Trypanosomal and leishmanial infections claim tens of thousands of lives each year. The metabolism of these unicellular eukaryotic parasites differs from the human host and their enzymes thus constitute promising drug targets. Tryparedoxin (Tpx) from Trypanosoma brucei is the essential oxidoreductase in the parasite's hydroperoxide‐clearance cascade. In vitro and in vivo functional assays show that a small, selective inhibitor efficiently inhibits Tpx. With X‐ray crystallography, SAXS, analytical SEC, SEC‐MALS, MD simulations, ITC, and NMR spectroscopy, we show how covalent binding of this monofunctional inhibitor leads to Tpx dimerization. Intra‐ and intermolecular inhibitor–inhibitor, protein–protein, and inhibitor–protein interactions stabilize the dimer. The behavior of this efficient antitrypanosomal molecule thus constitutes an exquisite example of chemically induced dimerization with a small, monovalent ligand that can be exploited for future drug design.  相似文献

Single‐walled carbon nanotubes (SWCNTs) have been functionalized with poly(γ‐benzyl‐L ‐glutamate) (PBLG) by ring‐opening polymerizations of γ‐benzyl‐L ‐glutamic acid‐based N‐carboxylanhydrides (NCA‐BLG) using amino‐functionalized SWCNTs (SWCNT‐NH2) as initiators. The SWCNT functionalization has been verified by FTIR spectroscopy and transmission electron microscopy. The FTIR study reveals that surface‐attached PBLGs adopt random‐coil conformations in contrast to the physically absorbed or bulk PBLGs, which exhibit α‐helical conformations. Raman spectroscopic analysis reveals a significant alteration of the electronic structure of SWCNTs as a result of PBLG functionalization. The PBLG‐functionalized SWCNTs (SWCNT‐PBLG) exhibit enhanced solubility in DMF. Stable DMF solutions of SWCNT‐PBLG/PBLG with a maximum SWCNTs concentration of 259 mg L?1 can be readily obtained. SWCNT‐PBLG/PBLG solid composites have been characterized by differential scanning calorimetry, thermogravimetric analysis, wide/small‐angle X‐ray scattering (W/SAXS), scanning electron microscopy, and polarized optical microscopy for their thermal or morphological properties. Microfibers containing SWCNT‐PBLG and PBLG can also be prepared via electrospinning. WAXS characterization reveals that SWCNTs are evenly distributed among PBLG rods in solution and in the solid state where PBLGs form a short‐range nematic phase interspersed with amorphous domains. © 2010 Wiley Periodicals, Inc. Synthesis and Properties of the Layered Perovskite Phase Sr3Mo1.5Zn0.5O7‐δ The new layered perovskite phase Sr3Mo1.5Zn0.5O7‐δ was synthesized by solid state reaction using a Zn/ZnO oxygen buffer. The crystal structure was refined from X‐ray powder pattern by the Rietveld method. The compound crystallizes tetragonal in the space group I4/mmm (no. 139) with the lattice parameters a = 3.9631(3) Å, c = 20.583(1) Å. An oxygen deficiency corresponding to δ ≈ 0.25 was determinated, indicating the presence of molybdenum in mixed valence (Mo4+ and Mo6+).  相似文献

Six 4‐alkyl‐4‐azaheptane‐1,7‐diamines, characterized as their solid bis(D ‐gluconamides), were prepared in a two‐step synthesis: bis(cyanoethylation) of the primary alkylamines (C8–C18, even‐numbered) followed by an efficient lithium aluminum hydride reduction of the resulting bisnitriles. The azadiamines were then used as monomers in condensation polymerizations with methyl D ‐glucarate 1,4‐lactone in a methanol solution, yielding polyhydroxypolyaminopolyamides isolated directly as white solids with varying hydrophobic/hydrophilic character. © 2000 John Wiley & Sons, Inc. An amphiphilic multiblock copolymer [poly(ethylene oxide)‐b‐polystyrene]n [(PEO‐b‐PS)n] is synthesized by using trithiocarbonate‐embedded PEO as macro‐RAFT agent. PEO with four inserted trithiocarbonate (Mn = 9200 and Mw/Mn = 1.62) groups is prepared first by condensation of α, ω‐dihydroxyl poly(ethylene oxide) with S, S′‐Bis(α, α′‐dimethyl‐α″‐acetic acid)‐trithiocarbonate (BDATC) in the presence of pyridine, then a series of goal copolymers with different St units (varied from 25 to 218 per segment) are obtained by reversible addition‐fragmentation chain transfer (RAFT) polymerization. The synthesis process is monitored by size exclusion chromatography (SEC), 1H NMR and FT‐IR. The self‐assembled morphologies of the copolymers are strongly dependent of the length of PS block chains when the chain length of PEO is fixed, some new morphologies as large leaf‐like aggregates (LLAs), large octopus‐like aggregates (LOAs), and coarse‐grain like micelles (CGMs) are observed besides some familiar aggregates as large compound vesicles (LCVs), lamellae and rods, and the effect of water content on the morphologies is also discussed. © 2006 Wiley Periodicals, Inc.
