Reaction of the title compound with hydrazine in the presence of air gives the 1-unsubstituted parent system via oxidative dehydrazination of the 1-hydrazino intermediate. The latter can be obtained in high yield by carrying out the hydrazinolysis step under inert gas, and it is smoothly converted into [1,2,4]-triazolo[4',3':1,6]pyridazino[4,5- b]indoles.  相似文献

以2,6-二氯吡啶为起始原料, 经肼基化、还原、硝化、Nietzki-Dietschy环合4步反应得到5-氨基-6-硝基-[1,2,5]噁二唑并[3,4-b]吡啶-1-氧化物。 结合反应机理讨论了还原、硝化、Nietzki-Dietschy环合反应的影响因素,获得了合成5-氨基-6-硝基-[1,2,5]噁二唑并[3,4-b]吡啶-1-氧化物的最佳工艺条件,目标产物的总收率为59.2%。 用1H NMR、MS和IR谱对5-氨基-6-硝基-[1,2,5]噁二唑并[3,4-b]吡啶-1-氧化物的结构进行了表征。  相似文献

A series of new calix[4]phenols, calix[4]resorcinols and calix[4]pyrogallols with acetylhydrazide substitutes has been␣synthesized with high yields by hydrazinolysis of ester group containing calix[4]arenes. The synthesized calix[4]phenols adopt the cone conformation while the calix[4]resorcinol and calix[4]pyrogallol derivatives prefer the boat conformation. The amide fragment of the hydrazide groups predominantly exists in the trans-conformation. The binding ability of synthesized calix[4]arenes toward transition and alkali metals by solvent extraction has been investigated.  相似文献

4‐Hydrazino‐2‐methylpyrimidino[4′,5′:4,5]thiazolo[3,2‐a]benzimidazole ( 4 ) was obtained from hydrazinolysis of the 4‐chloro derivative 3 with hydrazine hydrate. The hydrazino derivative 4 was further cyclized to the corresponding pyrazole 5 , pyrazolone 6 and 5‐methyl‐1,2,4‐triazolo[1″,5″:3′,4′]pyrimidino[5′,6′:5,4]‐thiazolo[3,2‐a]benzimidazole ( 9 ) and 5‐methy‐1,2,4‐triazolo[4″,3″:3′,4′]pyrimidino[5′,6′:5,4]thiazolo‐[3,2‐a]benzimidazole ( 10 ), respectively. The long-range goal of the present study is the development of a general approach for in vivo dosimetry of reactive metabolites of polycyclic aromatic hydrocarbons (PAHs), to be used as a tool in cancer risk assessment. With benzo[a]pyrene (BaP) chosen as indicator and a model of PAHs this study aims at the development of a method for the determination of adducts to histidine (His) in hemoglobin (Hb) and serum albumin (SA) of reactive metabolites of BaP. The predominantly mutagenic metabolite of BaP has been shown to be a diolepoxide isomer, +(anti)r-7, t-8-dihydroxy-t-9,10-epoxy-7,8, 9,10-tetrahydrobenzo[a]pyrene (+BPDE). In comparison with other methods for protein degradation, hydrazinolysis was found to be sufficiently effective and mild. The His adduct isolated after protein hydrazinolysis, with protection by tert-butyloxycarbonyl (Boc) of the hydrazide and alpha-amino groups, was shown to be N(im)- +/- (r-7, t-8, t-9-trihydroxy-7, 8, 9, 10-tetrahydrobenzo[a]pyren-c-10-yl)-N(alpha), N(2)-bis(tert-butyloxycarbonyl)-L-histidinehydrazide. Isomers of this compound, used as references, were synthesized and characterized by liquid chromatography/tandem mass spectrometry (LC/MS/MS). Adducts in Hb and SA from in vitro treatment with BPDE were characterized after hydrazinolysis by HPLC-UV/MS, muHPLC/MS/MS and gas chromatography/mass spectrometry (GC/MS). Approximately 70 and 10% of the isolated BPDE adducts from SA and Hb, respectively, were His adducts. Other products were released as BaP tetrols and BaP triols. For the purpose of enrichment/purification of BPDE-His adducts, C(18) and cation exchange solid phase extraction (SPE) were utilized. The sensitivity obtained by this new approach, based on hydrazinolysis of protein, enrichment by SPE and analysis with muHPLC/MS/MS (APCI), is in the low-fmole range.  相似文献

Some new 7,9-disubstituted 7H-1,2,3,4-tetrazolo[1,5-c]pyrrolo[3,2-e]pyrimidines 5 have been synthesized either by diazotization of 4-hydrazino-5,7-disubstituted-7H-pyrrolo[2,3-d]pyrimidines 4 obtained by hydrazinolysis of 4-chloro-5,7-disubstituted-7H-pyrrolo[2,3-d]pyrimidines 3 or via a substitution reaction between 3 and sodium azide. 5,7-Disubstituted-7H-pyrrolo[2,3-d]pyrimidin-4(3H)-ones 2 were obtained by cyclocondensation of 2-amino-3-cyano-1,4-disubstituted pyrroles 1 with formic acid which on chlorination using phosphorus oxychloride afforded 3 . A novel route for the synthesis of 4-amino-5,7-disubstituted-7H-pyrrolo[2,3-d]pyrimidines 6 by the reductive ring cleavage of 5 has been reported.  相似文献

Some fluoroaryl substituted 2-amino-3-cyanopyrroles 2 were synthesized from the reaction between (2-bromo-1-arylalkylidene)propanedinitriles 1 and fluoroaryl substituted aromatic amines under Gewald reaction condition, which on reaction with formamide and formic acid gave 4-aminopyrrolo[2,3-d]pyrimidines 3 and pyrrolo[2,3-d]-pyrimidin-4(3H)-ones 4 respectively. 4-Chloropyrrolo[2,3-d]pyrimidines 5 were prepared by chlorination of 4 with phosphorus oxychloride, which on hydrazinolysis provided 4-hydrazinopyrrolo[2–3-d]pyrirnidines 6 .  相似文献

The reaction of 4-dialkylamino-6-chloro-2-methylthiopyrimidine-5-carbaldehydes with ethyl mercaptoacetate in the presence of triethylamine gives the corresponding ethyl esters of thieno[2,3-d]pyrimidine-6-carboxylic acids. Methods were developed for the synthesis of ω-mercaptoalkoxy derivatives of thiacalix[4]-arenes in the 1,3-alternate conformation containing different numbers (2–5) of methylene spacers at the lower rim. 以2-苄硫基-5-甲基-7-羟基-1,2,4-三唑并[1,5-a]嘧啶为原料,经醚化、肼解、成盐、关环和席夫碱反应合成了20个新型的含1,2,4-三唑-5-硫酮席夫碱的1,2,4-三唑并[1,5-a]嘧啶类化合物,通过IR,1H NMR,MS和元素分析对所合成的化合物进行了结构表征.初步生物活性测试结果表明,部分化合物表现出一定的抑菌或较好的抗烟草花叶病毒(TMV)活性.在500μg/mL浓度下,化合物6b,6f和6p对TMV的抑制率分别为41%,43%和40%.  相似文献

Two novel 2,3-dihydro-1H-imidazo[1,2-b]pyrazole derivatives 7 and 8 have been prepared by hydrazinolysis with 2,4-dinitrophenylhydrazine of 1-(benzylideneamino)-2-(2-ethoxycarbonyl-2-nitromethylidene)-imidazolidine 4 . The precursor 4 was conveniently prepared from ethyl nitroacetate and 1-(benzylidene-amino)-2-(methylthio)imidazoline 3 . Two examples are presented in which ethyl aceto(and trifluoroaceto)acetate 2-nitrophenyl-hyrazone, 9 and 10 , both of which also feature the β-hydrazinoacrylate arrangement, are refluxed in ethanol containing hydrochloric acid and thereby converted into pyrazolones 11 and 12 , pyrazoles 13 and 14 , and ketone 2-nitrophenylhydrazones 15 and 16 , respectively.  相似文献

Summary Isomerization of available amidinoyl isothiocyanates has been elaborated into a viable synthetic approach to sec. amine substituted tetrazolo[1,5-c]quinazolines. The method involves isomerization to quinazoline-4-thiones, their hydrazinolysis, followed by a nitrosation to give the title tetrazoloquinazolines. Infrared spectra of both solid samples and their solutions were used to assess the stability of the tetrazole ring.

Zusammenfassung Die Isomerisierung von gut zugänglichen Amidinoylisothiocyanaten wurde zu einer synthetischen Methode für die durch sek. Amine substituierten Tetrazolo[1,5-c]chinazoline entwickelt. Die Methode beinhaltet Isomerisierung von Chinazolin-4-thionen, deren Hydrazinolyse und anschließende Nitrosierung. Die Stabilität des Tetrazolringes in den Tetrazolochinazolinen wurde aus den Infrarotspektren abgeschätzt.

New de-tert-butyltetrathiacalix[4]arenes with acetylhydrazide substituents in cone and 1,3-alternate conformations have been synthesised with good yield by the hydrazinolysis of calix[4]arene ester derivatives. The recognition ability of synthesised macrocycles towards transition and alkali metals has been investigated by the picrate extraction method. The stoichiometry of complexes and the extraction constants have been determined. It has been found that tetrahydrazides do not extract alkali metal ions, but show an excellent affinity towards transition and heavy metal cations. The 1,3-alternate conformer of de-tert-butyltetrathiacalix[4]arene has revealed a remarkable selectivity for Ni2+ and Ag+ in the row of d-elements and for Cd2+ ion among the toxic heavy metals. The experimental data show that the removal of tert-butyl groups from tetrathiacalix[4]arene framework leads to the drastic improvement of extraction efficiency and selectivity of new tetrahydrazides.  相似文献

Chiral pseudo[1]rotaxanes and [1]rotaxanes constructed from macrocyclic arenes still remain a big challenge mainly owing to the lack of such chiral macrocycles. In this work, a new system of chiral pseudo[1]rotaxanes formed by self-inclusion of helic[6]arene containing amide linked with the terminal tertiary amines was first discovered. Based on an atom-economic stopping strategy, a pair of chiral [1]rotaxanes were conveniently obtained in almost quantitative yields by blocking the pseudo[1]rotaxanes with monobenzyl bromide of tetraphenylethene. The structures of pseudo[1]rotaxanes and [1]rotaxanes were characterized by 2D NMR spectra in solution, combined with DFT calculations. The photophysical properties further revealed the efficient chirality transfer of helic[6]arene to the tetraphenylethene moiety, compared to their unthreaded chiral isomers. The discovery of the chiral pseudo[1]rotaxanes allows for a wide and available synthesis of chiral [1]rotaxanes, and also opening a new avenue to the design of chiral supramolecular materials.  相似文献

Photocyclization of 3-chloro-N-phenylbenzo[b]thiophene-2-carboxamide 10 afforded [1]benzothieno[2,3-c]-quinolin-6(5H)-one 11 which was chlorinated to 6-chloro[1]benzothieno[2,3-c]quinoline 12 followed by dechlorination to give [1]benzothieno[2,3-c]quinoline 5 . A series of 6-substituted alkoxy and thioalkoxy[1]benzothieno[2,3-c]quinoline derivatives were prepared along with the N-methyl quaternary salt 13 of 5 . 6-Chloro[1]-benzothieno[2,3-c]quinoline 12 was converted into 6-hydrazino[1]benzothieno[2,3-c]quinoline 23 which upon treatment with formic acid yielded [1]benzothieno[2,3-c][1,2,4]triazolo[4,3-a]quinoline 6 . Treatment of 23 with nitrous acid resulted in [1]benzothieno[2,3-c]tetrazolo[1,5-a]quinoline 7 . Compounds 6 and 7 are novel heterocyclic ring systems.  相似文献
