A Pd‐catalyzed direct cross‐coupling of two distinct aryl bromides mediated by tBuLi is described. The use of [Pd‐PEPPSI‐IPr] or [Pd‐PEPPSI‐IPent] as catalyst allows for the efficient one‐pot synthesis of unsymmetrical biaryls at room temperature. The key for this selective cross‐coupling is the use of an ortho‐substituted bromide that undergoes lithium–halogen exchange preferentially.  相似文献

A Pd‐catalyzed cross‐coupling of aryl trifluoroborates with arylsulfonyl chlorides has been successfully achieved. This transformation is a new method for the Suzuki–Miyaura‐type reaction of aryl trifluoroborates via the cleavage of C? S bond, thus providing an alternative synthesis of biaryls. The reported cross‐coupling reactions are tolerant to many common functional groups regardless of electron‐donating or electron‐withdrawing nature, making these transformations attractive alternatives to the traditional Suzuki–Miyaura coupling approaches. Synthesis of di‐substituted aryl olefins via a Pd(0)‐catalyzed cross‐coupling reaction of biphenyl ketones/aldehydes, tosylhydrazide, and aryl bromides (or benzyl halides) was developed. This methodology was achieved by one‐pot two‐step reactions involving the preparation of N ‐tosylhydrazones by reacting tosylhydrazide with biphenyl ketones/aldehydes, followed by coupling with aryl bromides (or benzyl halides) in the presence of Pd(PPh3 )4 and lithium t ‐butoxide to produce various di‐substituted aryl olefins in moderate to good yields.  相似文献

A novel class of compounds bearing indole and benzimidazole rings was designed and easily synthesized from 2‐indolecarboxylic acid and o‐phenylenediamine. The catalytic system derived from a 2‐indolylbenzimidazole‐based ligand and Pd(OAc)2 in situ could lead to complete conversion of aryl bromides at 0.5 mol% Pd loading under mild reaction conditions. In the presence of a catalyst, sterically hindered biaryls were selectively generated in excellent yields by adjusting reaction parameters through the coupling of arylboronic acids with aryl halides. The efficiency of this reaction was demonstrated by its compatibility with various functional groups.  相似文献

A copper(0)‐promoted direct reductive gem‐difluoromethylenation of unactivated aryl or alkenyl halides with benzo‐1,3‐azolic (oxa‐, thia‐ or aza‐) difluoromethyl bromides or 2‐bromodifluoromethyl‐1,3‐oxazoline has been developed for the construction of pharmaceutically important gem‐difluoromethylene‐linked twin molecules. The unique π‐conjugated aryl‐fused 1,3‐azolic moiety in difluoromethyl bromide substrates could stabilise the reaction intermediates, which promotes the reactivities, providing facile access to the cross‐coupling products in good to excellent yields, and allowing significant functional group tolerance. The reaction exhibits an enhanced neighbouring‐group‐participation effect. This method could provide a new strategy for the construction of gem‐difluoromethylene‐linked identical or nonidentical twin drugs through further functionalisation of 1,3‐azolic skeletons.  相似文献

An efficient Pd‐catalyzed method for C—O cross‐coupling of ketoximes and chalcone oximes with activated aryl bromides and bromo‐chalcones has been developed. All oxime ethers were obtained in good to excellent yields by [(π‐allyl)PdCl]2/tBuXPhos ( L7 ) catalyst system. TrixiePhos ( L11 ) was also found to be effective for the oxime coupling. This method offers an easy and smooth coupling of chalcone oximes with activated aryl bromides and bromo‐chalcones, which has not been previously explored.  相似文献

cis,cis,cis‐1,2,3,4‐Tetrakis(diphenylphosphinomethyl)cyclopentane–[PdCl(C3H5)]2 efficiently catalyses the Heck reaction of alk‐1‐en‐3‐ol with a variety of aryl halides. In the presence of hex‐1‐en‐3‐ol or oct‐1‐en‐3‐ol, the β‐arylated carbonyl compounds were selectively obtained. Turnover numbers up to 84 000 can be obtained for this reaction. Linalool and 2‐methylbut‐3‐en‐2‐ol led regio‐ and stereoselectively to the corresponding (E)‐1‐arylalk‐1‐en‐3‐ol derivatives. A minor electronic effect of the substituents of the aryl bromide was observed. Quite similar reaction rates were generally observed in the presence of activated aryl bromides such as bromoacetophenone and deactivated aryl bromides such as bromoanisole, indicating that, with these alkenols and this catalyst, the oxidative addition of aryl bromides to palladium is not the rate‐limiting step. It should be noted that this reaction also proceeds with sterically very congested aryl bromides such as 9‐bromoanthracene or 2,4,6‐triisopropylbromobenzene or with a vinyl bromide. Air‐stable bis(imino)pyridine palladium(II) complexes were synthesized and complex 12 proved to be a highly efficient catalyst for the Suzuki cross‐coupling reaction between aryl bromides and arylboronic acids in air using water as solvent. The coupling reaction proceeded smoothly under mild conditions to provide biaryls in excellent yields and Pd black was not observed. The recycling of the catalysts was also investigated, for up to three cycles, and complex 12 still exhibited good activity. Copyright © 2009 John Wiley & Sons, Ltd.  相似文献

A new protocol for the NiCl2 ‐catalyzed cross‐electrophile coupling of aryl bromides with pyrimidin‐2‐yl tosylates to give the corresponding C2 ‐arylation pyrimidine derivatives has been developed. This study provides an improvement over previous methods by using pyrimidin‐2‐yl tosylates instead of halides as coupling partners that are stable and easily available.  相似文献

Arylzinc reagents, prepared from aryl halides/zinc powder or aryl Grignard reagents/zinc chloride, were found to undergo coupling with aryl and alkenyl halides without the aid of transition‐metal catalysis to give biaryls and styrene derivatives, respectively. In this context, we have already reported the corresponding reaction using aryl Grignard reagents instead of arylzinc reagents. Compared with the Grignard cross‐coupling, the present reaction features high functional‐group tolerance, whereby electrophilic groups such as alkoxycarbonyl and cyano groups are compatible as substituents on both the arylzinc reagents and the aryl halides. Aryl halides receive a single electron and thereby become activated as the corresponding anion radicals, which react with arylzinc reagents, thus leading to the cross‐coupling products.  相似文献

A method for enantioselective desymmetrization of 1,1‐diborylalkanes through a stereoselective Pd‐catalyzed Suzuki–Miyaura cross‐coupling has been thoroughly optimized. The most effective ligand was found to be a α,α,α,α‐tetra‐aryl‐1,3‐dioxolane‐4,5‐dimethanol (TADDOL)‐derived phosphoramidite. Results show that in order to achieve high selectivity, a suitable balance between the sterics of the aryl groups and the amino group on the ligand must be achieved. While the base has been known to facilitate transmetallation in cross‐coupling reactions, mechanistic studies on this desymmetrization process reveal that the base, in the presence of KHF2, likely plays an additional role in the hydrolysis of the pinacol boronates to the corresponding boronic acids. Through an in depth optimization of the chiral ligand and mechanistic studies, it was possible to obtain ee values over 90 % for several aryl bromides and to develop a reliably scalable process (up to one gram of 1,1‐diborylalkane substrate).  相似文献

An agro waste‐derived, ‘water extract of pomegranate ash’ (WEPA), has been utilized for the first time as a renewable medium for Pd(OAc)2‐catalysed Suzuki–Miyaura cross‐coupling at room temperature. This method offers a simple and sustainable synthesis of biaryls from aryl halides and arylboronic acids under ligand‐ and external base‐free aerobic and ambient conditions. This method has been found effective for both activated and unactivated aryl halides in the production of biaryls with moderate to nearly quantitative yields. The protocol shows high chemoselectivity over identical/similar reactive sites in aryl halides (i.e. selectivity over identical halogens or different halogens of aryl halides). This method exhibits high regioselectivity, i.e. the selective reactivity of a halogen over other identical halogens at different positions on the aromatic nucleus. Therefore, we disclose here a clean, benign, substantial chemo‐ and regioselective and highly economic alternative method for the palladium‐assisted synthesis of biaryls using an agro waste‐derived medium.  相似文献

本文报道了有机-无机杂化材料固载的钯催化的无铜Sonogashira偶联反应。在3-[N,N-双(二苯基膦)氨基)]丙基功能化的硅胶固载的钯催化下,末端炔烃和碘代芳烃、溴代芳烃的偶联反应生成高产率相应的偶联产物。反应条件包括使用乙二醇为溶剂,三乙胺为碱。而且硅胶负载的膦钯催化剂和溶剂乙二醇经简单处理,可循环使用6次不降低活性。  相似文献

A simple and efficient C? N cross‐coupling method of aryl halides with various heterocycles was reported, by using 10 mol% of CuI as catalyst and 1.2 equiv. NaH as base. Aryl iodides, aryl bromides and many substituted aryl chlorides could efficiently react with heterocycles, providing variety of N‐arylated products in good to excellent yields. The ligand‐free catalyst system was stable in air and could be readily reused.  相似文献

A series of differently substituted Chugaev‐type palladium bis(acyclic diaminocarbene) complexes was screened to identify the most active catalyst for Mizoroki–Heck coupling reactions of aryl bromides with styrene. The best catalyst, which contains three methyl groups on the bis(carbene) ligand, gives excellent coupling yields at 120 °C for both activated and deactivated aryl bromides. However, activity with aryl chlorides is limited to electron‐deficient examples. The optimized catalyst demonstrates limited air and moisture stability, giving reduced yields in couplings of activated aryl bromides in open‐flask conditions. The modular synthesis of this class of catalysts should allow further fine‐tuning of activity in Mizoroki–Heck and related coupling reactions. Copyright © 2012 John Wiley & Sons, Ltd.  相似文献

A nickel‐mediated intermolecular reductive cross‐coupling reaction of unactivated alkyl bromides and aryl iodides at room temperature has been developed and successfully extended to less explored intramolecular versions and tandem cyclization‐intermolecular cross‐coupling. Highly stereoselective (or stereospecific) synthesis of linear‐fused perhydrofuro[2,3‐b]furan (pyran) and spiroketal skeletons allows rapid access to these useful building blocks, which would be potentially valuable in the synthesis of relevant natural products. A rational explanation for the formation of contiguous stereogenic centers is given.  相似文献

A novel PdCl2/bis(2‐pyridylmethyl)amine‐based ligand ( 1 ) catalytic system, which is water‐soluble and air‐stable, has been successfully synthesized and applied for Suzuki‐Miyaura cross‐coupling reaction. In the presence of catalytic amount of PdCl2/ 1 system, arylboronic acids can couple with a wide range of aryl halides, including aryl bromides and aryl chlorides. The reactions proceed under mild conditions to give excellent yields, and a wide range of functionalities is tolerated.  相似文献

In this work, ortho‐palladated complexes [Pd(µ‐Cl)(C6H4CH2 NRR′‐κ2‐C,N)]2 and [Pd(C6H4CH2NH2‐2‐C,N)Cl(Y)] were tested in the Suzuki–Miyaura cross‐coupling reaction. Cyclopalladated Pd(II) complexes as thermally stable catalysts can activate aryl bromides and chlorides. These complexes were active and efficient catalysts for the Suzuki–Miyaura reaction of aryl bromides and even less reactive aryl chlorides. The cross‐coupled products of a variety of aryl bromides and aryl chloride with phenylboronic acid in methanol as solvent at 60 °C were produced in excellent yields. Pd/C‐catalyzed Suzuki–Miyaura cross‐coupling between aryl bromides and arylboronic acids in 50% methanol aqueous solution proceeded smoothly in the presence of 18‐crown‐6. Various aryl bromides bearing electron‐withdrawing groups and electron‐donating groups coupled with arylboronic acid in high yields. 我们发展一种在催化体系可回收和无配体条件下溴化四丁基铵(TBAB)中钯催化卤代芳烃与芳基硼酸的Suzuki-Miyaura交叉偶联反应方法。我们发现水的量对反应有很大的影响。当水的用量为1 %(质量比)时,反应的结果最好。在3 mol%的醋酸钯和1.5 g的TBAB(含1%的水),一系列卤代芳烃与芳基硼酸的顺利地发生Suzuki-Miyaura交叉偶联反应,得到中等及良好的产率。而且在溴代芳烃和活泼的氯代芳烃的交叉反应中,Pd(OAc)2/TBAB催化体系可以回收重复使用多次,并且催化活性基本不变。  相似文献
