研制了紫外、可见、近红外光谱区通用光谱透射比较标准物质。以高纯熔融石英为基片材料,镍铬合金的镀膜材料,采用双光楔对称光胶结构以消除同色杂散光的影响,对镀膜进行光胶处理以保证膜层的强度、稳定性及光学中性,采用高精度分光光度测量装置作为定值手段的以保证准确的定值,设计定位保护架以消除多次反射误差。该光度标准适用的光谱范围的200-2600nm,光谱透射比定值不确定度为0.5%。  相似文献

采用重量-容量法制备丙酮中除虫脲溶液标准物质。准确称量国家二级标准物质除虫脲溶解到色谱纯丙酮中,通过A级容量瓶定容至500 mL,摇匀后分装到2 mL安瓿瓶内共460瓶,每瓶为1 mL,保持低温迅速封口,配制过程中室温保持在(20±2)℃。采用液相色谱法进行均匀性、稳定性检验和定值结果验证。从样品中随机抽取16瓶进行均匀性检验,经F检验表明,在95%的置信区间范围内该标准物质均匀性良好;采用t检验对标准物质稳定性进行检验,标准物质在12月内稳定性良好。对该标准物质的不确定度进行了评定,研制的丙酮中除虫脲溶液标准物质定值结果为100μg/mL,相对扩展不确定度为2%(k=2)。结果表明,该标准物质均匀性与稳定性良好,量值准确,可用于日常分析检测中的方法评价和仪器校准。  相似文献

大豆蛋白在各领域的应用已得到广泛的关注,因此大豆蛋白及其改性材料在结构性能方面的研究显得越来越重要。中红外光谱(mid-infrared spectroscopy,MIR)和近红外光谱(near-infrared spectroscopy,NIR)正是对蛋白质进行定性定量分析的有力手段。中红外光谱可以有效地分析大豆蛋白在溶液和薄膜中的二级结构以及大豆衍生材料内蛋白质的结构变化情况。近红外光谱则在蛋白质定量分析方面有着独特的优势。本文介绍了运用这两种光谱技术进行研究的一些工作,这些实例表明了中红外和近红外光谱在大豆蛋白研究领域的重要应用价值。  相似文献

采用称量法制备并计算定值,研制(10~5000)μmol.mol-1氮中二氧化氮气体标准物质。考察了气体标准物质随贮存时间和钢瓶压力变化的稳定性,将制备的气体标准物质与GBW 08180进行比对验证,结果表明研制的气体标准物质定值的扩展不确定度优于3%(k=3),有效期限为12个月。  相似文献

介绍甲醇中甲苯溶液标准物质的制备方法。以重量–容量法制备甲醇中甲苯溶液标准物质,以计算值作为标准物质的特性量值。采用气相色谱法、卡尔·费休法、电感耦合等离子体质谱法等分别对原料中的主成分、金属元素及水分含量准确定量。方差分析和线性拟合结果表明,该溶液标准物质具有良好的均匀性和稳定性。甲醇中甲苯溶液标准物质特性值为996.73μg/mL,相对扩展不确定度为2%(k=2)。  相似文献

以藏药诃子为原料,研制诃黎勒酸国家标准样品. 诃子经60%乙醇提取,大孔树脂富集,制备液相色谱技术得到诃黎勒酸标准品. 采用高效液相色谱(HPLC)、液相色谱-质谱(LC-MS)和薄层色谱(TLC)等手段进行纯度分析. 采用紫外光谱(UV)、红外光谱(IR)、高分辨质谱(MS)和核磁共振波谱(NMR)技术进行结构确认. 最后进行均匀性、稳定性和联合定值. 结果表明,诃黎勒酸样品均匀性良好,4 ℃储存,24个月内稳定. 联合定值确定诃黎勒酸纯度标准值为99.33%,符合国家标准品的要求. 研制出的诃黎勒酸(GSB 11-3724-2020)国家标准样品,可用于含量测定、检测方法评定、相关产品的检测与质量控制.  相似文献

A suite of three ginkgo-containing dietary supplement Standard Reference Materials (SRMs) has been issued by the National Institute of Standards and Technology (NIST) with certified values for flavonoid aglycones, ginkgolides, bilobalide, and selected toxic trace elements. The materials represent a range of matrices (i.e., plant, extract, and finished product) that provide different analytical challenges. The constituents have been determined by at least two independent analytical methods with measurements performed by NIST and at least one collaborating laboratory. The methods utilized different extractions, chromatographic separations, modes of detection, and approaches to quantitation. The SRMs are primarily intended for method validation and for use as control materials to support the analysis of dietary supplements and related botanical materials. As part of a collaboration with the National Institutes of Health’s Office of Dietary Supplements and the Food and Drug Administration’s Center for Drug Evaluation and Research, the National Institute of Standards and Technology has developed two standard reference materials (SRMs) representing different forms of saw palmetto (Serenoa repens), SRM 3250 Serenoa repens fruit and SRM 3251 Serenoa repens extract. Both of these SRMs have been characterized for their fatty acid and phytosterol content. The fatty acid concentration values are based on results from gas chromatography with flame ionization detection (GC-FID) and mass spectrometry (GC/MS) analysis while the sterol concentration values are based on results from GC-FID and liquid chromatography with mass spectrometry analysis. In addition, SRM 3250 has been characterized for lead content, and SRM 3251 has been characterized for the content of β-carotene and tocopherols. SRM 3250 (fruit) has certified concentration values for three phytosterols, 14 fatty acids as triglycerides, and lead along with reference concentration values for four fatty acids as triglycerides and 16 free fatty acids. SRM 3251 (extract) has certified concentration values for three phytosterols, 17 fatty acids as triglycerides, β-carotene, and γ-tocopherol along with reference concentration values for three fatty acids as triglycerides, 17 fatty acids as free fatty acids, β-carotene isomers, and δ-tocopherol and information values for two phytosterols. These SRMs will complement other reference materials currently available with concentrations for similar analytes and are part of a series of SRMs being developed for dietary supplements. Well-characterized reference materials are needed by laboratories in the food testing and nutrition communities to facilitate compliance with nutritional labeling laws, to provide traceability for food exports needed for acceptance in many foreign markets, and to improve the accuracy of nutrition information that is provided to assist consumers in making sound dietary choices. As a result of the enactment of the Nutrition Labeling and Education Act of 1990 and the Infant Formula Act of 1980, the National Institute of Standards and Technology (NIST) has developed a suite of food-matrix Standard Reference Materials (SRMs) characterized for nutrient concentrations. These include SRM 1544 Fatty Acids and Cholesterol in a Frozen Diet Composite, SRM 1546 Meat Homogenate, SRM 1548a Typical Diet, SRM 1566b Oyster Tissue, SRM 1846 Infant Formula, SRM 1946 Lake Superior Fish Tissue, SRM 2383 Baby Food Composite, SRM 2384 Baking Chocolate, SRM 2385 Spinach, and SRM 2387 Peanut Butter. Many of these materials were developed at the request of the food industry to populate a nine-sectored fat-protein-carbohydrate triangle developed by AOAC International. With the completion of SRM 2387, SRMs representing each sector of the triangle are now available. These food-matrix reference materials are intended primarily for validation of analytical methods for the measurement of proximates, fatty acids, vitamins, minerals, and so on in foods of similar composition. They may also be used as "primary control materials" in the value-assignment of in-house, secondary, control materials to confirm accuracy as well as to establish traceability to NIST.  相似文献

With the emergence of highly sensitive analytical techniques, the microanalysis of natural-matrix materials employing smaller sample sizes is increasingly more common, which subsequently warrants a homogeneity assessment for the individual components at the appropriate sampling level. Pressurized liquid extraction (PLE) in combination with gas chromatography/mass spectrometry (GC/MS) has been used to determine the sampling constants and evaluate the relative homogeneity of trace levels of polycyclic aromatic hydrocarbons (PAHs) for two previously certified particulate standard reference materials, SRM 1649a Urban Dust and SRM 1650b Diesel Particulate Matter, in the milligram sampling range. Fluoranthene, pyrene, benz[a]anthracene and benzo[e]pyrene within SRM 1650b Diesel Particulate Matter were deemed to be homogeneous, based on relatively small sampling constants (K S<100 mg), whereas the larger sampling constants (K S>100 mg) obtained for all PAHs in SRM 1649a Urban Dust suggest more material heterogeneity. The material heterogeneity of ten individual PAHs (phenanthrene, anthracene, fluoranthene, pyrene, benz[a]anthracene, benzo[k]fluoranthene, benzo[e]pyrene, benzo[a]pyrene, indeno[1,2,3-cd]pyrene and benzo[ghi]perylene) was also described via nonlinear relationships (i.e., power law) between subsampling error S s (%) and sample mass, which are used to predict analyte-specific minimum sample masses that result in a specific level of analytical uncertainty. Two new standard reference materials (SRM) for drugs of abuse in human hair have been developed. SRM 2379 consists of hair spiked with cocaine, benzoylecgonine, cocaethylene, phencyclidine, amphetamine, and methamphetamine. SRM 2380 consists of hair spiked with codeine, morphine, monoacetylmorphine, and tetrahydrocannabinol (THC). The SRMs were prepared by soaking the hair in a solution of the target analytes in water-dimethylsulfoxide. The concentration of each analyte was determined using two methods, one based upon gas chromatography/mass spectrometry (GC/MS) and one based upon liquid chromatography/mass spectrometry (LC/MS). Both methods used 0.1 M HCl for extraction of all the analytes from the hair, except for THC, which was extracted with 1 M NaOH. For isolation of the analytes from the extracts, the GC/MS-based methods used different clean-up procedures from those used for the LC/MS-based methods. The results from the two methods were in good agreement with mean differences for the analytes ranging from 4% to 16%. These materials will enable laboratories performing analyses of hair for drugs of abuse to test the accuracy of their methods.  相似文献

The National Institute of Standards and Technology (NIST), the Food and Drug Administration (FDA) Center for Drug Evaluation and Research (CDER) and Center for Food Safety and Applied Nutrition (CFSAN), and the National Institutes of Health (NIH), Office of Dietary Supplements (ODS) are collaborating to produce a series of standard reference materials (SRMs) for dietary supplements. Standard reference material (SRM) 3276 Carrot Extract in Oil is one in this series, with values assigned for trans-alpha-carotene, trans- and total beta-carotene, delta- and gamma-tocopherol, and twelve fatty acids. Results for carotenoids and tocopherols were obtained by use of combinations of liquid chromatography (LC), on columns of differing selectivity, with absorbance and mass spectrometric (MS) detection. Fluorescence detection also was used for the tocopherols. Results for fatty acids were obtained by use of gas chromatography (GC) with both flame-ionization and mass-spectrometric detection. This material is intended for use as a primary control material when assigning values to in-house (secondary) control materials and for validation of analytical methods for measurement of these analytes in similar matrices.  相似文献
