An Mg2+-dependent low ATPase activity can be detected in erythrocyte "white membranes," in addition to that of the well known (Ca2+ + Mg2+)-ATPase. The thiol oxidizing agent diamide affects both activities. The oxidation of neighboring thiols seems to leave the mechanism of the (Ca2+ + Mg2+)-ATPase amplification system evoked by Ca2+ largely unaffected. The perturbation caused by diamide in the membranes seems to affect primarily a step of the ATP hydrolysis mechanism that is common to both ATPase activities. The effectiveness of diamide seems to be the same when either Ca2+ and Mg2+, or Mg2+ alone are present during the reagent action. Reduction of disulfide bonds by DTE after diamide treatment restores the (Ca2+ + Mg2+)-ATPase activity but is unable to take the Mg2+-ATPase activity back to the original level. The hypothesis is discussed that the redox state of one (or more than one) couple of --SH close to each other and possibly connected to the active site, may be an important factor in optimizing the efficiency of Ca action on the (Ca2+ + Mg2+)-ATPase.  相似文献

This study was aimed at evaluating the preventive role of the ethanolic extract of Lagenaria siceraria (Mol) fruit on membrane-bound enzymes, such as sodium potassium-dependent adenosine triphosphatase (Na(+)/K(+) ATPase), calcium-dependent adenosine triphosphatase (Ca(2+) ATPase) and magnesium-dependent adenosine triphosphatase (Mg(2+) ATPase) on isoproterenol (ISO)-induced myocardial infarction (MI) in rats. Male albino Wistar rats were pretreated with the ethanolic extract of L. siceraria (Mol) fruit (125, 250 and 500?mg?kg(-1) body weight) for a period of 30 days. Acridine orange (AO), a weakly basic fluorescent dye, is permeable to plasma and vesicle membranes and preferentially remains in intracellular acidic regions. Using fluorescence microscopy, we observed dynamic changes in AO-loaded cultured malignant melanoma cells during illumination with blue light. Immediately after the start of the illumination, the successive disruption of vesicles was observed as a flash of fluorescence, and shortly after that, blebs were formed on the plasma membrane. These cells died within 5 min. Vesicle disruption was completely inhibited when cells were treated with the vacuolar H(+)-ATPase inhibitor bafilomycin A1 followed by loading with AO, but not when bafilomycin A1 was treated after AO loading. Thus, the filling of AO in the vesicle, which is driven by vacuolar H(+)-ATPase, is initially required for vesicle disruption. In contrast, bafilomycin A1 did not prevent plasma membrane blebbing, indicating that the blebs are formed independently of the vesicle disruption. Acute cell death was inhibited by treatment with bafilomycin A1 before but not after AO loading. Thus, AO- and blue light-induced acute cell death is associated with vesicle disruption rather than bleb formation. Both the vesicle disruption and the formation of plasma membrane blebs were inhibited by removal of oxygen from the cell environment and by singlet oxygen scavengers, sodium azide, ascorbic acid, and L-histidine, but not inhibited by the hydroxyl radical scavenger dimethyl thiourea. Acute cell death was also prevented by singlet oxygen scavengers but not by dimethyl thiourea. Thus, these phenomena are likely caused at least in part by the generation of singlet oxygen. The photosensitive features of plasma and vesicle membranes observed in the present study may be based on the use of the photodynamic effect, such as cancer therapy.  相似文献

Novel gadolinium complexes (KMR-series: KMR-K and KMR-Mg), which have a bis-15-crown-5 ether or a charged beta-diketone structure as a recognition site, have been designed, synthesized and applied for the detection of K(+) or of Mg(2+) and Ca(2+) using MRI or NMR techniques. The measurements are based on the modulation of the longitudinal relaxation time (T(1)) of water protons in proximity of the gadolinium complexes. Relaxivity measurements of KMR-K1 in aqueous solution showed that the initial longitudinal relaxivity value (r(1)) of 5.05 mM(-1) s(-1) is monotonously decreasing with increasing K(+) concentrations, reaching a final value of 4.78 mM(-1) s(-1). This decrease is attributed to a change in the second sphere of hydration of the gadolinium (Gd(3+)) complex (KMR-K), resulting in a K(+) concentration-dependent contrast in MR images. From stoichiometric analysis using mass spectrometry and UV/VIS spectrometry, a 1 : 1 complex formation between KMR-K1 and K(+) in a sandwich-type manner with a log K of 3.20 was confirmed. In the case of KMR-Mg, the initial r(1) value of 4.98 mM(-1) s(-1) is monotonously decreasing with increasing Mg(2+) or Ca(2+) concentrations, reaching a final value of 3.95 or 4.16 mM(-1) s(-1), respectively, resulting in Mg(2+) and Ca(2+) concentration-dependent contrast in MR images. The formation of a 1:1 complex with a log K of 2.33 for Mg(2+) and 1.91 for Ca(2+) was confirmed. KMR-K1 and KMR-Mg are the first ion-selective or ion-sensitive gadolinium complexes for K(+) or Mg(2+) and Ca(2+), respectively.  相似文献

A molecular simulation study is reported for CO(2) adsorption in rho zeolite-like metal-organic framework (rho-ZMOF) exchanged with a series of cations (Na(+), K(+), Rb(+), Cs(+), Mg(2+), Ca(2+), and Al(3+)). The isosteric heat and Henry's constant at infinite dilution increase monotonically with increasing charge-to-diameter ratio of cation (Cs(+) < Rb(+) < K(+) < Na(+) < Ca(2+) < Mg(2+) < Al(3+)). At low pressures, cations act as preferential adsorption sites for CO(2) and the capacity follows the charge-to-diameter ratio. However, the free volume of framework becomes predominant with increasing pressure and Mg-rho-ZMOF appears to possess the highest saturation capacity. The equilibrium locations of cations are observed to shift slightly upon CO(2) adsorption. Furthermore, the adsorption selectivity of CO(2)/H(2) mixture increases as Cs(+) < Rb(+) < K(+) < Na(+) < Ca(2+) < Mg(2+) ≈ Al(3+). At ambient conditions, the selectivity is in the range of 800-3000 and significantly higher than in other nanoporous materials. In the presence of 0.1% H(2)O, the selectivity decreases drastically because of the competitive adsorption between H(2)O and CO(2), and shows a similar value in all of the cation-exchanged rho-ZMOFs. This simulation study provides microscopic insight into the important role of cations in governing gas adsorption and separation, and suggests that the performance of ionic rho-ZMOF can be tailored by cations.  相似文献

The fluorinated anti-psychotic drug trifluoperazine (TFP) has been shown to be a K(+)-competitive inhibitor of gastric H(+)/K(+)-ATPase, a membrane-embedded therapeutic target for peptic ulcer disease. This paper describes how variable contact time (19)F cross-polarization magic angle spinning (VCT-CP/MAS) NMR has been used to probe the inhibitory interactions between TFP and H(+)/K(+)-ATPase in native gastric membranes. The (19)F CP/MAS spectra for TFP in H(+)/K(+)-ATPase enriched (GI) gastric membranes and in control membranes containing less than 5 nmol of the protein indicated that the drug associates with the membranes independently of the presence of H(+)/K(+)-ATPase. The (19)F peak intensities in the VCT-CP/MAS experiment confirmed that TFP undergoes slow dissociation (k(off) < 100 s(-1)) from binding sites in GI membranes, and more rapid dissociation (k(off) < 100 s(-1)) from control membranes. The spectra showed that up to 40% of bound TFP was displaced from GI membranes by 100 mM K(+) and by the K(+)-competitive inhibitor TMPIP, but TFP was not displaced from the control membranes. Hence the spectra of TFP in GI membranes represent the drug bound to the K(+)-competitive inhibitory site of H(+)/K(+)-ATPase and to other non-specific sites. The affinity of TFP for the K(+)-competitive site (K(D) = 4 mM) was determined from a binding curve of (19)F peak intensity versus TFP concentration after correction for non-specific binding. The K(D) was much higher than the IC(50) for ATPase inhibition (8 microM), which suggests that the substantial non-specific binding of TFP to the membranes contributes to ATPase inhibition. This novel approach to probing ligand binding can be applied to a wide range of membrane-embedded pharmaceutical targets, such as G-protein coupled receptors and ion channels, regardless of the size of the protein or strength of binding.  相似文献

New 5-chloro-8-hydroxyquinoline (CHQ)-substituted aza-18-crown-6 (4), diaza-18-crown-6 (1), diaza-21-crown-7 (2), and diaza-24-crown-8 (3) ligands, where CHQ was attached through the 7-position, and aza-18-crown-6 (11) and diaza-18-crown-6 (10) macrocycles, where CHQ was attached through the 2-position, were prepared. Thermodynamic quantities for complexation of these CHQ-substituted macrocycles with alkali, alkaline earth, and transition metal ions were determined in absolute methanol at 25.0 degrees C by calorimetric titration. Two isomers, 1 and 10, which are different only in the attachment positions of the CHQ to the parent macroring, exhibit remarkable differences in their affinities toward the metal ions. Compound 1 forms very stable complexes with Mg(2+), Ca(2+), Cu(2+), and Ni(2+) (log K = 6.82, 5.31, 10.1, and 11.4, respectively), but not with the alkali metal ions. Ligand 10 displays strong complexation with K(+) and Ba(2+) (log K = 6.61 and 12.2, respectively) but not with Mg(2+) or Cu(2+). The new macrocycles and their complexes have been characterized by means of UV-visible and (1)H NMR spectra and X-ray crystallography. New peaks in the UV spectrum of the Mg(2+)-1 complex could allow an analytical determination of Mg(2+) in very dilute solutions in the presence of other alkali and alkaline earth metal cations. (1)H NMR spectral and X-ray crystallographic studies indicate that ligand 10 forms a cryptate-like structure when coordinated with K(+) and Ba(2+), which induces an efficient overlap of the two hydroxyquinoline rings. Such overlapping forms a pseudo second macroring that results in a significant increase in both complex stability and cation selectivity.  相似文献

The metabolism of 1-(2-methyl-4-methoxyphenyl)-4-[(3-hydroxypropyl)amino]-6-methyl-2,3-dihydropyrrolo[3,2c]quinoline (DBM-819), a new H(+)/K(+) ATPase inhibitor, has been studied by HPLC with spectrometric detection and on-line LC-electrospray mass spectrometry. In vitro incubation of DBM-819 with rat liver microsomes in the presence of NADPH resulted in the production of four metabolites (M1-4), whereas DBM-819 was oxidized to two metabolites, M2 and M4, by human liver microsomes. M2, M3 and M4 were identified as O-demethyl-DBM-819, 8-hydroxy-DBM-819 and N-dehydroxypropyl-DBM-819, respectively, based on LC/MS/MS analysis with authentic standards. M1 was tentatively identified as 1-(hydroxy-2-methyl-4-methoxyphenyl)-4-[(3-hydroxypropyl)amino]-6-methyl-2,3-dihydropyrrolo[3,2c]quinoline. Rat liver CYP1A1/2 catalyzed the oxidation of DBM-819 to 8-hydroxy-DBM-819 and N-dehydroxypropyl-DBM-819. Human CYP3A4 was a major isozyme for the formation of O-demethyl-DBM-819 as well as N-dehydroxypropyl-DBM-819.  相似文献

Cation interactions with π-systems are a problem of outstanding contemporary interest and the nature of these interactions seems to be quite different for transition and main group metal ions. In this paper, we have systematically analyzed the contrast in the bonding of Cu(+) and main group metal ions. The molecular structures and energetics of the complexes formed by various alkenes (A = C(n)H(2n), n = 2-6; C(n)H(2n- 2), n = 3-8 and C(n)H(2n + 2), n = 5-10) and metal ions (M = Li(+), Na(+), K(+), Ca(2+), Mg(2+), Cu(+) and Zn(2+)) are investigated by employing ab initio post Hartree-Fock (MP2/6-311++G**) calculations and are reported in the current study. The study, which also aims to evaluate the effect of the size of the alkyl portion attached to the π-system on the complexation energy, indicates a linear relationship between the two. The decreasing order of complexation energy with various metal ion-alkene complexes follows the order Zn(2+)-A > Mg(2+)-A > Ca(2+)-A > Cu(+)-A > Li(+)-A > Na(+)-A > K(+)-A. The increased charge transfer and the electron density at (3,-1) intermolecular bond critical point corroborates well with the size of the π-system and the complexation energy. The observed deviation from the linear dependency of the Cu(+)-A complexes is attributed to the dπ→π* back bonding interaction. An energy decomposition analysis via the reduced variational space (RVS) procedure was also carried out to analyze which component among polarization, charge transfer, coulomb and exchange repulsion contributes to the increase in the complexation energy. The RVS results suggest that the polarization component significantly contributes to the increase in the complexation energy when the alkene size increases.  相似文献

Small hydrocarbon complexes (X@cage) incorporating cage-centered endohedral atoms and ions (X = H(+), H, He, Ne, Ar, Li(0,+), Be(0,+,2+), Na(0,+), Mg(0,+,2+)) have been studied at the B3LYP/6-31G(d) hybrid HF/DFT level of theory. No tetrahedrane (C(4)H(4), T(d)()) endohedral complexes are minima, not even with the very small hydrogen atom or beryllium dication. Cubane (C(8)H(8), O(h)()) and bicyclo[2.2.2]octane (C(8)H(14), D(3)(h)()) minima are limited to encapsulating species smaller than Ne and Na(+). Despite its intermediate size, adamantane (C(10)H(16), T(d)()) can enclose a wide variety of endohedral atoms and ions including H, He, Ne, Li(0,+), Be(0,+,2+), Na(0,+), and Mg(2+). In contrast, the truncated tetrahedrane (C(12)H(12), T(d)()) encapsulates fewer species, while the D(4)(d)() symmetric C(16)H(16) hydrocarbon cage (see Table of Contents graphic) encapsulates all but the larger Be, Mg, and Mg(+) species. The host cages have more compact geometries when metal atoms, rather than cations, are inside. This is due to electron donation from the endohedral metals into C-C bonding and C-H antibonding cage molecular orbitals. The relative stabilities of endohedral minima are evaluated by comparing their energies (E(endo)) to the sum of their isolated components (E(inc) = E(endo) - E(cage) - E(x)) and to their exohedral isomer energies (E(isom) = E(endo) - E(exo)). Although exohedral binding is preferred to endohedral encapsulation without exception (i.e., E(isom) is always exothermic), Be(2+)@C(10)H(16) (T(d)(); -235.5 kcal/mol), Li(+)@C(12)H(12) (T(d)(); 50.2 kcal/mol), Be(2+)@C(12)H(12) (T(d)(); -181.2 kcal/mol), Mg(2+)@C(12)H(12) (T(d)(); -45.0 kcal/mol), Li(+)@C(16)H(16) (D(4)(d)(); 13.3 kcal/mol), Be(+)@C(16)H(16) (C(4)(v)(); 31.8 kcal/mol), Be(2+)@C(16)H(16) (D(4)(d)(); -239.2 kcal/mol), and Mg(2+)@C(16)H(16) (D(4)(d)(); -37.7 kcal/mol) are relatively stable as compared to experimentally known He@C(20)H(20) (I(h)()), which has an E(inc) = 37.9 kcal/mol and E(isom) = -35.4 kcal/mol. Overall, endohedral cage complexes with low parent cage strain energies, large cage internal cavity volumes, and a small, highly charged guest species are the most viable synthetic targets.  相似文献

Erythrocyte membranes prepared by three different procedures showed (Mg2+ + Ca2+)-ATPase activities differing in specific activity and in affinity for Ca2+. The (Mg2+ + Ca2+)-ATPase activity of the three preparations was stimulated to different extents by a Ca2+-dependent protein activator isolated from hemolysates. The Ca2+ affinity of the two most active preparations was decreased as the ATP concentration in the assay medium was increased. Lowering the ATP concentration from 2 mM to 2-200 microM or lowering the Mg:ATP ratio to less than one shifted the (Mg2+ + Ca2+)-ATPase activity in stepwise hemolysis membranes from mixed "high" and "low" affinity to a single high Ca2+ affinity. Membranes from which soluble proteins were extracted by EDTA (0.1 mM) in low ionic strength, or membranes prepared by the EDTA (1-10 mM) procedure, did not undergo the shift in the Ca2+ affinity with changes in ATP and MgCl2 concentrations. The EDTA-wash membranes were only weakly activated by the protein activator. It is suggested that the differences in properties of the (Mg2+ + Ca2+)-ATPase prepared by these three procedures reflect differences determined in part by the degree of association of the membrane with a soluble protein activator and changes in the state of the enzyme to a less activatable form.  相似文献

A series of 4-substituted 8-[(2-benzimidazolyl)sulfinylmethyl]-1,2,3,4-tetrahydroquinolin es was synthesized and examined for their (H+ + K+)adenosine triphosphatase (ATPase)-inhibitory and antisecretory activities against histamine-induced gastric acid secretion in rats. Many compounds tested were potent inhibitors of (H+ + K+)ATPase. Most compounds showed antisecretory activity. The antiulcer activity against water-immersion stress-induced gastric ulcer, aspirin-induced gastric ulcer and gastric necrosis induced by hydrochloric acid also were tested in the rat. Some of these compounds, in particular, 4-(N-allyl-N-methylamino)-1-ethyl-8-[(5-fluoro-6-methoxy-2-benzimidazoly l) sulfinylmethyl]-1-ethyl-1,2,3,4-tetrahydroquinoline (XVIIx) were found to have potent activity. Lithium is used (in the form of soluble salts) to treat bipolar disorder and has been considered as a possible drug in treating chronic neurodegenerative diseases such as Alzheimer's, Parkinson's, and Huntington's diseases. One of the proposed mechanisms of Li(+) action involves a competition between the alien Li(+) and native Mg(2+) for metal-binding sites and subsequent inhibition of key enzymes involved in specific neurotransmission pathways, but not vital Mg(2+) proteins in the cell. This raises the following intriguing questions: Why does Li(+) replace Mg(2+) only in enzymes involved in bipolar disorder, but not in Mg(2+) proteins essential to cells? In general, what factors allow monovalent Li(+) to displace divalent Mg(2+) in proteins? Specifically, how do the composition, overall charge, and solvent exposure of the metal-binding site as well as a metal-bound phosphate affect the selectivity of Li(+) over Mg(2+)? Among the many possible factors, we show that the competition between Mg(2+) and Li(+) depends on the net charge of the metal complex, which is determined by the numbers of metal cations and negatively charged ligands, as well as the relative solvent exposure of the metal cavity. The protein itself is found to select Mg(2+) over the monovalent Li(+) by providing a solvent-inaccessible Mg(2+)-binding site lined by negatively charged Asp/Glu, whereas the cell machinery was found to select Mg(2+) among other competing divalent cations in the cellular fluids such as Ca(2+) and Zn(2+) by maintaining a high concentration ratio of Mg(2+) to its biogenic competitor in various biological compartments. The calculations reveal why Li(+) replaces Mg(2+) only in enzymes that are known targets of Li(+) therapy, but not in Mg(2+) enzymes essential to cells, and also reveal features common to the former that differ from those in the latter proteins.  相似文献

A three-dimensional potential energy surface is developed to describe the structure and dynamical behavior of the Mg(+)-H(2) and Mg(+)-D(2) complexes. Ab initio points calculated using the RCCSD(T) method and aug-cc-pVQZ basis set (augmented by bond functions) are fitted using a reproducing kernel Hilbert space method [Ho and Rabitz, J. Chem. Phys. 104, 2584 (1996)] to generate an analytical representation of the potential energy surface. The calculations confirm that Mg(+)-H(2) and Mg(+)-D(2) essentially consist of a Mg(+) atomic cation attached, respectively, to a moderately perturbed H(2) or D(2) molecule in a T-shaped configuration with an intermolecular separation of 2.62 A? and a well depth of D(e) = 842 cm(-1). The barrier for internal rotation through the linear configuration is 689 cm(-1). Interaction with the Mg(+) ion is predicted to increase the H(2) molecule's bond-length by 0.008 A?. Variational rovibrational energy level calculations using the new potential energy surface predict a dissociation energy of 614 cm(-1) for Mg(+)-H(2) and 716 cm(-1) for Mg(+)-D(2). The H-H and D-D stretch band centers are predicted to occur at 4059.4 and 2929.2 cm(-1), respectively, overestimating measured values by 3.9 and 2.6 cm(-1). For Mg(+)-H(2) and Mg(+)-D(2), the experimental B and C rotational constants exceed the calculated values by ~1.3%, suggesting that the calculated potential energy surface slightly overestimates the intermolecular separation. An ab initio dipole moment function is used to simulate the infrared spectra of both complexes.  相似文献

The effect of regucalcin, a calcium-binding protein isolated from rat liver cytosol, on Ca2+-adenosine triphosphatase (ATPase) activity in hepatic microsomes was investigated. Mg2+-ATPase activity was clearly increased by the presence of 50 microM Ca2+. Regucalcin (1.0-4.0 microM) caused a remarkable elevation (about 3-fold) of Ca2+-ATPase activity. Also, Mg2+-ATPase activity was increased (about 1.6-fold) by the presence of regucalcin (2.0 and 4.0 microM). Guanosine-5'-O-(3-thiotriphosphate) (GTPrs; 10(-5) and 10(-4) M) and nicotinamide adenine dinucleotide phosphate oxidized form (NADP+; 10(-5) to 10(-3) M) or reduced form (NADPH; 10(-4) and 10(-3) M) significantly increased Ca2+-ATPase activity. These increases were not enhanced by the presence of regucalcin (2.0 microM). Of various metal ions, a comparatively low concentration of V5+ (10(-5) M) or Cd2+ (10(-6) M) significantly increased Ca2+-ATPase activity, while Hg2+, Zn2+, Cu2+ and Mn2+ did not have such an effect. Regucalcin (2.0 microM) did not enhance the effect of V5+ and Cd2+ on Ca2+-ATPase activity. The present finding, that regucalcin activates hepatic microsomal Ca2+-ATPase, suggests a cell physiological role of regucalcin as an activator in the microsomal Ca2+-pump activity. This action of regucalcin may not be influenced by other regulators.  相似文献

A series of new diethylenetriaminepentaacetic acid (DTPA)-bisamide chelators has been prepared and characterized for application as zinc sensors. We have designed and synthesized (GdL(a))(2-), which contains a DTPA-bisamide moiety. The R(1) relaxivity of (GdL(a))(2-) solution decreased monotonically on the addition of Zn(2+). Moreover, (GdL(a))(2-) showed high selectivity for Zn(2+) against Ca(2+) and Mg(2+). We also measured the UV-visible spectra and the coldspray ionization (CSI) MS spectra and concluded that the 1-to-1 Zn(2+) complex of (GdL(a))(2-) is stable at higher concentrations of Zn(2+). These complexes should provide the basis for creating a superior Zn(2+)-sensitive MRI contrast agent and are excellent candidates for incorporation into sensors designed for selective detection of Zn(2+) in biological applications.  相似文献

Ultrasound is a special physical stimulus that has a variety of biological effects. This study provides a first systemic investigation on the ultrasound-induced oxidation and protection actions of the antioxidant defense system in Porphyridium cruentum. The hydroxyl radical and superoxide anion radical scavenging ability of the cells and the electrolyte leakage of the cell membrane were examined. The change of glutathione and carotenoids produced with/without ultrasonic processing were measured; the enzyme activities of superoxide dismutase, catalase, and membrane bound ATPases (Na(+)/K(+)-ATPase, Ca(2+)/Mg(2+)-ATPase) were evaluated for either ultrasound-treated or untreated P. cruentum. The hydroxyl radical and superoxide anion radical scavenging ability of ultrasound-treated P. cruentum increase 49.8 and 76.0%, respectively, of which the electrolyte leakage and malonyldialdehyde accumulation are also found increased 48.6 and 48.0%, respectively, indicating a state of oxidative stress. A significant enhancement of the activities of superoxide dismutase by 53.5%, catalase, membrane bound ATPases (Na(+)/K(+)-ATPase, Ca(2+)/Mg(2+)-ATPase increased by 67.7 and 69.3%, respectively), and the increment of glutathione and carotenoids production are also observed. These results suggested that oxidative stress manifested by elevated reactive oxygen species levels and malonyldialdehyde contents might be resulted from the biophysical responses of P. cruentum to the physical stimuli, and most likely the enhanced antioxidation ability of the algal cells stimuli by ultrasonic comes from the enhancement of enzymatic and nonenzymatic preventive substances as observed in this work.  相似文献

Reactions between Mg(+) and O(3), O(2), N(2), CO(2) and N(2)O were studied using the pulsed laser photo-dissociation at 193 nm of Mg(C(5)H(7)O(2))(2) vapour, followed by time-resolved laser-induced fluorescence of Mg(+) at 279.6 nm (Mg(+)(3(2)P(3/2)-3(2)S(1/2))). The rate coefficient for the reaction Mg(+) + O(3) is at the Langevin capture rate coefficient and independent of temperature, k(190-340 K) = (1.17 ± 0.19) × 10(-9) cm(3) molecule(-1) s(-1) (1σ error). The reaction MgO(+) + O(3) is also fast, k(295 K) = (8.5 ± 1.5) × 10(-10) cm(3) molecule(-1) s(-1), and produces Mg(+) + 2O(2) with a branching ratio of (0.35 ± 0.21), the major channel forming MgO(2)(+) + O(2). Rate data for Mg(+) recombination reactions yielded the following low-pressure limiting rate coefficients: k(Mg(+) + N(2)) = 2.7 × 10(-31) (T/300 K)(-1.88); k(Mg(+) + O(2)) = 4.1 × 10(-31) (T/300 K)(-1.65); k(Mg(+) + CO(2)) = 7.3 × 10(-30) (T/300 K)(-1.59); k(Mg(+) + N(2)O) = 1.9 × 10(-30) (T/300 K)(-2.51) cm(6) molecule(-2) s(-1), with 1σ errors of ±15%. Reactions involving molecular Mg-containing ions were then studied at 295 K by the pulsed laser ablation of a magnesite target in a fast flow tube, with mass spectrometric detection. Rate coefficients for the following ligand-switching reactions were measured: k(Mg(+)·CO(2) + H(2)O → Mg(+)·H(2)O + CO(2)) = (5.1 ± 0.9) × 10(-11); k(MgO(2)(+) + H(2)O → Mg(+)·H(2)O + O(2)) = (1.9 ± 0.6) × 10(-11); k(Mg(+)·N(2) + O(2)→ Mg(+)·O(2) + N(2)) = (3.5 ± 1.5) × 10(-12) cm(3) molecule(-1) s(-1). Low-pressure limiting rate coefficients were obtained for the following recombination reactions in He: k(MgO(2)(+) + O(2)) = 9.0 × 10(-30) (T/300 K)(-3.80); k(Mg(+)·CO(2) + CO(2)) = 2.3 × 10(-29) (T/300 K)(-5.08); k(Mg(+)·H(2)O + H(2)O) = 3.0 × 10(-28) (T/300 K)(-3.96); k(MgO(2)(+) + N(2)) = 4.7 × 10(-30) (T/300 K)(-3.75); k(MgO(2)(+) + CO(2)) = 6.6 × 10(-29) (T/300 K)(-4.18); k(Mg(+)·H(2)O + O(2)) = 1.2 × 10(-27) (T/300 K)(-4.13) cm(6) molecule(-2) s(-1). Vanadate stimulated the release of rat hepatic lipase activity from liver slices into an incubation medium in a time- and dose-dependent manner. Insulin, however, failed to have this stimulatory action, and the release by heparin was recognized, but was not additive to that by vanadate. Amiloride, an inhibitor of tyrosine kinase in some receptors and of the Na+/H+ exchange system suppressed the vanadate-stimulated release. Biochanin A, a different type of tyrosine kinase inhibitor than amiloride, also suppressed the effect of vanadate. The stimulation by vanadate was clearly preserved in Na(+)-, K(+)-, or Ca(2+)-free medium, suggesting that neither the Na+/H+ exchange system, Na+, K(+)-adenosine triphosphatase, nor Ca(2+)-influx into cells is involved in the action of this substance. These results suggest that vanadate-stimulated release of the enzyme activity is associated with the activation of the tyrosine kinase activity.  相似文献

Glycogen synthase kinase 3β (GSK3β) is a serine/threonine kinase that requires two cofactor Mg(2+) ions for catalysis in regulating many important cellular signals. Experimentally, Li(+) is a competitive inhibitor of GSK3β relative to Mg(2+), while this mechanism is not experienced with other group I metal ions. Herein, we use native Mg(2)(2+)-Mg(1)(2+) GSK3β and its Mg(2)(2+)-M(1)(+) (M = Li, Na, K, and Rb) derivatives to investigate the effect of metal ion substitution on the mechanism of inhibition through two-layer ONIOM-based quantum mechanics/molecular mechanics (QM/MM) calculations and molecular dynamics (MD) simulations. The results of ONIOM calculations elucidate that the interaction of Na(+), K(+), and Rb(+) with ATP is weaker compared to that of Mg(2+) and Li(+) with ATP, and the critical triphosphate moiety of ATP undergoes a large conformational change in the Na(+), K(+), and Rb(+) substituted systems. As a result, the three metal ions (Na(+), K(+), and Rb(+)) are not stable and depart from the active site, while Mg(2+) and Li(+) can stabilize in the active site, evident in MD simulations. Comparisons of Mg(2)(2+)-Mg(1)(2+) and Mg(2)(2+)-Li(1)(+) systems reveal that the inline phosphor-transfer of ATP and the two conserved hydrogen bonds between Lys85 and ATP, together with the electrostatic potential at the Li(1)(+) site, are disrupted in the Mg(2)(2+)-Li(1)(+) system. These computational results highlight the possible mechanism why Li(+) inhibits GSK3β.  相似文献
