The incidence of fungal infections, in particular candidiasis and aspergillosis, has considerably increased during the last three decades. This is mainly due to advances in medical treatments and technologies. In high risk patients (e.g. in haematology or intensive care), the prognosis of invasive candidiasis is relatively poor. Therefore, a rapid and correct identification of the infectious agent is important for an efficient and prompt therapy. Most clinical laboratories rely on conventional identification methods that are based on morphological, physiological and nutritional characteristics. However, these have their limitations because they are time-consuming and not always very accurate. Moreover, molecular methods may be required to determine the genetic relationship between the infectious strains, for instance in Candida outbreaks. In addition, the latter methods require time, expensive consumables and highly trained staff to be performed adequately. In this study, we have applied the FTIR spectroscopic approach to different situations encountered in routine mycological diagnosis. We show the potentials of this phenotypic approach, used in parallel with routine identification methods, for the differentiation of 3 frequently encountered Candida species (C. albicans, C. glabrata and C. krusei) by using both suspensions and microcolonies. This approach, developed for an early discrimination, may help in the initial choice of antifungal treatment. Furthermore, we demonstrate the feasibility of the method for intraspecies comparison (typing) of 3 Candida species (C. albicans, C. glabrata and C. parapsilosis), particularly when an outbreak is suspected.  相似文献

1H NMR spectra in CDCl3 of poly(N-vinylpyrrolidone), epoxidized poly(N-vinyl-pyrrolidone), and products derived from the latter by modification with amino acids (glycine, β-alanine, γ-aminobutyric acid, and ε-aminocaproic acid) were examined. The 1H NMR spectra of the modified polymers contain signals for water protons due to different centers of water sorption. These signals differ in chemical shift and integral intensity and indicate a changed spatial packing of the polymer as the result of its modification. Published in Russian in Izvestiya Akademii Nauk. Synchrotron FTIR can provide high spatial resolution (<10 μm pixel size) in situ biochemical analyses of intact biotissues, an area of increasing importance in the post-genomic era, as gene functions and gene networks are coming under direct scrutiny. With this technique, we can simultaneously assess multiple aspects of cell biochemistry and cytoplasmic composition. In this paper, we report the first results of our synchrotron FTIR examination of hyphae of three important fungal model systems, each with sequenced genomes and a wealth of research: Aspergillus, Neurospora, and Rhizopus. We have analyzed the FTIR maps of Aspergillus nidulans cells containing the hypA1 allele, a well-characterized single-gene temperature-sensitive morphogenetic mutation. The hypA1 cells resemble wildtype at 28 °C but have growth defects at 42 °C. We have also investigated Neurospora and Rhizopus cultures grown in media with optimal or elevated pH. Significant differences between the spectra of the three fungi are likely related to differences in composition and structure. In addition, high spatial resolution synchrotron FTIR spectroscopy provides an outstanding method for monitoring subtle subcellular changes that accompany environmental stress. The antimicrobial properties of light is an area of increasing interest. This study investigates the sensitivity of the significant foodborne pathogen Listeria monocytogenes to selected wavelengths of visible light. Results demonstrate that exposure to wavelength region 400–450 nm, at sufficiently high dose levels (750 J cm?2), induced complete inactivation of a 5 log10 population. Exposure to wavelengths longer than 450 nm did not cause significant inactivation. Analysis of 10 nm bandwidths between 400 and 450 nm confirmed 405(±5) nm light to be most effective for the inactivation of L. monocytogenes, with a lesser bactericidal effect also evident at other wavelengths between 400 and 440 nm. Identification of the optimum bactericidal wavelength enabled the comparison of inactivation using 405(±5) nm filtered light and a 405 nm light‐emitting diode (LED) array (14 nm FWHM). Results demonstrate similar inactivation kinetics, indicating that the applied dose of 405 nm light is the important factor. Use of the 405 nm LED array for the inactivation of L. monocytogenes and other Listeria species resulted in similar kinetics, with up to 5 log10 reductions with a dose of 185 J cm?2. Comparative data for the 405 nm light inactivation of L. monocytogenes and other important foodborne pathogens, Escherichia coli, Salmonella enteritidis and Shigella sonnei, are also presented, with L. monocytogenes showing higher susceptibility to inactivation through 405 nm light exposure.  相似文献

Listeria monocytogenes is a facultative intracellular Gram-positive bacterium that is widely distributed in the environment. Despite being pathogenic at the species level, L. monocytogenes in fact comprises a diversity of strains from pathogenic ones that can result in disease and/or mortality to others that are relatively avirulent. The main goal of the current study was to answer the question on whether enhanced binding or attachment of L. monocytogenes to inert surfaces bears any relationship to pathogenicity in food-borne isolates. To answer this question, the nanoscale adhesion forces of eight L. monocytogenes strains that vary in their pathogenicity levels to a model surface of silicon nitride were quantified using atomic force microscopy. The strains used were the highly pathogenic (EGDe, 874, 1002, ATCC 19115), the intermediate pathogenic (ATCC 19112, ATCC 19118), and the non pathogenic (ATCC 15313 and HCC25). Our results indicate that the average nanoscale adhesion (in nN) and the 50% lethal dose (LD50) of strain virulence quantified in mice are logarithmically correlated according to: (nN) = −0.032 ln (LD50) + 1.040, r2 = 0.96. Such correlation indicates that nanoscale adhesion could potentially be used as a design criterion to distinguish between virulent and avirulent L. monocytogenes strains. Finally, stronger adhesion of virulent strains to inert surfaces modeled by silicon nitride might be a way for pathogenic strains to survive better in the environment and thus increase their likelihood of infecting animals or humans.  相似文献

Oxidation of (+)-sabinol, (1S,3R,5S)-1-isopropyl-4-methylidenebicyclo[3.1.0]hexan-3-ol, by active MnO2 afforded not the expected sabinone but only its [4+2]-cyclodimer. The molecular structure of the latter was established by X-ray diffraction analysis. The 1H and 13C NMR spectra of this cyclodimer were interpreted using 2D NMR spectroscopy.  相似文献

Enterocin LR/6, a purified bacteriocin, exhibited broad inhibitory spectrum both against related as well as some food-borne pathogens such as Listeria monocytogenes, Yersinia enterocolitica, Aeromonas sp., Shigella sp., and Bacillus licheniformis. In this investigation, we have focused on L. monocytogenes as the target organism, as it is not only an important pathogen but can also survive over a wide range of environmental conditions such as refrigeration temperature, low pH, and high-salt concentration. This allows the pathogen to overcome many food preservation and safety barriers and poses a potential risk to human health. The enterocin LR/6 showed a bactericidal action against L. monocytogenes and completely inhibited the growth on agar plates, supplemented with 200 AU/ml of enterocin LR/6. The effectiveness of enterocin LR/6 in completely killing a population of acid-adapted (pH 5.2, 2 h) L. monocytogenes exposed to different temperatures (437 °C), pH (2.5–8.0), and osmotic (up to 30% NaCl) stress is reported here. This paper focuses on the key issue of killing of the acid-adapted L. monocytogenes cells under adverse environmental conditions.  相似文献

The chemical composition of the essential oils of Achillea clavennae L., Achillea holosericea Sibth. & Sm., Achillea lingulata W. & K., and Achillea millefolium L. from the Balkans was determined by GC and GC/MS analyses. The main components were 1,8-cineole in A. holosericea, camphor in A. clavennae, β-pinene in A. millefolium, and τ-cadinol in A. lingulata. Different extraction methods for the subsequent gas chromatographic determination of the composition of essential oils and related compounds from marjoram (Origanum majorana L.), caraway (Carum carvi L.), sage (Salvia officinalis L.), and thyme (Thymus vulgaris L.) have been compared. The comparison was also discussed with regard to transformation processes of genuine compounds, particularly in terms of expenditure of time. Hydrodistillation is the method of choice for the determination of the essential oil content of plants. For investigating the composition of genuine essential oils and related, aroma-active compounds, hydrodistillation is not very useful, because of discrimination and transformation processes due to high temperatures and acidic conditions. With cold solvent extraction, accelerated solvent extraction, and supercritical fluid extraction, discrimination of high and non-volatile aroma-active components as well as transformation processes can be diminished, but non-aroma-active fats, waxes, or pigments are often extracted, too. As solid-phase microextraction is a solvent-free fully automizable sample preparation technique, this was the most sparing to sensitive components and the most time-saving method for the rapid determination of the aroma compounds composition in marjoram, caraway, sage, and thyme. Finally, solid-phase microextraction could be successfully optimized for the extraction of the aroma components from the plants for their subsequent gas chromatographic determination.  相似文献

There are many changes, both qualitative and quantitative, in eucalypt waste during growth and fructification of Lentinula edodes. Wet chemical analysis and near-infrared (NIR) spectroscopy were used in conjunction with multivariate regression and principal components analysis to monitor biodegradation of eucalyptus waste during growth of several L. edodes strains. Weight and component losses of eucalypt residue after biodegradation by L. edodes strains were compared for periods of 1 to 5 mo. Decrease in cellulose, hemicellulose, and lignin contents occurred, however it was not concomitant. Measurement of lignin degradation by NIR and wet chemical analysis indicated its attack in the early stages of biodegradation. Selective lignin degradation by L. edodes was observed up to 2 mo of biodegradation for strains DEBIQ and FEB-14. One group of degraded substrate was identified based on the principal component analysis (PCA) of the data on their biodegradation time. Samples treated for 5 months by L. edodes strains (DEBIQ, UFV or FEB-14) differed from other, but no discrimination was observed among them. By the end of 5 mo, NIR analyses showed decrease of about 18–47% cellulose, 35–47% polyose and 39–60% lignin. These data were used for comparison with those obtained by wet chemical method for the degradation of the substrate by other five L. edodes strains cultivated at the same conditons. NIR calibration developed in this study was proven to be perfectly suitable as an analytical method to predict the changes in lignocellulose composition during biodegradation.  相似文献

The infrared spectra of monothiodiacetamide (MTDA, CH3CONHCSCH3) and its N-deuterated compound in solution, solid state and at low temperature are measured. Normal coordinate analysis for the planar vibrations ofMTDA-d 0 and –d 1 have been performed for the two most probablecis-trans-CONHCS-or-CSNHCO-conformers using a simpleUrey-Bradley force function. The conformation ofMTDA derived from the vibrational spectra is supported by the all valence CNDO/2 molecular orbital method. The vibrational assignments and the electronic structure ofMTDA are also given.

Zusammenfassung Das Infrarotspektrum von Monothiodiacetamid (MTDA, CH3CONHCSCH3) und der entsprechenden N-deuterierten Derivate wurde in Lösung, im Festzustand und bei tiefer Temperatur gemessen. Die Normalkoordinatenanalyse der möglichen planaren Konformeren wurde mittels einer einfachenUrey-Bradley-Funktion durchgeführt. Die dabei abgeleitete Konformation vonMTDA steht im Einklang mit einer semiempirischen quantenchemischen Rechnung (CNDO/2). Die Zuordnung der Schwingungen und die Elektronendichteverteilung inMTDA werden angegeben.

EPR spectral investigations have been carried out on four edible leafy vegetables of India, which are used as dietary component in day to day life. In Rumex vesicarius leaf sample, EPR spectral investigations at different temperatures indicate the presence of anti-ferromagnetically coupled Mn(IV)–Mn(IV) complexes. EPR spectra of Trigonella foenum graecum show the presence of Mn ions in multivalent state and Fe3+ ions in rhombic symmetry. EPR spectra of Basella rubra indicate the presence of Mn(IV)–O–Mn(IV) type complexes. The EPR spectra of Basella rubra have been studied at different temperatures. It is found that the spin population for the resonance signal at g = 2.06 obeys the Boltzmann distribution law. The EPR spectra of Moringa oliefera leaves show the presence of Mn2+ ions. Radiation induced changes in free radical of this sample have also been studied. The FT-IR spectra of Basella rubra and Moringa oliefera leaves show the evidences for the protein matrix bands and those corresponding to carboxylic CO bonds.  相似文献

The ability of adsorbed biosurfactants (Pf and Lb) obtained from gram-negative bacterium (Pseudomonas fluorescens) or gram-positive bacterium (Lactobacillus helveticus) to inhibit adhesion of four listerial strains to stainless steel was investigated. These metallic surfaces were characterized using the following complementary analytical techniques: contact-angle measurements (CAM), atomic force microscopy (AFM), polarization modulation-infrared reflection-adsorption spectroscopy (PM-IRRAS) and X-ray photoelectron spectroscopy (XPS). Contact-angles with polar liquids (water and formamide) indicated that the stainless steel surface covered with adsorbed biosurfactant was more hydrophilic and electron-donating than bare stainless steel. The surface characterization by XPS and PM-IRRAS revealed that conditioning the stainless steel changes the substrate in two ways, by modifying the surface alloy composition and by leaving an thin adsorbed organic layer. AFM observations enabled to say that the layer covered entirely the surface and was probably thicker (with patches) in the case of Pf-conditioned surfaces compared to the Lb-conditioned ones, which seemed to be less homogeneous. Though the added layer was thin, significant chemical changes were observed that can account for drastic modifications in the surface adhesive properties. As a matter of fact, adhesion tests showed that both used biosurfactants were effective by decreasing strongly the level of contamination of stainless steel surfaces by the four strains of Listeria monocytogenes. The more important decrease concerned the CIP104794 and CIP103573 strains (>99.7%) on surface conditioned by L. helveticus biosurfactant. A less reduced phenomenon (75.2%) for the CIP103574 strain on stainless steel with absorbed biosurfactant from P. fluorescens was observed. Whatever the strain of L. monocytogenes and the biosurfactant used, this antiadhesive biologic coating reduced both total adhering flora and viable and cultivable adherent bacteria on stainless steel surfaces. This study confirms that biosurfactants constitute an effective strategy to prevent microbial colonization of metallic surfaces by pathogenic bacteria like the food-borne pathogen L. monocytogenes.  相似文献

The adhesion energies between pathogenic Listeriamonocytogenes EGDe to a model surface of silicon nitride were quantified using atomic force microscopy (AFM) in water for cells grown in pure media (as the control) and in media of four different ionic strengths of added NaCl (IS of 0.05 M, 0.1 M, 0.3 M and 0.5 M NaCl). The physiochemical properties of L. monocytogenes EGDe surface brushes were shown to have a strong influence on the adhesion of the microbe to the silicon nitride surface. The transitions in the adhesion energies, physiochemical properties, and the structure of bacterial surface polyelectrolyte brushes were observed for the cells grown in the media of 0.1 M added NaCl. Our results suggested that the highest long-range electrostatic repulsion which was partially balanced by the Liftshitz-van der Waals attraction for the cells grown at 0.1 M was responsible for the highest energy barrier to adhesion for these cells as predicted by the soft-particle analysis of DLVO theory and the lower adhesion measured by AFM.  相似文献

采用微波固相法制备了CaWO_4∶xEu~(3+),ySm~(3+),zLi~+红色荧光粉。测量样品的XRD图、激发谱、发射谱及发光衰减曲线,研究并分析了Eu~(3+)、Sm~(3+)、Li~+的掺杂浓度,对样品微结构、光致发光特性、能量传递及能级寿命的影响。结果表明,Eu~(3+)、Sm~(3+)、Li~+掺杂并未引起合成粉体改变晶相,仍为CaWO_4单一四方晶系结构。Eu~(3+)、Sm~(3+)共掺样品中,Sm~(3+)掺杂为3%时,Sm~(3+)对Eu~(3+)的能量传递最有效。Li~+掺杂起到了助熔剂和敏化剂的作用,使样品发光更强。在394 nm激发下,与CaWO_4∶3%Eu~(3+)样品比较,3%Eu~(3+)、3%Sm~(3+)共掺CaWO_4及3%Eu~(3+)、3%Sm~(3+)、1%Li~+共掺CaWO_4样品的发光分别增强2倍及2.4倍。同一激发波长下,单掺Eu~(3+)样品寿命最短,Sm~(3+)、Eu~(3+)共掺样品随Sm~(3+)浓度增加,寿命先减小后增加,且掺杂了Li~+的样品比不掺Li~+的样品~5D_0能级寿命有所增加。  相似文献

Oxidation of amalgamated magnesium metal with 3,6-di-tert-butyl-o-benzoquinone (1) in different aprotic organic solvents afforded magnesium catecholate and bis-o-semiquinolate complexes. The catecholate derivatives of magnesium CatMgL2 (Cat is the 3,6-di-tert-butyl-o-benzoquinone dianion, L = THF or Py) were synthesized in high yields in pyridine and tetrahydrofuran, respectively. The reactions in diethyl ether or dimethoxyethane produced hexacoordinated metal bis-o-semiquinolates SQ2MgLn (SQ is the 3,6-di-tert-butyl-o-benzoquinone radical anion, L = Et2O, n = 2; L = DME, n = 1). The reaction with the use of toluene as the solvent gave a magnesium bis-o-semiquinolate complex containing the coordinated unreduced o-quinone molecule. The molecular structures of the [CatMgPy2]2 and SQ2Mg·DME complexes were established by X-ray diffraction. Published in Russian in Izvestiya Akademii Nauk. Formononetin and the new isoflavonoid glycosides formononetin-7-O-β-D-galactopyranoside and inermin-3-O-β-D-galactopyranoside were isolated from Trifolium pratense L. roots. Ion/molecule reactions of four coordinateSchiff base complexes under negative ion chemical ionization conditions have been studied. The complex metal ions consisted of cobalt(II), nickel(II) and copper(II).Schiff base ligands with different donor strengths were employed. The gas mixtures used contained 90% methane and 10% of the gases O2, NO or CO. The spectra showed intense molecular negative ions, formed by secondary electron capture processes. Secondary ions were formed via ion/molecule reactions between the parent molecular negative ion and added gas molecules to giveMLX ,X=O2, NO, CO;L=Schiff base ligand,M=Co(II) or Ni(II). Consistent with former investigations, secondary ion formation was not found for the copper compounds. Influence of the central metal ion as well as the ligand donor strength on the ion/molecule reactions are discussed. From the results obtained a mechanism of the secondary ion formation is suggested.
