Synthesis of a Resorcylic Acid Lactone (RAL) Library Using Fluorous‐Mixture Synthesis and Profile of its Selectivity Against a Panel of Kinases

A library of resorcylic acid lactones (RAL) containing a cis‐enone moiety targeting kinases bearing a cysteine residue within the ATP‐binding pocket was prepared using a fluorous‐mixture synthesis and evaluated against a panel of 19 kinases thus providing important structure–activity trends. Two new analogues were then profiled for their selectivity against a panel of 402 kinases providing the broadest evaluation of this pharmacophores’ selectivity.     

Resorcylic acid lactones: A pluripotent scaffold with therapeutic potential

The recent discoveries of potent HSP90 and MAP kinase inhibitors amongst the resorcylic acid lactones (RALs) have revived interest in this family of natural products. Both HSP90 and MAP kinase inhibition hold tremendous therapeutic potential, particularly in the treatment of cancer. Our synthetic efforts towards the RALs and, in particular, selective inhibitors of HSP90 and kinases are reviewed.     

Total Synthesis of Resorcylic Acid Lactone (−)-Neocosmosin A Using Palladium-Catalyzed α-Arylation of Enones as the Key Step

The total synthesis of the 14-membered resorcylic acid lactone neocosmosin A is described. The key step in the synthesis is the palladium-catalyzed α-arylation of TES-enol ethers of enones. The employment of the α-arylation approach to this class of resorcylic acid lactones is a new approach, which has the scope of being generalized into a unified approach for the synthesis of this class of natural products.     

Brominated resorcylic acid lactones from the marine-derived fungus Cochliobolus lunatus induced by histone deacetylase inhibitors

Chemical epigenetic manipulation was applied to the marine-derived fungus Cochliobolus lunatus (TA26-46) with histone deacetylase inhibitors, resulting in significant changes of the secondary metabolites. Two new 14-membered resorcylic acid lactones characterized with bromine substitution, 5-bromozeaenol (1) and 3,5-dibromozeaenol (2), together with four known analogues (3–6), were isolated from the culture treated with sodium butyrate. The absolute configurations of 1 and 2 were assigned by CD spectra and chemical conversion. Compounds 1 and 2 represent the first example of natural brominated resorcylic acid lactones. These brominated lactones were exclusively obtained from C. lunatus (TA26-46) following epigenetic modifying treatments.     

Chemistry and biology of resorcylic acid lactones

While resorcylic acid lactones (RALs) have been known for a long time, the more recent discoveries that radicicol is a potent and selective HSP90 inhibitor while other members such as hypothemycin, LL-Z1640-2 and LL-783,277 are potent kinase inhibitors have stimulated a renewed interest in this family of natural products. The recent developments regarding the chemistry and biology of RALs are reviewed.     

Ultra-high-pressure liquid chromatography–tandem mass spectrometry for the analysis of six resorcylic acid lactones in bovine milk

This work reports a rapid, reliable and sensitive multi-residue method for the simultaneous determination of six resorcylic acid lactones in bovine milk by ultra-high-pressure liquid chromatography–tandem mass spectrometry (UHPLC–MS/MS). The resorcylic acid lactones were extracted, purified, and concentrated from milk samples in one step using a solid-phase extraction (SPE) cartridge that contained a polymeric mixed-mode anion-exchange sorbent. The analysis was performed on a Waters Acquity BEH C₁₈ column utilizing a gradient elution profile. Each LC run was completed in 3.5 min. The analytes were detected by multiple reaction monitoring (MRM) using electrospray ionization (ESI) negative mode. Mean recoveries from fortified samples ranged from 92.6% to 112.5%, with relative standard deviations lower than 11.4%. Using 5 mL bovine milk, the limits of detection and quantification for resorcylic acid lactones were in the ranges of 0.01–0.05 and 0.05–0.2 μg/L, respectively. The application of this newly developed method was demonstrated by analyzing bovine milk samples from markets.     

Resorcylic acid lactones as new lead structures for kinase inhibition

The naturally occurring resorcylic acid lactones LL-Z1640-2 (1), hypothemycin (2), L-783277 (3), radicicol A (4) and, to a somewhat lesser extent, aigialomycin D (5) have recently emerged as new lead structures for kinase inhibition. Total syntheses have now been reported for all of these natural products, most of which are based on macrocyclization through ester bond formation. However, RCM-based approaches have also been described and a variety of strategies have been pursued to obtain the requisite seco acids or dienes, respectively, as precursors for macrocyclization.     

Access to resorcylic acid lactones via phosphonate based intramolecular olefination

An approach to resorcylic acid lactones is described, exploiting an intramolecular olefination reaction for the generation of the 14-membered macrolactone. The synthetic route gave zearalenone precursors, and the preparations of other RAL analogues, trans- and cis-resorcylides are included, the latter being prepared by photoisomerization of the trans-isomer. β-Haloketone derivatives were also prepared in a highly stereoselective manner by conjugate addition of chloride or bromide to the E-enone using boron trichloride and boron tribromide, respectively.     

Chemical syntheses of the cochliomycins and certain related resorcylic acid lactones

The cochliomycins (7–12) are a group of six resorcylic acid lactones that have recently been isolated from culture broths of marine fungi found in the South China Sea. These natural products have attracted attention as synthetic targets because of (in certain instances) their novel structural features and their capacities to suppress biofouling. This short review summarizes the synthesis of these and some related compounds that have been reported to date, including those developed in the authors’ laboratories.     

An efficient and enantioselective total synthesis of naturally occurring L-783277

Naturally occurring L-783277 which belongs to 14-membered resorcylic acid lactones (RALs) turned out to be a potent kinase inhibitor against MEK (MAP kinase kinase). We successfully accomplished efficient and enantioselective total synthesis of L-783277 based on convergent assembly of one aromatic unit and two chiral building blocks with efficient orthogonal protection-deprotection strategy. Three key steps composed of olefin cross metathesis, addition of acetylene derivative to aldehyde, and Yamaguchi macrolactonization were subsequently employed to construct the framework of L-783277. The optical rotation value of L-783277 is for the first time presented in this Letter.     

液相色谱-串联质谱法测定谷物类饲料中的41种激素

建立了谷物类饲料中10种雄性激素、11种孕激素、10种糖皮质激素、5种雌性激素以及5种二羟基苯甲酸内酯类药物的液相色谱-串联质谱(LC-MS/MS)检测方法。均质样品经乙酸乙酯提取,以改良的QuEChERS吸附剂分散于提取液中实现快速净化。采用C18色谱柱(150 mm×2.1 mm, 3.0 μm)分离,分别在正、负电喷雾串联质谱多反应监测模式下检测。在优化条件下,41种化合物在5.0～200.0 μg/kg含量范围内有良好的线性相关性,相关系数均不低于0.99,定量限(S/N≥10)为0.123～2.72 μg/kg。在5、40、200 μg/kg或10、40、200 μg/kg 3个添加水平下,豆粕类饲料中各化合物的回收率为70.3%～118.1%,相对标准偏差(RSD)为0.82%～19.5%;玉米类饲料中各化合物的回收率为76.1%～122.8%, RSD为1.3%～15.0%。该方法简便、快速、准确,可用于谷物类饲料中雄性激素、孕激素、糖皮质激素、雌性激素以及二羟基苯甲酸内酯类激素的筛查和测定。     

Rapid and scalable synthesis of chiral bromolactones as precursors to α-exo-methylene-γ-butyrolactone-containing sesquiterpene lactones

The sesquiterpene lactones cover a diverse and pharmacologically important diversity space. In particular, the electrophilic α-exo-methylene-γ-butyrolactone moiety that is preponderant in this natural product family has been shown to readily engage in covalent inhibition via conjugate addition of cysteine residues in target proteins. However, the synthetic accessibility of sesquiterpenes or related probes to investigate their mode of action remains laborious. Herein, we present a rapid and scalable route to chiral bromolactones as enabling precursors in the synthesis of sesquiterpene lactones.     

Inhibition of Hsp90 with synthetic macrolactones: synthesis and structural and biological evaluation of ring and conformational analogs of radicicol

A series of benzo-macrolactones of varying ring size and conformation has been prepared by chemical synthesis and evaluated by structural and biological techniques. Thus, 12- to 16-membered lactones were obtained by concise routes, involving ring-closing metathesis as a key step. In enzyme assays, the 13-, 15-, and 16-membered analogs are good inhibitors, suggesting that they can adopt the required conformation to fit in the ATP-binding site. This was confirmed by cocrystallization of 13-, 14-, and 15-membered lactones with the N-terminal domain of yeast Hsp90, showing that they bind similarly to the "natural" 14-membered radicicol. The most active compounds in the ATPase assays also showed the greatest growth-inhibitory potency in HCT116 human colon cancer cells and the established molecular signature of Hsp90 inhibition, i.e., depletion of client proteins with upregulation of Hsp70.     

Development and in-house validation of an LC-MS/MS method for the determination of stilbenes and resorcylic acid lactones in bovine urine

Stilbenes and zeranol are nonsteroidal estrogenic growth promoters which are banned in the European Union (EU) for use in food-producing animals by Council Directive 96/22/EC. A liquid chromatography–tandem mass spectrometry (LC-MS/MS) method was developed for the screening and confirmation of stilbenes (diethylstilbestrol, dienestrol, hexestrol) and resorcylic acid lactones (zeranol and its metabolites taleranol and zearalanone as well as the mycotoxins α-zearalenol, β-zearalenol and zearalenone) in bovine urine. The method permits the confirmation and quantification of stilbenes and resorcylic acid lactones at levels below 1 μg L⁻¹ and 1.5 μg L⁻¹, respectively. The validation was carried out according to Commission Decision 2002/657/EC, Chap. 3.1.3 “alternative validation” by a matrix-comprehensive in-house validation concept. Decision limit CCα, detection capability CCβ, recovery, repeatabiliy, within-laboratory reproducibility and the uncertainty of measurement were calculated. Furthermore, a factorial effect analysis was carried out to identify factors that have a significant influence on the method. Factors considered to be relevant for the method in routine analysis (e.g. operator, matrix condition, storage duration of the extracts before measurement, different cartridge lots, hydrolysis conditions) were systematically varied on two levels. The factorial analysis showed that different cartridge lots, storage durations and matrix conditions can exert a relevant influence on the method.     

Total synthesis of greensporone C

The first total synthesis of greensporone C, a cytotoxic 14-membered resorcylic acid lactone, has been accomplished via a longest linear sequence of 16 steps in 3.3% overall yield. The key features of the synthesis include Mitsunobu esterification and ring-closing metathesis to construct the macrocycle and establish the (E)-olefin geometry, respectively. Our synthesis also confirmed the absolute stereochemistry of the natural product.     

Soluble-polymer supported synthesis of a prostanoid library: identification of antiviral activity

[formula: see text] The prostaglandins are potent natural products taking part in many biological processes. The "convergent generation of diversity" from a "toolbox" of prostanoid components, augmented with additional polymer-supported transformations, can enable construction of valuable libraries. A parallel-pool strategy was used to assemble a small library of prostanoids. The inhibition of a herpes-family virus demonstrated the potential for new drug discovery.     

Synthesis,kinase activity and molecular modeling of a resorcylic acid lactone incorporating an amide and a trans-enone in the macrocycle

Details for the synthesis of a resorcylic acid lactone (RAL) incorporating a trans-enone and an amide in the macrocyclic ring are provided herein. The sequence included the assembly of three fragments by esterification, olefination, and lactamization. The RAL with the lactam was less potent as an inhibitor of kinases than other RALs investigated. The biological results were rationalized by docking and molecular dynamics simulations of the lactam bound to human ERK2 and comparison with hypothemycin.     

Determination of Six Resorcylic Acid Lactones in Feed by GC–MS

A method was developed to simultaneously detect six resorcylic acid lactones in feed by GC–MS. Samples were extracted with methanol followed by a two step liquid–liquid extraction and an HLB SPE clean-up. The samples were derivatized with BSTFA + TMCS (99/1; v/v) and determined by GC–MS. For all analytes, the ranges of recoveries were 81.2–98.2%, with RSDs of 3.2–15.2%, and the LODs were 0.2–0.6 μg kg⁻¹.
