Intrahepatic cholangiocarcinoma (ICC), a malignant tumor derived from the intrahepatic bile duct epithelium, has a poor prognosis and is refractory to conventional chemotherapy and radiation therapy. Thus, there is an urgent need to develop new effective therapeutic strategies for this disease. We previously found that L1 cell adhesion molecule (L1CAM) plays an important role in tumor progression of ICC, and we generated a murine mAb, A10-A3 (IgG1), that binds to the Ig1 domain of L1CAM. In the present study, we further characterized A10-A3, constructed a chimeric A10-A3 antibody (cA10-A3) containing the constant regions of human IgG1, and evaluated the therapeutic potential in a human ICC xenograft nude mice model. The affinities (KD) of A10-A3 and cA10-A3 for soluble L1CAM were 1.8 nM and 1.9 nM, respectively, as determined by competition ELISA. A10-A3 inhibited L1CAM homophilic binding and was slowly internalized into the tumor cells, but it did not significantly inhibit proliferation of ICC cells in vitro. cA10-A3 mediated antibody- dependent cell-mediated cytotoxicity in vitro and displayed anti-tumor activity in the ICC animal model. These results suggest that the humanized A10-A3 antibody may have potential as an anticancer agent for the treatment of ICC.  相似文献

Accurate static dipole polarizabilities and hyperpolarizabilities are calculated for the ground states of the Al, Si, P, S, Cl, and Ar atoms. The finite-field computations use energies obtained with various ab initio methods including Moller-Plesset perturbation theory and the coupled cluster approach. Excellent agreement with experiment is found for argon. The experimental alpha for Al is likely to be in error. Only limited comparisons are possible for the other atoms because hyperpolarizabilities have not been reported previously for most of these atoms. Our recommended values of the mean dipole polarizability (in the order Al-Ar) are alpha/e(2)a(0) (2)E(h) (-1)=57.74, 37.17, 24.93, 19.37, 14.57, and 11.085 with an error estimate of +/-0.5%. The recommended values of the mean second dipole hyperpolarizability (in the order Al-Ar) are gamma/e(4)a(0) (4)E(h) (-3)=2.02 x 10(5), 4.31 x 10(4), 1.14 x 10(4), 6.51 x 10(3), 2.73 x 10(3), and 1.18 x 10(3) with an error estimate of +/-2%. Our recommended polarizability anisotropy values are Deltaalpha/e(2)a(0) (2)E(h) (-1)=-25.60, 8.41, -3.63, and 1.71 for Al, Si, S, and Cl respectively, with an error estimate of +/-1%. The recommended hyperpolarizability anisotropies are Deltagamma/e(4)a(0) (4)E(h) (-3)=-3.88 x 10(5), 4.16 x 10(4), -7.00 x 10(3), and 1.65 x 10(3) for Al, Si, S, and Cl, respectively, with an error estimate of +/-4%.  相似文献

A series of five Fe(III) phosphonate clusters with four different topologies is reported. The choice of coligand carboxylate plays an important role in directing the structure of the molecule. [Fe9(O)4(O2CCMe3)13(C10P)3] (1) and [Fe9(O)2(OH)(CO2Ph)10(C10P)6(H2O)2](CH3CN)7 (2; camphyl phosphonic acid, C10H17PO3H2 = C10PH2) represent two unprecedented nonanuclear Fe(III) cages having Fe9O4 and Fe9(O)2(OH) core structures, respectively. Whereas [Fe6O2(O)2(O2CCMe3)8(C10P)2 (H2O)2](CH3CN)4 (3) is a peroxo-bridged hexameric compound with an Fe6(O)2(O2) core. [Fe4(O)(O2CCMe3)4(C10P)3(Py)4](CH3CN)3 (4) and [Fe4(O)(O2CPh)4(C10P)3(Py)4](Py)3(CH3CN)2 (5; Py = pyridine) represents two tetranuclear clusters with the same Fe4O core structure.  相似文献

Treatment of 3-methoxy-17β-acetoxyestra-2,5(10)-diene with perchloryl fluoride in aqueous dioxane or tetrahydrofuran yielded a 2-fluoro-3-ketosteroid with unusual properties. Unlike the known 2-fluoro-19-nortestosterone, this fluoroketone possessing a 5(10) double bond easily lost hydrogen fluoride to form estradiol 17-acetate; aromatization took place on melting, on exposure to pyridine or acid or even during chromatography on silica gel or Florisil. Because of the Δ-5(10) structure and the absence of the C19 Me group, it is difficult to infer the exact orientation of the 2-fluorine substituent from its effect on the NMR spectrum, CD or the CO absorption in the IR. However, the properties of certain derivatives suggested that the fluorine substituent was 2β. Treatment of the unsaturated fluorketone with peracid gave a mixture of epoxides which on basic hydrolysis furnished two 2-fluoro-10-hydroxy-17-acetoxyestr-4-en-3-ones epimeric at C10. The UV and NMR spectra of the 10-hydroxy epimer indicated an equatorial 2β-orientation of fluorine, whereas the 10β-hydroxy epimer showed an axial 2β-fluorine substituent which could be isomerized by acid to the more stable equatorial 2-configuration.  相似文献

Background

BTBD10 binds to Akt and protein phosphatase 2A (PP2A) and inhibits the PP2A-mediated dephosphorylation of Akt, thereby keeping Akt activated. Previous studies have suggested that BTBD10 plays an important role in preventing motor neuronal death and accelerating the growth of pancreatic beta cells. Because levels of BTBD10 expression are much lower in many non-nervous tissues than nervous tissues, there may be a relative of BTBD10 that has BTBD10-like function in non-neuronal cells.

Results

A 419-amino-acid BTBD10-like protein, named KCTD20 (potassium channel tetramerization protein domain containing 20), was to found to bind to all Akt isoforms and PP2A. Overexpression of KCTD20 increased Akt phosphorylation at Thr308, as BTBD10 did, which suggests that KCTD20 as well as BTBD10 positively regulates the function of Akt. KCTD20 was ubiquitously expressed in non-nervous as well as nervous tissues.

Conclusions

KCTD20 is a positive regulator of Akt and may play an important role in regulating the death and growth of some non-nervous and nervous cells.

The life time of aromatic radical cations is limited by reactions like β-elimination, dimerization, and addition to the solvent. Here we show that the attachment of such a radical cation to the C-terminal end of an α-/3(10)-helical peptide further reduces its life time by two orders of magnitude. For PPII-helical peptides, such an effect is only observed if the peptide contains an adjacent electron donor like tyrosine, which enables electron transfer (ET) through the peptide. In order to explain the special role of α-/3(10)-helical peptides, it is assumed that the aromatic radical cation injects a positive charge into an adjacent amide group. This is in accord with quantum chemical calculations and electrochemical experiments in the literature showing a decrease in the amide redox potentials caused by the dipole moments of long α-/3(10)-helical peptides. Rate measurements are in accord with a mechanism for a multi-step ET through α-/3(10)-helical peptides that uses the amide groups or H-bonds as stepping stones.  相似文献

The CH2Cl + CH3 (1) and CHCl2 + CH3 (2) cross-radical reactions were studied by laser photolysis/photoionization mass spectroscopy. Overall rate constants were obtained in direct real-time experiments in the temperature region 301-800 K and bath gas (helium) density (6-12) x 10(16) atom cm(-3). The observed rate constant of reaction 1 can be represented by an Arrhenius expression k1 = 3.93 x 10(-11) exp(91 K/T) cm3 molecule(-1) s(-1) (+/-25%) or as an average temperature-independent value of k1= (4.8 +/- 0.7) x 10(-11) cm3 molecule(-1) s(-1). The rate constant of reaction 2 can be expressed as k2= 1.66 x 10(-11) exp(359 K/T) cm3 molecule(-1) s(-1) (+/-25%). C2H4 and C2H3Cl were detected as the primary products of reactions 1 and 2, respectively. The experimental values of the rate constant are in reasonable agreement with the prediction based on the "geometric mean rule." A separate experimental attempt to determine the rate constants of the high-temperature CH2Cl + O2 (10) and CHCl2 + O2 (11) reaction resulted in an upper limit of 1.2 x 10(-16) cm(3) molecule(-1) s(-1) for k10 and k11 at 800 K.  相似文献

This work reports that ammonia (NH(3)) can be used as an efficient co-reactant for tris(2,2'-bipyridine)ruthenium(II) (Ru(bpy)(3)(2+)) electrochemiluminescence (ECL) in ionic liquids (ILs), on the basis of which a signal-on ECL sensor for directly detecting gaseous NH(3) has been developed. The NH(3) ECL sensor has a very high sensitivity, with a detection limit of 10 ppt NH(3) (at signal-to-noise ratio of 3) without any preconcentration. The high sensitivity is mainly due to the zero ECL background of Ru(bpy)(3)(2+) in the ILs, strong co-reactant ECL activity of NH(3), and high solubility of NH(3) in imidazolium-based ILs. Additionally, the ECL sensor shows an excellent selectivity against common interfering gases and a wide linear response range from 10 ppt to 10 ppm.  相似文献

A rapid method was developed to identify and quantify the azide ion (N(3)(-)) in gastric fluid and urine. N(3)(-) in diluted biological fluids was reacted with NaAuCl(4) to produce Au(N(3))(2)(-), which was extracted with octanol. Five microliters of the extract were flow-injected into an electrospray ionization tandem mass spectrometric instrument. Quantification of N(3)(-) was performed by selected reaction monitoring of the product ion Au(N)(N(3))(-) at m/z 253, which was derived from the precursor ion Au(N(3))(2)(-) at m/z 281, using 50 μL of aqueous solution within 10 min. This method was found to be linear up to 10(-5) M, to have a limit of quantification of 10(-7) M, a limit of detection of 3.0?×?10(-8) M, and a coefficient of variation of ≦10% at 10(-7) M. In the case of urine, 50 μL of urine were spiked with N(3)(-), this was diluted 10-fold and passed through 1 mL of a resin, and finally diluted to 100-fold of the original. This method was linear up to 10(-3) M, had a limit of quantification of 10(-5) M, a limit of detection of 3.0?×?10(-6) M, and coefficient of variation of ≦8.8% for an original urine concentration of 10(-5) M. The practical applicability of this method was checked by diluting 1 μL of a suspected suicide victim's gastric fluid 20,000-fold and 1 μL of the victim's urine 5,000-fold and then measuring the N(3)(-) levels. These levels were found to be (7.5?±?1.0)?×?10(-2) M and (3.2?±?0.4)?×?10(-3) M, respectively.  相似文献

The synthesis of N-methyl-3-aza[10]paracyclophane is reported which represents the first example of this ring system being formed via an acyloin reaction. This 3-aza[10]paraeyclophane ring system behaves physiochemically inbetween the normal [9]- and [10]paracyclophane ring systems. Reductive desulfurization of N-methyl-3-aza[10]paracyclophane-6-ethylene thioketal in ethanol provides a small amount of the title compound and an unexpected, ring-opened product, N-ethyl-N-methyl-p-heptylphenethylamine. A possible mechanism for the ring-opening process is suggested.  相似文献

Convenient, high-yield routes have been developed to [Fe 10(OMe) 20(O 2CR) 10] ( 1) "ferric wheels" involving the alcoholysis of [Fe 3O(O 2CR) 6(H 2O) 3] (+) salts in MeOH in the presence of NEt 3. Reactivity studies have established [Fe 10(OMe) 20(O 2CMe) 10] ( 1a) to undergo clean carboxylate substitution with a variety of other RCO 2H groups to the corresponding [Fe 10(OMe) 20(O 2CR) 10] product. In contrast, the reaction with phenol causes a nuclearity change to give a smaller [Fe 8(OH) 4(OPh) 8(O 2CR) 12] ( 2) wheel. Similarly, reactions of [Fe 10(OMe) 20(O 2CR) 10] with the bidentate chelate ethylenediamine (en) cause a structural change to give either [Fe 8O 5(O 2CMe) 8(en) 8](ClO 4) 6 ( 3) or [Fe 2O(O 2CBu (t))(en) 4](NO 3) 3 ( 4), depending on conditions. Complex 3 possesses a "Christmas-star" Fe 8 topology comprising a central planar [Fe 4(mu 4-O)] (10+) square subunit edge-fused to four oxide-centered [Fe 3(mu 3-O)] (7+) triangular units. Variable-temperature, solid-state dc and ac magnetization studies on complexes 1a- 4 in the 5.0-300 K range established that all the complexes possess an S = 0 ground state. The magnetic susceptibility data for 4 were fit to the theoretical chi M versus T expression derived by the use of an isotropic Heisenberg spin Hamiltonian and the Van Vleck equation, and this revealed an antiferromagnetic exchange parameter with a value of J = -107.7(5) cm (-1). This value is consistent with that predicted by a previously published magnetostructural relationship. Theoretically computed values of the exchange constants in 3 were obtained with the ZILSH method, and the pattern of spin frustration within its core and the origin of its S = 0 ground state have been analyzed in detail.  相似文献

Carbon---hydrogen bond cleavage at the terminal 6-position occurs when hex-5-en-2-one (CH2=CHCH2CH2COMe) oxidatively adds to [Os3(CO)10(MeCN)2] to give [Os3H(μ-CH=CHCH2CH2COMe)(CO)10], which is completely analogous to the simple vinyl complex [Os3H(μ-CH=CH2)(CO)10]. A minor product from the reaction is [Os3(CH3CH=CHCH2COMe)(CO)10], an isomer in which double-bond migration has occurred to give the βγ-unsaturated ketone; stabilisation occurs through chelation and ketone coordination. [Os3H2(CO)10] reacts with CH2=CHCH2CH2COMe in refluxing cyclohexane to give a third isomer, [Os3H(CH3CH2C=CHCOMe)(CO)10], in which further double bond migration has occurred to give the β-unsaturated ketone. Metallation at the β-site gives an Os---C bond as part of a 5-membered chelate ring. Thermolysis of each of the three isomeric decarbonyl species in refluxing cyclohexane or heptane leads to the elimination of an Os(CO)4 group to give the dinuclear compound [Os2H(EtC=CHCOMe)(CO)6] in varying yield. Pathways from γδ to the βγ and finally the β unsaturated ketones may be mapped out.  相似文献

The interaction between the osmium clusters [Os3(CO)12], [Os3(CO)10(mu-H)2], [Os3(CO)10(mu-H)(mu-OH)], and a series containing a free functional group, viz., [Os3(CO)10(mu-H)(mu-S--EH)] (where -- =alkylwedge chain or an aromatic ring, E=COO, S or O), with SiO2, ZnO and In2O3, was examined by ToF-SIMS and IR spectroscopy. To confirm how different anions influence sup-ramolecular self-assembly of lanthanide-organic frameworks (LnOFs) as well as their luminescent properties, a new flexible achiral tripodal ligand, 1,1,1-tris-{[(2'-benzylaminoformyl)phenoxyl]methyl}ethane (L) and the LnOFs {[EuL(NO(3))(3)]·1.5CHCl(3)}(n) and [EuL(pic)(3)](n) have been designed and assembled. In the two LnOFs, {[EuL(NO(3))(3)]·1.5CHCl(3)}(n) demonstrates an unprecedented chiral noninterpenetrated two-dimensional (2D) honeycomblike (6,3) (hcb, Schla?fli symbol 6(3), vertex symbol 6·6·6) topological network, and [EuL(pic)(3)](n) confirms an unusual chiral LnOF with three-dimensional (3D) (10,3)-a (srs, SrSi(2), Schla?fli symbol 10(3), vertex symbol 10(2)·10(4)·10(4)) topological framework. Also the anion-induced structures and energy transfer processes in the luminescence behavior of the two LnOFs were discussed in detail.  相似文献

The rhenacarborane salt Cs[Re(CO)3(eta5-7,8-C2B9H11)] (1) has been used to synthesize the tetranuclear metal complex [[ReAg(mu-10-H-eta5-7,8-C2B9H10)(CO)3]2[mu-Ph2P(CH2)2PPh2]] (3) where two [ReAg(mu-10-H-eta5-7,8-C2B9H10)(CO)3] fragments have been shown by X-ray crystallography to be bridged by a single 1,2-bis(diphenylphosphino)ethane ligand. Reaction of 1 with Ag[BF4] in the presence of the ligands bis- or tris(pyrazol-1-yl)methane yields the complexes [ReAg(mu-10-H-eta5-7,8-C2B9H10)(CO)3[kappa2-CH2(C3H3N2-1)2]] (4) or [[ReAg(mu-10-H-eta5-7,8-C2B9H10)(CO)3]2[mu-kappa1,kappa2-CH(C3H3N2-1)3]] (5), respectively. From X-ray studies, the former comprises a Re-Ag bond bridged by the carborane cage and with the bis(pyrazol-1-yl)methane coordinating the silver(I) center in an asymmetric kappa(2) mode. Complex 5 was unexpectedly found to contain a tris(pyrazol-1-yl)methane bridging two [ReAg(mu-10-H-eta5-7,8-C2B9H10)(CO)3] fragments in a kappa1,kappa2 manner. Treatment of 1 with Ag[BF4] in the presence of 2,2'-dipyridyl and 2,2':6',2' '-terpyridyl yields [ReAg(mu-10-H-eta5-7,8-C2B9H10)(CO)3[kappa2-(C5H4N-2)(2)]] (6) and [ReAg(mu-10-H-eta5-7,8-C2B9H10)(CO)3[kappa3-C5H3N(C5H4N-2)2-2,6]] (7). The X-ray structure determination of 7 revealed an unusual pentacoordinated silver(I) center, asymmetrically ligated by a kappa3-2,2':6',2' '-terpyridyl molecule. The same synthetic procedure using N,N,N',N'-tetramethylethylenediamine gave a tetranuclear metal complex [[ReAg(mu-10-H-eta5-7,8-C2B9H10)(CO)3]2[mu-Me2N(CH2)2NMe2]2] (8) which is believed, in the solid state, to be bridged between the silver atoms by two of the diamine molecules. The salt 1 with Ag[BF4] in the absence of any added ligand gave the tetrameric cluster [ReAg[mu-5,6,10-(H)3-eta5-7,8-C2B9H8](CO)3]4 (9) where, in the solid state, four [ReAg(mu-10-H-eta5-7,8-C2B9H10)(CO)3] units are held together by long interunit B-H right harpoon-up Ag bonds.  相似文献

A PVC membrane electrode for aluminium ion based on bis(5-phenyl azo salicylaldehyde) 2,3-naphthalene diimine (5PHAZOSALNPHN) as an ion carrier was developed. The electrode exhibits a Nernstian slope of 19.3+/-0.8 mV per decade and a linear range of 5.0x10(-6)-1.0x10(-2) M for Al(NO(3))(3). The limit of detection is 2.5x10(-6) M. It has a fast response time of about 10 s and can be used for at least 10 weeks without observing any deviation. The proposed membrane sensor revealed good selectivity for Al(3+) over a wide variety of other metal ions and could be used in pH range of 2.9-5.0. It was used as an indicator electrode in potentiometric titration of aluminium ion.  相似文献
