Simultaneous multicomponent analysis is usually carried out by multivariate calibration models such as partial least squares (PLS) that utilize the full spectrum. It has been demonstrated by both experimental and theoretical considerations that better results can be obtained by a proper selection of the spectral range to be included in calculations. A genetic algorithm is one of the most popular methods for selecting variables for PLS calibration of mixtures with almost identical spectra without loss of prediction capacity. In this work, a simple and precise method for rapid and accurate simultaneous determination of sulfide and sulfite ions based on the addition reaction of these ions with new fuchsin at pH 8 and 25°C by PLS regression and using a genetic algorithm (GA) for variable selection is proposed. The concentrations of sulfide and sulfite ions varied between 0.05–2.50 and 0.15–2.00 μg/mL, respectively. A series of synthetic solutions containing different concentrations of sulfide and sulfite were used to check the prediction ability of GA-PLS models. The root mean square error of prediction with PLS on the whole data set was 0.19 μg/mL for sulfide and 0.09 μg/mL for sulfite. Two spectrophotometric methods were applied to the simultaneous assay of chlorhexidine hydrochloride (CHL) and lidocaine hydrochloride (LIH) in pharmaceutical formulations. Using derivative spectrophotometry, CHL was determined by measurement of its first derivative signal at 290 nm (peak to zero amplitude) in the concentration range 5–9 μg/mL, and LIH was analysed by measurement of its second derivative signals at 272 and 276 nm (peak to peak amplitude) in the concentration range 160–480 μg/mL. With the partial least-squares (PLS-2), the experimental calibration matrix was constructed using 9 samples. The concentration ranges considered were 5–7 μg/mL for CHL and 220, 240, 260 μg/mL for LIH. The absorbances were recorded between 240 and 310 nm at every 5 nm.  相似文献

A simple, sensitive and selective spectrophotometric method for simultaneous determination of tretinoin and minoxidil using partial least square (PLS) calibration and H-point standard addition method (HPSAM) is described. The results of the H-point standard addition method show that minoxidil and tretinoin can be determined simultaneously with the concentration ratio of tretinoin to minoxidil varying from 2: 1 to 1: 33 in mixed samples. A partial least squares multivariate calibration method for the analysis of binary mixtures of tretinoin and minoxidil was also developed. The total relative standard error for applying the PLS method to eleven synthetic samples in the concentration range of 0–10 μg mL−1 tretinoin and 0–32 μg mL−1 minoxidil was 2.59 %. Both proposed methods (PLS and HPSAM) were also successfully applied in the determination of tretinoin and minoxidil in several synthetic pharmaceutical solutions.  相似文献

A new, simple, and rapid capillary electrophoresis (CE) method employing hexadimethrine bromide (HDB) as electroosmotic flow (EOF) modifier was developed for the identification and quantitative determination of two pharmaceutically active constituents—gallic acid (GA) and salidroside (S)—in extracts of Rhodiola root and its medicinal preparation. The optimum separation was achieved at pH 11.00 with the use of 10 mM borate buffer containing 0.001% (w/v) of HDB. The applied voltage was ∼15 kV and the capillary temperature was kept constant at 25°C. m-Phthalic acid was used as an internal standard for quantification. The calibration dependences exhibited good linearity for the ratios of the concentrations of standard samples and internal standard and the ratios of the peak area of samples and internal standard over the concentration range from 24 to 1200 μg/mL for GA and 2.4 to 72 μg/mL for S. The correlation coefficients were 0.9999 and 0.9997, and the detection limits of the CE method corresponding to a signal-to-noise ratio of three were 6 and 2 μg/mL for GA and S, respectively. The relative standard deviations of the relative migration time and the relative peak area of samples were 0.5 and 4.0% for GA and 1.9 and 5.3% for S. The effects of buffer pH and the concentration of HDB on the resolution were studied systematically. The contents of these two active compounds in Rhodiola root and its preparation were successfully determined over 6 min with satisfactory repeatability and recovery. A method is described for measuring the concentrations of both glucose and glutamine in binary mixtures from near infrared (NIR) absorption spectra. Spectra are collected over the range from 5000–4000/cm (2.0–2.5μm) with a 1-mm optical path length. Glucose absorbance features at 4710, 4400, and 4300/cm and glutamine features at 4700, 4580, and 4390/cm provide the analytical information required for the measurement. Multivariate calibration models are generated by using partial least squares (PLS) regression alone and PLS regression combined with a preprocessing digital Fourier filtering step. The ideal number of PLS factors and spectral range are identified separately for each analyte. In addition, the optimum Fourier filter parameters are established for both compounds. The best overall analytical performance is obtained by combining Fourier filtering and PLS regression. Glucose measurements are established over the concentration range from 1.66–59.91 mM, with a standard error of prediction (SEP) of 0.32 mM and a mean percent error of 1.84%. Glutamine can be measured over the concentration range from 1.10–30.65 mM with a SEP of 0.75 mM and a mean percent error of 6.67%. These results demonstrate the analytical utility of NIR spectroscopy for monitoring glucose and glutamine levels in mammalian and insect cell cultures.  相似文献

Simple, accurate, rapid, and sensitive spectrofluorimetric methods for the determination of levosulpiride in pharmaceutical formulation were developed utilizing its fluorescence reaction with Fe3+ (method A) and Al3+ (method B). The calibration curves were found to be linear in the concentration range 0.239–3.44 μg/mL and 0.310–2.730 μg/mL with limit of detection 0.005 μg/mL and 0.0032 μg/mL, respectively, for method A and method B. The reaction conditions were studied and optimized. In addition, the complexation of Mg2+ and Ca2+ was also studied. In all cases, an enhancement in fluorescence emission of levosulpiride upon formation of complex with metal ions was observed. A 2: 1 (drug: metal) stoichiometry for all the complexes was established. Benesi-Hildebrand method was applied for calculation of association constant at 25 and 35°C. The thermodynamic parameters obtained in this study revealed that the interaction process was spontaneous and mainly ΔS-driven.  相似文献

A new simple, sensitive and rapid catalytic-spectrophotometric method for the determination of oxalic acid has been described based on its catalytic effect on the redox reaction between dichromate and Brilliant cresyl blue in acidic media by means of a flow injection analysis method. The color change of Brilliant cresyl blue due to its oxidation was monitored spectrophotometrically at 625 nm. The calibration graph was linear in the range of 0.020–4.70 μg/mL oxalic acid with a limit of detection 0.005 μg/mL of oxalic acid. The relative standard deviation for ten replicate measurements of 0.020 μg/mL and 0.900 μg/mL was 2.2% and 1.7%, respectively. No serious interference was identified. The catalytic effect of Se(IV) was first observed in the reaction of the reduction of Nile Blue with sulfide ions. Optimal conditions for the determination of selenium by this reaction were found. The dependence of the reaction rate on the concentration of Se(IV) was linear in the concentration range 0.008-0.16 μg/mL. Solvent extraction with dithiophosphoric acids was proposed for the separation of selenium from the interferents. A procedure for the extraction-kinetic determination of selenium with the detection limit 0.006 μg/mL was developed.  相似文献

In this article, a sensitive cloud point extraction procedure for the preconcentration of trace amounts of palladium, gold and nickel prior to their determination by flame atomic absorption spectrometry has been developed. The cloud point extraction method is based on the complexation of Pd(II), Au(II), and Ni(II) ions with 1-(2-pyridylazo)-2-naphthol and entrapping in non-ionic surfactant Triton X-114. The main factors affecting cloud point extraction efficiency, such as pH of sample solution, concentration of 1-(2-pyridylazo)-2-naphthol and Triton X-114, equilibration temperature and time, were investigated in detail. Under the optimized conditions, calibration curves were constructed for the determination of palladium, gold and nickel according to the general procedure. Linearity was maintained from 0.01 to 1.0 μg/mL for palladium, 10.0 μg/mL to 1.5 μg/mL for gold, and 10.0 μg/mL to 0.5 μg/mL for nickel. Detection limits based on three times the standard deviation of the blank divided by the slope of analytical curve (3Sb/m) for Pd(II), Au(III), and Ni(11) ions were 3.4, 3.9, and 2.4 μg/mL, respectively. Seven replicate determination of a mixture of 0.5 μg/mL palladium and gold and 0.2 μg/mL nickel gave a mean absorbance of 0.174, 0.150, and 0.201 with relative standard deviation ±1.5, ±1.3, and ±1.8%, respectively. The high efficiency of cloud point extraction to carry out the determination of analytes in complex matrices was demonstrated. The proposed method has been applied for determination of trace amount of palladium, gold and nickel in certified reference material and water samples with satisfactory results.  相似文献

 Two different mass spectrometric methods, negative thermal ionization isotope dilution mass spectrometry (NTI-IDMS) and inductively coupled plasma mass spectrometry (ICP-MS), off-line and on-line coupled with anion exchange chromatography, have been developed for simultaneous bromide and bromate determinations in water samples. The detection limits of these methods are in the range of 0.03–0.09 μg/L using a 50 mL sample.The results are independent of the content of other anions, which could be demonstrated by the analyses of six mineral waters containing chloride and sulfate of up to 160 mg/L and 1500 mg/L, respectively. Bromide has been analyzed by the NTI-IDMS method in the range of 10–500 μg/L and bromate in the range of 1–50 μg/L with relative standard deviations of 0.3–1.2% and 0.4–6%. Quantification for the ICP-MS method was carried out by the standard addition technique, which resulted in relative standard deviations of 5.5% for bromide at the 500 μg/L level and of 13% for bromate at the level of about 3 μg/L. These results are compared with those described in the literature for ion chromatographic (IC) and other methods and those obtained in this work by IC using UV detection, which allows high concentrations of chloride in the bromate fraction. The detection limits of this IC method are 6 μg/L for bromide and 30 μg/L for bromate. NTI-IDMS and ICP-MS therefore fit the recommendations of the European Union (detection limit<2.5 μg/L; precision and accuracy better than 25% at the 10 μg/L level) for methods analyzing the carcinogenic bromate much better than IC and other methods applied up to now. As a definitive but time consuming method, NTI-IDMS is preferably applicable as a calibration technique, whereas ICP-MS, with relatively short analysis times, due to on-line coupling with chromatography, can be used as a sensitive and powerful routine method for trace bromide and bromate species in water samples. A new liquid chromatography–mass spectrometry (LC/MS) method has been developed for the quantitative analysis of plasmalogens in human plasma using a nonendogenous plasmalogen (1-O-1′-(Z)-tricosenyl-2-oleoyl-rac-glycero-3-phosphocholine, PLS 23:0/18:1) as an internal standard. 1-O-1′-(Z)-Tricosenyl glyceryl ether was prepared by reacting lithioalkoxyallyl with 1-iodoeicosane as the key intermediate in the formation of PLS 23:0/18:1. In LC/MS analyses, PLS 23:0/18:1 generated significant fragment ions in positive and negative modes. In positive ion mode, the [M+H]+ of PLS 23:0/18:1 yielded unique fragments with cleavages at the sn-1 and sn-2 positions of the glycerol backbone. In negative ion mode, the [M+CH3COO] of PLS 23:0/18:1 resulted in characteristic fragmentation at the sn-2 and sn-3 positions. 1-O-1′-(Z)-Hexadecenyl-2-linoleoyl-rac-glycero-3-phosphocholine (PLS 16:0/18:2) and 2-arachidonoyl-O-1′-(Z)-hexadecenyl-rac-glycero-3-phosphocholine (PLS 16:0/20:4) were chemically synthesized as PLS 23:0/18:1. The calibration curves obtained for PLS 16:0/18:2 and PLS 16:0/20:4 were linear throughout the calibration range (0.04–1.60 pmol). The LOD (S/N = 5:1) was 0.008 pmol and the LOQ (S/N = 6:1) was 0.01 pmol for both PLS 16:0/18:2 and PLS 16:0/20:4. Plasma concentrations of PLS 16:0/18:2 and PLS 16:0/20:4 were 4.0 ± 1.3 μM and 3.5 ± 1.2 μM (mean ± SD), respectively, in five healthy volunteers.  相似文献

An analytical method for detecting and quantifying cefotaxime in plasma and several tissues is described. The method was developed and validated using plasma and tissues of rats. The samples were analyzed by reversed phase liquid chromatography (HPLC) with UV detection (254 nm). Calibration graphs showed a linear correlation (r > 0.999) over the concentration ranges of 0.5–200 μg/mL and 1.25–25 μg/g for plasma and tissues, respectively. The recovery of cefotaxime from plasma standards prepared at the concentrations of 25 μg/mL and 100 μg/mL was 98.5 ± 3.5% and 101.8 ± 2.2%, respectively. The recovery of cefotaxime from tissue standards of liver, fat and muscle, prepared at the concentration of 10 μg/g was: 89.8 ± 1.2% (liver), 103.9 ± 6.5% (fat) and 97.8 ± 2.1% (muscle). The detection (LOD) and quantitation (LOQ) limits for plasma samples were established at 0.11 μg/mL and 0.49 μg/mL, respectively. The values of these limits for tissues samples were approximately 2.5 times higher: 0.3 μg/g (LOD) and 1.25 μg/g (LOQ). For plasma samples, the deviation of the observed concentration from the nominal concentration was less than 5% and the coefficient of variation for within-day and between-day assays was less than 6% and 12%, respectively. The method was used in a pharmacokinetic study of cefotaxime in the rat and the mean values of the pharmacokinetic parameters are given. A novel method is proposed for the extraction-thermal lens quantification of cobalt with Nitroso-R-Salt based on the distribution of the colored complex in a two-phase aqueous system on the basis of poly-ethylene glycol (PEG) and an ammonium sulfate solution followed by its thermal lens detection in the extract. The limit of detection is 0.3 μM (20 ng/mL); the lower limit of the analytical range is 0.7 μM (40 ng/mL); the relative standard deviation for the concentrations 1–50 μM makes 1–3% (n = 6, P = 0.95). In the determination of cobalt by spectrophotometry under the same conditions, the detection limit is 10 μM (0.6 μg/mL) and the lower limit of the analytical range is 40 μM (2.5 μg/mL). The precision of thermal lens measurements in PEG solutions is higher in comparison to that in aqueous ones because of the weaker interference of convection in aqueous solutions of PEG.  相似文献

In the present study, a new sensitive and simple kinetic-spectrophotometric method for the determination of the insecticide dimethoate [O,O-dimethyl-S-(N-methyl-carbomoylmethyl)-phosphoro-dithioate] is developed. The method is based on the inhibition effect of dimethoate on the oxidation of malachite green (MG) by potassium periodate (KIO4) in the presence of Mn(II) ions. Inhibition kinetics of this catalytic reaction was investigated in the presence of dimethoate and the possibility of its analytical application was evaluated. The important variables that affected the reaction rate were investigated and the optimum conditions giving maximum sensitivity were established. Dimethoate was determined with linear calibration graph in the interval from 4.58 to 41.22 μg/mL. The optimized conditions yielded a theoretical detection limit of 1.24 μg/mL based on the 3S b criterion. The RSDs of the method (n = 5) were 1.2–4.9% for the concentration interval of dimethoate from 4.58 to 41.22 μg/mL. The reaction was monitored spectrophotometrically by measuring the change in absorbance over time at 615 nm. The method was applied to the determination of dimethoate in waters and milk, and was compared with the spectrophotometric method. The quantitive method developed on the basis of inhibition kinetics is practical, fast and economical. For this reason, it is open for new application fields.  相似文献

Near infrared (NIR) spectra in the 1300– 1850 nm region were measured for control serum solutions containing both albumin and γ-globulin of various concentrations. Partial least squares two (PLS2) regression was applied to the NIR spectra to determine simultaneously the concentrations of both proteins. For albumin, the correlation coefficient (R) of 0.988, the standard error of calibration (SEC) of 1.61 g/L, the standard error of prediction (SEP) of 1.29 g/L, the relative standard deviation (RSD) of 0.026 and the ratio of standard deviation of reference data in prediction to SEP (RPD) of 12.2 were obtained. For γ-globulin, the corresponding values were 0.997, 1.36 g/L, 1.35 g/L, 0.0365 and 8.66, respectively. The regression coefficients (RCs) of PLS factors were compared between albumin and γ-globulin, and the observed differences in the RCs were discussed based upon the differences in the hydration between albumin and γ-globulin. In order to explore the effects of various metabolites such as glucose, and cholesterol on the chemometrics models, the RCs for albumin and γ-globulin in the control serum solutions were also compared with those for albumin and γ-globulin in phosphate buffer solutions previously studied. The results of our experiments show that NIR spectroscopy with the use of PLS2 regression has considerable promise in nondestructive determination of the concentrations of blood serum proteins. A procedure was developed for the determination of the following thioamides based on thiazole, 1,3,4-thiadiazole, 1,2,4-triazole, and tetrazole: 2-mercaptothiazole (I), 2-mercapto-1,3,4-thiadiazole (II), 2-mercapto-5-methyl-1,3,4-thiadiazole (III), 3-mercapto-1,2,4-triazole (IV), 3-mercapto-4-methyl-1,2,4-triazole (V), and 5-mercapto-1-methyltetrazole (VI). The determination was performed by reversed-phase HPLC on a column (150 × 4 mm) packed with Diaspher-110-C18 (5 μm) using elution with an acetonitrile-acetate buffer solution (pH 4.70) mixture (5: 95). Detection was performed at the light absorption maximums of compounds I (320 nm), II (305 nm), III (310 nm), IV (260 nm), V (254 nm), and VI (245 nm). The calibration graphs were linear over the following concentration ranges (μg/mL): 0.47–11.72 (I), 0.47–11.82 (II), 0.53–13.22 (III), 0.40–10.11 (IV), 0.46–11.52 (V), and 0.46–11.62 (VI). The limits of detection were 0.45, 0.43, 0.50, 0.37, 0.41, and 0.42 μg/mL for compounds IVI, respectively. Conditions for the separation of a mixture of compounds I and IIIV and for the quantitative determination of compounds IVI by capillary zone electrophoresis (CZE) were optimized. CZE was performed on a quartz capillary of size 60 cm (effective length of 50 cm) × 75 μm at a voltage of 20 kV with a borate buffer solution (pH 9.18). The procedure allowed us to evaluate the concentrations of substances in the ranges of 1.17–93.75 (I), 1.18–94.54 (II), 1.32–105.76 (III), 1.01–101.13 (IV) 1.15–115.16 (V), and 1.16–116.15 (VI) μg/mL with the detection limits of 1.10, 1.11, 1.20, 0.96, 1.01, and 1.02 μg/mL for compounds IVI, respectively.  相似文献

A simple, economical, and automated spectrophotometric method for the determination of chlorpromazine hydrochloride by sequential injection analysis using ammonium metavanadate as colorimetric reagent is proposed. The various chemical and physical conditions that affected the reaction have been thoroughly investigated. The calibration curve was linear within the range 10–100 μg/mL. The detection limit (S/N = 3) was 0.7 μg/mL and the limit of quantification (S/N = 10) was 2.3 μg/mL. The sampling frequency was 22 h−1. The method has been used for the determination of chlorpromazine hydrochloride in pure form and pharmaceutical formulations. The t-test has revealed that there is no evidence of significant differences between the obtained results at the 95% confidence level. The method can be applied to the quantitative determination of chlorpromazine hydrochloride. It is also applicable in the quality control of chlorpromazine hydrochloride preparations. For the first time, a simple, selective and accurate high-performance liquid chromatography method with ultraviolet detection was developed and validated to quantify simultaneously three structurally related antiepileptic drugs; carbamazepine, oxcarbazepine, and the recently launched eslicarbazepine acetate and their main metabolites, carbamazepine-10,11-epoxide, 10,11-trans-dihydroxy-10,11-dihydro-carbamazepine, and licarbazepine. The method involves a solid-phase extraction and a reverse-phase C18 column with 5 cm length. The mobile phase consisting of water, methanol, and acetonitrile in the ratio 64:30:6 was selected as the best one and pumped at 1 mL/min at 40 °C. The use of this recent column and an aqueous mobile phase instead of buffers gives several advantages over the method herein developed; namely the fact that the chromatographic analysis takes only 9 min. The method was validated according to the guidelines of the Food and Drug Administration, showing to be accurate (bias within ±12%), precise (coefficient variation <9%), selective and linear (r 2 > 0.997) over the concentration range of 0.05–30 μg/mL for carbamazepine; 0.05–20 μg/mL for oxcarbazepine; 0.15–4 μg/mL for eslicarbazepine acetate; 0.1–30 μg/mL for carbamazepine-10,11-epoxide; 0.1–10 μg/mL for 10,11-trans-dihydroxy-10,11-dihydro-carbamazepine, and 0.1–60 μg/mL for licarbazepine. It was also shown that this method can adequately be used for the therapeutic drug monitoring of the considered antiepileptic drugs, carbamazepine, oxcarbazepine, eslicarazepine acetate, and their metabolites.  相似文献

A method for sulfur determination in diesel fuel employing near infrared spectroscopy, variable selection and multivariate calibration is described. The performances of principal component regression (PCR) and partial least square (PLS) chemometric methods were compared with those shown by multiple linear regression (MLR), performed after variable selection based on the genetic algorithm (GA) or the successive projection algorithm (SPA). Ninety seven diesel samples were divided into three sets (41 for calibration, 30 for internal validation and 26 for external validation), each of them covering the full range of sulfur concentrations (from 0.07 to 0.33% w/w). Transflectance measurements were performed from 850 to 1800 nm. Although principal component analysis identified the presence of three groups, PLS, PCR and MLR provided models whose predicting capabilities were independent of the diesel type. Calibration with PLS and PCR employing all the 454 wavelengths provided root mean square errors of prediction (RMSEP) of 0.036% and 0.043% for the validation set, respectively. The use of GA and SPA for variable selection provided calibration models based on 19 and 9 wavelengths, with a RMSEP of 0.031% (PLS-GA), 0.022% (MLR-SPA) and 0.034% (MLR-GA). As the ASTM 4294 method allows a reproducibility of 0.05%, it can be concluded that a method based on NIR spectroscopy and multivariate calibration can be employed for the determination of sulfur in diesel fuels. Furthermore, the selection of variables can provide more robust calibration models and SPA provided more parsimonious models than GA.  相似文献
