Alkylphosphonic acids (APAs) are degradation products and chemical markers of organophosphorous (OP) nerve agents (chemical warfare agents). Anion exchange disk-based solid phase extraction (SPE) has been combined with in vial solid phase derivatization (SPD) and GC–MS analysis for the determination of APAs in aqueous samples. The optimization of critical method parameters, such as the SPD reaction, was achieved using statistical experimental design and multivariate data analysis. The optimized method achieved quantitative recoveries in the range from 83% to 101% (n = 13, RSD from 4% to 10%). The method was sensitive, with LODs in SIM mode of 0.14 ppb, and demonstrated excellent linearity with an average R2 ≥ 0.99 over the concentration range of 0.07–1.4 ppm in full scan mode and from 0.14 ppb to 14 ppb in SIM mode. For forensic applications, aqueous samples containing APAs at concentrations exceeding 14 ppb were concentrated and target analytes were successfully identified by spectral library and retention index matching. Method robustness was evaluated using aqueous samples from the official OPCW Proficiency Test (round 19) and all APAs present in the sample were conclusively identified. The SPE disk retained the underivatized APAs in a stable condition for extended periods of time. No significant losses of APAs from the disk were observed over a 36-day period. Overall, the method is well suited to the qualitative and quantitative analysis of degradation markers of OP nerve agents in aqueous matrices with simplicity, a low risk of cross-contamination and trace level sensitivity.  相似文献

建立了高效液相色谱同时测定化妆品中12种紫外吸收剂含量的方法。样品用甲醇提取,高速离心,过滤,以SB-C8柱(250 mm×4.6 mm, 5 μm)为分离色谱柱,甲醇和0.1%(v/v)甲酸水溶液为流动相,梯度洗脱,以311 nm为检测波长进行定性、定量分析。该方法前处理简单、易操作,12种紫外吸收剂分离效果良好;在1.0~500 mg/L范围内呈线性关系,相关系数大于0.9995;方法检出限为0.002~0.1 mg/L;实际样品中的加标回收率为97.4%~107.5%,相对标准偏差为1.54%~4.98%。该方法简便、准确,能够满足化妆品中紫外吸收剂的检测要求。  相似文献

A simple and rapid analytical method was developed for the determination of phthalates, usually employed in nail cosmetic products. The method is based on an ultrasonic extraction of the sample with ethanol-water (90:10, v/v) followed by HPLC separation and quantitation. HPLC was carried out using a C18 column and spectrophotometric detection at 254 nm. A linear gradient elution was performed with ethanol-water starting from 50 to 95% ethanol in 30 min. Standard calibration curves were linear for all the analytes over the concentration range 5-200 μg ml−1 with LOD values of about 0.5 μg ml−1.The proposed green analytical method has been successfully applied for the analysis of commercial samples in order to check the presence of phthalates and to determine their concentration.  相似文献

In this work, a non-chromatographic procedure for the on-line determination of ultratraces of V(V) and V(IV) is presented. The method involves a solid phase extraction-flow injection system coupled to electrothermal atomic absorption spectrometry (SPE-FI-ETAAS). The system holds two microcolumns (MC) set in parallel and filled with lab-made mesoporous silica functionalized with 3-aminopropyltriethoxy silane (APS) and mesoporous silica MCM-41, respectively. The pre-concentration of V(V) is performed by sorption onto the first MC (C1) filled with APS at pH 3, whilst that of V(IV) is performed by sorption onto the second column (C2) filled with mesoporous silica MCM-41 at pH 5. Aqueous samples containing both analytes are loaded and, after pre-concentration (pre-concentration factor PCF = 10, sorption flow rate = 1 mL min−1, sorption time = 10 min), they are eluted in separate vessels with hydroxylammonium chloride (HC) 0.1 mol L−1 in HCl 0.5 mol L−1 (elution volume = 1 mL, elution flow rate = 0.5 mL min−1). Afterwards, both analytes are determined through ETAAS with graphite furnace. Under optimized conditions, the main analytical figures of merit for V(V) and V(IV) are, respectively: detection limits (3 s): 0.5 and 0.6 μg L−1, linear range: 2-100 μg L−1 (both analytes), sensitivity: 0.015 and 0.013 μg−1 L and sample throughput: 6 h−1 (both analytes). Recoveries of both species were assayed in different water samples. Validation was performed through certified reference materials for ultratraces of total vanadium in river water.  相似文献

Benzophenones (BPs) are a family of compounds widely used to protect the skin and hair from UV irradiation. Despite human exposure to BPs through dermal application of products containing sunscreen agents and the increasing evidence that BPs are able to interfere with endocrine systems, few studies have examined the occurrence of BPs in humans. In this work, we propose a new liquid chromatography-tandem mass spectrometry (LC-MS/MS) method to determine six BPs, namely, benzophenone-1 (BP-1), benzophenone-2 (BP-2), benzophenone-3 (BP-3), benzophenone-6 (BP-6), benzophenone-8 (BP-8) and 4-hydroxybenzophenone (4-OH-BP) in human placental tissue samples. The method involves an extraction step of the analytes from the samples using ethyl acetate, followed by a clean-up step using centrifugation prior to their quantification by LC-MS/MS using an atmospheric pressure chemical ionization (APCI) interface in the positive mode. Benzophenone-d10 (BP-d10) was used as surrogate. Found detection limits (LOD) ranged from 0.07 to 0.3 ng g−1 and quantification limits (LOQ) from 0.3 to 1.0 ng g−1, while inter- and intra-day variability was under 5%. The method was validated using standard addition calibration and a recovery assay. Recovery rates for spiked samples ranged from 98 to 104%. This method was satisfactorily applied for the determination of BPs in 16 placental tissue samples collected from women who live in Granada (Spain).  相似文献

A fully automated cold fiber solid phase microextraction device has been developed by coupling to a GERSTEL multipurpose (MPS 2) autosampler and applied to the analysis of volatiles and semi-volatiles in aqueous and solid matrices. The proposed device was thoroughly evaluated for its extraction performance, robustness, reproducibility and reliability by gas chromatograph/mass spectrometer (GC/MS). With the use of a septumless head injector, the entire automated setup was capable of analyzing over 200 samples without any GC injector leakages. Evaluation of the automated cold fiber device was carried out using a group of compounds characterized by different volatilities and polarities. Extraction efficiency as well as analytical figures of merit was compared to commercial solid phase microextraction fibers. The automated cold fiber device showed significantly improved extraction efficiency compared to the commercial polydimethylsiloxane (PDMS) and cold fiber without cooling for the analysis of aqueous standard samples due to the low temperature of the coating. Comparing results obtained from cold fiber and commercial divinylbenzene/carboxen/polydimethylsiloxane (DVB/CAR/PDMS) fiber temperature profile demonstrated that the temperature gap between the sample matrix and the coating improved the distribution coefficient and therefore the extraction amount. The linear dynamic range of the cold fiber device was 0.5 ng mL−1 to 100 ng mL−1 with a linear regression coefficient ≥0.9963 for all compounds. The limit of detection for all analytes ranged from 1.0 ng mL−1 to 9.4 ng mL−1. The newly automated cold fiber device presents a platform for headspace analysis of volatiles and semi-volatiles for large number of samples with improved throughput and sensitivity.  相似文献

The development and performance evaluation of an analytical method dedicated to the comprehensive determination of the most relevant antioxidants and their metabolites in aqueous environmental samples is presented. This was achieved by a miniaturised solid-phase extraction (SPE) with 10 mg Oasis HLB cartridges, which allow to achieve a concentration factor of 200, reducing organic solvent wastes (1 mL of ethyl acetate suffices for complete elution) and SPE costs and eliminating the need for solvent evaporation that otherwise compromises the recoveries of butylated hydroxytoluene (BHT) and 2,6-di-tert-butylcyclohexa-2,5-diene-1,4-dione (BHT-Q). Analytes were then determined by gas chromatography–mass spectrometry (GC–MS) after derivatisation with N-methyl-N-(tert-butyldimethylsilyl)-trifluoroacetamide (MTBSTFA) in a single run. BHT-d7 and n-propyl-paraben-d4 (PrP-d4) were used as surrogate internal standards. These surrogates allowed obtaining relative recoveries in the 80–110% range for all analytes even with complex wastewater samples and LODs at the 2–44 ng L−1 level taking into account blank issues often associated to antioxidants analysis. The method was applied to sewage and river waters, showing that the seven analytes could be detected in raw wastewater. BHT and BHT-Q were the most concentrated species in that type of sample (in the 275–871 ng L−1 range). On the other hand two metabolites of BHT, 3,5-di-tert-butyl-4-hydroxybenzaldehyde (BHT-CHO) and 3,5-di-tert-butyl-4-hydroxybenzoic acid (BHT-COOH) appeared to be the most ubiquitous species, being found in all samples in the 10–150 ng L−1 concentration range.  相似文献

A novel method combining dispersive liquid-liquid microextraction (DLLME) and heart-cutting multidimensional gas chromatography coupled to mass spectrometry was developed for the determination of free and total bisphenol A (BPA) and bisphenol B (BPB) in human urine samples. The DLLME procedure combines extraction, derivatization and concentration of the analytes into one step. Several important variables influencing the extraction efficiency and selectivity such as nature and volume of extractive and dispersive solvents as well as the amount of acetylating reagent were investigated. The temperature and time to hydrolyze BPA and BPB conjugates with a β-glucuronidase and sulfatase enzyme preparation were also studied. Under the optimized conditions good efficiency extraction (71-93%) and acceptable total DLLME yields (56-77%) were obtained for both analytes. Matrix-matched calibration curves were linear with correlation coefficients higher than 0.996 in the range level 0.1-5 μg/l, and the relative standard deviations (%RSD) were lower than 20% (n = 6). The limits of detection were 0.03 and 0.05 μg/l for BPA and BPB, respectively. The applicability of the proposed method for determining urinary free and total BPA and BPB was assessed by analyzing the human urine of a group of 20 volunteers. Free BPA was detected in 45% of the sample whereas total BPA was detected in 85% of the samples at concentrations ranging between 0.39 and 4.99 μg/l. A capillary electrophoresis (CE) and a high performance liquid chromatography (HPLC) method are described for the simultaneous determination of ethylenediaminetetraacetic acid (EDTA), S,S′-ethylenediaminedisuccinic acid (EDDS) and R,S-iminodisuccinic acid (IDS) complexing agents as their Fe(III) complexes in cosmetics like shower cream and foam bath. The non-biodegradable EDTA is used in combination with biodegradable analogues like EDDS and IDS in many commercial products. The HPLC method involves separation by reversed-phase ion pair chromatography on a C18 column using methanol-formate buffer (20 mM tetrabutylammonium hydrogen sulfate, 15 mM sodium formate adjusted to pH 4.0 with formic acid) (10:90, v/v) as mobile solvent at a flow rate of 0.8 mL min−1 at 24 °C using UV detection at 240 nm. The CE separation was performed in a fused silica capillary of 50 μm i.d. with the total length of 50 cm with a 10 mM MES and MOPSO (pH 5.5) at an applied voltage of −25 kV. The samples were introduced by applying a 50 mbar pressure for 2 s. Absorbances at 215 and 225 nm were monitored for the detection of the complexes. The methodology performance of the two methods was evaluated in terms of linearity, limit of detection (LOD), limit of quantitation (LOQ) and reproducibility. The LOD values obtained from HPLC are low when compared with CE. The applicability of both the methods was demonstrated for the analysis of cosmetic products such as shower cream and foam bath. The results obtained by both CE and HPLC were found to be comparable and in good agreement.  相似文献

A gas chromatography method with atomic emission detection (GC-AED) for the determination of dimethylarsinic acid (DMA), monomethylarsonic acid (MMA) and inorganic arsenic was optimized. The analytes were derivatized in the sample solutions with methyl thioglycolate (TGM) and the products were extracted into cyclohexane before an aliquot of this organic phase was directly injected into the chromatograph. The procedure was applied to the analysis of seawaters, wines, beers and infant foods, the last requiring an additional enzymatic reaction prior to analyte derivatization. Detection limits in seawaters and beverages were 0.05, 0.15 and 0.8 ng mL−1 for DMA, MMA and inorganic arsenic, respectively. In infant foods the detection limits were 1, 10 and 25 ng g−1 for DMA, MMA and inorganic arsenic, respectively. Inorganic arsenic was detected in some of the seawater samples and three of the wines analyzed at concentration levels in the range 1-40 ng mL−1, and DMA in several of the infant foods in the range 20-80 ng g−1. The method was validated by analyzing a certified reference material and by recovery studies. All the samples were also analyzed by hydride generation and atomic fluorescence spectrometry (HG-AFS), which provided data for the total arsenic content.  相似文献

Fast liquid chromatography coupled to triple-quadrupole tandem mass spectrometry was employed for the determination of six UV filters in seawater. The separation of the analytes was achieved in less than 5 min; polarity switching was used as four of the analytes were ionized in positive mode and the remaining two in negative mode. Two ionization sources were employed and compared: atmospheric pressure chemical ionization (APCI) gave better results than electrospray ionization (ESI) for all analytes, with higher reproducibility and lower detection limits. Therefore APCI was chosen for the determination of the analytes in seawater samples using stir bar sorptive extraction-liquid desorption (SBSE-LD).Quantitative analysis was performed in multiple reaction monitoring (MRM) mode; fragmentation pathways of the analytes with regard to the formation of the MRM ions were also proposed.For the analysis of seawater samples, calibration curves were drawn using SBSE in spiked seawater. All figures of merit of the method were satisfactory; limits of detection were particularly low for the four analytes ionized in positive mode, being in the range 8-31 ng/L. The method was applied to the determination of the six UV filters in seawater samples from Liguria, Italy. Only benzophenone-3 (BP-3) and ethylhexyl methoxycinnamate (EHMC) were measured in the analyzed samples; some of the remaining analytes were also detected but always below the limit of quantitation.  相似文献

A high throughput liquid chromatography-tandem mass spectrometric (LC-MS/MS) method for the determination of valproic acid, an antiepileptic drug, in human plasma is described. It is a rapid and sensitive isocratic reversed-phase liquid chromatography-tandem mass spectrometric method equipped with turbo ion spray (TIS) source, operating in the negative ion and pseudo selective reaction monitoring (SRM) acquisition mode to quantify valproic acid. The extraction of valproic acid and hydrochlorothiazide (IS) from the plasma involved sample treatment with phosphoric acid followed by solid-phase extraction using Waters hydrophilic-lipophilic balance (HLB) cartridge giving extracts free from endogenous interferences. Sample preparation by this method yielded very good and consistent mean recoveries of 99.73 and 74.47% for valproic acid and IS, respectively. The method was linear over the dynamic range of 2.0-200.0 μg/ml (covering entire therapeutic range) with a correlation coefficient r ≥ 0.9989. The coefficient of variance (CV, %) was 7.03% at 2.0 μg/ml (LLOQ). This method was fully validated for its accuracy, precision, recovery and matrix effect especially because the pattern of elution of all the analytes may appear as flow injection type. The analyte stability was examined under conditions mimicking the sample storage, handling and analysis procedures. The method was successfully applied for bioequivalence studies in human subject samples after oral administration of 500 mg formulations.  相似文献

Pollutants such as human pharmaceuticals and synthetic hormones that are not covered by environmental legislation have increasingly become important emerging aquatic contaminants. This paper reports the development of a sensitive and selective multi-residue method for simultaneous determination and quantification of 23 pharmaceuticals and synthetic hormones from different therapeutic classes in water samples. Target pharmaceuticals include anti-diabetic, antihypertensive, hypolipidemic agents, β2-adrenergic receptor agonist, antihistamine, analgesic and sex hormones. The developed method is based on solid phase extraction (SPE) followed by instrumental analysis using liquid chromatography-electrospray ionization-tandem mass spectrometry (LC–ESI-MS/MS) with 30 min total run time. River water samples (150 mL) and (sewage treatment plant) STP effluents (100 mL) adjusted to pH 2, were loaded into MCX (3 cm3, 60 mg) cartridge and eluted with four different reagents for maximum recovery. Quantification was achieved by using eight isotopically labeled internal standards (I.S.) that effectively correct for losses during sample preparation and matrix effects during LC–ESI-MS/MS analysis. Good recoveries higher than 70% were obtained for most of target analytes in all matrices. Method detection limit (MDL) ranged from 0.2 to 281 ng/L. The developed method was applied to determine the levels of target analytes in various samples, including river water and STP effluents. Among the tested emerging pollutants, chlorothiazide was found at the highest level, with concentrations reaching up to 865 ng/L in STP effluent, and 182 ng/L in river water.  相似文献

A sequential injection UV method was developed to determine benzophenone-4 (BZ4) and phenylbenzimidazole sulphonic acid (PBS) simultaneously, these being the most commonly used UV-filters in aqueous formulations used as sunscreen sprays. The selective elution of both was performed by on-line solid-phase extraction, by retention on a SAX microcolumn and separation by varying the pH of elution. The sensitivity obtained was 0.042±0.001 ml μg−1 for PBS and 0.0159±0.0003 ml μg−1 for BZ4. The limit of detection was 1.6 μg ml−1 for PBS and 0.6 μg ml−1 for BZ4. The R.S.D. of the results was 1-6% for PBS and 1-12% for BZ4. The method was validated using commercial sunscreen formulations with concentrations determined by a liquid chromatographic procedure. The two procedures gave comparable results. Automation of the method means the amount of reagents used and residues generated are decreased. The system allows the required analysis sequence to be programmed using suitable software.  相似文献

Capillary zone electrophoresis (CZE) with indirect UV detection was used in developing a method for the simultaneous determination of inorganic anions, aliphatic and heterocyclic organic acids in various processed samples. The analytes were determined simultaneously in 10 min using an electrolyte containing 20 mM 2,3-pyrazine dicarboxylic acid, 65 mM tricine, 2 mM BaCl2, 0.5 mM cetyltrimethylammonium bromide, and 2 M urea at pH 8.06. Linear plots for the analytes were obtained in the concentration range of 2–150 mg L−1. Relative standard deviations (RSDs) of peak areas during a 3-day analysis period varied from 5.5% for glycolate to 9.5% for oxalate. RSDs of migration times varied between 0.4% and 1.1%. The detection limit (at S/N 3) was 1 mg L−1 for all the analytes studied. The proposed method was successfully demonstrated for the determination of carboxylic acids in eight oxygen treated samples of commercial softwood and hardwood kraft lignin and two red wine samples of Pinot Noir grapes. In the kraft lignin samples the concentrations of carboxylic acids correspond to the oxidation time. The acid concentrations of wine varied considerable.  相似文献

建立了化妆品中11种二苯酮类紫外线吸收剂的高效液相色谱分析方法。采用四氢呋喃-甲醇-水或二氯甲烷-水体系提取化妆品中二苯酮类紫外线吸收剂,经离心(5000 r/min),上清液过0.22 μm滤膜后,进入高效液相色谱分析。采用C18色谱柱进行分离,以0.1% (体积分数)甲酸水溶液(含10 mmol/L乙酸铵)为流动相A,以0.1% (体积分数)甲酸甲醇为流动相B,梯度洗脱。方法加标回收率(n=7)为93.4%~103.8%,相对标准偏差为0.1%~4.2%,方法的检出限为4.0~30 μg/g,方法的定量限为15~100 μg/g。采用该方法对42种市售化妆品检测分析发现,有5种二苯酮类紫外线吸收剂被检出,其中防晒隔离液中二苯酮-3和香水中二苯酮-2的检出量分别为2785 μg/g和2106 μg/g。研究结果表明,所建立的方法具有良好的回收率、重现性和较高的灵敏度,可用于化妆品中多种二苯酮类紫外线吸收剂的分析。  相似文献

No previous publications about percutaneous absorption of polyethylene glycol 25 p-aminobenzoic acid (PEG-25 PABA) have been found in the literature and the expected levels to be found in human urine after sunscreens use are unknown. The method proposed here is suitable to determine PEG-25 PABA in the urine of sunscreens users in order to carry out studies on body accumulation/excretion. It is based on solid-phase extraction (SPE) with size-exclusion liquid chromatography determination. Solid-phase extraction allows the analyte to be retained and subsequently eluted for a clean-up, using a silica-based cartridge. The size-exclusion liquid chromatography of the eluted allows the rest of matrix interferences to be avoided. Fluorescence intensity was measured at λem = 350 nm (λexc = 300 nm). The sensitivity of the proposed method is in the order of 450 ± 5 mL ng−1 and the detection limit (3 Sy/x/b) in the measured solutions is in the order of 13 ng mL−1, that is 2.6 ng mL−1 in urine samples. The method enables PEG-25 PABA to be determined in both, spiked and unspiked human urine samples. Results obtained for spiked human urine samples (11-100 ng mL−1) demonstrated the accuracy of the method. The mean relative standard deviation of the results was in the order of 3-10%. Three volunteers applied a sunscreen lotion containing a 8% PEG-25 PABA sunscreen cream and their urinary excretion was controlled from the moment of application until the excreted amounts were no longer detectable.  相似文献

A higher monitoring rate is highly desirable in the labs, but this goal is typically limited by sample throughput. In this study, we sought to assess the real-world applicability of fast, low-pressure GC–time-of-flight MS (LP-GC/TOFMS) for the identification and quantification of 150 pesticides in tomato, strawberry, potato, orange, and lettuce samples. Buffered and unbuffered versions of QuEChERS (which stands for “quick, easy, cheap, effective, rugged, and safe”) using dispersive solid-phase extraction (d-SPE) and disposable pipette extraction (DPX) for clean-up were compared for sample preparation. For clean-up of all sample types, a combination of 150 mg MgSO4, 50 mg primary secondary amine (PSA), 50 mg C18, and 7.5 mg graphitized carbon black (GCB) per mL extract was used. No significant differences were observed in the results between the different sample preparation versions. QuEChERS took <10 min per individual sample, or <1 h for two chemists to prepare 32 pre-homogenized samples, and using LP-GC/TOFMS, <10 min run time and <15 min cycle time allowed >32 injections in 8 h. Overall, >126 analytes gave recoveries (3 spiking levels) in the range of 70–120% with <20% RSD. The results indicate that LP-GC/TOFMS for GC-amenable analytes matches UHPLC–MS/MS in terms of sample throughput and turnaround time for their routine, concurrent use in the analysis of a wide range of analytes in QuEChERS extracts to achieve reliable quantification and identification of pesticide residues in foods.  相似文献

In this study, a simple, rapid and efficient method, dispersive liquid-liquid microextraction (DLLME) combined gas chromatography-electron capture detection (GC-ECD), for the determination of chlorobenzenes (CBs) in water samples, has been described. This method involves the use of an appropriate mixture of extraction solvent (9.5 μl chlorobenzene) and disperser solvent (0.50 ml acetone) for the formation of cloudy solution in 5.00 ml aqueous sample containing analytes. After extraction, phase separation was performed by centrifugation and the enriched analytes in sedimented phase were determined by gas chromatography-electron capture detection (GC-ECD). Our simple conditions were conducted at room temperature with no stiring and no salt addition in order to minimize sample preparation steps. Parameters such as the kind and volume of extraction solvent, the kind and volume of disperser solvent, extraction time and salt effect, were studied and optimized. The method exhibited enrichment factors and recoveries ranging from 711 to 813 and 71.1 to 81.3%, respectively, within very short extraction time. The linearity of the method ranged from 0.05 to 100 μg l−1 for dichlorobenzene isomers (DCB), 0.002-20 μg l−1 for trichlorobenzene (TCB) and tetrachlorobenzene (TeCB) isomers and from 0.001 to 4 μg l−1 for pentachlorobenzene (PeCB) and hexachlorobenzene (HCB). The limit of detection was in the low μg l−1 level, ranging between 0.0005 and 0.05 μg l−1. The relative standard deviations (R.S.D.s) for the concentration of DCB isomers, 5.00 μg l−1, TCB and TeCB isomers, 0.500 μg l−1, PeCB and HCB 0.100 μg l−1 in water by using the internal standard were in the range of 0.52-2.8% (n = 5) and without the internal standard were in the range of 4.6-6.0% (n = 5). The relative recoveries of spiked CBs at different levels of chlorobenzene isomers in tap, well and river water samples were 109-121%, 105-113% and 87-120%, respectively. It is concluded that this method can be successfully applied for the determination of CBs in tap, river and well water samples.  相似文献
