On reaction of different copper(II) salts with 3,4-bis(2-pyridylmethylthio)toluene (L) having neutral tetradentate NSSN donor set in different chemical environments, two mononuclear copper(II), one dinuclear copper(I) and one dinuclear copper(II) complexes, formulated as [CuII(L)(H2O)2](NO3)2 (1), [CuII(pic)2] (2), [CuI2(L)2](ClO4)2 (3) and [CuII2(L)2Cl2](ClO4)2 (4), respectively, were isolated in pure form [where pic = picolinate]. All the complexes were characterized by physicochemical and spectroscopic methods. The product of the reactions are dependent on the counter anion of copper(II) salts used as reactant and on the reaction medium. Complexes 1 and 4 were obtained with nitrate and perchlorate copper(II) salts, respectively. On the other hand, C–S bond cleavage was observed in the reaction of L with copper(II) chloride to form in situ picolinic acid and complex 2. Dinuclear complexes 3 and 4 were separated out when copper(II) perchlorate was allowed to react with L in methanol and in acetonitrile, respectively, under aerobic condition. The X-ray diffraction analysis of the dinuclear complex 3 shows a highly distorted tetrahedral geometry about each copper ion. Complex 4 is converted to 3 in acetonitrile in presence of catechol. The spectral study of complex 4 with calf thymus DNA is indicative of a groove binding mode interaction.  相似文献

Schiff base mixed-ligand copper complexes [CuL1(phen)Cl2], [CuL1(bipy)Cl2], [Cu(L1)2Cl2], [Cu(L2)2Cl2], [CuL2(bipy)Cl2], and [CuL2(phen)Cl2] (where L1?=?4-[3,4-dimethoxy-benzylidene]-1,5-dimethyl-2-phenyl-1,2-dihydro-pyrazole-3-one; L2?=?4-[3-hydroxy-4-nitro-benzylidene]-1,5-dimethyl-2-phenyl-1,2-dihydro-pyrazole-3-one; phen?=?1,10-phenanthroline; and bipy?=?2,2′-bipyridine) have been synthesized and characterized. Their DNA-binding properties have been studied by electronic absorption spectra, viscosity, and electrochemical measurements. The absorption spectral and viscosity results suggest that the copper(II) complexes bind to DNA via partial intercalation. The addition of DNA resulting in the decrease of the peak current of the copper(II) complexes indicates their interaction. Interaction between the complexes and DNA has also been investigated by submarine gel electrophoresis. The copper complexes cleave supercoiled pUC19 DNA to nicked and linear forms through hydroxyl radical and singlet oxygen in the presence of 3-mercaptopropionic acid as the reducing agent. These copper complexes promote the photocleavage of pUC19 DNA under irradiation at 360?nm. Mechanistic study reveals that singlet oxygen is likely to be the reactive species responsible for the cleavage of plasmid DNA by the synthesized complexes. The in vitro antimicrobial study indicates that the metal chelates have higher activity against the bacterial and fungal strains than the free ligands.  相似文献

A mononuclear Cu(II) complex, [Cu(FA)2(NO3)2], in which FA is ferulic acid ((E)-3-(4-hydroxy-3-methoxy-phenyl)prop-2-enoic acid), was synthesized and characterized by spectroscopic methods. The main structures of the ligand and its complexes with Cu2+ were optimized by QM calculations. The calculations on the structures of the [Cu(FA)2(NO3)2] complexes forms and the intercalating with DNA profile were undertaken by UHF/PM6 and MMFF94 methods, respectively. In vitro studies (UV-vis spectroscopy, emission titration, circular dichroism techniques, and viscometry) under physiological conditions (Tris-HCl buffer solutions, pH 7.4) showed that the complex interacts with calf-thymus DNA (ct-DNA) via an intercalative binding mode. The thermodynamic parameters, enthalpy change (ΔH), and entropy change (ΔS) showed that the acting forces between Cu(II) complex and ct-DNA mainly included van der Waals interactions and hydrogen bonds. Methylene blue (MB) displacement studies revealed that Cu(II) complex can substitute MB probe in the MB-DNA complex which was indicative of intercalative binding. The theoretical data confirm the experimental results with respect to the mechanism of binding.  相似文献

A Schiff base bis(N-salicylidene)-3-oxapentane-1,5-diamine (H2L) and its Cu(II) complex, [Cu2(L)2]?CHCl3, have been synthesized and characterized by physicochemical and spectroscopic methods. Single-crystal X-ray analysis revealed that the complex is a centrosymmetric binuclear neutral entity, in which Cu(II) is a five-coordinate in a distorted trigonal bipyramidal geometry. The DNA-binding properties of the free ligand and the complex have been investigated by electronic absorption, fluorescence, and viscosity measurements. The results suggest that the H2L and the complex to DNA via the intercalation mode and the binding affinity of the complex were higher than that of the H2L. The intrinsic binding constants Kb of the ligand and the complex are 2.2 × 104 and 2.7 × 104 M?1, respectively. Antioxidant assay in vitro shows the Cu(II) complex possesses significant antioxidant activities and better scavenging activity than the H2L and other antioxidants.  相似文献

A new water-soluble copper(II) complex, Cu(TACNA)Br?·?0.375H2O (1) [TACNA?=?1,4,7-triazacyclononane-N-acetate], has been synthesized to serve as artificial nucleases. The X-ray crystal structure of 1 indicates that one bromide and an oxygen from acetate pendant coordinate to copper(II) in addition to the nitrogen atoms in the TACN macrocycle, resulting in a five-coordinate complex with square-pyramidal geometry. The interaction of 1 with calf thymus DNA (ct-DNA) has been investigated by UV absorption and fluorescence spectroscopies, and the mode of ct-DNA binding for 1 has been proposed. In the absence of external agents, supercoiled plasmid DNA cleavage by 1 was performed under aerobic condition; the influences on DNA cleavage of different complex concentrations and reaction times were also studied. The cleavage of plasmid DNA likely involves oxidative mechanism.  相似文献

Hydrolysis of DNA is an important enzymatic reaction , but it is exceedingly difficult to mimic in the laboratory because of the stability of hydrolysis of DNA. In this paper, the cleavage activity of complexes formed between Cu(Ⅱ) and four different amino acid or amino acid methyl ester on DNA is studied by gel elec-trophoresis. It is found that DNA could be cleaved by Cu(Ⅱ)-L-His and Cu(Ⅱ)-L-His methyl ester complexes and the efficiency of cleavage is largely dependent on the metal ion-to-ligand ratio. Further experiments show that the cleavage of DNA mediated by Cu(Ⅱ)-L-His complexes occurs via a hydrolytic mechanism and the active chemical species that affects DNA cleavage is proposed to be MI2H and ML2H22 .  相似文献

By reacting thiosemicarbazides substituted on the aminic nitrogen with 5-formyluracil, several new 5-formyluracil thiosemicarbazones (H3ut) derivatives were synthesised and characterized. These ligands, treated with copper chloride and nitrate, afforded two different kinds of compounds. In the complexes derived from copper chloride the metal atom is pentacoordinated, being surrounded by the neutral ligand binding through SNO donor atoms and by two chlorines, while the nitrate derivatives consist of monocations and nitrate anions. The copper coordination (4 + 2) involves the SNO ligand atoms, two water oxygens and an oxygen atom of a monodentate nitrate group. On varying the substituents on the thiosemicarbazidic moiety, remarkable modifications of the coordination geometry are not observed for the complexes with the same counterion. For all the compounds, interactions with DNA (calf thymus) were studied using UV–Vis spectroscopy; the nuclease activity was verified on plasmid DNA pBR 322 by electrophoresis.  相似文献

One nonlinear and one linear trinuclear copper(II) complex [Cu3(dien)2(pdc)2CH3OH]2?·?6CH3OH (1) and [Cu3(pdc)2(CH3OH)6(H2O)4] (2) were prepared and characterized structurally, where dien is diethylenetriamine and pdc3? the trianion of 3,5-pyrazoledicarboxylic acid. Both complexes consist of 3,5-pyrazoledicarboxylato-bridged trinuclear copper(II) centers. In 1, copper(II) ions are five-coordinate in distorted square pyramids with bond angles 164.78° for Cu(1)–Cu(2)–Cu(3) and 164.51° for Cu(4)–Cu(5)–Cu(6). In 2, the three copper(II) ions are six-coordinate with elongated octahedral geometry. The trinuclear units of 1 and 2 interact through hydrogen bonds to form 3-D and 2-D supramolecular networks, respectively. Variable temperature magnetic susceptibility measurements show that 1 and 2 are antiferromagnetically coupled with J values of ?11.2 and ?13.3?cm?1.  相似文献

The reaction of Cu(ClO4)2·6H2O and Cd(ClO4)2 with di-(2-picolyl)sulfur (dps) leads to the formation of mononuclear complexes [Cu(dps)(H2O)(ClO4)](ClO4) (1) and [Cd(dps)2](ClO4)2 (2). The crystal structure of 1 exhibits a distorted square pyramidal geometry, coordinated by one sulfur and two nitrogen atoms from the dps ligand, one water molecule and one perchlorate oxygen atom. For 2, the environment around cadmium atom is in a distorted octahedron with four nitrogen and two sulfur atoms from the dps ligand. Cyclic voltammetric data show that complexes undergo two waves of a one-electron transfer corresponding to M(II)/M(III) and M(II)/M(I) processes. Spectral and electrochemical behaviors of the complexes are also discussed.  相似文献

An asymmetric bidentate Schiff-base ligand (2-hydroxybenzyl-2-furylmethyl)imine (L–OH) was prepared. Three complexes derived from L–OH were synthesized by treating an ethanolic solution of the appropriate ligand with an equimolar amount of metallic salt. Three complexes, Cu2(L–O?)2Cl2 (1), Ni(L–O?)2 (2) and Co(L–O?)3 (3), have been structurally characterized through elemental analysis, IR, UV spectra and thermogravimetric analysis. Single crystal X-ray diffraction shows metal ions and ligands reacted with different proportions 1?:?1, 1?:?2 and 1?:?3, respectively, so copper(II), nickel(II), and cobalt(III) have different geometries.  相似文献

Complexes CuL3Cl2, PdL2Cl2 and PtL2Cl2, where L is a novel ligand from the series of 2-substituted 5-aminotetrazoles, namely 5-amino-2-tert-butyltetrazole (1), have been synthesized by the reaction of metal(II) chlorides with 1 and characterized by IR spectroscopy, thermal and X-ray analyses. The crystallographic structural analysis of these complexes revealed that 1 acts as a monodentate ligand coordinated to the metal via endocyclic N4 atom. Platinum complex demonstrates promising cytotoxicity against human cervical carcinoma cells with IC50 value average between those of cisplatin and carboplatin.  相似文献

A new complex, [Cu(valcyte)2(NO3)2], in which valcyte served as a valganciclovir drug, was synthesized and characterized by different physicochemical methods. Optimization of ligand structures and their complexes with Cu2+ were performed by semi-empirical and DFT methods. Binding interaction of this complex with calf-thymus DNA (ct-DNA) was explored by emission, absorption, circular dichroism and viscosity techniques. Additionally, cell line targeting was studied and cytotoxic effects of [Cu(valcyte)2(NO3)2] (0.0–160 μg) on AGS and MCF-7 cell lines were reported. Percentage of Cell Viability and Apoptotic Index were assessed. The complex displayed significant binding properties to ct-DNA. Undertaking fluorometric studies, the binding mode of the complex with ct-DNA was explored utilizing Hoechst as a fluorescence probe, indicating the binding to be of groove mode. The DNA viscosity altered slightly in presence of the complex. Enthalpy and entropy changes during the interaction showed that the process is endothermic, with the complex mainly bound to ct-DNA by hydrophobic attraction. Values of ΔG revealed a spontaneous reaction between DNA and the complex. Optimized docked model of DNA–complex mixture confirmed the experimental results. The results of MMFF94 calculations indicated stability of [Cu(valcyte)2(NO3)2] after docking with the modeled DNA profile, as compared to the DNA profile and valganciclovir results before the docking process. Cytotoxicity studies showed that an increase in [Cu(valcyte)2(NO3)2] may result in a significant decrease in cell viability and increase apoptosis index in the treated cells, as compared to valganciclovir treated cells (p < 0.05). The findings further showed that [Cu(valcyte)2(NO3)2] has potential for use in cancer therapy.  相似文献

Four mononuclear copper(II) complexes of two new carboxamide derivatives formulated as [Cu(L1)2](ClO4)2 (1a), [Cu(L1)2](NO3)2 (1b), [Cu(L2)2(H2O)2](ClO4)2 (2a), and [Cu(L2)2(H2O)](NO3)2 (2b) have been isolated in pure form from the reaction of L1 and L2 [where L1 = N-(furan-2-ylmethyl)-2-pyridinecarboxamide and L2 = N-(thiophen-2-ylmethyl)-2-pyridinecarboxamide] with copper(II) salts of perchlorate and nitrate. All the complexes were characterized by physicochemical and spectroscopic tools along with single-crystal X-ray diffraction studies. The structural analyses showed that 1 is monomeric of square planar geometry with copper(II) chelated by two L1 ligands. Complex 2 differs in coordination geometry, being octahedral and distorted square pyramidal. Two L2 ligands occupy the equatorial positions of the octahedral 2a and the basal sites of the pyramidal 2b, with water molecules that complete the coordination sphere in each case. Electrochemical studies using cyclic voltammetry showed a reversible redox behavior of the copper(II) in 1 and 2. The electronic spectroscopic behavior and the trend of one electron equivalent redox potential corresponding to a CuII/CuI couple have also been confirmed by density functional theory calculations. The spectroscopic and viscosity measurement study in tris–HCl buffer suggested an intercalative interaction of 1a and 2 with calf thymus DNA likely due to the stacking between the non-coordinated furan and thiophene chromophore with the base pairs of DNA.  相似文献

Two new copper(II) complexes with aminothioether ligands, [Cu(L1)(ClO4)](ClO4) · 0.5H2O (1) and [Cu(L2)(H2O)](ClO4)2 · H2O (2) (L1 = 2-benzyl-1,3-bis(aminoethylthio)propane and L2 = 2-(4-butylbenzyl)-1,3-bis(aminoethylthio)propane), have been synthesized and characterized. The single crystal X-ray diffraction analysis reveals that both 1 and 2 adopt distorted square pyramidal geometries. The binding modes of both complexes with calf thymus DNA were investigated by UV–Vis and CD spectroscopies. The results show that both complexes mainly adopt an electrostatic attraction binding mode with DNA and the binding constants are (1.62 ± 0.02) × 103 and (2.02 ± 0.02) × 103 M−1, respectively. Both complexes are able to cleave pBR322 plasmid DNA efficiently in the presence of ascorbic acid and the activity of 2 is higher than that of 1. The DNA cleavage by 1 and 2 were inhibited strongly in the presence of DMSO and tert-butyl alcohol, which suggests that hydroxyl radicals are the reactive oxygen species for the cleavage.  相似文献

Two new usymmetric bidentate Schiff-base ligands (2-pyridyl-2-furylmethyl)imine (L1) and (2-pyridyl-phenylmethyl)imine (L2) were prepared. The crystal structures of two chloro-bridged complexes [Cu2(μ-Cl)2(L1)2Cl2] (1) and [Mn (μ-Cl)2(L2)] (2) derived from the each ligand have been confirmed by single-crystal X-ray diffraction analysis. The complexes were characterized by IR, elemental analysis and spectroscopic methods. In complex 1, the two copper atoms are five-coordinate involving a square-pyramidal geometry having a N2Cl3 donor set with the two chlorine atoms bridging the two copper atoms. In complex 2, the manganese atoms are both six-coordinate. In contrast to 1, all chlorine atoms in 2 are bridging chlorides and link adjacent manganese atoms together forming 1-D infinite chains.  相似文献

Two new potentially octadentate N2O6 Schiff-base ligands 2-((E)-(2-(2-(2-((E)-2-hydroxy-3-methoxybenzylideneamino)phenoxy)phenoxy)phenylimino)methyl)-6-methoxyphenol H2L1 and 2-((E)-(2-(2-(2-((E)-2-hydroxy-3-methoxybenzylideneamino)phenoxy)-4-tert-butylphenoxy)phenylimino)methyl)-6-methoxyphenol H2L2 were prepared from the reaction of O-Vaniline with 1,2-bis(2′-aminophenoxy)benzene or 1,2-bis(2′-aminophenoxy)-4-t-butylbenzene, respectively. Reactions of H2L1 and H2L2 with copper(II) and zinc(II) salts in methanol in the presence of N(Et)3 gave neutral [CuL1]?·?0.5CH2Cl2, [CuL2], [ZnL1]?·?0.5CH2Cl2, and [ZnL2] complexes. The complexes were characterized by IR spectra, elemental analysis, magnetic susceptibility, ESI–MS spectra, molar conductance (Λm), UV-Vis spectra and, in the case of [ZnL1]?·?0.5CH2Cl2 and [ZnL2], with 1H- and 13C-NMR. The crystal structure of [ZnL1]?·?0.5CH2Cl2 has also been determined showing the metal ion in a highly distorted trigonal bipyramidal geometry. The electrochemical behavior of H2L2 and its Cu(II) complex, [CuL2], was studied and the formation constant of [CuL2] was evaluated using cyclic voltammetry. The logarithm value of formation constant of [CuL2] is 21.9.  相似文献

Four new substituted amino acid ligands, N-(3-hydroxybenzyl)-glycine acid (HL1), N-(3-hydroxybenzyl)-alanine acid (HL2), N-(3-hydroxybenzyl)-phenylalanine acid (HL3), and N-(3-hydroxybenzyl)-leucine acid (HL4), were synthesized and characterized on the basis of 1H NMR, IR, ESI-MS, and elemental analyses. The crystal structures of their copper(II) complexes [Cu(L1)2]·2H2O (1), [Cu(L2)2(H2O)] (2), [Cu(L3)2(CH3OH)] (3), and [Cu(L4)2(H2O)]·H2O (4) were determined by X-ray diffraction analysis. The ligands coordinate with copper(II) through secondary amine and carboxylate in all complexes. In 2, 3, and 4, additional water or methanol coordinates, completing a distorted tetragonal pyramidal coordination geometry around copper. Fluorescence titration spectra, electronic absorption titration spectra, and EB displacement indicate that all the complexes bind to CT-DNA. Intrinsic binding constants of the copper(II) complexes with CT-DNA are 1.32?×?106?M?1, 4.32?×?105?M?1, 5.00?×?105?M?1, and 5.70?×?104?M?1 for 1, 2, 3, and 4, respectively. Antioxidant activities of the compounds have been investigated by spectrophotometric measurements. The results show that the Cu(II) complexes have similar superoxide dismutase activity to that of native Cu, Zn-SOD.  相似文献
