脂分析提供了一种高通量筛选方法,对于低丰度心磷脂的检测具有很大潜力。  相似文献

Three metastable transitions are observed with 31 5-t-butyldimethylsiloxy-dehydrosterone (BDMS-DHEA) by detecting their daughter ion at m/z 269 in a double focussing mass spectrometer. Because of their specificity, the transitions of molecular ion could be served as quantitation routes for the determination of the steroid in human blood plasma using direct insertion probe with ionization chamber temperature ≥230°C. Thus, as is in MS/MS it would significantly reduce sample work-up time and possibly eliminate the need for a GC separation in detecting the level of this steroid in human blood.  相似文献

常用的气体分析质谱仪使用四极杆质谱作为分析器,分辨率一般低于300,无法解决同质量数离子带来的干扰问题.本实验自行研制了一种小型高分辨气体分析质谱仪,它采用电子轰击离子源反射式飞行时间质量分析器.仪器腔体总长45 cm,在m/z 28的位置,质量分辨率达到3000(Full width at half maximum,FWHM),实现了CO和N2的半峰谷分离;在m/z 69的位置,仪器分辨率达到5000(FWHM).在直接大气压进样条件下,可以检测到空气中136Xe(含量7.8 μ g/m3)和80Kr(含量2.8 μg/m3).使用ADC采集时,仪器的动态范围为1 06.该仪器将作为高端气体质谱仪,应用于过程监测在线分析、环境有机挥发物研究、热分析质谱及催化反应监测等领域.  相似文献

Native mass spectrometry (MS) with electrospray ionization (ESI) has evolved as an invaluable tool for the characterization of intact native proteins and non-covalently bound protein complexes. Here we report the structural characterization by high resolution native top-down MS of human thrombin and its complex with the Bock thrombin binding aptamer (TBA), a 15-nucleotide DNA with high specificity and affinity for thrombin. Accurate mass measurements revealed that the predominant form of native human α-thrombin contains a glycosylation mass of 2205 Da, corresponding to a sialylated symmetric biantennary oligosaccharide structure without fucosylation. Native MS showed that thrombin and TBA predominantly form a 1:1 complex under near physiological conditions (pH 6.8, 200 mM NH4OAc), but the binding stoichiometry is influenced by the solution ionic strength. In 20 mM ammonium acetate solution, up to two TBAs were bound to thrombin, whereas increasing the solution ionic strength destabilized the thrombin–TBA complex and 1 M NH4OAc nearly completely dissociated the complex. This observation is consistent with the mediation of thrombin-aptamer binding through electrostatic interactions and it is further consistent with the human thrombin structure that contains two anion binding sites on the surface. Electron capture dissociation (ECD) top-down MS of the thrombin–TBA complex performed with a high resolution 15 Tesla Fourier transform ion cyclotron resonance (FTICR) mass spectrometer showed the primary binding site to be at exosite I located near the N-terminal sequence of the heavy chain, consistent with crystallographic data. High resolution native top-down MS is complementary to traditional structural biology methods for structurally characterizing native proteins and protein–DNA complexes.

A high resolution Fourier transform ion cyclotron resonance (FTICR) mass spectrometer is used for characterizing the fragmentation of chlorophyll-a. Three tandem mass spectrometry (MS/MS) techniques, including electron-induced dissociation (EID), collisionally activated dissociation (CAD), and infrared mutiphoton dissociation (IRMPD) are applied to the singly protonated chlorophyll-a. Some previously unpublished fragments are identified unambiguously by utilizing high resolution and accurate mass value provided by the FTICR mass spectrometer. According to this research, the two long aliphatic side chains are shown to be the most labile parts, and favorable cleavage sites are proposed. Even though similar fragmentation patterns are generated by all three methods, there are much more abundant peaks in EID and IRMPD spectra. The similarities and differences are discussed in detail. Comparatively, cleavage leading to odd electron species and H? loss both seem more common in EID experiments. Extensive loss of small side groups (e.g., methyl and ethyl) next to the macrocyclic ring was observed. Coupling the high performance FTICR mass spectrometer with contemporary MS/MS techniques, especially IRMPD and EID, proved to be very promising for the structural characterization of chlorophyll, which is also suitable for the rapid and accurate structural investigation of other singly charged porphyrinic compounds.   相似文献

建立了利用高分辨气相色谱-高分辨质谱定量测定多溴联苯醚(PBDEs)的方法,参与了测定PBDEs的国际比对实验,实验结果表明该法是可行的。对不同地区(北京、山东临沂、上海)的3个污水处理厂的活性污泥进行了索氏提取、多层复合硅胶柱分离,然后用所建立的方法测定了19种多溴联苯醚单体的含量。结果表明,北京某污水处理厂的活性污泥中PBDEs的总量高于其他两者。  相似文献

Abstract

Methods have been developed for the quantification of low-microgram levels of the extractable organic matter contained in the atmospheric fine aerosol fraction. Extract quantification is accomplished by computer-assisted high-resolution gas chromatography (HRGC) used in conjunction with a procedural recovery mixture containing perdeuterated compounds of differing polarity and molecular weight. Recovery data for these species indicate that relative volatility rather than functional group classification is the primary factor affecting overall recovery. Routine quality control analysis is performed on a per-sample basis by high-resolution gas chromatography/mass spectrometry (HRGC/MS) for confirmation of the standard components and for identification of procedural contaminants. By the way of illustration, fine aerosol samples have been analyzed from Anaheim. California. The absolute solvent extract yields range from 49 to 346μg of organic carbon, and demonstrate a seasonal variation with winter maximum and summer minimum concentrations.  相似文献
