Three Ru(II) complexes [Ru(bpy)2(1-IQTNH)](ClO4)2 (1), [Ru(bpy)2(2-QTNH)](ClO4)2 (2) and [Ru(bpy)2(3-IQTNH)](ClO4)2 (3) (bpy = 2,2′-bipyridine, 1-IQTNH = 6-(isoquinolin-1-yl)-1,3,5-triazine- 2,4-diamine, 2-QTNH = 6-(quinolin-2-yl)-1,3,5-triazine- 2,4-diamine, 3-IQTNH = 6-(isoquinolin-3-yl)-1,3,5-triazine-2,4-diamine) have been synthesized and characterized by elemental analysis, 1H NMR spectroscopy, electrospray ionization mass spectrometry and X-ray crystallography. The electrochemical and spectroscopic properties of the complexes differ from those of [Ru(bpy)3]2+ owing to the structural differences between the ligands and their complexes.  相似文献

Two new Ruthenium (II) polypyridyl complexes [Ru(dmp)2(ipbp)](ClO4)2 (1) (dmp = 2,9-dimethyl-1,10-phenanthroline, ipbp = 3-(1H-imidazo[4,5-f][1,10]phenanthrolin-2-yl)-4H-1-banzopyran-2-one) and [Ru(dmb)2(ipbp)](ClO4)2 (2) (dmb = 4,4′-dimethyl-2,2′-bipyridine) have been synthesized and characterized by elemental analysis, FAB-MS, ES-MS and 1H NMR and cyclic voltammetric methods. The DNA-binding behaviors of these complexes were investigated by spectroscopic titration, viscosity measurements, and thermal denaturation. Absorption titration and thermal denaturation studies reveal that these complexes are moderately strong binders of calf thymus DNA (CT-DNA). Viscosity measurements show that the complexes 1 and 2 interact with CT-DNA by intercalative mode. The DNA-binding affinity of the complex 2 is larger than that of complex 1.  相似文献

设计并合成了3个含有席夫碱基团的配体(BLn,n=1~3)及相应的双核联吡啶钌配合物[Ru (BLn)(bpy)2]2(ClO44,其中bpy=2,2''-联吡啶,BLn=((PyCHN)-Ph-O-C6H42R (PyCHN=N-2-吡啶亚甲基,R=none (Ru1),—C (CH32Ru2),—SO2Ru3))。通过元素分析、核磁氢谱、红外光谱、质谱等方法对配体和配合物进行了表征。通过体外细胞毒性实验研究了Ru1~Ru3对宫颈癌细胞(Hela)、胃癌细胞(BGC823)、胃癌细胞(SGC-7901)三种肿瘤细胞和人正常胚肺成纤维细胞MRC-5的细胞毒性,实验结果表明Ru3的抗肿瘤活性最好,而Ru1~Ru3均对BGC823具有选择性。  相似文献

Two novel RuII complexes [Ru(dppt)(bpy)Cl]ClO4 (1) and [Ru(pta)(bpy)Cl]ClO4 (2)[dppt, pta and bpy = 3-(1,10-phenanthrolin-2-yl)-5,6-diphenyl-as-triazine, 3-(1,10-phenanthrolin-2-yl)-as-triazino[5,6-f]acenaphthylene and 2,2-bipyridine, respectively] were synthesized and characterized by elemental analysis and electrospray mass spectrometry, 1H-n.m.r., and u.v.–vis spectroscopy. The redox properties of the complexes were examined using cyclic voltammetry. Due to the strong -accepting character of asymmetric ligands, the MLCT bands of (1) and (2) are shifted significantly to lower energies by comparison with [Ru(tpy)(bpy)Cl]+.  相似文献

Three novel unsymmetric tridentate ligands, namely, ptmi (ptmi = 3-(1,10-phenanthroline-2-yl)-as-triazino[5,6-f]-5-methoxyisatin), pti (pti = 3-(1,10-phenanthroline-2-yl)-as-triazino-[5,6-f]isatin), ptni (ptni = 3-(1,10-phenanthroline-2-yl)-as-triazino[5,6-f]-5-nitroisatin), and their complexes [Ru(tpy)(ptmi)](ClO4)2 (tpy = 2,2′:6′,2″-terpyridine) (1), [Ru(tpy)(pti)](ClO4)2 (2), and [Ru(tpy)(ptni)](ClO4)2 (3) were prepared and characterized by elemental analysis, 1H NMR, ES–MS. The electrochemical behaviors were studied by cyclic voltammetry. The DNA-binding properties of these complexes were investigated by the spectroscopic method, viscosity measurements, and thermal denaturation. Theoretical studies on these complexes were also performed with the density functional theory (DFT) method. The experimental results showed that these complexes bind to calf thymus (CT-DNA) in an intercalative mode. The order of DNA-binding affinities (A) of these complexes is A(1) < A(2) < A(3). The trend in the DNA-binding affinities of this series of complexes can be reasonably explained by the DFT calculations.  相似文献

This article deals with the hitherto unexplored metal complexes of deprotonated 6,12‐di(pyridin‐2‐yl)‐5,11‐dihydroindolo[3,2‐b]carbazole (H2L). The synthesis and structural, optical, electrochemical characterization of dimeric [{RuIII(acac)2}2(μ‐L.?)]ClO4 ([ 1 ]ClO4, S=1/2), [{RuII(bpy)2}2(μ‐L.?)](ClO4)3 ([ 2 ](ClO4)3, S=1/2), [{RuII(pap)2}2(μ‐L2?)](ClO4)2 ([ 4 ](ClO4)2, S=0), and monomeric [(bpy)2RuII(HL?)]ClO4 ([ 3 ]ClO4, S=0), [(pap)2RuII(HL?)]ClO4 ([ 5 ]ClO4, S=0) (acac=σ‐donating acetylacetonate, bpy=moderately π‐accepting 2,2’‐bipyridine, pap=strongly π‐accepting 2‐phenylazopyridine) are reported. The radical and dianionic states of deprotonated L in isolated dimeric 1 +/ 2 3+ and 4 2+, respectively, could be attributed to the varying electronic features of the ancillary (acac, bpy, and pap) ligands, as was reflected in their redox potentials. Perturbation of the energy level of the deprotonated L or HL upon coordination with {Ru(acac)2}, {Ru(bpy)2}, or {Ru(pap)2} led to the smaller energy gap in the frontier molecular orbitals (FMO), resulting in bathochromically shifted NIR absorption bands (800–2000 nm) in the accessible redox states of the complexes, which varied to some extent as a function of the ancillary ligands. Spectroelectrochemical (UV/Vis/NIR, EPR) studies along with DFT/TD‐DFT calculations revealed (i) involvement of deprotonated L or HL in the oxidation processes owing to its redox non‐innocent potential and (ii) metal (RuIII/RuII) or bpy/pap dominated reduction processes in 1 + or 2 2+/ 3 +/ 4 2+/ 5 +, respectively.  相似文献

Abstract

Two new ruthenium(II) complexes, [Ru(bpy)2(DClPIP)](ClO4)2 (1) and [Ru(phen)2(DClPIP)](ClO4)2 (2) (bpy = 2,2′-bipyridine, phen = 1,10-phenanthroline, and DClPIP = 2-(2,4-dichlorophenyl)-1H-imidazo[4,5-f][1, 10]phenanthroline), have been prepared in high yield by using microwave-assisted synthesis technology. The anticancer activity of the two ruthenium(II) complexes against A549, C6, CNE-1 and MDA-MB-231 cell lines has been evaluated by MTT assay and results showed that 2 exhibited higher antitumor activity than 1 toward all the selected tumor cell lines. Besides, A549 cell line was sensitive to both ruthenium(II) complexes, especially to 2 (IC50 = 8.01?±?0.36?μM). Meanwhile, 2 showed low toxicity against MCF-10A human normal cells. Furthermore, the DNA-binding properties of the two new ruthenium(II) complexes with CT-DNA have been investigated by electronic absorption titration, luminescence spectra, circular dichroism spectra and viscosity measurements. The results suggested that 1 and 2 were able to interact with CT-DNA via intercalative mode with a strong binding affinity in the order 2?>?1. All of these results suggested that anticancer activity of both ruthenium(II) complexes could be closely related to their interaction with DNA.  相似文献

Abstract

Two mononuclear heteroleptic copper complexes, [Cu(±trans-dach)(bpy)](ClO4)2 1a and [Cu(±trans-dach)(phen)](ClO4)2 2a [dach?=?1,2-diaminocyclohexane, bpy?=?2,2′-bipyridine and phen?=?1,10-phenanthroline], were synthesized and analyzed by CHN analysis, electronic absorption, FT-IR spectroscopy, EPR, and SXRD. The molecular structures of 1a and 2a showed octahedral geometry around Cu(II). Both complexes interacted with phosphoesters and DNA. Their binding affinities with diphenylphosphate, di n-butylphosphate, trimethylphosphate, and triphenylphosphate were studied by UV–vis spectroscopy. For understanding the stereochemical role of dach ligand toward DNA interaction, enantiopure DACH complexes [Cu(R,R-trans-dach(bpy)](ClO4)2 1b, [Cu(S,S-trans-dach)(bpy)](ClO4)2 1c, [Cu(cis-dach)(bpy)](ClO4)2 1d, [Cu(R,R-trans-dach)(phen)](ClO4)2 2b, [Cu(S,S-trans-dach)(phen)](ClO4)2 2c, and [Cu(cis-dach)(phen)](ClO4)2 2d were synthesized and analyzed. All complexes interacted with calf thymus-DNA (CT-DNA) as studied by UV–vis spectroscopy. The nature of binding to CT-DNA was groove/electrostatic as supported by circular dichroism, cyclic voltammetry, and docking studies. Complexes were able to cleave plasmid DNA at 12.5 µM (1ad) and 6 µM (2ad), where 2d showed 64% Form II and 36% Form III. The in vitro cytotoxic studies of two different cancer cell lines showed inhibition with low IC50 value in comparison to reference control (cisplatin). These complexes are efficient in inducing apoptosis in cancer cells, making them viable for potent anticancer activity.  相似文献

A rare example of a mononuclear complex [(bpy)2Ru(L1?H)](ClO4), 1 (ClO4) and dinuclear complexes [(bpy)2Ru(μ‐L1?2H)Ru(bpy)2](ClO4)2, 2 (ClO4)2, [(bpy)2Ru(μ‐L2?2H)Ru(bpy)2](ClO4)2, 3 (ClO4)2, and [(bpy)2Ru(μ‐L3?2H)Ru(bpy)2](ClO4)2, 4 (ClO4)2 (bpy=2,2′‐bipyridine, L1=2,5‐di‐(isopropyl‐amino)‐1,4‐benzoquinone, L2=2,5‐di‐(benzyl‐amino)‐1,4‐benzoquinone, and L3=2,5‐di‐[2,4,6‐(trimethyl)‐anilino]‐1,4‐benzoquinone) with the symmetrically substituted p‐quinone ligands, L, are reported. Bond‐length analysis within the potentially bridging ligands in both the mono‐ and dinuclear complexes shows a localization of bonds, and binding to the metal centers through a phenolate‐type “O?” and an immine/imminium‐type neutral “N” donor. For the mononuclear complex 1 (ClO4), this facilitates strong intermolecular hydrogen bonding and leads to the imminium‐type character of the noncoordinated nitrogen atom. The dinuclear complexes display two oxidation and several reduction steps in acetonitrile solutions. In contrast, the mononuclear complex 1 + exhibits just one oxidation and several reduction steps. The redox processes of 1 1+ are strongly dependent on the solvent. The one‐electron oxidized forms 2 3+, 3 3+, and 4 3+ of the dinuclear complexes exhibit strong absorptions in the NIR region. Weak NIR absorption bands are observed for the one‐electron reduced forms of all complexes. A combination of structural data, electrochemistry, UV/Vis/NIR/EPR spectroelectrochemistry, and DFT calculations is used to elucidate the electronic structures of the complexes. Our DFT results indicate that the electronic natures of the various redox states of the complexes in vacuum differ greatly from those in a solvent continuum. We show here the tuning possibilities that arise upon substituting [O] for the isoelectronic [NR] groups in such quinone ligands.  相似文献

Mononuclear and trinuclear zinc(II) complexes (1 and 2) with tridentate NNO Schiff-base ligands (HL1?=?N-2-pyridiylmethylidene-4-chloro-2-hydroxy-phenylamine, HL2?=?N-2-pyridiylmethylidene-2-hydroxy-5-chloro-phenylamine) have been synthesized and characterized by single-crystal X-ray diffraction and elemental analysis. The binding properties of zinc(II) complexes with calf thymus DNA (CT-DNA) and HSA were investigated by UV–visible, fluorescence, and circular dichroism spectra. The zinc(II) complexes bind significantly to CT-DNA by intercalation and bind to protein HSA through a static quenching mechanism. The in vitro cytotoxicity of the complexes on human tumor cells lines was assessed by 3-(4,5-dimathylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide, Hoechst 33258 staining experiments.  相似文献

An intercalative ligand, ppip (ppip = {2-(4-(piperidin-1-yl)phenyl)-1H-imidazo[4,5-f][1,10]phenanthroline}), and its mononuclear Ru(II) polypyridyl complexes, [Ru(phen)2(ppip)]2+ (1) (phen=1,10-phenanthrolene), [Ru(bpy)2(ppip)]2+ (2) (bpy=2,2′-bipyridine) and [Ru(dmb)2(ppip)]2+ (3) (dmb=4,4′-dimethyl-2,2′-bipyridine), have been synthesized and characterized by elemental analysis and spectroscopic techniques such as UV–vis, IR, 1H, as well as 13C NMR and ESI-MS. The interaction of these complexes with DNA/BSA (bovine serum albumin) was investigated using absorption, emission spectroscopy, viscosity measurements and molecular docking studies. The docking study infers that the binding strength (Kb) of these complexes was in agreement with results from absorption and emission techniques. These studies reveal that these three Ru(II) polypyridyl complexes bind to DNA/BSA. The binding ability of these complexes in the presence of different ions and solvents were also reported. All complexes were effectively cleaving pBR322 DNA in different forms and follows order which is similar to absorption and emission studies. These complexes were effective exhibiting the antimicrobial activity against different microbes Bacillus subtilis, Escherichia coli and Staphylococcus aureus.  相似文献

The binding of the ruthenium(II) complexes [Ru(bpy)2(ITAP)](ClO4)2 (bpy = 2,2’-bipyridine) and [Ru(phen)2(ITAP)](ClO4)2 (phen = 1,10-phenanthroline, ITAP = isatino[1,2-b]-1,4,8,9-tetraazatriphenylene) to calf thymus DNA (CT-DNA) have been investigated with UV–visible and emission spectroscopy, viscosity measurements, thermal denaturation, and photoactivated cleavage. The experimental results indicate that the two complexes bind to CT-DNA through an intercalative mode. The two Ru(II) complexes in the presence of plasmid pBR322 DNA have been found to give rise to nicking of DNA upon irradiation.  相似文献

Preparation of the ligands HL1 = 2,6-[(N-phenylpiperazin-1-yl)methyl]-p-ethylphenol; HL2 = 2,6-[(N-phenylpiperazin-1-yl)methyl]-p-methoxyphenol and HL3 = 2,6-[(N-phenylpiperazin-1-yl)methyl]-p-nitrophenol are described together with their Cu(II) complexes with different bridging units. The exogenous bridges incorporated into the complexes are: hydroxo [Cu2L(OH)(H2O)2](ClO4)2.H2O (L1=1a, L2 =1b, L3 =1c), acetato [Cu2L(OAc)2]ClO4.H2O (L1 =2a, L2 =2b, L3 =2c) and nitrito [Cu2L1(NO2)2(H2O)2]ClO4.H2O (L1=3a, L2 =3b, L3 =3c). Complexes1a,1b,1c and2a,2b,2c contain bridging exogenous groups, while3a,3b,3c possess only open μ-phenolate structures. Both the ligands and complexes were characterized by spectral studies. Cyclic voltammetric investigation of these complexes revealed that the reaction process involves two successive quasireversible one-electron steps at different potentials. The first reduction potential is sensitive to electronic effects of the substituents at the aromatic ring of the ligand system, shifting to positive potentials when the substituents are replaced by more electrophilic groups. EPR studies indicate very weak interaction between the two copper atoms. Various covalency parameters have been calculated.  相似文献

The new compounds [(acac)2Ru(μ‐boptz)Ru(acac)2] ( 1 ), [(bpy)2Ru(μ‐boptz)Ru(bpy)2](ClO4)2 ( 2 ‐(ClO4)2), and [(pap)2Ru(μ‐boptz)Ru(pap)2](ClO4)2 ( 3 ‐(ClO4)2) were obtained from 3,6‐bis(2‐hydroxyphenyl)‐1,2,4,5‐tetrazine (H2boptz), the crystal structure analysis of which is reported. Compound 1 contains two antiferromagnetically coupled (J=?36.7 cm?1) RuIII centers. We have investigated the role of both the donor and acceptor functions containing the boptz2? bridging ligand in combination with the electronically different ancillary ligands (donating acac?, moderately π‐accepting bpy, and strongly π‐accepting pap; acac=acetylacetonate, bpy=2,2′‐bipyridine pap=2‐phenylazopyridine) by using cyclic voltammetry, spectroelectrochemistry and electron paramagnetic resonance (EPR) spectroscopy for several in situ accessible redox states. We found that metal–ligand–metal oxidation state combinations remain invariant to ancillary ligand change in some instances; however, three isoelectronic paramagnetic cores Ru(μ‐boptz)Ru showed remarkable differences. The excellent tolerance of the bpy co ‐ ligand for both RuIII and RuII is demonstrated by the adoption of the mixed ‐ valent form in [L2Ru(μ‐boptz)RuL2]3+, L=bpy, whereas the corresponding system with pap stabilizes the RuII states to yield a phenoxyl radical ligand and the compound with L=acac? contains two RuIII centers connected by a tetrazine radical‐anion bridge.  相似文献

The interaction between the complexes and calf thymus DNA (CT-DNA) has been investigated by spectroscopic methods and viscosity measurements. The results indicate that the complexes can bind to CT-DNA in an intercalative mode. In addition, both complexes promote the photocleavage of plasmid pBR322 DNA under irradiation. The mechanistic studies reveal that singlet oxygen 1O2 plays a significant role in DNA photocleavage.  相似文献

Abstract

Dinuclear ruthenium complexes [Ru2(bpy)4BL](ClO4)2 (Ru-1), where bpy = 2,2′-bipyridine and BL = 2,2′-((1E,1′E)-((E)-diazene-1,2-diyl-bis(2,1-phenylene))-bis(azanylylidene))bis(methanylylidene))diphenol (a bidentate bridging ligand), and mononuclear ruthenium complexes [Ru(bpy)2L](ClO4) (Ru-2), where L = (E)-2-((phenylimino)methyl)phenol, were synthesized and characterized by elemental analysis and electrospray ionization mass spectrometry. Their photophysical and electrochemical properties were also studied. The cytotoxicity of the two complexes in vitro was evaluated using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. The results indicated that Ru-1 and Ru-2 exhibited significant dose-dependent cytotoxicity to human breast cancer (MCF-7), gastric cancer (SGC-7901), cervical cancer (Hela), and lung cancer (A549) tumor cell lines. Ru-1 showed excellent antitumor effects in a cellular study (IC50 values of 3.61 μM for MCF-7 human breast cancer cells in vitro). However, Ru-2 exhibited the highest cytotoxicity to Hela cells; the IC50 value is 3.71 μM. The results reveal that Ru-1 and Ru-2 have obvious selectivity and might be a potential anticancer agent that could improve the efficacy of common anticancer therapies.  相似文献

Three new Cu(II) complexes, [Cu(HL1)(pyridine)(H2O)](ClO4)2·2MeOH (1), [Cu2(HL1)2(NO3)2](NO3)2·3H2O (2) and [Cu(HL2)(NO3)2]·MeCN (3), have been synthesized from two Schiff base ligands [HL1 = 1-phenyl-3-((2-(piperazin-4-yl)ethyl)imino)but-1-en-1-ol and HL2 = 4-((2-(piperazin-1-yl)ethyl)imino)pent-2-en-2-ol] using the chair conformer of a flexible piperazinyl moiety. Structural analysis reveals that 1 and 3 are monomeric Cu(II) complexes consisting of five- and six-coordinate Cu(II), respectively, whereas 2 is a dinuclear Cu(II) complex consisting of two different Cu(II) centers, one square planar with the other distorted octahedral. Screening tests were conducted to quantify the binding of 13 towards DNA and BSA as well as the DNA cleavage activity of these complexes using gel electrophoresis. Enzyme kinetic studies were also performed for the complexes mimicking catecholase-like activities. Antibacterial activities of these complexes were also examined towards Methicillin-Resistant Staphylococcus aureus bacteria. The results reflect that 2 is more active than the monomeric complexes, which is further corroborated by density functional theory study.  相似文献

Two ruthenium(III) complexes containing ethylenediaminetetraacetate(edta), viz. [{Ru(Hedta)}2L]·xH2O L = 4,4′-bipyridine(bpy) (1) and 4,4′-azopyridine(Azpy) (2), have been synthesized by the reaction between K[Ru(Hedta)Cl]·1.5H2O and the corresponding N-heterocycles. Complex 1 was determined by single-crystal X-ray diffraction. The products were characterized by IR, UV–vis, cyclic voltammetry, and magnetic techniques. Their DNA-binding activities were investigated using electronic absorption spectroscopic methods and ?uorescence quenching; the experimental results show that these two ruthenium complexes may bind to CT-DNA through intercalation modes.  相似文献
