Phosphorylation is a post‐translational modification that is involved in many basic cellular processes and diseases, but is difficult to detect in real time with existing technologies. A label‐free detection of phosphorylation is reported in real time with self‐assembled nano‐oscillators. Each nano‐oscillator consists of a gold nanoparticle tethered to a gold surface with a molecular linker. When the nanoparticle is charged, the nano‐oscillator can be driven into oscillation with an electric field and detected with a plasmonic imaging approach. The nano‐oscillators measure charge change associated with phosphorylation of peptides attached onto a single nanoparticle, allowing us to study the dynamic process of phosphorylation in real time without antibodies down to a few molecules, from which Michaelis and catalytic rate constants are determined.  相似文献

Over the last few years, one of the most important and complex problems facing our society is treating infectious diseases caused by multidrug‐resistant bacteria (MDRB), by using current market‐existing antibiotics. Driven by this need, we report for the first time the development of the multifunctional popcorn‐shaped iron magnetic core–gold plasmonic shell nanotechnology‐driven approach for targeted magnetic separation and enrichment, label‐free surface‐enhanced Raman spectroscopy (SERS) detection, and the selective photothermal destruction of MDR Salmonella DT104. Due to the presence of the “lightning‐rod effect”, the core–shell popcorn‐shaped gold‐nanoparticle tips provided a huge field of SERS enhancement. The experimental data show that the M3038 antibody‐conjugated nanoparticles can be used for targeted separation and SERS imaging of MDR Salmonella DT104. A targeted photothermal‐lysis experiment, by using 670 nm light at 1.5 W cm?2 for 10 min, results in selective and irreparable cellular‐damage to MDR Salmonella. We discuss the possible mechanism and operating principle for the targeted separation, label‐free SERS imaging, and photothermal destruction of MDRB by using the popcorn‐shaped magnetic/plasmonic nanotechnology.  相似文献

We report on bottom-up assembly routes for fabricating plasmonic structures and metamaterials composed of colloidal gold and silver nanostructures, such as nanoparticles ("metatoms") and shape-controlled nanocrystals. Owing to their well-controlled sizes/shapes, facile surface functionalization, and excellent plasmonic properties in the visible and near-infrared regions, these nanoparticles and nanocrystals are excellent building blocks of plasmonic structures and metamaterials for optical applications. Recently, we have utilized two kinds of bottom-up techniques (i.e., multiple-probe-based nanomanipulation and layer-by-layer self-assembly) to fabricate strongly coupled plasmonic dimers, one-dimensional (1D) chains, and large-scale two-dimensional/three-dimensional (2D/3D) nanoparticle supercrystals. These coupled nanoparticle/nanocrystal assemblies exhibit unique and tunable plasmonic properties, depending on the material composition, size/shape, intergap distance, the number of composing nanoparticles/nanocrystals (1D chains), and the nanoparticle layer number in the case of 3D nanoparticle supercrystals. By studying these coupled nanoparticle/nanocrystal assemblies, the fundamental plasmonic metamaterial effects could be investigated in detail under well-prepared and previously unexplored experimental settings.  相似文献

Observation of single plasmonic nanoparticles in reconstituted biological systems allows us to obtain snapshots of dynamic processes between molecules and nanoparticles with unprecedented spatiotemporal resolution and single‐molecule/single‐particle‐level data acquisition. This Concept is intended to introduce nanoparticle‐tethered supported lipid bilayer platforms that allow for the dynamic confinement of nanoparticles on a two‐dimensional fluidic surface. The dark‐field‐based long‐term, stable, real‐time observation of freely diffusing plasmonic nanoparticles on a lipid bilayer enables one to extract a broad range of information about interparticle and molecular interactions throughout the entire reaction period. Herein, we highlight important developments in this context to provide ideas on how molecular interactions can be interpreted by monitoring dynamic behaviors and optical signals of laterally mobile nanoparticles.  相似文献

Plasmonic nanoparticles such as those of gold or silver have been recently investigated as a possible way to improve light absorption in thin film solar cells. Here, a simple method for the preparation of spherical plasmonic gold nanoparticles in the form of a colloidal solution is presented. The nanoparticle diameter is controlled in the range from several nm to tens of nm depending on the synthesis parameters with the size dispersion down to 14 %. The synthesis is based on thermal decomposition and reduction of the chloroauric acid in the presence of a stabilizing capping agent (surfactant) that is very slowly injected into the hot solvent. The surfactant prevents uncontrolled nanoparticle aggregation during the growth process. The nanoparticle size and shape depend on the type of the stabilizing agent. Surfactants with different lengths of the hydrocarbon chains such as Z-octa-9-decenylamine (oleylamine) with AgNO3 and polyvinylpyrrolidone with AgNO3 were used for the steric stabilization. Hydrodynamic diameter of the gold nanoparticles in the colloidal solution was determined by dynamic light scattering while the size of the nanoparticle metallic core was found by small-angle X-ray scattering. The UV-VIS-NIR spectrophotometer measurements revealed a plasmon resonance absorption in the 500–600 nm range. Self-assembled nanoparticle arrays on a silicon substrate were prepared by drop casting followed by spontaneous evaporation of the solvent and by a modified Langmuir-Blodgett deposition. The degree of perfection of the self-assembled arrays was analyzed by scanning electron microscopy and grazing-incidence small-angle X-ray scattering. Homogeneous close-packed hexagonal ordering of the nanoparticles stretching over large areas was evidenced. These results document the viability of the proposed nanoparticle synthesis for the preparation of high-quality plasmonic templates for thin film solar cells with enhanced power conversion efficiency, surface enhanced Raman scattering, and other applications.  相似文献

A real time multimodal data acquisition imaging setup is developed for the electrophoretic movement of plasmonic nanoparticles. Movement of the nanoparticles is recorded by time-lapse digital imaging at 8-megapixel resolutions. The analysis of the moving nanoparticle band is performed using threshold image and color extraction at respective time frames. The migration dynamics is sensitive to size, nature and the extent of the surface conjugation to the nanoparticle. The dynamics of color intensity of the nanoparticles is shown to be dependent on the extent of stabilization of nanoparticle (by a given agent). The stability of nanoparticle is determined by stationary nature and also the relative proportions of pixel intensities in respective color planes (R, G and B). Detergents stabilize nanoparticles in a concentration dependent fashion. In case of neutral polymers the extent of stabilization depends on the relative proportion of the polymer and also on the nature of the same, e.g., PEG (polyethylene glycol) at low concentration imparts higher stability as compared to PVP (polyvinylpyrilidone). The ascending or declining temporal dynamics of color profiles observed in case of citrate stabilized or amino acid conjugated nanoparticles, represent enrichment of plasmonic particles, or their diffusion resulting from loss of charge during migration. The higher dimensional imaging technique thus can be exploited for discriminating the nanoparticles on the basis of their migration behavior and their stability as reflected from color dynamics. The technique is applicable to other nano-sized colored objects, e.g. proteins like hemoglobin where the protein color has important clinical value.  相似文献

Surface plasmon resonances of metal nanoparticles have shown significant promise for the use of solar energy to drive catalytic chemical reactions. More importantly, understanding and monitoring such catalytic reactions at single‐nanoparticle level is crucial for the study of local reaction processes. Herein, using plasmonic photoluminescence (PL) spectroscopy, we describe a novel sensing method for catalytic ethanol oxidation reactions at the single‐nanoparticle level. The Au nanorod monitors the interfacial interaction with ethanol during the catalytic reaction through the PL intensity changes in the single‐particle PL spectra. The analysis of energy relaxation of excited electron–hole pairs indicates the relationship between the PL quenching and ethanol oxidation reaction on the single Au nanorod.  相似文献

We report the development of bioconjugated plasmonic vesicles assembled from SERS-encoded amphiphilic gold nanoparticles for cancer-targeted drug delivery. This new type of plasmonic assemblies with a hollow cavity can play multifunctional roles as delivery carriers for anticancer drugs and SERS-active plasmonic imaging probes to specifically label targeted cancer cells and monitor intracellular drug delivery. We have shown that the pH-responsive disassembly of the plasmonic vesicle, stimulated by the hydrophobic-to-hydrophilic transition of the hydrophobic brushes in acidic intracellular compartments, allows for triggered intracellular drug release. Because self-assembled plasmonic vesicles exhibit significantly different plasmonic properties and greatly enhanced SERS intensity in comparison with single gold nanoparticles due to strong interparticle plasmonic coupling, disassembly of the vesicles in endocytic compartments leads to dramatic changes in scattering properties and SERS signals, which can serve as independent feedback mechanisms to signal cargo release from the vesicles. The unique structural and optical properties of the plasmonic vesicle have made it a promising platform for targeted combination therapy and theranostic applications by taking advantage of recent advances in gold nanostructure based in vivo bioimaging and photothermal therapy and their loading capacity for both hydrophilic (nucleic acids and proteins) and hydrophobic (small molecules) therapeutic agents.  相似文献

A micro‐beam scanning X‐ray photoelectron spectroscopy (XPS) has been utilized to obtain information about the attachment of ligand to nanoparticles in solution by measuring the ligand components relative to nanoparticle components. CdSe/ZnS core/shell nanoparticles capped with three different kinds of ligand molecules are drop‐casted on a substrate and led to dry into self‐agglomerated micropatterns. Those specimens are mapped out by element‐sensitive XPS imaging. The spatial correlation between surface ligand molecules and core nanoparticle atoms is statistically analysed using Pearson correlation function, which provides with a useful assessment of the ligand binding to nanoparticle surfaces. The influence of capping agents on the oxidation of silver nanoparticles was studied by using the electrochemical techniques of anodic stripping voltammetry and anodic particle coulometry (“nano‐impacts”). Five spherical silver nanoparticles each with a different capping agent (branched polyethylenimine (BPEI), citrate, lipoic acid, polyethylene glycol (PEG) and polyvinylpyrrolidone (PVP)) were used to perform comparative experiments. In all cases, regardless of the capping agent, complete oxidation of the single nanoparticles was seen in anodic particle coulometry. The successful quantitative detection of the silver nanoparticle size displays the potential application of anodic particle coulometry for nanoparticle characterisation. In contrast, for anodic stripping voltammetry using nanoparticles drop casting, it was observed that the capping agent has a very significant effect on the extent of silver oxidation. All five samples gave a low oxidative charge corresponding to partial oxidation. It is concluded that the use of anodic stripping voltammetry to quantify nanoparticles is unreliable, and this is attributed to nanoparticle aggregation.  相似文献

How many of the several attributes of the bulk metallic state persist in a nanoparticle containing a finite number of atoms of a metallic element? Do all those attributes emerge suddenly at a well‐defined cluster size or do they rather evolve at different rates and in a broad size range? These fundamental questions have been addressed through a conjoint experimental/theoretical investigation of zinc clusters. We report the observation of novel coexistence phenomena involving different electronic phases: for some sizes, metallic and insulating electronic states coexist within a single, Janus‐like, nanoparticle; for the rest of sizes, we report the coexistence of two weakly interacting metallic phases with different dimensionalities, localized at the shell and the core of the nanoparticle. These fascinating features are due to an anomalously long core–shell separation that equips the shell and core regions with largely independent structural, vibrational, and thermal properties.  相似文献

We report core-satellites (Au-Ag) coupled plasmonic nanoassemblies based on bottom-up, high-density assembly of molecular-scale silver nanoparticles on a single gold nanoparticle surface, and demonstrate direct observation and quantification of enhanced plasmon coupling (i.e., intensity amplification and apparent spectra shift) in a single particle level. We also explore metal ion sensing capability based on our coupled plasmonic core-satellites, which enabled at least 1000 times better detection limit as compared to that of a single plasmonic nanoparticle. Our results demonstrate and suggest substantial promise for the development of coupled plasmonic nanostructures for ultrasensitive detection of various biological and chemical analytes.  相似文献

Fluorescence barcoding based on nanoparticles provides many advantages for multiparameter imaging. However, creating different concentration‐independent codes without mixing various nanoparticles and by using single‐wavelength excitation and emission for multiplexed cellular imaging is extremely challenging. Herein, we report the development of quantum dots (QDs) with two different SiO2 shell thicknesses (6 and 12 nm) that are coated with two different lanthanide complexes (Tb and Eu). FRET from the Tb or Eu donors to the QD acceptors resulted in four distinct photoluminescence (PL) decays, which were encoded by simple time‐gated (TG) PL intensity detection in three individual temporal detection windows. The well‐defined single‐nanoparticle codes were used for live cell imaging and a one‐measurement distinction of four different cells in a single field of view. This single‐color barcoding strategy opens new opportunities for multiplexed labeling and tracking of cells.  相似文献

Metamolecules and crystals consisting of nanoscale building blocks offer rich models to study colloidal chemistry, materials science, and photonics. Herein we demonstrate the self‐assembly of colloidal Ag nanoparticles into quasi‐one‐dimensional metamolecules with an intriguing self‐healing ability in a linearly polarized optical field. By investigating the spatial stability of the metamolecules, we found that the origin of self‐healing is the inhomogeneous interparticle electrodynamic interactions enhanced by the formation of unusual nanoparticle dimers, which minimize the free energy of the whole structure. The equilibrium configuration and self‐healing behavior can be further tuned by modifying the electrical double layers surrounding the nanoparticles. Our results reveal a unique route to build self‐healing colloidal structures assembled from simple metal nanoparticles. This approach could potentially lead to reconfigurable plasmonic devices for photonic and sensing applications.  相似文献

The transition from molecular to plasmonic behaviour in metal nanoparticles with increasing size remains a central question in nanoscience. We report that the giant 246‐gold‐atom nanocluster (2.2 nm in gold core diameter) protected by 80 thiolate ligands is surprisingly non‐metallic based on UV/Vis and femtosecond transient absorption spectroscopy as well as electrochemical measurements. Specifically, the Au246 nanocluster exhibits multiple excitonic peaks in transient absorption spectra and electron dynamics independent of the pump power, which are in contrast to the behaviour of metallic gold nanoparticles. Moreover, a prominent oscillatory feature with frequency of 0.5 THz can be observed in almost all the probe wavelengths. The phase and amplitude analysis of the oscillation suggests that it arises from the wavepacket motion on the ground state potential energy surface, which also indicates the presence of a small band‐gap and thus non‐metallic or molecular‐like behaviour.  相似文献

Plasmon enhanced electrochemistry (PEEC), where specific electrochemical reactions are promoted due to the reduced energy barrier of the reaction processes by the light excited “hot carriers” of the plasmonic nanoparticles, has aroused tremendous interest in recent years. A deep understanding of the PEEC process becomes a key issue for facilitating PEEC catalyst design and improving PEEC performance. This concept article begins with a brief discussion of the macroscopic electrochemical method of PEEC study of the plasmonic nanoparticle ensembles. Following that, we highlight two electrochemical techniques that may possess single nanoparticle sensitivity, i. e., scanning electrochemical microscope and nano-impact electrochemistry. The pros and cons of each technique are discussed and an outlook is given. We hope to provide the readers with the current status of PEEC to evoke reflections regarding the reaction mechanisms, performance improvement, and the utilizations to important systems.  相似文献

等离子体纳米颗粒(PNPs)因其独特的物理、化学、光学和生物学特性而被广泛地应用于材料科学、生物学和医药学等研究领域。PNPs的光学性质是可以通过改变其组成、形状和大小来进行调控的,所以利用可控合成的方式能够筛选出适合的光散射探针。在单分子水平上实时研究PNPs的动态行为对于理解细胞及活体组织的生命活动机制、制备功能型纳米材料和开发新型化学生物传感器等有着重要的意义。基于传统的暗场显微镜(DFM),通过对光源、检测器及其它光学元件的择优组装和调试,我们开发出了一系列具有高灵敏度、高时空分辨率和高通量的等离子体光散射成像技术,并将其应用于单分子检测、多颗粒传感、单细胞成像以及生物过程示踪等领域。基于具有光学各向异性的PNPs,我们还研制出了活细胞三维扫描成像系统和超连续激光光片成像与高速毛细管电泳联用系统,推进了单分子光谱方面的研究。本文将总结近十年来本课题组在PNP单颗粒分析及成像中的工作,并为该领域未来的发展提出一些新的思路。  相似文献

A wide-field localized surface plasmon resonance (LSPR) imaging method using a liquid crystal tunable filter (LCTF) is used to measure the scattering spectra of multiple Ag nanoparticles in parallel. This method provides the ability to characterize moving Ag nanoparticles by measuring the scattering spectra of the particles while simultaneously tracking their motion. Consequently, single particle diffusion coefficients can be determined. As an example, several single Ag nanoprisms are tracked, the LSPR scattering spectrum of each moving particle is obtained, and the single particle diffusion coefficient is determined from its trajectory. Coupling diffusion information with spectral information in real time is a significant advance and addresses many scientific problems, both fundamental and biological, such as cell membrane protein diffusion, functional plasmonic distributions, and nanoparticle growth mechanisms.  相似文献

Biomaterials for in vivo fluorescence imaging are required to be biocompatible, nontoxic, photostable and highly fluorescent. Fluorescence must be in the near infrared (NIR) region of the electromagnetic spectrum to avoid absorption and autofluorescence of endogenous tissues. NIR fluorescent polystyrene nanoparticles may be considered ideal biomaterials for in vivo imaging applications. These NIR nanoparticles were prepared by a swelling process of polystyrene template nanoparticles with a hydrophobic NIR dye dissolved in a water‐miscible swelling solvent, a method developed for preparation of nonbiodegradable nanoparticles, for NIR fluorescent bioimaging applications. This method overcomes common problems that occur with dye entrapment during nanoparticle formation such as loss of fluorescence and size polydispersity. Fluorescence intensity of the nanoparticles was found to be size dependent, and was optimized for differently sized nanoparticles. The resulting NIR nanoparticles were also found to be more fluorescent and highly photostable compared to the free dye in solution, showing their potential as biomaterials for in vivo fluorescence imaging.  相似文献

以单层SiO2胶体微球为模板, 利用Au/SiO2/Au交替沉积结合后退火处理的方法, 制备了一种垂直堆叠且均一取向的等离子体二聚体结构. 该方法具有很大的自由度, 可以通过调节实验流程来制备大面积取向相同的同质或异质纳米粒子二聚体. 所制备的纳米粒子的等离子体杂化效应明显, 在消光光谱中可以观察到成键及反键共振峰. 由于所得纳米粒子二聚体具有垂直堆叠的特殊规整取向, 还可以观察到所得样品等离子体吸收峰的角度依赖特性. 此外, 还探讨了Au/SiO2/Au同质二聚体和Au/SiO2/Ag异质二聚体的光学特性差异, 发现与Au/SiO2/Au同质二聚体相比, Au/SiO2/Ag异质二聚体由于Ag偶极等离子体模式与Au带间吸收的耦合而呈现Fano共振峰. 所得结果提供了一个调节贵金属等离子体光学共振峰位、 强度和波形的策略, 在纳米光子学领域有着广阔的应用前景, 对今后的实验和理论研究具有重要参考价值.  相似文献
