Sixteen caerulein-type peptides have been isolated from the skin secretions of the Australian Blue Mountains tree frog Litoria citropa. There are four groups of these peptides. The first is based on the structure of the known neuropeptide caerulein [pEQDY(SO(3))TGWMDF-NH(2)], now renamed caerulein 1.1. Examples of peptides of the other groups are as follows: caerulein 2.1 [pEQDY(SO(3))TGAHMDF-NH(2)], caerulein 3.1 [pEQDY(SO(3))GTGWMDF-NH(2)] and caerulein 4.1 [pEQDY(SO(3))TGSHMDF-NH(2)]. All of these peptides are accompanied by the associated peptide where Phe replaces Met, and all eight of the caerulein peptides are accompanied by the desulfated analogues. Negative ion electrospray mass spectrometry (ES-MS) is used to determine the molecular weights of the caeruleins 1-4 [from their [M - H](-) ions], while the sequences of the peptides are determined from the B and Y + 2 cleavage ions in the mass spectra of the [MH(+) - SO(3)](+) ions. 相似文献

The secretion from the dorsal glands of the frog Litoria rothii contains a series of new peptides including rothein 1 (SVSNIPESIGF-OH, a neuropeptide which contracts smooth muscle), a number of inactive rothein 2 and 3 peptides (e.g. rothein 2.1, AGGLDDLLEPVLNSADNLVHGL-OH), and a new proline rich peptide, named rothein 4.1 (AEILFGDVRPPWMPPPIFPEMP-OH), which shows neither antimicrobial nor neuronal nitric oxide synthase (nNOS) activity. Two known neuropeptides of the caerulein family [e.g. caerulein, pEQDY(SO3)TGWMDF-NH2] together with a series of known caerin 1 antibiotic and nNOS-inhibiting peptides (e.g. caerin 1.1, GLLSVLGSVAKHVLPHVVPVIAEHL-NH2) were also identified. Positive ion electrospray mass spectrometry (ES-MS) was used as the primary method to investigate the sequences of the new peptides. Negative ion ES-MS was used to fill in any gaps in the positive ion data and, finally, Edman automated sequencing was used to differentiate between Leu and Ile and to confirm the sequences determined by mass spectrometry. 相似文献

Eleven dahlein peptides are present in the skin secretion of the Australian aquatic frog Litoria dahlii. All peptides have been sequenced using a combination of electrospray mass spectrometry (ES-MS) and Lys-C digestion/MS, with each sequence confirmed by automated Edman sequencing. The 13-residue dahlein 1 peptides (e.g. dahlein 1.1 GLFDIIKNIVSTL-NH(2)) exhibit weak wide-spectrum antimicrobial activity but no significant activity in the anticancer testing program of the National Cancer Institute (Washington). There are no potent antimicrobial peptides present in the glandular secretion, but the dahleins 5 strongly inhibit the formation of NO by neuronal nitric oxide synthase (e.g. dahlein 5.1 GLLGSIGNAIGAFIANKLKP-OH). 相似文献

Sixteen aurein peptides are present in the host defence secretion from the granular dorsal glands of the Green and Golden Bell Frog Litoria aureus and seventeen from those of the related Southern Bell Frog Litoria raniformis. All peptides have been sequenced using a combination of electrospray mass spectrometry and Lys-C digestion, with each sequence confirmed by automated Edman sequencing. The peptides are named in five groups, viz. aureins 1-5. Ten of these peptides are common to both species of frog. Thirteen of the aurein peptides show wide-spectrum antibiotic and anticancer activity. Amongst the more active peptides are aurein 1.2 (GLFDIIKKIAESF-NH2), the smallest peptide from an anuran reported to have both antibiotic and anticancer activity; aurein 2.2 (GLLDIVKKVIGAFGSL-NH2) and aurein 3.1 (GLFDIVKKIAGHIAGSI-NH2). The aurein 4 and 5 peptides, e.g. aurein 4.1 (GLIQTIKEKLKELAGGLVTGIQS-OH) and aurein 5.1 (GLLDIVTGLLGNLIVDVLKPKTPAS-OH), show neither antibacterial nor anticancer activity. 相似文献

Many species of frogs of the genus Litoria secrete bioactive peptides from their skin glands. These peptides are normally host‐defence compounds and may have one, or more of the following activities; smooth muscle contraction, analgesic, antimicrobial, antiviral, lymphocyte proliferator (immunomodulator) and neuronal nitric oxide synthase (nNOS) inactivation. Two frog species of the Litoria rubella Group that have been studied before, namely, Litoria electrica and Litoria rubella, are different from other species of the genus Litoria in that they produce small peptides that show neither membrane, lymphocyte nor nNOS activity. In this study we have used electrospray mass spectrometry together with Edman sequencing to identify eight skin peptides of the third member of this Group, Litoria dentata: surprisingly, none of these peptides show activity in our biological screening program. However, two major peptides (FPWL‐NH2 and FPWP‐NH2) from L. electrica and L. rubella are opioids at the micromolar concentration. Copyright © 2009 John Wiley & Sons, Ltd. 相似文献

The glandular skin secretion of the Eastern Dwarf Tree Frog Litoria fallax contains nine peptides named fallaxidins. The sequences of these peptides were elucidated using a combination of positive and negative electrospray mass spectrometry together with Edman sequencing. Among these peptides are: (i) fallaxidins 1.1 and 2.1 which have the sequences YFPIPI-NH2 and FWPFM-NH2. The activities of these peptides are unknown, but it has been shown that they are not smooth muscle active, opioids or antimicrobially active, nor do they effect proliferation of lymphocytes; (ii) two weakly active antibiotics, fallaxidins 3.1 and 3.2 (e.g. fallaxidin 3.1, GLLDLAKHVIGIASKL-NH2), and a moderately active antibiotic fallaxidin 4.1 (GLLSFLPKVIGVIGHLIHPPS-OH). Fallaxidin 4.1 has an unusual sequence for an antibiotic, containing three Pro residues together with a C-terminal CO2H group. cDNA cloning has confirmed the identity of the nine isolated peptides from L. fallax, together with five additional peptides not detected in the peptide profile. The pre-regions of the nine preprofallaxidins are conserved and similar to those of the caerin peptides from L. caerulea and L. splendida, suggesting that the fallaxidin and caerin peptides, although significantly different in sequence, originated from a common ancestor gene. 相似文献

Collision-induced fragmentations of deprotonated maculatin 1 peptides provide significant sequencing information. When the peptide lacks those residues which can fragment through their alpha side chains (e.g. Thr, Ser, Glu and Gln in this study) the basic alpha and beta' backbone cleavages occur from the [Mbond;H](-) anion. When Thr, Ser, Glu and Gln are present, the ease of side-chain fragmentation of these residues is: Thr (loss of MeCHO) > Ser (CH(2)O) > Glu (H(2)O) > Gln (NH(3)). When one of more of these residues is (are) present, the alpha and beta' cleavages often occur from a fragment rather than the [Mbond;H](-) anion, e.g. for Thr, the [(Mbond;H)(-)bond;MeCHO](-) anion. These four residues also initiate gamma backbone cleavage reactions. The relative abundances of peaks resulting from gamma cleavage are Glu > Ser = Thr > Gln for maculatin 1 spectra. An unusual Gln19/Ile17 cyclisation/cleavage reaction occurs in maculatin spectra: the peptide [Mbond;H](-) anion must adopt a helical conformation in order for these two groups to interact. Analogous fragmentations have been reported previously in the negative ion spectra of the caerin 1 peptides. 相似文献

Positive and negative ion electrospray mass spectrometry together with Edman sequencing (when appropriate) has been used to sequence the host‐defence peptides secreted from skin glands of the tree frog Litoria peronii. The peptide profiles are different in winter and summer. In winter, the frog produces small amounts of the known caerin 1.1 [GLLSVLGSVAKHVLPHVVPVIAEHL‐NH2] (a wide‐spectrum antibiotic) and caerin 2.1 [GLVSSIGRALGGLLADVVKSKQPA‐OH], a narrow‐spectrum antibiotic and an inhibitor of neuronal nitric oxide synthase. The major peptides produced throughout the year are the pGlu‐containing peroniins 1.1 to 1.5 (e.g. peroniin 1.1 [pEPWLPFG‐NH2], a smooth muscle contractor from 10?7 M), and caerulein [pEQDY(SO3H)TGWMDF‐NH2], a known and potent smooth muscle contractor from 10?10 M. There are also some precursors to the peroniin 1 peptides, only detected in the skin secretion in summer, which are inactive and appear to be all (or part) of the spacer peroniin 1 peptides, e.g. peroniin 1.1b [SEEEKRQPWLPFG‐NH2]. There are three members of the Litoria peronii Group of tree frogs classified in Australia, namely, L. peronii, L. rothii and L.tyleri. A comparison of the skin peptide profiles of L. peronii with those reported previously for L. rothii suggests that either these two species of tree frog are not as closely related as determined previously on morphological grounds, or that skin peptide divergence in tree frogs of this Group is more extensive than in others that have been studied. The combination of electrospray ionization (ESI) and quadrupole time-of-flight (Q-Tof) mass spectrometry presents a powerful tool to verify/determine the sequence of oligonucleotides. An ESI-Q-Tof instrument provides better sensitivity and much higher resolution compared with either ESI-triple quadrupole or ESI-ion trap devices. With high-resolution capability, the quadrupole time-of-flight instrument can provide an isotope pattern to support the charge state assignment. This will improve the reliability of the assignments of sequence-related w or a-Base series ions and lead to accurate determination of the oligonucleotide sequence. 相似文献

A combination of positive and negative ion electrospray mass spectrometry (ES-MS) together with automated Edman sequencing has been used to determine the amino acid sequences of the host-defence peptides from the skin glands of the froglet Crinia riparia. The peptides are called riparins. Of the eight peptides isolated, five are neuropeptides containing intramolecular disulfide linkages; e.g. the major peptide riparin 1.4 (FFLPPCAYKGTC-OH). Positive ion ES-MS identifies the five residues of riparin 1.4 outside the disulfide moiety, but provides no information on the sequence within the disulfide ring. In contrast, the negative ion dissociations of the [M-H]- ion of riparin 1.4 identify the --S-S-- link by loss of H2S2 from the [M-H]- ion, and also provide the sequence within the disulfide unit. Other peptides are riparin 2.1 [(IIEKLVNTALGLLSGL-NH2), a narrow-spectrum antibiotic], signiferin 3.1 [(GIAEFLNYIKSKA-NH2), an nNOS inhibitor] and riparin 5.1 [IVSYPDDAGEHAHKMG-NH2], which shows no neuropeptide, antibiotic or nNOS activity. 相似文献

Negative-ion electrospray tandem mass spectrometry (ES-MS/MS) with collision-induced dissociation (CID) is attempted for sequence determination of alginate oligosaccharides, derived from polyanionic alginic acid, polymannuronate, and polyguluronate by partial depolymerization using either alginate lyase or mild acid hydrolysis. Sixteen homo- and hetero-oligomeric fragments were obtained after fractionation by gel-filtration and strong anion exchange high performance liquid chromatography. The product-ion spectra of these alginate oligosaccharides were dominated by intense B-, C-, Y-, and Z-type ions together with (0,2)A- and (2,5)A-ions of lower intensities. Internal mannuronate residues (M) produce weak but specific decarboxylated Z(int)-ions (Z(int) - 44 Da; int: denotes internal), which can be used for distinction of M and a guluronate residue (G) at an internal position. A reducing terminal M or G, although neither gives rise to a specific ion, can be identified by differences in the intensity ratio of fragment ions of the reducing terminal residue [(2,5)A(red)]/[(0,4)A(red)] (red: denotes reducing terminal). 相似文献

Alkaline metal adduct ions of sphingomyelin were formed by electrospray ionization in positive ion mode. Under low energy collisionally activated dissociation (CAD), the product ion spectra yield abundant fragment ions representative of both long chain base and fatty acid which permit unequivocal determination of the structure. Tandem spectra obtained by constant neutral loss scanning permit identification of sphingomyelin class and specific long chain base subclass in the mixture. The fragmentation pathways under CAD were proposed, and were further confirmed by source CAD tandem mass spectrometry. The total analysis of sphingomyelin mixtures from bovine brain, bovine erythrocytes, and chicken egg yolk is also presented. 相似文献

Immobilized metal ion affinity chromatography (IMAC) has been widely used for the enrichment of phosphopeptides, whereas no report exists describing the use of IMAC columns for the enrichment of sulfopeptides. In this study, we used IMAC-Ga microcolumns for the enrichment of sulfopeptides from a complex mixture of peptides, extracted from skin secretions of the Pachymedusa dacnicolor frog. The enriched fraction obtained by IMAC-Ga was analyzed by liquid chromatograpy/electrospray ionization mass spectrometry (LC/ESI-MS) in an Orbitrap XL and by matrix-assisted laser desorption/ionization time-of-flight time-of-flight (MALDI-TOF/TOF) in an ABI 4800 instrument. From this fraction, different sulfated and non-sulfated peptides belonging to the caerulin and bradykinin families were structurally characterized. Other interesting negatively charged groups, such as phosphate adducts of dermaseptins and pyridoxal phosphate attached to a protease inhibitor, were also characterized. Unexpectedly, some dermaseptin antimicrobial peptides were also enriched by IMAC-Ga and a Sauvatine-like peptide was also fully sequenced. Furthermore, neutral loss of sulfated peptides and their fragmentation patterns in the gas phase were also compared using collision-induced dissociation (CID) and high-energy collision dissociation (HCD). Our present study provides evidence that IMAC-Ga enrichment is a fast, useful and promising method for high-throughput analysis of sulfated-peptides, since high-resolution mass spectrometers can be used for this purpose. 相似文献

Polymerization in hexavalent uranium solutions was measured by electrospray ionization time‐of‐flight mass spectrometry in three different acidic media at pH values from 3 to 5.3 in order to detect all hydrolysis species present in solution. The aqueous solutions were directly measured without further dilution in organic solvents. At high uranyl concentrations ([U(VI)] = 10? 3 M) artifacts were observed due to the presence of more than one solution species per formed microdroplet. Those artifacts were composed of ions and neutral species being present in the same droplet. However, by analyzing the detected species carefully, the origin of the artifacts could be traced back to the physically meaningful species. Still, only general trends of the hydrolysis behavior can be deduced from the measurements at [U(VI)] = 1 ? 10? 3 M. The solutions at [U(VI)] = 5 ? 10? 5 M did not show any comparable artifact formation. The detected species distributions resemble the expected trends calculated from the equilibrium constants published in the Nuclear Energy Agency Thermodynamic Database (NEA‐TDB). The neutral (UO2)(CO3)0 species present in solution causes, if located in the same microdroplet as a charged species, the apparent formation of dimeric and trimeric ternary hydroxo carbonate complexes at pH 5.3. As the uncharged species is not repelled from the ionic species, it might remain in the same droplet during the droplet fission process. By dividing those detected species into the uncharged (UO2)(CO3)0 and a second ionic species, the relative abundances of the solution species can be corrected, leading to a good agreement with the predictions of the published equilibrium constants. Twenty proteinogenic amino acids (AAs) were determined without derivatization using flow injection analysis followed by electrospray ionization mass spectrometry and tandem mass spectrometry (ESI-MS and ESI-MS/MS) and electrospray ionization high-field asymmetric waveform ion mobility mass spectrometry and tandem mass spectrometry (ESI-FAIMS-MS and ESI-FAIMS-MS/MS), in positive and negative ionization modes. Three separate sets of ESI-FAIMS conditions were used for the separation and detection of the 20 AAs. Typically ESI-FAIMS-MS showed somewhat improved sensitivity and significantly better signal-to-noise ratios than ESI-MS mainly due to the elimination of background noise. However, the difference between ESI-FAIMS-MS and ESI-MS/MS was significantly less. ESI-FAIMS was able to partially or completely resolve all the isobaric amino acid overlaps such as leucine, isoleucine and hydroxyproline or lysine and glutamine. Detection limits for the amino acids in ESI-FAIMS-MS mode ranged from 2 ng/mL for proline to 200 ng/mL for aspartic acid. Overall, ESI-FAIMS-MS is the preferred method for the quantitative analysis of AAs in a hydrolyzed yeast matrix. 相似文献

Most complexes of azides and transition metals involve the N(3)(-) azide anion as a ligand other than an organic azide. Complexes of organic azides with metals are involved in biological applications and in the deposition of nitrenes on metal surfaces, producing nitride layers for semi-conductors preparation; this makes the study of these interactions an important issue. This work describes a study of the complexation of nickel and cobalt by 3-azidopropionitrile by means of electrospray ionization mass spectrometry (ESI-MS). Complexes were obtained from solutions of NiCl(2) and CoCl(2) in methanol/water. In the case of nickel, other NiX(2) salts were investigated (where X = Br or NO(3)) and other solvents were also studied (notably ethanol/water). All complexes detected were single positively charged, with various stoichiometries, some resulted from the fragmentation of the ligand, the loss of N(2), and HCN being quite common. The most abundant cations observed were [Ni(II)AzAzX](+), where X = Cl, Br, NO(3). Some of the complexes showed solvation with methanol/ethanol/water. Metal reduction was observed in complexes where a radical was lost, resulting from the homolytic cleavage of a metal-nitrogen bond. Collision induced dissociation (CID) experiments followed by tandem mass spectrometry (MS-MS) analysis were not absolutely conclusive about the coordination site. However, terminal ions observed from the fragmentation routes were explained by a proposed gas-phase mechanism. Density functional theory calculations were carried out and provided structures for some complexes, pointing to the possibility of 3-azidopropionitrile acting as a mono- or a bidentate ligand. 相似文献

Arsenic pollution of public water supplies has been reported in various regions of the world. Recently, some cancer patients are treated with arsenite (AsIII); most Japanese people consume seafoods containing large amounts of negligibly toxic arsenic compounds. Some of these arsenic species are metabolized, but some remain intact. For the determination of toxic AsIII, a simple, rapid and sensitive method has been developed using electrospray ionization mass spectrometry (ESI-MS). AsIII was reacted with a chelating agent, pyrrolidinedithiocarbamate (PDC, C4H8NCSS-) and tripyrrolidinedithiocarbamate-arsine, As(PDC)3, extracted with methyl isobutyl ketone (MIBK). A 1 μL aliquot of MIBK layer was directly injected into ESI-MS instrument without chromatographic separation, and was detected within 1 min. Arsenate (AsV) was reduced to AsIII with thiosulfate, and then the total inorganic As was quantified as AsIII. This method was validated for the analysis of urine samples. The limit of detection of As was 0.22 μg L−1 using 10 μL of sample solution, and it is far below the permissible limit of As in drinking water, 10 μg L−1, recommended by the WHO. Results were obtained in < 10 min with a linear calibration range of 1-100 μg L−1. Several organic arsenic compounds in urine did not interfere with AsIII detection, and the inorganic As in the reference materials SRM 2670a and 1643e were quantified after the reduction of AsV to AsIII. 相似文献

Mass spectrometry of ochratoxin A (OTA) and B (OTB) under electrospray ionization (ESI) and atmospheric pressure chemical ionization (APCI) was studied. ESI offers higher sensitivities and less fragmentation than APCI. A sensitive LC/MS/MS method for the determination of ochratoxin A (OTA) in human plasma samples was developed. The absolute minimum detection limit was around 10-20 pg per injection, corresponding to 0.5 ppb in an injection equivalent to 20-40microg of human plasma. Ochratoxin B (OTB) was used as an internal standard and its absence in real-life samples was carefully checked before samples were spiked with the internal standard. It was found that these two ochratoxins are susceptible to sodium adduct formation. Fragment ions from the [M + H](+) and [M + Na](+) ions of both OTA and OTB were monitored in the multiple reaction monitoring mode. Three quantitative approaches, standard addition method, internal standard method (using ochratoxin B as an internal standard) and external standard method, were compared in the analysis of human blood plasma. Results from the mass spectrometric method were comparable to those from a conventional LC/fluorescence method. The LC/MS/MS method was also applied to the analysis of contaminated coffee samples. 相似文献

Rapid and simple chiral analysis of thalidomide solutions is demonstrated by using electrospray ionization tandem mass spectrometry and analysis of cluster ion dissociation by the kinetic method. The electrospray (ES) behaviour of selected Remazol textile dyes, their hydrolysis products and the latters' reaction, following elution from a strong anion-exchange cartridge, with 30% concentrated HCl in MeOH, is studied and applied to the direct analysis of dye containing effluent. For unambiguous identification and determination of these textile dyes in effluents, it is necessary to resort to ES utilising MS-MS and MS3. Further, a tabular review of recent applications of HPLC-ES-MS and, to a lesser extent, CE-ES-MS with reference to drug and pesticide analysis is presented. 相似文献
